Species differences in carcinogenicity: the role of metabolism and pharmacokinetics in risk assessment.

Attempts to improve quantitative risk assessments inevitably lead to the use of additional biological data in the risk calculation. The need for more data increases further when differences in response between laboratory animals result in uncertainty in the choice of either the species or the tumour incidence on which to base the risk assessment. Of the many stages of carcinogenesis, the first stage, which involves the uptake and activation of the chemical, is probably the most understood and is by far the easiest to measure experimentally. A review of the use of metabolism and pharmacokinetic

Role of cobalt, iron, lead, manganese, mercury, platinum, selenium, and titanium in carcinogenesis.

The possible carcinogenicity of cobalt, iron, lead, manganese, mercury, platinum, selenium, and titanium is reviewed, taking into account epidemiological data, the results of animal experimental studies, data on mutagenic effects and on other in vitro test systems. Of the great variety of occupations where exposure to one of these metals may occur, only haematite mining has been clearly shown to involve an increased human cancer risk. While the possibility that haematite might in some way act as a carcinogen has to be taken into consideration it is more likely that other carcinogens are respon

Mechanisms of anti-carcinogenesis by indole-3-carbinol: detailed in vivo DNA binding dose-response studies after dietary administration with aflatoxin B1.

Several recent reports have described inhibitor-mediated reductions in the covalent binding of various carcinogens to DNA in vivo. The majority of these studies show inhibitory effects after testing at one inhibitor and one carcinogen dose level only. Consequently, the detailed relationships between inhibitor dose, carcinogen dose, and in vivo inhibitory potency have not been clearly delineated in any species. To systematically determine these relationships in vivo, rainbow trout (Salmo gairdneri) were exposed to a range of carcinogen (aflatoxin B1, AFB1) and inhibitor (indole-3-carbinol, I3C)

Enhancing effects of various gastric carcinogens on development of pepsinogen-altered pyloric glands in rats.

To assess the possibility of establishing an in vivo, medium-term bioassay system for gastric carcinogens and promoters, a total of 220 male WKY rats were divided into two groups. Group 1 animals were treated first with a single dose of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) (160 mg/kg body wt) and starting 2 weeks later administrated one of five gastric carcinogens, a gastric promoter, one of five non-gastric carcinogens or no treatment, as a control, for 14 weeks. Saturated sodium chloride solution (1 ml) treatments were given by gastric intubation at weeks 4, 6, 8 and 10. Group 2 rats

Human fecal fractions can produce nuclear damage in the colonic epithelial cells of mice.

Nuclear toxicity of several known carcinogens and fecal fractions obtained from 10 healthy individuals was investigated in the colonic nuclear aberration (NA) assay using an intrarectal administration. Two known colon carcinogens, MNNG and DMAB, and a carcinogen of organs other than the colon, B(a)P, induced NA in a dose-related manner. Chromatographic fractions of feces from 10 donors were tested for their ability to produce NA. The dichloromethane fraction for several was active and yielded a significantly positive response which was dose-related. Our study demonstrated that the feces of som

DNA repair-deficient Xpa and Xpa/p53+/- knock-out mice: nature of the models.

Xeroderma pigmentosum (XP) is a rare autosomal recessive disease in which repair of ultraviolet (UV)-induced DNA damage is impaired or is totally absent due to mutations in genes controlling the DNA repair pathway known as nucleotide excision repair (NER). XP is characterized, in part, by extreme sensitivity of the skin to sunlight, and XP patients have a more than 1000-fold increased risk of developing cancer at sun-exposed areas of the skin. To study the role of NER in chemical-induced tumorigenesis in more detail, the authors developed Xpa-/- homozygous knockout mice with a complete defect

Usefulness and rapidity of screening for the toxicity and carcinogenicity of chemicals in medaka, Oryzias latipes.

The toxicity and carcinogenicity of number of chemicals, for which there are toxicity and/or carcinogenicity data in mouse and rat, were tested in medaka, a small aquarium fish (Oryzias latipes). The water-soluble chemicals were dissolved in the water and the water-insoluble chemicals were incorporated into the diet. Observation for 24 wk revealed induction of liver cell carcinoma by 6 chemicals; aflatoxins B1 and G1, sterigmatocystin, ortho-aminoazotoluene, methylazoxymethanol acetate and N-nitrosodiethylamine. In medaka fed PCB or DDT only preneoplastic liver cell nodules were observed. No e

Triesters of glycerin and aliphatic acids, known generically as glyceryl triesters and specifically as Trilaurin, etc., are used in cosmetic products as occlusive skin-conditioning agents and/or nonaqueous viscosity-increasing agents. Hundreds of glyceryl triesters are used in a wide variety of cosmetic products at concentrations ranging from a few tenths of a percent to 46%. Glyceryl triesters are also known as triglycerides; ingested triglycerides are metabolized to monoglycerides, free fatty acids, and glycerol, all of which are absorbed in the intestinal mucosa and undergo further metaboli

A comprehensive review of mechanistic insights into formaldehyde-induced nasal cavity carcinogenicity.

According to the International Agency for Research on Cancer classification, formaldehyde is a human carcinogen that targets the nasal cavity. In humans and rats, inhaled formaldehyde is primarily deposited in the nasal cavity mucosa, metabolized to the less toxic formic acid, and finally excreted into the urine or exhaled. Thus, formaldehyde-induced nasal carcinogenicity may be a direct effect of formaldehyde itself, although the underlying mechanisms remain unclear. With regard to cytotoxicity, degeneration and necrosis of nasal respiratory cells occur in rats after short exposure to formald

Suitability of the ICH S1B(R1) Weight-of-Evidence Framework to Evaluate Glucagon-Like Protein-1 Receptor Agonist (GLP-1RA) Carcinogenicity Risk.

Glucagon-like peptide 1 receptor agonists (GLP-1RAs) are an established treatment for management of type 2 diabetes mellitus (T2DM) and obesity. While these agents provide clinical benefits, chronic rodent bioassays have shown a consistent increase in thyroid C-cell tumors, raising regulatory concerns for human carcinogenicity risk and resulting in a boxed warning for medullary thyroid carcinoma (MTC). However, the human relevance of the rodent tumors is unclear due to species differences in GLP-1 receptor (GLP-1R) biology. Carcinogenicity testing of pharmaceuticals is described in ICH S1 guid

Transgenerational effects from exposure to environmental toxic substances.

Exposure of mouse germ cells to radiation and chemicals results in mutation, malformation, cancer and other adverse effects (e.g., functional disorders) in the offspring, though these findings have not been proven in human studies. Environmental toxic substances such as urethane (ethyl carbamate) which had been injected subcutaneously to 50 million people as a co-solvent of analgesics and dioxin (an endocrine disruptor) have been found to be associated with adverse effects in the progeny of mice after parental exposures. There are some reports on congenital malformations in the progeny of fath

Medium-term bioassay system for detection of carcinogens and modifiers of hepatocarcinogenesis utilizing the GST-P positive liver cell focus as an endpoint marker.

We have developed a medium-term bioassay system of 8 weeks duration utilizing male Fischer 344 (F344) rats for detection of liver carcinogens and modifiers of hepatocarcinogenesis. The system consists of a single intraperitoneal injection of diethylnitrosamine (DEN, 200 mg/kg), 6-weeks-administration of test chemical beginning 2 weeks after the DEN injection, and 2/3 partial hepatectomy (PH) performed at week 3. Carcinogenic potency of test chemicals is predicted based on the results of quantitative analyses of immunohistochemically-demonstrated glutathione S-transferase placental form (GST-P)

Deregulation of cancer-related pathways in primary hepatocytes derived from DNA repair-deficient Xpa-/-p53+/- mice upon exposure to benzo[a]pyrene.

The current method to predict carcinogenicity of chemicals or drugs is the chronic 2-year rodent bioassay, which has disadvantages in duration, animal use, and specificity. An attractive alternative is the DNA repair-deficient Xpa(-/-)p53(+/-) mouse model that is sensitive to both genotoxic and nongenotoxic carcinogens. A next step in alternative carcinogenicity testing is the development of reliable in vitro systems. We investigated the use of primary hepatocytes, isolated from wild-type (WT) and Xpa(-/-)p53(+/-) mice, in combination with transcriptome analyses for their usefulness to predict

CoFe2O4/TiO2-Nb2O5 magnetic catalysts synthesized by chemical and physical mixture applied in the reduction of Cr(VI).

Among heavy metals, chromium stands out, given the high toxicity and carcinogenicity of Cr(VI). The use of magnetic photocatalysts has opened up new possibilities for their large-scale application, given the ease of catalyst separation. The present work was dedicated to the synthesis and characterization of CoFe2O4/TiO2-Nb2O5 magnetic catalysts and their application in the photocatalytic reduction of chromium. Cobalt ferrite and TiO2-Nb2O5 were combined through chemical (sol-gel synthesis) or physical (mortar and pestle) mixing. In preliminary tests to assess the catalytic activity, approximat

A quantitative weight-of-evidence review of preclinical studies examining the potential carcinogenicity of acetaminophen.

Introduced in the 1950s, acetaminophen is one of the most widely used antipyretics and analgesics worldwide. In 1999, the International Agency for Research on Cancer (IARC) judged the available data inadequate to determine carcinogenicity, and in 2024 an IARC Advisory Group recommended high-priority re-evaluation. Given this context, we conducted a quantitative weight-of-evidence (QWoE) analysis of in vivo rodent carcinogenicity studies using a previously developed framework for biological relevance and methodological quality (Kougias et al., 2025a; Kougias et al., 2025b). A total of 192 QWoE

28-Day somatic gene mutation study of 1-bromopropane in female Big Blue® B6C3F1 mice via whole-body inhalation: Support for a carcinogenic threshold.

A 2-year inhalation rat and mouse cancer study by the National Toxicology Program (NTP) on 1-bromopropane, a brominated solvent most commonly used as a vapor degreaser, showed significant increase in tumors in the lung of female mice and in the large intestine of male and female rats. The most sensitive endpoint was lung tumors in female mice. Mice of both sexes had hyperplasia and inflammation of the nose and showed regeneration of lung tissue. The NTP assumed that these tumors were due to genotoxic effects and that a linear dose-response relationship was appropriate. It is plausible that, si

Electrochemical behavior and determination of tumor inhibiting or promoting activities of flavonoids.

This paper deals with determination of the tumor inhibiting or promoting activities of 11 flavonoids performed by DC polarography. Flavonoids were tested in the presence of polyaromatic carcinogen 7,12-dimethylbenz(a)anthracene (DMBA) or in combination with DMBA and 12-O-tetradecanoylphorbol-13-acetate (TPA), which is known as a specific tumor promoter for epidermal carcinogenesis. We found that in this experimental system the promotory or inhibitory activities of studied flavonoids depend on the number and position of hydroxyl groups in their chemical structures and are related to the polarog

Factors contributory to early death of young CD-1 mice in carcinogenicity studies.

It is often difficult to determine the cause of early or sporadic deaths in toxicity studies, making interpretation difficult. To help in the interpretation of early deaths, it is critical that the background incidence of factors contributing to death be recorded and archived. Information was gathered from the control groups of 20 CD-1 mouse carcinogenicity studies. Route of administration was either dietary or oral gavage, and animals were housed in groups of one to four in wired or solid bottom cages. Analysis was confined to sporadic deaths in control groups occurring during the first 50 we

Defining early carcinogenic markers for the prediction of the hazard potential of mineral fibres in an in vitro human 3D bronchial model.

This study aimed to identify early carcinogenic markers to predict the hazard potential of mineral fibres using a human 3D bronchial model. Traditional 2D cell models inadequately mimic lung complexity, prompting the use of more physiologically relevant 3D systems. We investigated the effects of crocidolite (CRO) and two size fractions of chrysotile (≤ and > 5 μm length: CHR S and CHR L) to elucidate early toxicological mechanisms and relate them to the IARC key characteristics (KCs) of carcinogens. Initial analyses (MTT, TEER, and histology) at 24-48 h showed a transient acute toxicity and ti
