Cytoplasmic Delivery of an Antibiotic, Trimethoprim, with a Simple Bidentate Catechol Analog as a Siderophore Mimetic.

Concerns about antibiotic-resistant Gram-negative pathogens are escalating, and accordingly siderophore-based intracellular antibiotic delivery is attracting more attention as an effective means to overcome these infections. Despite the successful clinical translation of this strategy, the delivery potential of siderophores has been limited to periplasm targeting, and this has appreciably restricted the repertoire of applicable antibiotics. To overcome this shortcoming of the current technology, this study focused on investigating the capability of simple bidentate catechol analogs to function

Catechol siderophores framed on 2,3-dihydroxybenzoyl-L-serine from Streptomyces varsoviensis.

Enterobactin is an archetypical catecholate siderophore that plays a key role in the acquisition of ferric iron by microorganisms. Catechol moieties have been shown to be promising siderophore cores. Variants of the conserved 2,3-dihydroxybenzoate (DHB) moiety with structural modifications expand the bioactivity. Streptomyces are characterized by metabolites with diverse structures. The genomic sequence of Streptomyces varsoviensis indicated that it possessed a biosynthetic gene cluster for DHB containing siderophores and metabolic profiling revealed metabolites correlated with catechol-type n

Integrated Mass Spectrometry Reveals Celastrol As a Novel Catechol-O-methyltransferase Inhibitor.

Natural product celastrol is known to have various biological activities, yet its molecular targets that correspond to many activities remain unclear. Here, we used multiple mass-spectrometry-based approaches to identify catechol-O-methyltransferase (COMT) as a major binding target of celastrol and characterized their interaction comprehensively. Celastrol was found to inhibit the enzymatic activity of COMT and increased the dopamine level in neuroendocrine chromaffin cells significantly. Our study not only revealed a novel binding target of celastrol but also provided a new scaffold and cyste

Catechol-Based Functionalizable Ligands for Gallium-68 Positron Emission Tomography Imaging.

Four tris-bidentate catecholamide (CAM) ligands were synthesized, characterized, and evaluated as ligands for radiolabeling of gallium-68 for positron emission tomography (PET). Three of those ligands, 2,2-Glu-CAM, 3,3-Glu-CAM, and TREN-bisGlyGlu-CAM, incorporate ligand caps that contain a pendant carboxylic group for further conjugation to targeting moieties. The acyclic ligands all exhibited high (>80%) radiolabeling yields after short reaction times (<10 min) at room temperature, a distinct advantage over macrocyclic analogues that display slower kinetics. The stabilities of the four GaIII

Enhanced chemiluminescence of the fluorescein-KIO4 system by CdTe quantum dots for determination of catechol.

In this paper, a novel chemiluminescence (CL) system was introduced based on the use of CdTe quantum dots (QDs) with the mixture solutions of fluorescein and potassium periodate (KIO4 ) in alkaline medium. The CL signal of an ultra-weak system was strongly enhanced in the presence of QDs. The application of CdTe QDs-fluorescein-KIO4 system is reported for the first time. It was found that catechol had a diminishing effect on the CL reaction. Under optimal experimental conditions, CL intensity decreased linearly in a 1 to 100 μM catechol concentration range, with a 0.18 μM detection limit. A po

Synthesis and assessment of catechol diether compounds as inhibitors of trypanosomal phosphodiesterase B1 (TbrPDEB1).

Human African trypanosomiasis (HAT) is a parasitic neglected tropical disease that affects 10,000 patients each year. Current treatments are sub-optimal, and the disease is fatal if not treated. Herein, we report our continuing efforts to repurpose the human phosphodiesterase 4 (hPDE4) inhibitor piclamilast to target trypanosomal phosphodiesterase TbrPDEB1. We prepared a range of substituted heterocyclic replacements for the 4-amino-3,5-dichloro-pyridine headgroup of piclamilast, and found that these compounds exhibited weak inhibitory activity of TbrPDEB1.

Catechol-O-methyltransferase gene variation: impact on amygdala response to aversive stimuli.

The functional catechol-O-methyltransferase (COMT) val158met polymorphism has been found to be associated with anxiety disorders and depression as well as with neural correlates of emotional processing, with, however, contradictory results. Thus, the aim of the present study was to re-evaluate the impact of the COMT val158met variant on neural activation correlates of emotional face processing in a sample of healthy probands. In 85 healthy subjects genotyped for the COMT val158met polymorphism, amygdala responses were assessed by means of fMRI. Participants were presented with anger- and fear-

Catechol-O-methyltransferase inhibitors in preclinical models as adjuncts of L-dopa treatment.

Long-term L-dopa treatment is limited by the development of motor complications, such as motor fluctuations and dyskinesias. These motor complications are postulated to arise from a non-physiological intermittent or pulsatile stimulation of striatal dopamine (DA) receptors that normally receive tonic stimulation. The concept of continuous dopaminergic stimulation (CDS) proposes that therapies providing more continuous stimulation of brain dopaminergic receptors are associated with a reduced risk of motor complications. One approach to the CDS is to prolong the half-life of L-dopa inhibiting it

Catechol-O-methyltransferase: effects of the val108met polymorphism on protein turnover in human cells.

A single nucleotide polymorphism in the human COMT (catechol-O-methyltransferase) gene has been associated with increased risk for breast cancer and several CNS diseases and disorders. The G to A polymorphism causes a valine (val) to methionine (met) substitution at codon 108 soluble - (S)/158 membrane - (MB)-COMT, generating alleles encoding high and low-activity forms of the enzyme, COMT H and COMT L, respectively. Tissues and cells with a COMT LL genotype have decreased COMT activity compared to COMT HH cells. Previously, we reported that the decreased activity was due to decreased amounts

Comprehensive assessment of genetic variation of catechol-O-methyltransferase and breast cancer risk.

Because catechol-O-methyltransferase (COMT) catalyzes the addition of methyl groups to stabilize catechol estrogens that may induce DNA damage, genetic variants could influence breast cancer risk. To comprehensively characterize genetic variation in this gene, we selected haplotype-tagging single nucleotide polymorphisms (htSNP) in COMT. A total of 11 htSNPs (including COMT Val(158)Met) were selected based on the resequencing and dense genotyping approach of the Breast and Prostate Cancer Cohort Consortium. htSNPs were genotyped in a population-based, case-control study in Poland (1,995 cases

Crystal structures of the reaction intermediate and its homologue of an extradiol-cleaving catecholic dioxygenase.

BphC derived from Pseudomonas sp. strain KKS102 is an extradiol-cleaving catecholic dioxygenase. This enzyme contains a non-heme iron atom and plays an important role in degrading biphenyl/polychlorinated biphenyls (PCBs) in the microbe. To elucidate detailed structures of BphC reaction intermediates, crystal structures of the substrate-free form, the BphC-substrate complex, and the BphC-substrate-NO (nitric oxide) complex were determined. These crystal structures revealed (1) the binding site of the O(2) molecule in the coordination sphere and (2) conformational changes of His194 during the c

Zinc/Catechol Resin-Based Microsphere Coating for Durable Antibacterial Cotton Fabrics.

Zinc oxide nanoparticles (ZnO NPs) exhibit strong and broad-spectrum antibacterial properties, making them a promising agent for textile applications. However, their weak adhesion to fibers and poor washing durability have hindered practical use. In this work, we report zinc/catechol resin-based microspheres (Zn/CFRs) synthesized via a one-pot hydrothermal route and applied to cotton fabric through a pad-dry-cure process. The resulting Zn/CFRs exhibit a monodisperse spherical morphology, with zinc ions concentrated on the surface and ZnO NPs encapsulated within the resin matrix. The finished f

Spasmolytic Effect of Flopropione Does Not Involve Catechol-O- methyltransferase (COMT) Inhibition.

Flopropione (Flo) has been used for gallstone and urolithiasis as a spasmolytic agent almost exclusively in Japan. According to the package insert, its main mechanism is catechol-O-methyltransferase (COMT) inhibition and anti-serotonergic effect. This is obviously contrary to pharmacological common sense, but it is described that way in pharmacology textbooks and occurs in questions in the National Examination for Pharmacists in Japan. As this is a serious problem in education, we re-examined the action of Flo. The guinea pig ureter was hardly contracted by serotonin, but noradrenaline (NA) el

Natural and Nature-Inspired Catechol Siderophores: A Promising Strategy for Rice Blast Management.

The rice-blast fungus Pyricularia oryzae poses a significant threat to rice production worldwide. Ferroptosis, an iron-dependent form of regulated cell death, has recently been reported to be involved in P. oryzae pathogenicity during plant-fungal interactions. Ferroptosis regulates the developmental cell death of conidia necessary for appressorium maturation. In this study, we have established that a series of benzamides containing a chelating catechol moiety suppresses the formation/maturation of appressoria, which are essential for host infection by the rice blast fungus. Moreover, for the

Catechol-Isolated Atomically Dispersed Nanocatalysts for Self-Motivated Cocatalytic Tumor Therapy.

Nanocatalytic tumor therapy based on Fenton nanocatalysts has attracted considerable attention because of its therapeutic specificity, enhanced outcomes, and high biocompatibility. Nevertheless, the rate-determining step in Fenton chemistry, which involves the transition of a high-valence metallic center (FeIII ) to a Fenton-active low-valence metallic center (FeII ), has hindered advances in nanocatalyst-based therapeutics. In this study, we constructed mesoporous single iron atomic nanocatalysts (mSAFe NCs) by employing catechols from dopamine to coordinate and isolate single iron atoms. The

Increasing the antioxidant capacity of ceria nanoparticles with catechol-grafted poly(ethylene glycol).

Ceria nanoparticles are remarkable antioxidants due to their large cerium(III) content and the possibility of recovering cerium(III) from cerium(IV) after reaction. Here we increase the cerium(III) content of colloidally stable nanoparticles (e.g., nanocrystals) using a reactive polymeric surface coating. Catechol-grafted poly(ethylene glycols) (PEG) polymers of varying lengths and architectures yield materials that are non-aggregating in a variety of aqueous media. Cerium(IV) on the ceria surface both binds and oxidizes the catechol functionality, generating a dark-red colour emblematic of su

In vitro identification and characterisation of iron chelating catechol-containing natural products and derivatives.

Iron is an essential component for multiple biological processes. Its regulation within the body is thus tightly controlled. Dysregulation of iron levels within the body can result in several disorders associated with either excess iron accumulation, including haemochromatosis and thalassaemia, or iron deficiency. In cases of excess body iron, therapy involves depleting body iron levels either by venesection, typically for haemochromatosis, or using iron chelators for thalassemia. However, the current chelation options for people with iron overload are limited, with only three iron chelators a
