Antihypertensive Effects, Molecular Docking Study, and Isothermal Titration Calorimetry Assay of Angiotensin I-Converting Enzyme Inhibitory Peptides from Chlorella vulgaris.

The aim of this work is to explore angiotensin I-converting enzyme (ACE) inhibitory peptides from Chlorella vulgaris (C. vulgaris) and discover the inhibitory mechanism of the peptides. After C. vulgaris proteins were gastrointestinal digested in silico, several ACE inhibitory peptides with C-terminal tryptophan were screened. Among them, two novel noncompetitive ACE inhibitors, Thr-Thr-Trp (TTW) and Val-His-Trp (VHW), exhibited the highest inhibitory activity indicated by IC50 values 0.61 ± 0.12 and 0.91 ± 0.31 μM, respectively. Both the peptides were demonstrated stable against gastrointesti

Estimation of parvalbumin Ca(2+)- and Mg(2+)-binding constants by global least-squares analysis of isothermal titration calorimetry data.

The use of competitive isothermal titration calorimetry (ITC) to measure high-affinity binding constants has been largely restricted to systems with a single binding site or multiple identical sites. This study demonstrates the extension of this approach to proteins with two nonequivalent EF-hand Ca(2+)-binding sites--rat beta parvalbumin and the S55D/E59D variant of rat alpha parvalbumin. The method involves simultaneous (global) least-squares analysis of titrations with Ca(2+), with Mg(2+), with Ca(2+) in the presence of Mg(2+), and with Ca(2+) or Mg(2+) in the presence of a competitive chel

Optimization of freezing and thawing protocols for human ovarian tissue cryopreservation through thermophysical characterisation of freezing medium by differential scanning calorimetry.

Fertility preservation should be offered to patients facing gonadotoxic therapy. The method for preserving prepubescent girls' fertility, which is also suitable for women, is ovarian tissue cryopreservation (OTC). Although 200 births have been reported worldwide with this approach, significant improvements are needed. The literature indeed reports numerous protocols for freezing and thawing ovarian tissue, with no clear rationale for selection criteria. This study aims to optimize human OTC protocols by characterizing the thermodynamic properties of freezing medium. The freezing medium associa

Thermal and structural analysis of binary mixtures of pyrimidine liquid crystals using modulated differential calorimetry and synchrotron x-ray diffraction measurements.

We present a systematic experimental dataset on the temperature dependence of specific heat capacity in a binary mixture of the second and seventh homologous series of 5-alkyloxy-2-(4-nonyloxy-phenyl) pyrimidine (PhP) liquid crystal compound. These binary mixtures exhibit nematic, smectic-A, and smectic-C phases within a concentration range ofxPhP1= 0-0.45. The liquid crystalline phases are structurally characterized using synchrotron x-ray diffraction. We determine the apparent molecular length in the nematic phase, smectic layer spacing, average distance between the long axes of molecules, c

To measure or to estimate? A comparison between indirect calorimetry and predictive equations for resting metabolic rate in institutionalized older adults: a systematic review.

INTRODUCTION: Population aging and institutionalization require accurate energy estimates for the maintenance of health and vitality. Resting Metabolic Rate (RMR) is frequently estimated using predictive equations; however, their accuracy in institutionalized older adults remains uncertain, which may compromise nutritional management and worsen clinical outcomes. OBJECTIVE: To systematically evaluate the accuracy of RMR predictive equations compared to the gold standard of indirect calorimetry in residents of Long-Term Care Facilities (LTCFs). METHODS: A systematic review was conducted in ac

Liver exhibits thermal variations according to the stage of fibrosis progression: A novel use of modulated-differential scanning calorimetry for research in hepatology.

AIM: Liver fibrosis results in a disproportion of the hepatic composition and architecture, characterized by a progressive accumulation of fibrillar proteins at the liver parenchyma. Modulated-differential scanning calorimetry (mDSC) is an experimental methodology able to determine the specific thermal signature from any biological substance, based on the variation in heat flow and heat capacity. As these physicochemical properties are directly influenced by compositional and structural changes, we decided to study the thermal behavior of the liver during fibrosis using mDSC. METHODS: Liver f

Combination of chemometrically assisted voltammetry, calorimetry, and circular dichroism as a new method for the study of bioinorganic substances: application to selenocystine metal complexes.

Selenium-containing compounds play an important role in antioxidant defense systems, binding to toxic metals, preventing their uptake into cells, and thus protecting cells from metal-induced formation of reactive oxygen species. Here, we present a proposal for a relatively new method as a complement to the more usual methods used in selenium studies. A systematic study of the metal-binding properties of selenocystine (SeCyst) in the presence of divalent metal cations (Cd, Co, Hg, Ni, and Zn) is reported. Isothermal titration calorimetry provides thermodynamic parameters of the systems. Titrati

Differential Inhibitory Mechanisms of Myricetin and Dihydromyricetin on α-Glucosidase: A Combined Molecular Docking, Isothermal Titration Calorimetry and Surface Plasmon Resonance Study.

α-Glucosidase inhibitors (AGIs) significantly regulate blood glucose by delaying carbohydrate digestion and slowing glucose absorption, thus playing a critical role in glycemic control. Structurally, dihydromyricetin (Unless otherwise stated, the term dihydromyricetin used throughout this manuscript refers to trans-(2R,3R)-(+)-dihydromyricetin.) differs from myricetin in that the C2=C3 double bond in the C-ring is saturated, resulting in a dihydroflavonol instead of a flavonol. This study investigated the inhibition mechanism of α-glucosidase by the C2=C3 double bond structure using a set of i

Hemi-synthesis of novel chiral benzodiazepine derivatives from Eucalyptus citriodora essential oil: 2D NMR experiments and differential scanning calorimetry study of diastereoisomers.

An efficient in situ condensation of citronellal, the main constituent of Eucalyptus citriodora essential oil (51%), with different amine derivatives of 2,3-diaminomaleonitrile and 3-[(2-aminoaryl)amino]dimedone has led to novel chiral benzodiazepine structures. All reactions were precipitated in ethanol and pure products were obtained in good yields (58-75%) without any purification. The synthesized benezodiazepines were characterized by spectroscopic techniques, namely 1H-NMR, 13C-NMR, 2D NMR and FTIR. Differential Scanning Calorimetry (DSC) and HPLC were used to confirm the formation diaste

Label-free characterization of carbonic anhydrase-novel inhibitor interactions using surface plasmon resonance, isothermal titration calorimetry and fluorescence-based thermal shift assays.

This work describes the development of biophysical unbiased methods to study the interactions between new designed compounds and carbonic anhydrase II (CAII) enzyme. These methods have to permit both a screening of a series of sulfonamide derivatives and the identification of a lead compound after a thorough study of the most promising molecules. Interactions data were collected using surface plasmon resonance (SPR) and thermal shift assay (TSA). In the first step, experiments were performed with bovine CAII isoform and were extended to human CAII. Isothermal titration calorimetry (ITC) experi

Ethanol perturbs lipid organization in models of stratum corneum membranes: An investigation combining differential scanning calorimetry, infrared and (2)H NMR spectroscopy.

Ethanol is used in a variety of topical products. It is known to enhance the permeability of the skin by altering the ability of the stratum corneum (SC) intercellular membranes to form an effective barrier. In addition, ethanol and other alcohols are key components of antiseptic gels currently used for hand wash. Using infrared and deuterium NMR spectroscopy as well as calorimetry, we have investigated the effect of ethanol on a model membrane composed of lipids representing the three classes of SC lipids, an equimolar mixture of N-palmitoylsphingosine (ceramide), palmitic acid and cholestero

Advancing Accurate Quantification of Protein-Ligand Interactions: Differential Scanning Calorimetry as a Precision Screening Tool Using BCL-2 as a Model System.

Accurate and reliable quantification of protein-ligand energetics at the screening stage is often complicated by ligand aggregation, hydrophobicity-driven artifacts, and the need for cosolvents. Here, differential scanning calorimetry (DSC) as a quantitative, label-free screening method is evaluated using BCL-2 as a model oncogenic target. Nine inhibitors (i.e., venetoclax, navitoclax; and seven previously prioritized BCL-2 hit inhibitors by our research group) are profiled across solvent systems, including neat DMSO, 10% DMSO, and a ternary matrix (S3: 10% DMSO, 90% sulfobutylether-β-cyclodex

Blood compatibility of poly(propylene glycol diester) and its water structure observed by differential scanning calorimetry and 2H-nuclear magnetic resonance spectroscopy.

Recently, we applied solution 2H-nuclear magnetic resonance spectroscopy (2H NMR) to analyze the water (deuterium oxide, D2O) structure in several biopolymers at ambient temperature. We established that polymers with good blood compatibility (i.e. poly(2-methoxyethyl acrylate) (PMEA)) have water observed at high magnetic fields (upfield) compared with bulk water. Polymers containing poly(propylene glycol) (PPG) or poly(propylene oxide) (PPO) exhibit good compatibility; however, the reason for this remains unclear. In addition, reports on the blood compatibility of PPO/PPG are limited. Therefor

Expanding the use of indirect calorimetry in critical care settings to determine the impact on nutrition interventions and compare to predictive equations: A quality improvement initiative.

BACKGROUND: Critical illness leads to a hypermetabolic state, increasing the risk of malnutrition, infectious complications, and poor outcomes. Current guidelines recommend early enteral nutrition (EN) support; however, determining appropriate caloric provision remains challenging. Indirect calorimetry (IC) is the gold standard for assessing resting energy expenditure (REE) in critically ill patients, yet its use is limited due to various constraints. METHODS: A quality improvement initiative was launched at the Mayo Clinic to increase the use of IC among critically ill adults. The primary ob

Agreement between predictive equations and indirect calorimetry in estimating resting metabolic rate among free-living older adults with sarcopenia: A prospective observational study.

Resting metabolic rate (RMR) accounts for the largest proportion of daily total energy expenditure. The purpose of this study is to systematically evaluate the agreement of predictive equations for RMR and indirect calorimetry (IC) in a population with sarcopenia. The study included 43 individuals with sarcopenia aged 60 and above (diagnosed according to the AWGS 2019 criteria for sarcopenia). One-on-one measurements and questionnaires were used to obtain information including height, weight, body composition, gait speed, and grip strength. A total of 11 different predictive equations were use

Total energy expenditure and its components in healthy young adults from Southwest China using doubly labeled water, indirect calorimetry, accelerometry, and energy intake.

BACKGROUND: Energy requirement (ER) in young adults is commonly estimated from predicted resting metabolic rate (RMR) and assumed physical activity level (PAL). These assumptions may not fully reflect free-living total energy expenditure (TEE) in healthy young adults from Southwest China with low-activity daily routines. OBJECTIVE: This study compared doubly labeled water (DLW)-measured TEE with factorial TEE based on measured RMR and with equation-estimated TEE, interpreted as estimated ER, derived from Chinese and FAO/WHO/UNU RMR prediction equations. Measured and predicted RMR, TEE partiti

Transition state characterization of the low- to physiological-temperature nondenaturational conformational change in bovine adenosine deaminase by slow scan rate differential scanning calorimetry.

Bovine adenosine deaminase undergoes a nondenaturational conformational change at 29 degrees C upon heating which is characterized by a large increase in heat capacity. We have determined the transition state thermodynamics of the conformational change using a novel application of differential scanning calorimetry (DSC) which employs very slow scan rates. DSC scans at the conventional, and arbitrary, scan rate of 1 degree C/min show no evidence of the transition. Scan rates from 0.030 to 0.20 degrees C/min reveal the transition indicating it is under kinetic control. The transition temperature

Determining cold strain in cold air: a comparison of two methods of partitional calorimetry to calculate heat storage and debt in cold air with mild hypothermia.

We compared two methods of partitional calorimetry to calculate heat storage and heat debt during cold air (0°C) exposure causing mild core cooling. Twelve participants performed a 5 min baseline in thermoneutral conditions (∼22.0°C, ∼50% relative humidity) followed by cold air exposure (∼0°C) until rectal temperature was reduced by ∆-0.5°C. Partitional calorimetry was used to calculate avenues of heat exchange (radiative, convective, and evaporative), heat storage, and heat debt continuously throughout cold exposure. We compared deriving these variables using prediction equations based on env
