A novel biosensor MDC@N-MMCNs to selective detection and elimination of foodborne bacterial pathogens.

BACKGROUND: Infections caused by foodborne pathogens pose a major threat to human health. Traditional bacterial detection methods, such as plate culture and polymerase chain reaction, cannot meet the growing demand for fast and accurate detection. In contrast, colorimetric sensors have the characteristics of convenience, speed, and visualization, but their specific sensitivity is relatively poor. Therefore, it is necessary to develop a biosensor with selective identification of foodborne pathogens, high sensitivity, and early detection of foodborne pathogen contamination in food. RESULTS: We

Common carp (Cyprinus carpio) blood cells are modulated by a parasitic antimicrobial peptide, A-2S and Edwardsiella spp. strains.

Aquaculture industry faces significant challenges due to edwardsiellosis, a bacterial disease caused by Edwardsiella species, particularly E. tarda, which leads to high mortality in farmed fish and consequent financial losses. This pathogen, opportunistic in nature, becomes virulent under unsuitable conditions for fish, causing severe tissues damage in the host. Antibiotic treatments are increasingly ineffective due to Edwardsiella antibiotic resistance, necessitating alternative solutions. This for, we tested the antimicrobial efficiency of the cecropin anisaxin-2S (A-2S), an antimicrobial pe

Antibiofilm property and multiple action of peptide PEW300 against Pseudomonas aeruginosa.

Pseudomonas aeruginosa (P. aeruginosa), an opportunistic pathogen, is often associated with difficulties in treating hospital-acquired infections. Biofilms formed by P. aeruginosa significantly improve its resistance to antimicrobial agents, thereby, posing a great challenge to the combat of P. aeruginosa infection. Antimicrobial peptides (AMPs) have recently emerged as promising antibiofilm agents and increasingly attracting the attention of scientists worldwide. However, current knowledge of their antibiofilm behavior is limited and their underlying mechanism remains unclear. In this study,

Characterisation of the cellular and proteomic response of Galleria mellonella larvae to the development of invasive aspergillosis.

BACKGROUND: Galleria mellonella larvae were infected with conidia of Aspergillus fumigatus and the cellular and humoral immune responses of larvae to the pathogen were characterized as invasive aspergillosis developed. RESULTS: At 2 h post-infection there was an increase in hemocyte density to 7.43 ± 0.50 × 106/ml from 0.98 ± 0.08 × 106/ml at 0 h. Hemocytes from larvae immune primed for 6 h with heat killed A. fumigatus conidia displayed superior anti-fungal activity. Examination of the spread of the fungus by Cryo-imaging and fluorescent microscopy revealed dissemination of the fungus throug

Changes in the Anopheles arabiensis transcriptome and gut microbiota profiles associated with the endosymbiotic Microsporidia MB.

BACKGROUND: Microsporidia MB is an endosymbiotic microbe found in Anopheles mosquito populations. This symbiont can block Plasmodium transmission in mosquitoes, and it can spread through mosquito populations and be sustained over generations by vertical and horizontal transmission. These characteristics make Microsporidia MB a potential candidate for symbiont-based malaria vector control. However, the mechanistic basis of interactions between Microsporidia MB and Anopheles arabiensis is poorly characterized. We investigated how the presence of Microsporidia MB affects transcriptomics profiles

Unveiling the hidden threat: flupyradifurone soluble liquid's toxic impact on silkworms.

Bombyx mori Linnaeus (Lepidoptera: Bombycidae) is an economically significant insect that consistently contributes to the enhancement of the national economy in all countries with predominant sericulture industries. Flupyradifurone, a butenolide insecticide, is recognized for its minimal toxic effects on bees when applied at standard dosages; however, its potential toxicity or threat to B. mori has not been fully elucidated. This study aims to investigate and evaluate the toxicity of flupyradifurone soluble liquid (SL) on B. mori by administering various concentrations to third instar silkworm

Assessing the antiviral activity of antimicrobial peptides Caerin1.1 against PRRSV in Vitro and in Vivo.

The Porcine reproductive and respiratory syndrome (PRRS) causes severe financial losses to the global swine industry. Due to continuous virus evolution, the protection against the PRRS provided by current vaccines is limited. In order to find new antiviral strategies, this study investigated the antiviral potential of antimicrobial peptides (AMPs) against PRRSV. Given the diversity of PRRSV strains and the limited effectiveness of existing vaccines in controlling PRRSV, this study evaluated the inhibitory effects of KLAK, Cecropin B, Piscidin1, and Caerin1.1 on 3 strains of PRRSV (lineage 5 cl

Candida albicans increases the pathogenicity of Staphylococcus aureus during polymicrobial infection of Galleria mellonella larvae.

This study detailed the responses of Galleria mellonella larvae to disseminated infection caused by co-infection with Candida albicans and Staphylococcus aureus. Doses of C. albicans (1×105 larva-1) and S. aureus (1×104 larva-1) were non-lethal in mono-infection but when combined significantly (P<0.05) reduced larval survival at 24, 48 and 72 h relative to larvae receiving S. aureus (2×104 larva-1) alone. Co-infected larvae displayed a significantly higher density of S. aureus larva-1 compared to larvae infected solely with S. aureus. Co-infection resulted in dissemination throughout the host

Investigating specific bacterial resistance to AMPs by using a magainin I-resistant Escherichia coli model.

Antimicrobial peptides (AMPs) are multifunctional compounds that may show antimicrobial and immunomodulatory activities. With the rapid increase in the incidence of multidrug-resistant bacteria, there is an enormous interest in AMPs as templates for the production of new antibiotics. However, there are concerns that the therapeutic administration of AMPs can select resistant strains. In order to distinguish between resistant and non-resistant strains and verify resistance specificity to AMPs, in this study a magainin I-resistant Escherichia coli model was used. First, the identity of all strai

LPS modification promotes maintenance of Yersinia pestis in fleas.

Yersinia pestis, the causative agent of plague, can be transmitted by fleas by two different mechanisms: by early-phase transmission (EPT), which occurs shortly after flea infection, or by blocked fleas following long-term infection. Efficient flea-borne transmission is predicated upon the ability of Y. pestis to be maintained within the flea. Signature-tagged mutagenesis (STM) was used to identify genes required for Y. pestis maintenance in a genuine plague vector, Xenopsylla cheopis. The STM screen identified seven mutants that displayed markedly reduced fitness in fleas after 4 days, the ti

Interaction of cationic peptides with lipoteichoic acid and gram-positive bacteria.

Compounds with antiendotoxin properties have been extensively studied for their potential as therapeutic agents for sepsis attributable to gram-negative bacteria. However, with the increasing incidence of gram-positive sepsis, there is interest in identifying compounds with a broad spectrum of action against both gram-positive and gram-negative bacteria. A series of synthetic alpha-helical cationic peptides related to bee melittin and silk moth cecropin have previously been shown to bind lipopolysaccharide (LPS) with high affinity, inhibit LPS-induced tumor necrosis factor alpha (TNF-alpha) pr

Distinct roles of the Anopheles gambiae Imd/Rel2 and Toll/Rel1 pathways in resistance to systemic bacterial and fungal infections.

In the malaria vector Anopheles gambiae s.l. Toll and Imd immune pathways signal through Re1l and Rel2 NF-κB transcription factors, respectively. Toll/Rel1 and Imd/Rel2 have been characterized mostly in the context of defense against malaria parasites, exhibiting Plasmodium species-specific roles. There is also evidence associating Rel2 with mosquito resistance to bacterial infections, however, the role of Rel1 in this context remains questionable, especially that there have been no rigorous functional genetic studies assessing Rel1 and Rel2 contributions side-by-side to mosquito defense durin

Aedesin: structure and antimicrobial activity against multidrug resistant bacterial strains.

Multidrug resistance, which is acquired by both Gram-positive and Gram-negative bacteria, causes infections that are associated with significant morbidity and mortality in many clinical settings around the world. Because of the rapidly increasing incidence of pathogens that have become resistant to all or nearly all available antibiotics, there is a need for a new generation of antimicrobials with a broad therapeutic range for specific applications against infections. Aedesin is a cecropin-like anti-microbial peptide that was recently isolated from dengue virus-infected salivary glands of the

Preparation, characterization, and comparison of the release of antibacterial peptide CM11 from PLGA nanoparticles and microparticles.

OBJECTIVES: Pseudomonas aeruginosa (P. aeruginosa) is a Gram-negative bacterium capable of causing acute and chronic infections. Treating these infections is increasingly challenging as this bacterium develops antibiotic resistance. Antimicrobial peptides (AMPs) have emerged as promising therapeutic candidates due to their broad-spectrum activity. Cecropin-Melittin-11 (CM11), a cationic AMP, has shown antimicrobial activity against various pathogens. However, the progression of AMPs from preclinical research to clinical application is hindered by enzymatic degradation, short systemic half-life

Comprehensive analysis of pathogenic mechanisms of Enterobacter cancerogenus ECL9 in silkworms.

Bacterial diseases of silkworms (Bombyx mori L.) cause significant reductions in sericulture and result in huge economic loss. Enterobacter cancerogenus is usually known as an opportunistic host pathogen. The interactions between pathogenic bacteria and silkworms involve complex processes. This study aimed to systematically investigate the genetic traits of E. cancerogenus ECL9 and its effects on the intestinal microbiota, as well as the physiological and immune responses of silkworm intestines to infection. Bacterial genome sequencing indicated that ECL9 possessed multiple antibiotic-resistan

An entomopathogenic fungus exploits its host humoral antibacterial immunity to minimize bacterial competition in the hemolymph.

BACKGROUND: The insect hemolymph (blood-equivalent fluid), composed of a large number of hemocytes (blood cells) and a variety of soluble immune effectors, is hostile for pathogens including fungi. In order to survive in the insect hemocoel (body cavity), the entomopathogenic fungus (EPF) has evolved two classical coping strategies, namely evasion and suppression of the host immune reactions. However, it remains unclear whether EPF has other ways of coping with host immunity. RESULTS: In this study, we demonstrated that Metarhizium rileyi (an EPF) infection by injection of blastospores into t

In vitro and in vivo antimicrobial activity of antimicrobial peptide Jelleine-I against foodborne pathogen Listeria monocytogenes.

As a human foodborne pathogen, Listeria monocytogenes can cause severe human listeriosis and develop resistance to antibiotics. Antimicrobial peptides (AMPs) are produced from all kingdoms of life and regarded as promising alternatives to conventional antibiotics. Jelleine-I is an AMP identified from honeybees royal jelly. In this study, we explored the activity and action mechanism of Jelleine-I against L. monocytogenes. We found its minimum inhibitory concentration to be 12.5 μg/mL. Membrane permeability analysis revealed that Jelleine-I increased L. monocytogenes cell membrane permeability,

Antimicrobial peptide CAMA-syn expressed in pulmonary epithelium by recombination adenovirus inhibited the growth of intracellular bacteria.

BACKGROUND: Intracellular bacteria, especially Mycobacterium tuberculosis, are important pathogenic microorganisms that endanger human health. Purified and synthesized cecropin A-magainin 2 (CAMA-syn) can exhibit a higher antibacterial activity and lower cytotoxicity. To enhance such antimicrobial potential, it would be desirable to deliver CAMA-syn expressed in lung epithelial cells by an adenovirus vector using gene therapy. METHODS: A549 cells in vitro and lung epithelial cells in vivo were used to express CAMA-syn by transducing recombinant adenovirus Ad-SPC-CAMA/GFP, and the expression o

The Intestinal Roundworm Ascaris suum Releases Antimicrobial Factors Which Interfere With Bacterial Growth and Biofilm Formation.

Ascariasis is a widespread soil-transmitted helminth infection caused by the intestinal roundworm Ascaris lumbricoides in humans, and the closely related Ascaris suum in pigs. Progress has been made in understanding interactions between helminths and host immune cells, but less is known concerning the interactions of parasitic nematodes and the host microbiota. As the host microbiota represents the direct environment for intestinal helminths and thus a considerable challenge, we studied nematode products, including excretory-secretory products (ESP) and body fluid (BF), of A. suum to determine
