Bioavailability of a Capsaicin Lipid Multi-particulate Formulation in Rats.

BACKGROUND AND OBJECTIVE: Because of the stomach-burning sensation it induces, capsaicin has been used at relatively low doses as a nutritional supplement, which has limited its bioavailability. The objective of this study was to investigate the serum bioavailability of capsaicin supplementation with or without a lipid multi-particulate (LMP) formulation. METHODS: Thirty-five rats were divided into five groups and administered capsaicin at either 0.2 or 1 mg/kg with or without the LMP formulation. Capsaicin bioavailability was assessed based on the area under the concentation-time curve (AUC)

Assessment of intraoral mucosal pain induced by the application of capsaicin.

OBJECTIVE: To develop an objective method for assessing nociceptive behaviour in an animal model of capsaicin-induced intraoral pain. Changes in nociceptive responses were also examined after injury to the inferior alveolar nerve (IAN). DESIGN: Nociceptive responses evoked by the intraoral application of various doses of capsaicin were analyzed in lightly anaesthetized rats. The number of c-Fos protein-like immunoreactive (Fos-LI) neurons in the medullary dorsal horn (MDH) induced by the intraoral application of capsaicin was measured. Behavioural and c-Fos responses were also examined 14 day

Capsaicin delivery into the skin with lipidic nanoparticles for the treatment of psoriasis.

The study aims to explore the potential of solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs) in improving the topical delivery of capsaicin (CAP) by in vitro and in vivo studies. The lipidic nanoparticles were prepared by solvent diffusion method and were characterized for average particle size, zeta potential and entrapment efficiency. TEM photomicrographs revealed that the particles were nanometric in size. Higher amount of CAP can be encapsulated in the NLCs (87.4 ± 3.28) as compared with SLNs (79.7 ± 2.93%). The cumulative amounts of CAP permeated through the skin a

[Capsaicin and lidocaine usage in functional disorders of urinary bladder].

Most of the drugs in the treatment of functional disorders of the urinary bladder has a peripheral effect. Their work consists mainly in reducing detrusor contractility of the bladder, or effects on the afferent innervation. Anticholinergics are the first drugs of choice. An alternative pharmacological treatment is to eliminate the overactivity by acting on the bladder afferent innervation, while not inhibiting its contractility. One option is to modulate the pharmacological activity of sensory mechanisms governing the functioning of the bladder via the vanilloid receptor (TRPV1) and ancyrin (

Characterization of sensations induced by capsaicin in the upper gastrointestinal tract.

Intraluminal capsaicin induces perception in the jejunum, but chemosensitivity of proximal gastrointestinal regions is unclear. Our aim was to evaluate the quality of perception induced by intraluminal capsaicin in different regions of the upper gastrointestinal tract. Healthy volunteers received either an oral tube for distension and capsaicin perfusion of the mid-duodenum or jejunum or swallowed a capsule containing 0.75 mg capsaicin powder. Graded questionnaires evaluated quality and severity of sensations during distensions, capsaicin infusion and 30 min after ingestion of capsaicin capsul

The influence of ambroxol and capsaicin on the isolated rabbit bladder wall.

Unmyelinated C-fibers endings lye beneath the epithelial layer and release neuropeptides which regulate baseline transepithelial potential difference (PD) and changes of transepithelial potential difference during mechanical stimulation (dPD). Ambroxol can suppress reflexes such as the cough or the corneal reflex which are connected to stimulation of C fibre endings. The study aimed to estimate the influence of ambroxol (ABX) and capsaicin (CAPSA) on PD and dPD in isolated rabbit bladder. The experiments were carried out on 26 bladder specimens of. 2 cm2 surface area each, obtained from 13 rab

[Topical administration of capsaicin in dermatology for treatment of itching and pain].

BACKGROUND AND OBJECTIVES: Treatment of pruritus as observed in many dermatological and internal diseases may be very often disappointing. There are many reports describing the topical use of capsaicin, the pungent agent in red pepper. Long term administration of capsaicin depletes neuropeptides in unmyelinated, polymodal C-type and small myelinated A delta-type cutaneous nerves that conduct pruritus and pain. PATIENTS/METHODS: The aim of this study was to evaluate the efficacy, safety and practicability of capsaicin in the treatment of pruritus and pain of different origin in a total of 40 p

A capsaicin-receptor homologue with a high threshold for noxious heat.

Pain-producing heat is detected by several classes of nociceptive sensory neuron that differ in their thermal response thresholds. The cloned capsaicin receptor, also known as the vanilloid receptor subtype 1 (VR1), is a heat-gated ion channel that has been proposed to mediate responses of small-diameter sensory neurons to moderate (43 degrees C) thermal stimuli. VR1 is also activated by protons, indicating that it may participate in the detection of noxious thermal and chemical stimuli in vivo. Here we identify a structurally related receptor, VRL-1, that does not respond to capsaicin, acid o

Capsaicin increases the frequency of cryptorchidism in flutamide-treated rats.

Capsaicin, a specific neurotoxin that irreversibly results in degeneration of sensory C fibers of the peripheral nerves, was investigated for its ability to affect testicular descent. In particular, we investigated the ability of prenatal flutamide, postnatal capsaicin or combined prenatal flutamide with postnatal capsaicin to induce undescended testes in rats. Within these treatment groups the frequency of maldescent was 42% in the animals treated prenatally with flutamide, 32% with postnatal capsaicin, and 100% with prenatal flutamide and postnatal capsaicin. Of interest, it was found that a

[Effect of capsaicin on the migration of eosinophils into the bronchi of guinea pigs].

Substance P (SP), a potent neuropeptide, which is localized in the sensory nerves and released by many physiological stimuli has been implicated in airway neurogenic inflammation. We have studied the effects of capsaicin (CAP), which releases tachykinins (TK) from the sensory nerves, on eosinophil (EOS) recruitment in the airway in guinea pigs in vivo. Male guinea pigs were used. The respiratory resistance (Rrs) of the guinea pigs were measured by an oscillation technique and histological studies of the right main bronchus were carried out. Exposure to inhaled CAP resulted in a significant inc

The consequences of long-term topical capsaicin application in the rat.

Capsaicin has been used extensively as an experimental tool and in traditional and proprietary topical medications for acute soft tissue injuries. More recently it has been prescribed for several chronic pain conditions where it is usually administered topically for periods of several weeks. Here we have studied the consequences of this mode of application in the rat. Capsaicin cream (0.075% or 0.75%), or a vehicle cream, was applied twice daily to the hind paws of rats for a continuous period of 10 weeks. The hind paws treated with 0.75% capsaicin (but not 0.075%) because transiently hyperalg

The post-mastectomy pain syndrome and the effect of topical capsaicin.

Eighteen patients with the post-mastectomy pain syndrome (PMPS) form the basis of this study. PMPS probably occurs in a minority of women after mastectomy. The onset of persistent pain usually occurred immediately or very shortly after the operation. The pain location or sensory findings implied involvement of the territories of other cutaneous branches of the intercostal nerves as well as the intercostobrachial nerve. A variety of treatment approaches were unsatisfactory. Twelve of 14 patients completing treatment with topical 0.025% capsaicin showed improvement after 4 weeks and 8 (57%) were

Capsaicin inhibits cariogenic Streptococcus mutans-Candida albicans cross-kingdom biofilms.

OBJECTIVE: This research aimed to determine whether capsaicin (CAP), a phytochemical in chilli peppers, inhibits cariogenic Streptococcus mutans-Candida albicans cross-kingdom biofilms and their key virulence factors, and to explore underlying mechanisms. DESIGN: The anti-caries potential of CAP was assessed in S. mutans-C. albicans co-cultures by quantifying viable cell counts, biomass and biofilm metabolic activity. Biofilm microstructure was visualized by confocal laser scanning microscopy with COMSTAT quantification, and CAP-induced transcriptional changes were profiled by qRT-PCR. Matche
