Advancements, challenges, and future perspectives in developing feline herpesvirus 1 as a vaccine vector.

As the most prevalent companion animal, cats are threatened by numerous infectious diseases and carry zoonotic pathogens such as Toxoplasma gondii and Bartonella henselae, which are the primary causes of human toxoplasmosis and cat-scratch disease. Vaccines play a crucial role in preventing and controlling the spread of diseases in both humans and animals. Currently, there are only three core vaccines available to prevent feline panleukopenia, feline herpesvirus, and feline calicivirus infections, with few vaccines available for other significant feline infectious and zoonotic diseases. Feline

Rapid and sensitive detection of feline herpesvirus-1 using fluorescent microspheres as labels for immunochromatographic test strips.

Feline herpesvirus-1 is a local contact-infectious pathogen that causes acute upper respiratory tract infections in cats. This virus exclusively affects felines, with a clinical incidence rate of nearly 100%, particularly in young cats. To address this health concern, this study aimed to develop a rapid fluorescence microsphere immunochromatography assay (FM-ICA) for the direct detection of Feline herpesvirus-1 (FHV-1) antigen. This method utilizes a fusion protein and fluorescent nanoparticle-labelled monoclonal antibody to detect FHV-1 within 10 min, achieving a detection limit of 2.5 × 102

Etiological investigation of multiple respiratory infections in cats.

In order to evaluate the relevance of multiple infections in domestic cats with Upper Respiratory Tract Disease (URTD) one hundred animals with clinical signs were investigated for detection of Feline Herpesvirus type-1 (FHV-1), Chlamydophila felis, Feline Calicivirus (FCV) and Bordetella bronchiseptica from mucosal swabs. Forty-seven cats were positive for FCV, 42 cats for FHV-1, 26 for B. bronchiseptica and 8 for C. felis. Dual or multiple infections were found in 33 of examined animals. Our results document that FCV and FHV-1 are the major recognized cause of URTD, although infections assoc

Development of a Targeted NGS Assay for the Detection of Respiratory Pathogens including SARS-CoV-2 in Felines.

Acute respiratory diseases in felines can be attributed to a diverse range of pathogens. The recent emergence of novel viruses, particularly SARS-CoV-2 and its variants, has also been associated with respiratory ailments in cats and other pets, underscoring the need for a highly sensitive diagnostic assay capable of concurrently detecting multiple respiratory pathogens. In this study, we developed a targeted next generation sequencing panel using Ion Torrent Ampliseq technology to detect multiple respiratory pathogens, including recent SARS-CoV-2 variants and Feline herpesvirus-1, Feline calic

Surveillance using serological and molecular methods for the detection of infectious agents in captive Brazilian neotropic and exotic felids.

The aim of the current study was to investigate the exposure of captive wild felids to various infectious pathogens using serological and molecular methods. One hundred and fifty-nine neotropic felids and 51 exotic felids from 28 captive settings in Brazil were tested. While antibodies against Feline parvovirus and Feline coronavirus (FCoV), Feline calicivirus and Bartonella spp. were frequently detected by serologic tests, antibodies against Felid herpesvirus 1 or infection with hemotropic mycoplasmas were less prevalent. Serologic evidence of exposure to Ehrlichia spp., Feline immunodeficien

Effect of high-dose ciclosporin on the immune response to primary and booster vaccination in immunocompetent cats.

Ciclosporin (Atopica oral solution for cats 100 mg/ml; Novartis Animal Health) was recently approved for use in cats with feline hypersensitivity dermatitis. The immunosuppressant effect of ciclosporin on the ability of cats to mount an immune response following vaccination was determined. Thirty-two healthy, immunocompetent adult cats (16 cats/group) were treated with either ciclosporin for 56 days at a dose of 24 mg/kg once daily or sham dosed. Prior to treatment, cats had an adequate antibody response to primary vaccination against feline calicivirus (FCV), feline herpesvirus-1 (FHV-1), fel

A cross-sectional serosurvey of SARS-CoV-2 and co-infections in stray cats from the second wave to the sixth wave of COVID-19 outbreaks in Spain.

Severe Acute Respiratory Syndrome Coronavirus 2 is the causative agent of Coronavirus Disease 2019 in humans. Among domestic animals, cats are more susceptible to SARS-CoV-2 than dogs. The detection of anti-SARS-CoV-2 antibodies in seemingly healthy cats and/or infected cats which are in close contact with infected humans has been described. The presence of animals that tested positive by serology or molecular techniques could represent a potential transmission pathway of SARS-CoV-2 that can spill over into urban wildlife. This study analyses the seroprevalence variation of SARS-CoV-2 in stray

Exposure to Feline Viruses in European Wildcats (Felis s. silvestris) in Germany: Spatial Patterns and Environmental Risk Factors.

While viral diseases of domestic cats (Felis catus) can threaten the recovery of the European wildcat (Felis s. silvestris), their epidemiology in wildcat populations remains poorly understood. Here, we analyzed 428 road-killed wildcats from Western and Central Germany for the presence of antibodies/antigens of six feline viruses. The presence of at least one viral antigen or antiviral antibody was detected in 53.3% of the animals. Antibodies against feline parvovirus (FPV) showed the highest seroprevalence (29.2%), while feline leukemia virus (FeLV) antigens were detected in 22.2% of the anim

Temperature-dependent infectious stability of Severe fever with thrombocytopenia syndrome virus.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging tick-borne zoonotic pathogen of increasing concern in East Asia, including Japan. Not only humans but also domestic cats are highly susceptible and frequently develop severe disease. Despite its public and veterinary health significance, data on the environmental stability of SFTSV remain limited. In this study, we evaluated the stability profiles of SFTSV under various temperature and inactivation conditions. As comparative controls, Kabuto Mountain virus (KAMV) which belongs to the same class and order as SFTSV, Japanes

Genetic and pathogenicity analysis for the two FCV strains isolated from Eastern China.

In this study, the diversity and regularity of two new feline calicivirus (FCV) isolates, QD-7 and QD-164, were investigated. The genomes of these new strains were compared with 39 strains from the NCBI database including isolates from China, United States, Germany, South Korea, the United Kingdom and Japan. The nucleotide sequence identities ranged from 75-88%, indicating a high degree of variability. These variations were not related to distributions of the virus by time of isolation and geographical location. Cats that were experimentally infected with the new isolate QD-164 showed typical

Interstitial nephritis in cats inoculated with Crandell Rees feline kidney cell lysates.

Parenteral administration of Crandell Rees feline kidney (CRFK) cell lysates or feline herpesvirus 1, calicivirus, and panleukopenia virus-containing vaccines (FVRCP) grown on CRFK cells induces antibodies against CRFK cells. These antibodies also react with feline renal cell extracts. The purpose of this study was to determine whether interstitial nephritis would be detected in cats that were immunologically sensitized with CRFK lysates, boosted with CRFK lysates, and then biopsied 2 weeks after the booster. Cats (2 per group) were immunologically sensitized against CRFK lysates by administer

In vitro efficacy of povidone iodine and hydroxyethyl cellulose, alone and in combination, against common feline ocular pathogens.

Infectious ocular disease, such as conjunctivitis, is common in cats and can be caused by several viruses and bacteria, either as a single infection or as co-infections. In this study, povidone-iodine (PVP-I), alone or compounded with hydroxyethyl cellulose (HEC), was investigated for its efficacy against these pathogens in vitro. Whilst PVP-I alone was effective at inhibiting feline herpesvirus type 1 (FHV-1), Chlamydia felis, and Mycoplasma felis, PVP-I with HEC exerted a synergistic inhibitory effect against FHV-1 and C. felis. In contrast, only minimal inhibition of feline calicivirus was

A novel feline herpesvirus vector subunit FCV VP1 and FPV VP2 vaccine protects cats against FHV-1 and FPV challenge and induces serum neutralizing antibody responses against FCV.

Vaccines targeting feline parvovirus (FPV), feline calicivirus (FCV), and feline herpesvirus type 1 (FHV-1) are considered core vaccines and are widely recommended for feline immunoprophylaxis. Currently, trivalent feline vaccines used in clinical settings are primarily based on inactivated or modified live attenuated formulations. Among these pathogens, FHV-1 has demonstrated notable potential as a viral vector due to its large genome and immunogenic profile, making it an attractive platform for multivalent vaccine development. In this study, we developed a novel trivalent live attenuated vac

Evaluating triple inactivated vaccine-induced immunity from a large-scale study in feline population.

Feline panleukopenia virus (FPV), feline calicivirus (FCV), and feline herpesvirus-1 (FHV-1) substantially impact feline health. Field evidence on vaccine-induced population immunity remains limited. We evaluated humoral responses in 4,736 vaccinated domestic cats across 24 provinces in China (July 2024-June 2025), quantifying neutralizing antibody titers to FPV, FCV, and FHV-1 after a trivalent inactivated vaccine (Meowonder™). For cats aged 3 months or older, two primary doses achieved high antibody positivity rates across pathogens; a third dose yielded marginal additional benefit, while bo
