Development of Miniprotein-Type Inhibitors of Biofilm Formation in Candida albicans and Candida auris.

Candida auris is a pathogenic fungus associated with high-mortality infections and forms resilient biofilms on various surfaces. In this study, we introduced a novel antifungal strategy against C. auris by integrating an AI-powered protein design tool, ProteinMPNN, with classical molecular dynamics (MD) simulations to design artificial proteins from a miniprotein library. This combined approach accelerated and enhanced the design process, enabling the rapid development of effective miniprotein inhibitors specifically targeting C. auris biofilm formation. The miniproteins developed in this stud

Susceptibility of Candida auris and Candida albicans to 21 germicides used in healthcare facilities.

Candida auris is an emerging fungal pathogen that is often resistant to major classes of antifungal drugs. It is considered a serious global health threat because it can cause severe infections with frequent mortality in more than a dozen countries. It can survive on healthcare environmental surfaces for at least 7 days and can cause outbreaks in healthcare facilities. Clearly, infection prevention strategies, such as surface disinfection, will be essential to controlling Candida transmission. Unfortunately, data on the activity of antiseptics and disinfectants used in healthcare to inactivate

The role of annexin A1 in Candida albicans and Candida auris infections in murine neutrophils.

Annexin A1 (AnxA1) is an anti-inflammatory protein expressed in various cell types, especially macrophages and neutrophils. Because neutrophils play important roles in infections and inflammatory processes and the relationship between AnxA1 and Candida spp. infections is not well-understood, our study examined whether AnxA1 can serve as a target protein for the regulation of the immune response during fungal infections. C57BL/6 wild-type (WT) and AnxA1 knockout (AnxA1-/-) peritoneal neutrophils were coinfected with Candida albicans or Candida auris for 4 h. AnxA1-/- neutrophils exhibited a mar

Candida auris can acquire antifungal resistance without selection pressure.

The human fungal pathogen Candidozyma auris (formerly Candida auris) can cause prolonged infection outbreaks with high mortality rates in healthcare settings. Treatment failures of patients arise not only from antifungal drug resistance, but also from intra-species variability in pathogenicity as well as induced hypermutation events in response to clinical therapy. Whole genome sequencing was used to identify genetic mutations using the CDC mycoSNPs pipeline. Antifungal susceptibility testing was performed based on Clinical and Laboratory Standards Institute (CLSI) methods. We report here that

Validation of molecular detection of Candida auris from wastewater.

UNLABELLED: The high rates of multi-drug resistance and increasing transmission rates in healthcare environments reported for Candida auris (C. auris) led to its categorization as an urgent threat in the latest Centers for Disease Control and Prevention (CDC)'s Antibiotic Resistance Threats Report released in 2019. C. auris skin colonization and shedding contribute to its high transmissibility and allow for its recovery from wastewater. The incorporation of C. auris into wastewater surveillance workflows could aid in early detection of healthcare facilities with C. auris emergence and could le

Baicalein suppresses adhesion and biofilm formation in Candida auris.

Candida auris, an emerging multidrug-resistant fungal pathogen, poses a severe global public health threat owing to its high nosocomial transmissibility, considerable mortality, and widespread antifungal resistance. Baicalein (BE), a major bioactive constituent of Scutellaria baicalensis Georgi, exhibits notable antifungal potential, yet its specific molecular mechanisms against C. auris remain poorly elucidated. In this study, we determined the antifungal activity of BE against multiple C. auris isolates, assessed its effects on fungal growth, virulence attributes, adhesion, and biofilm devel

Candida auris undergoes adhesin-dependent and -independent cellular aggregation.

Candida auris is a fungal pathogen of humans responsible for nosocomial infections with high mortality rates. High levels of resistance to antifungal drugs and environmental persistence mean these infections are difficult to treat and eradicate from a healthcare setting. Understanding the life cycle and the genetics of this fungus underpinning clinically relevant traits, such as antifungal resistance and virulence, is of the utmost importance to develop novel treatments and therapies. Epidemiological and genomic studies have identified five geographical clades (I-V), which display phenotypic a

Azole resistance in Candida auris: mechanisms and combinatorial therapy.

Multidrug resistance Candida auris is a dangerous fungal pathogen that is emerging at an alarming rate and posing serious threats to public health. C. auris is associated with nosocomial infections that cause invasive candidiasis in immunocompromised patients. Several antifungal drugs with distinct mechanisms of action are clinically approved for the treatment of fungal infections. The high rates of intrinsic and acquired drug resistance, particularly to azoles, reported in characterized clinical isolates of C. auris make treatment extremely problematic. In systemic infections, azoles are the

Measuring Gene Expression via mRNA Abundance in Candida auris.

The recently emerged human pathogenic yeast Candida auris has become a major global threat. As compared to other Candida species, C. auris often displays a high level of resistance to commonly used antifungals and poses additional therapeutic challenges. There is a great need to understand the molecular basis of its success as a drug-resistant human pathogen. The study of condition-specific gene expression can provide good cues of regulatory circuitry governing high drug resistance. Here, we describe the protocol of quantitative reverse transcription PCR (RT-qPCR) which can be conveniently emp

Candida auris PCR for high-throughput infection control screening.

UNLABELLED: Candida auris has significant implications for infection control due to its multidrug resistance and spread in healthcare settings. Current culture-based screening methods are laborious and risk muco-cutaneous colonisation of laboratory staff. We describe the adaptation of a published real-time PCR for the identification of C. auris in skin swabs for high-throughput infection control screening. Two published primer and probe sets were analysed utilising serial 10-fold dilutions of 15 C. auris strains to assess the PCR limit of detection. One primer and probe set was compatible with

Innate immune responses against the fungal pathogen Candida auris.

Candida auris is a multidrug-resistant human fungal pathogen responsible for nosocomial outbreaks worldwide. Although considerable progress has increased our understanding of the biological and clinical aspects of C. auris, its interaction with the host immune system is only now beginning to be investigated in-depth. Here, we compare the innate immune responses induced by C. auris BJCA001 and Candida albicans SC5314 in vitro and in vivo. Our results indicate that C. auris BJCA001 appears to be less immunoinflammatory than C. albicans SC5314, and this differential response correlates with struc

Nine Things Genomics Can Tell Us About Candida auris.

Candida auris is a recently emerged multidrug-resistant fungal pathogen causing severe illness in hospitalized patients. C. auris is most closely related to a few environmental or rarely observed but cosmopolitan Candida species. However, C. auris is unique in the concern it is generating among public health agencies for its rapid emergence, difficulty to treat, and the likelihood for further and more extensive outbreaks and spread. To date, five geographically distributed and genetically divergent lineages have been identified, none of which includes isolates that were collected prior to 1996

Understanding Echinocandin Resistance in the Emerging Pathogen Candida auris.

Candida auris has simultaneously emerged on five continents as a fungal pathogen causing nosocomial outbreaks. The challenges in the treatment of C. auris infections are the variable antifungal susceptibility profiles among clinical isolates and the development of resistance to single or multiple classes of available antifungal drugs. Here, the in vitro susceptibility to echinocandin antifungal drugs was determined and FKS1 sequencing was performed on 106 C. auris clinical isolates. Four isolates were identified to be resistant to all tested echinocandins (MIC ≥ 4 mg/liter) and harbored an S63

Octenidine effectively reduces Candida auris colonisation on human skin.

Candidozyma (formerly Candida) auris (C. auris), a WHO critical priority pathogen known for its multi-drug resistance and strong skin tropism, is posing a significant health threat. This study evaluates the efficacy of commercial octenidine-based antiseptics in reducing C. auris colonisation on intact and wounded human skin. Using an established ex vivo human skin model to simulate clinical settings, skin samples from healthy donors were exposed to planktonic C. auris cells. Six hours post-contamination, two ready-to-use octenidine-based antiseptics were applied, and fungal colonisation was as

The high-osmolarity glycerol (HOG) pathway in Candida auris.

The emerging fungal pathogen Candida auris is known for its strong skin tropism and resilience against antifungal and disinfection treatment, posing a significant challenge for healthcare units. Although efforts to identify the effectors of its unique pathogenic behavior have been insightful, the role of the high-osmolarity glycerol (HOG) pathway in this context remains unexplored. The study by Shivarathri and co-workers (R. Shivarathri, M. Chauhan, A. Datta, D. Das et al., mBio 15:e02748-24, 2024, https://doi.org/10.1128/mbio.02748-24) sought to address this gap.
