Multiple structural states of S100A12: A key to its functional diversity.

S100A12 is a member of the S100 family of EF-hand calcium-binding proteins. Together with two other calgranulins, S100A8 and S100A9, it is mostly expressed in human granulocytes, although there is increasing evidence of expression in keratinocytes and psoriatic lesions. It is involved in host-parasite response, and linked to corneal autoimmune diseases connected with filarial parasite infestation. Interaction of S100A12 with a multiligand receptor for advanced glycation end products (RAGE) mediates inflammation. Human recombinant S100A12 was found to induce neuritogenesis of cultured hippocamp

Lack of S100A8 Impairs Lung-Protective Immunity Against Streptococcus pneumoniae.

BACKGROUND: We recently showed that S100A9 is indispensable for lung antibacterial immunity, making it crucial for the survival of pneumococcal pneumonia. However, the role of S100A8 in lung antibacterial immunity is ill-defined. METHODS: S100A8 levels in the bronchoalveolar lavage fluid (BALF) of patients with pneumonia were quantified by enzyme-linked immunosorbent assay. Wild type and S100A8 knockout mice were orotracheally infected with Streptococcus pneumoniae, and bacterial clearance, disease progression, lung histopathology, and leukocyte recruitment were analyzed at defined time point

S100A8/A9 blockade attenuates autoimmune neuroinflammation by regulating the heterogeneity and function of neutrophils.

Patients with multiple sclerosis (MS) exhibit elevated levels of calprotectin S100A8/A9 in the blood, bone marrow (BM), and feces. However, the potential of targeting S100A8/A9 as a therapeutic approach for MS treatment remains to be fully elucidated. In this study, using an experimental autoimmune encephalomyelitis (EAE) model of MS, we found that inhibiting S100A8/A9 using paquinimod attenuated the EAE disease severity, which is associated with decreased splenic neutrophils, particularly the immature Ly6GhiCD101-subset. Detailed analysis of neutrophil subsets revealed a decreased banded-to-s

S100A8/A9 Promotes Fibrosis in Iatrogenic Laryngotracheal Stenosis.

OBJECTIVE: Iatrogenic Laryngotracheal Stenosis (iLTS) is a pathologic narrowing of the laryngotracheal airway secondary to intubation injury. Inflammation plays a critical role in iLTS pathogenesis. However, the local immune mediators promoting inflammation and subsequent fibrosis remain poorly understood. Transcriptomic analysis of iLTS specimens using single-cell RNA sequencing has revealed increased expression of the damage-associated molecular pattern (DAMP), S100A8/A9. The goal of this study is to define the role of S100A8/A9 in iLTS pathogenesis in vivo. METHODS: S100A8/A9 expression wa

Myeloid-related Protein 8/14 Levels in Rheumatoid Arthritis: Marker of Disease Activity and Response to Methotrexate.

OBJECTIVE: Myeloid-related proteins (MRP) 8/14 belong to a family of calcium-binding proteins produced by myeloid cells. Baseline serum levels of MRP8/14 have been shown to predict response to biologicals in rheumatoid arthritis (RA). Because methotrexate (MTX) is the first-line therapy in RA, we studied whether MRP8/14 levels can predict response to MTX. METHODS: Patients with active RA disease who were naive to disease-modifying antirheumatic drugs were enrolled. All patients were treated with MTX only, to a maximum of 25 mg/week or the maximal tolerated dose. At 4 months, the European Leag

Lipopolysaccharide-mediated induction of calprotectin in the submandibular and parotid glands of mice.

S100A8 and S100A9 constitute a heterodimeric protein, calprotectin. The mRNAs of S100A8 and S100A9, being expressed at minimal levels in the submandibular and parotid glands (SMG and PG, respectively) of C3H/HeN mice, were induced strongly and transiently by lipopolysaccharide (LPS). Among the mRNAs of members of the S100 protein family examined, those of S100A8 and S100A9 were specifically induced by LPS in the salivary glands. The induction was assumed to be mediated via toll-like receptor 4 (TLR4), since their elevation was limited in C3H/HeJ mice, a TLR4-mutant strain. These proteins becam

Up-regulation of S100A8 and S100A9 protein in bronchial epithelial cells by lipopolysaccharide.

Increased serum levels of the S100A8 (MRP-8) protein have been reported in inflammatory conditions including bacterial infection, arthritis, and cystic fibrosis (CF). This protein is expressed constitutively with S100A9 (MRP-14) in neutrophils and is regulated by inflammatory stimulants. It has been hypothesized that increased inflammatory response to persistent bacterial infection is a major feature of CF lung disease. Therefore, the authors wished to determine the involvement of these two proteins in the innate defense response of the bronchial epithelium to lipopolysaccharide (LPS). Human b

Targeting S100A8 mitigates pathogenic neutrophil-driven tissue injury in bacterial epididymo-orchitis.

Bacterial epididymo-orchitis, primarily driven by uropathogenic Escherichia coli (UPEC), is a major cause of inflammatory testicular injury and male infertility. While our previous study indicates that neutrophils are rapidly recruited to the immune-privileged testis to contain the infection, their excessive activation can paradoxically exacerbate tissue destruction. However, the specific cellular states and molecular mediators driving this injurious inflammatory response remain poorly defined. To address this gap, we utilized single-cell transcriptomics in a murine orchitis model to map the d

Decreased expression of S100A8/A9 in V30M related ATTRv amyloidosis.

INTRODUCTION: Hereditary Transthyretin Amyloidosis is a rare, progressive and life-threatening systemic disease with predominant peripheral and autonomic nervous system involvement caused by mutation of the transthyretin protein. The most common TTR mutation regarding to ATTRv is a substitution of a Methionine for a Valine at position 30 that predisposes TTR to form aggregates and fibrils. METHODS: S100A8 protein levels were measured in plasma samples from ATTRV30M patients and healthy donors. Additionally, S100A8/9 levels were measured in Schwann cells after incubation with human WT or V30M

S100A8/A9, a potent serum and molecular imaging biomarker for synovial inflammation and joint destruction in seronegative experimental arthritis.

BACKGROUND: Seronegative joint diseases are characterized by a lack of well-defined biomarkers since autoantibodies are not elevated. Calprotectin (S100A8/A9) is a damage-associated molecular pattern (DAMP) which is released by activated phagocytes, and high levels are found in seronegative arthritides. In this study, we investigated the biomarker potential of systemic and local levels of these S100 proteins to assess joint inflammation and joint destruction in an experimental model for seronegative arthritis. METHODS: Serum levels of S100A8/A9 and various cytokines were monitored during dise

Association of Elevated Serum S100A8/A9 Levels and Cognitive Impairment in Patients With Systemic Lupus Erythematosus.

OBJECTIVE: Cognitive impairment (CI) is common in patients with systemic lupus erythematosus (SLE). Despite its prevalence, the immune mechanisms are not well understood. We previously reported elevated serum levels of S100A8/A9 and matrix metalloproteinase 9 (MMP-9) in patients with SLE and CI. This study aims to validate those findings by examining the relationship between serum levels and CI in patients with SLE at baseline and after one year. METHODS: We assessed cognitive function in 112 patients with SLE using the adapted American College of Rheumatology-Neuropsychological Battery, defi

Emerging roles of neutrophil-borne S100A8/A9 in cardiovascular inflammation.

Elevated neutrophil count is associated with higher risk of major adverse cardiac events including myocardial infarction and early development of heart failure. Neutrophils contribute to cardiac damage through a number of mechanisms, including attraction of other immune cells and release of inflammatory mediators. Recently, a number of independent studies have reported a causal role for neutrophil-derived alarmins (i.e. S100A8/A9) in inducing inflammation and cardiac injury following myocardial infarction (MI). Furthermore, a positive correlation between serum S100A8/A9 levels and major advers

Differential expression of the calcium-binding proteins MRP8 and MRP14 in granulomatous conditions: an immunohistochemical study.

MRP14 and MRP8 are well-characterized calcium-binding proteins present in myeloid cells and mononuclear phagocytes. These antigens can easily be visualized in paraffin-embedded tissue, making use of monospecific polyclonal antibodies. This study evaluates MRP14 and MRP8 expression in mononuclear phagocytes in various granulomatous conditions. MRP14 is strongly expressed in all granulomatous conditions. MRP8 is variably expressed. Mononuclear phagocytes in granulomas of foreign body type, cat-scratch disease and erythema nodosum strongly express MRP8. In contrast, MRP8 expression is weak or abs

Receptor for advanced glycation endproducts and progressive kidney disease.

PURPOSE OF REVIEW: Receptor for advanced glycation endproducts (RAGE) is a multiligand receptor of the immunoglobulin superfamily, which binds not only to advanced glycation endproducts, but also to high-mobility group box-1, S100/calgranulins, amyloid fibrils, and lipopolysaccharide. We discuss the pathophysiological role of RAGE in both diabetic and nondiabetic progressive renal diseases, and its potential use for therapeutic interventions in these devastating disorders. RECENT FINDINGS: Although initial studies about RAGE focused on diabetic nephropathy, a number of recent findings have re

Antagonism of RAGE suppresses peripheral nerve regeneration.

Axotomy of peripheral nerve triggers events that coordinate a limited inflammatory response to axonal degeneration and initiation of neurite outgrowth. Inflammatory and neurite outgrowth-promoting roles for the receptor for advanced glycation end products (RAGE) have been suggested, so we tested its role in peripheral nerve regeneration. Analysis of immunohistochemical localization of RAGE by confocal microscopy revealed that RAGE was expressed in axons and infiltrating mononuclear phagocytes upon unilateral sciatic nerve crush in mice. Administration of soluble RAGE, the extracellular ligand

S100-alarmins, antenatal corticosteroids and the risk of late-onset sepsis in preterm infants: A prospective cohort study.

OBJECTIVES: Antenatal corticosteroids (aCS) are an important measure improving the outcome of preterm infants. Their influence on late-onset sepsis (LOS) risk remains inconclusive. The alarmin S100A8/A9 protects from LOS by regulating innate immune responses. We examined whether aCS impact on postnatal S100A8/A9 serum-levels and consequently on LOS risk. STUDY DESIGN: In a prospective birth-cohort study of 162 preterm infants born before 32 gestational weeks, we determined postnatal S100A8/A9 serum-levels in relation to the timing of aCS and LOS incidence. RESULTS: aCS administration within

S100A8+S100A9+ transitional macrophages are associated with pulmonary fibrosis progression by integrating immunometabolism and fibrogenic crosstalk.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is a progressive interstitial lung disease with limited therapeutic options. Although macrophage heterogeneity has been implicated in pathogenesis, the contribution of transitional macrophage states remains poorly understood. METHODS: We integrated single-cell RNA-seq from 93 human lung specimens (44 IPF, 49 controls) and analyzed peripheral blood single-cell datasets (31 IPF, 17 controls). We examined differentiation trajectories and ligand-receptor networks and assessed metabolic programs. Functional relevance was evaluated in a bleomycin mous

Spatial atlas of upper tract urothelial carcinoma identifies SLC14A1 + and S100A8/9+ tumor cells as key determinants of malignant progression.

Upper tract urothelial carcinoma (UTUC) is characterized by a high incidence of muscle invasion and distinct molecular heterogeneity, with tumor microenvironment (TME) heterogeneity playing a pivotal role in its malignant progression. However, the spatial distribution patterns of functional cell populations in UTUC and their regulatory mechanisms driving high-grade (HG) and muscle-invasive (MI) progression remain largely unelucidated. Herein, we integrated single-cell RNA sequencing (scRNA-seq) and spatial transcriptome sequencing (ST-seq) to systematically characterize the cellular landscape,
