Role of Catalase in Oxidative Stress- and Age-Associated Degenerative Diseases.

Reactive species produced in the cell during normal cellular metabolism can chemically react with cellular biomolecules such as nucleic acids, proteins, and lipids, thereby causing their oxidative modifications leading to alterations in their compositions and potential damage to their cellular activities. Fortunately, cells have evolved several antioxidant defense mechanisms (as metabolites, vitamins, and enzymes) to neutralize or mitigate the harmful effect of reactive species and/or their byproducts. Any perturbation in the balance in the level of antioxidants and the reactive species result

Identification and characteristic analysis of the catalase gene from Locusta migratoria.

Catalase (CAT) is a ubiquitous antioxidant enzyme in almost all living organisms exposed to atmosphere, which involved in decomposing harmful hydrogen peroxide, into oxygen and water. In this study, a full-length cDNA (1524bp) encoding the catalase gene (LmCAT) from Locusta migratoria was cloned (accession number KT716445). The open reading frame of the LmCAT gene encoded 507 amino acids and shared 57.8%-97.8% amino acid identities with other insect CATs. The coding region was interrupted by 9 introns, while its promoter region contained 15 putative binding sites for 5 kinds of transcriptional

Promoter variant in the catalase gene is associated with vitiligo in Chinese people.

Vitiligo is an acquired depigmentation disorder, and reactive oxygen species have an important role in the physiology of cell damage. Reduced catalase enzyme activity and accumulation of excessive hydrogen peroxide have been observed in vitiligo. In a hospital-based case-control study of vitiligo patients (n=749) and age- and sex-matched healthy controls (n=763), we investigated three catalase (CAT) gene polymorphisms (-89A>T, 389C>T, and 419C>T) to examine whether CAT gene polymorphisms are associated with vitiligo susceptibility in the Chinese population. The case-control analysis revealed a

Inhibition of peritoneal dissemination of tumor cells by cationized catalase in mice.

To inhibit peritoneal dissemination of tumor cells by destroying hydrogen peroxide, ethylenediamine-conjugated catalase (ED-catalase), a cationized derivative, was injected into the peritoneal cavity of mice. ED-catalase had about a 6-fold longer retention time within the cavity than unmodified catalase. Peritoneal dissemination was evaluated after intraperitoneal inoculation of B16-BL6/Luc, a melanoma clone stably expressing firefly luciferase, by measuring luciferase activity. An intraperitoneal injection of ED-catalase just before tumor inoculation significantly reduced the number of tumor

Genotoxicity of bleomycin in human cell lines differing in catalase activity.

The influence of catalase on the genotoxic effect of bleomycin (BLM) has been evaluated in three cell lines which differ in catalase activity. CRL1307, cells from Xeroderma pigmentosum patient and CLV102, normal embryonic cells have catalase activity 3.5 and 5 times lower then CRL2088, normal skin fibroblasts. Genotoxicity of BLM (0.5-50 micrograms/ml, 2 h treatment) measured with in vitro micronucleus test did not differ in three tested lines. BLM at concentration range from 1 to 25 micrograms/ml (2 h treatment), tested in comet assay, caused similar degree of DNA damage in CLV102 and CRL2088

Mycobacterial FurA is a negative regulator of catalase-peroxidase gene katG.

In several bacteria, the catalase-peroxidase gene katG is under positive control by oxyR, a transcriptional regulator of the peroxide stress response. The Mycobacterium tuberculosis genome also contains sequences corresponding to oxyR, but this gene has been inactivated in the tubercle bacillus because of the presence of multiple mutations and deletions. Thus, M. tuberculosis katG and possibly other parts of the oxidative stress response in this organism are either not regulated or are controlled by a factor different from OxyR. The mycobacterial FurA is a homologue of the ferric uptake regula

Abnormality in catalase import into peroxisomes leads to severe neurological disorder.

Peroxisomal disorders are lethal inherited diseases caused by either defects in peroxisome assembly or dysfunction of single or multiple enzymatic function(s). The peroxisomal matrix proteins are targeted to peroxisomes via the interaction of peroxisomal targeting signal sequences 1 and 2 (PTS1 or PTS2) with their respective cytosolic receptors. We have studied human skin fibroblast cell lines that have multiple peroxisomal dysfunctions with normal packaging of PTS1 and PTS2 signal-containing proteins but lack catalase in peroxisomes. To understand the defect in targeting of catalase to peroxi

Stimulation by nitroxides of catalase-like activity of hemeproteins. Kinetics and mechanism.

The ability of stable nitroxide radicals to detoxify hypervalent heme proteins such as ferrylmyoglobin (MbFeIV) produced in the reaction of metmyoglobin (MbFeIII) and H2O2 was evaluated by monitoring O2 evolution, H2O2 depletion, and redox changes of the heme prosthetic group. The rate of H2O2 depletion and O2 evolution catalyzed by MbFeIII was enhanced by stable nitroxides such as 4-OH-2,2,6,6-tetramethyl-piperidinoxyl (TPL) in a catalytic fashion. The reduction of MbFeIV to MbFeIII was the rate-limiting step. Excess TPL over MbFeIII enhanced catalase-like activity more than 4-fold. During di

[Erythrocyte catalase activity in patients with endemic nephropathy and in their family members].

Etiology and pathogenesis of endemic nephropathy (EN) are unknown and in this work the possible pathogenetic role of disturbed antioxidant protection, estimated by the activity of erythrocyte catalase, was evaluated. In patients with EN as well as in clinically healthy members of their families a statistically significant increase in catalase activity (16.7 +/- 0.63 x 10(4) U/g Hb) compared to the control group of the blood bank donors from the city of Nish (11.7 +/- 0.69 x 10 U/g Hb) was found. Increased catalase activity, at the level of significance, was also found in clinically healthy mem

Contribution of superoxide dismutase and catalase activities to Shigella flexneri pathogenesis.

A Shigella flexneri serotype 5 strain deficient in the production of the iron-containing superoxide dismutase FeSOD (sodB) and a catalase-negative (katFG) S. flexneri serotype 5 strain were isolated. Both strains were examined for increased sensitivity to oxygen stress by using assays involving killing by mouse peritoneal macrophages and human polymorphonuclear leukocytes as well as infection of rabbit ileal loops. The sodB mutant was extremely sensitive to killing by phagocytes when compared with the wild-type parent, M90T. The catalase mutant also showed an increased sensitivity to killing,

The gene for catalase is assigned between the antigen loci MIC4 and MIC11.

Genetic analysis of the cells of a WAGR patient (W, predisposition to Wilms tumor; A, aniridia; G, genitourinary abnormalities; R, mental retardation), bearing a partial deletion of band 11p13, was performed with biochemical and antigenic 11p markers by using gene dosage, somatic hybridization, molecular hybridization, and indirect immunofluorescence techniques. These studies allowed the regional assignment of the gene for catalase, which is linked to the Wilms tumor locus, between MIC4 and MIC11, two loci encoding for membrane antigens previously mapped to band 11p13.

Isolation of a cDNA clone for murine catalase and analysis of an acatalasemic mutant.

We have investigated the genetic control of murine catalase expression by analyzing catalase transcription and translation products from the tissues of control (Csa) and acatalasemic (Csb) mouse strains. Csb animals possess nearly normal catalase enzyme activity levels in liver, while displaying approximately 20 and 1% of normal activity levels in kidney and red blood cells, respectively. Immunoblot analyses of catalase in these tissues have revealed reduced levels of immunologically reactive catalase protein in Csb kidney and red blood cells, paralleling the reduction of catalase enzyme activ

Catalase determination in various etiologic forms of Wilms' tumor and gonadoblastoma.

We have previously mapped the gene coding for catalase to 11p13 by gene dosage analysis. Deletion of this chromosomal region causes aniridia, mental retardation, and predisposition to Wilms' tumor (WT). In the present study, 22 patients with various etiologic forms of WT and/or aniridia were investigated. The catalase (CAT) level and karyotype were examined in order to determine the linkage and the gene ordering on chromosome number 11 of the different loci involved. The CAT concentration was normal in the 19 cases without detectable chromosomal abnormalities.

[Effect of 1,2,4-triaminotriazole on Candida mycoderma catalase in substrate oxidation].

The effect of 1,2,4-triaminotriazole on the activity of catalase was studied in the resting cells of Candida mycoderma when they oxidized glucose, ethanol and other alcohols. The compound was shown to inhibit the activity of catalase in the cells when the endogenous substrate and glucose were oxidized, but it had no effect on its activity during the oxidation of alcohols (C1-C4). Catalase is supposed to be involved in the oxidation of lower alcohols forming a complex with them and thus preventing the inhibiting action of 1,2,4-triaminotriazole.

Analysis of heme biosynthesis in catalase and cytochrome deficient yeast mutants.

Mutants of Saccharomyces cerevisiae, described as catalase and cytochromes deficient (Pachecka et al., 1974), have been analyzed for heme biosynthesis ability. Some enzymatic activities involved in protoheme synthesis were measured in acellular extracts, whereas whole cells were analyzed for cytochrome spectra and for possible accumulation of porphyrin synthesis intermediates. A good correlation was found between these in vitro and in vivo studies. Results show that two mutants were impaired in 5-aminolevulinate synthesis, two mutants were devoid of uroporphyrinogen I synthetase activity and o
