Chronic intermittent hypoxia (CIH), a hallmark feature of obstructive sleep apnea (OSA), is strongly implicated in the development of hepatic injury. However, the cellular mechanisms underlying CIH-induced liver dysfunction remain poorly understood. This study aimed to systematically characterize hepatic cellular responses to CIH at single-nucleus resolution. A rodent model of CIH was established and subjected to histological and transcriptomic analyses. Single-nucleus RNA sequencing (snRNA-seq) was performed to delineate transcriptional alterations across liver cell populations. Differentiall

The cochlear nucleus (CN) is the first central relay of auditory information, receiving input from spiral ganglion neurons (SGNs) through specialized synapses whose molecular regulation remains incompletely defined. Here, we examined the role of Progranulin (Grn) in a stem cell-based CN synaptogenesis using an in vitro coculture model. DsRed-expressing mouse embryonic stem cells were differentiated into SGN-like neurons (deSGNs) and cocultured for 4 days with primary CN neurons isolated from postnatal days 3-6 wild-type (Grn+/+) or Grn-deficient (Grn-/-) mice. Cocultures containing Grn+/+ mous

Nucleus pulposus (NP) cell senescence is a critical factor in the progression of intervertebral disc degeneration (IVDD). Our analysis demonstrates that FTO and YAP1 expression levels are significantly diminished in degenerative NP tissues from both human and rat models, which correlates with increased m6A modification of YAP1 transcripts. To investigate the underlying mechanisms, we utilized IL-1β to induce senescence in cultured NP cells. Our findings reveal that FTO knockdown leads to a decrease in YAP1 levels while simultaneously increasing senescence markers. In contrast, the overexpressi

In addition to its motor functions, the cerebellum is involved in emotional regulation, anxiety and affect. We found that suppressing the firing of cerebellar Purkinje cells (PCs) rapidly excites forebrain areas that contribute to such functions (including the amygdala, basal forebrain and septum), but that the classic cerebellar outputs, the deep cerebellar nuclei, do not directly project there. We show that PCs directly inhibit parabrachial nuclei (PBN) neurons that project to numerous forebrain regions. Suppressing the PC-PBN pathway influences many regions in the forebrain and is aversive.

OBJECTIVE: The nucleus ambiguus (NAmb) is a column of neurons in the medulla oblongata, involved in bulbar functions. Expression of Glial Cell-Derived Neurotrophic Factor (GDNF) and its receptors (GDNFR) is observed within the cell bodies during reinnervation following recurrent laryngeal nerve (RLN) injury. Little is known regarding GDNFR expression in the formation of the NAmb and the laryngeal innervation during embryogenesis. Understanding the timing and pattern of GDNFR expression in embryogenesis versus after RLN injury may provide insights into therapeutic targets for regeneration after

Parkinson's disease (PD) is a neurodegenerative disease with an impairment of movement execution that is related to age and genetic and environmental factors. 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is a neurotoxin widely used to induce PD models, but the effect of MPTP on the cells and genes of PD has not been fully elucidated. By single-nucleus RNA sequencing, we uncovered the PD-specific cells and revealed the changes in their cellular states, including astrocytosis and endothelial cells' absence, as well as a cluster of medium spiny neuron cells unique to PD. Furthermore, traje

BACKGROUND: The intervertebral disc is the largest avascular tissue in the human body. The nucleus pulposus (NP) consumes glucose and oxygen to generate energy to maintain cellular metabolism via nutrients that diffuse from the cartilage endplate. The microenvironment in the intervertebral disc becomes nutritionally deficient during degeneration, and nutritional deficiency has been shown to inhibit the viability and proliferation of NP cells.
METHODS: To investigate the molecular mechanism by which nutritional deficiency reduces viability and decreases proliferation, we created an in vitro mo

Cross-modal auditory influence on cell activity in the primary visual cortex emerging at short latencies raises the possibility that the first-order visual thalamic nucleus, which is considered dedicated to unimodal visual processing, could contribute to cross-modal sensory processing, as has been indicated in the auditory and somatosensory systems. To test this hypothesis, the effects of sound stimulation on visual cell activity in the dorsal lateral geniculate nucleus were examined in anesthetized rats, using juxta-cellular recording and labeling techniques. Visual responses evoked by light

BACKGROUND: The nucleus accumbens (NAc) is a brain region implicated in pathological motivated behaviors such as drug addiction and is composed predominantly of two discrete populations of neurons, dopamine receptor-1- and dopamine receptor-2-expressing medium spiny neurons (D1-MSNs and D2-MSNs, respectively). It is unclear whether these populations receive inputs from different brain areas and whether input regions to these cell types undergo distinct structural adaptations in response to the administration of addictive drugs such as cocaine.

While most adults are able to clear acute hepatitis B virus (HBV) infection, chronic HBV infection is recalcitrant to current therapy because of the persistence of covalently closed circular DNA in the nucleus. Complete clearance of the virus in these patients is rare, and long-term therapy with interferon and/or nucleoside analogues may be required in an attempt to suppress viral replication and prevent progressive liver damage. The difficulty of establishing HBV infection in cell culture and experimental organisms has hindered efforts to elucidate details of the HBV life cycle, but it has al

The cDNA corresponding to a fourth species of diacylglycerol (DG) kinase (EC 2.7.1.107) was isolated from cDNA libraries of rat retina and brain. This cDNA encoded a 929-aa, 104-kDa polypeptide termed DGK-IV. DGK-IV was different from previously identified mammalian DG kinase species, DGK-I, DGK-II, and DGK-III, in that it contained no EF-hand motifs but did contain four ankyrin-like repeats at the carboxyl terminus. These structural features of DGK-IV closely resemble the recently cloned, eye-specific DG kinase of Drosophila that is encoded by the retinal degeneration A (rdgA) gene. However,

Following transection of the vagal nerve on the neck, a dramatic loss of neurons occurs in the dorsal motor vagal nucleus (DMV) of the adult rabbit. The present study describes the light and electron microscopical characteristics of the accompanying glial cell response. The number of microglial cells was considerably increased a few days after nerve injury and remained at a higher than normal level throughout almost the whole period examined. Astrocytes increased to some extent at a later post-operative stage. Ultrastructurally, microglial cells hypertrophied, surrounded degenerating neurons,

Lgr5+ stem cells play crucial roles in maintaining intestinal epithelial cell homeostasis. However, the cellular heterogeneity and underlying regulatory programs of Lgr5+ small intestinal stem cells (ISCs) remain elusive. In this study, we profiled gene expression and chromatin accessibility of Lgr5+ ISCs at single-cell resolution to gain a deeper understanding of the lineage specification and early fate determining mechanisms. Our analysis identified a total of 6 subsets of Lgr5+ cell populations, which exhibited heterogeneity in gene expression and chromatin structure. We found that early fa

The objective of this study was to determine whether single-cell RNA sequencing (scRNA-seq) or single-nucleus RNA sequencing (snRNA-seq) was more effective for studying the goat pancreas. Pancreas tissues from three healthy 10-day-old female Xiangdong black goats were processed into single-cell and single-nucleus suspensions. These suspensions were then used to compare cellular composition and gene expression levels following library construction and sequencing. Both scRNA-seq and snRNA-seq were eligible for primary analysis but produced different cell identification profiles in pancreatic tis

BACKGROUND: The nucleus pulposus (NP) plays a central role in the pathogenesis of intervertebral disc degeneration (IVDD); however, its internal cellular heterogeneity and molecular mechanisms have not yet been elucidated.
METHODS: ScRNA-seq was used to evaluate the structure of the NP at different degenerative stages in the same individual with IVDD. Unsupervised clustering of cells based on gene expression profiles was performed using the Seurat package and passed to uniform manifold approximation and projection (UMAP) for cluster visualization. A rat disc degeneration model and an in vitro

Cell proliferation and matrix synthesis were compared for rat nucleus pulposus cells cocultured with mesenchymal stem cells (MSCs) or fresh whole bone marrow cells (BMCs), harvested by the perfusion or aspiration methods. Nucleus pulposus cells were isolated from tail intervertebral discs of F344/slc rats, and BMCs were obtained from femora. Proteoglycan synthesis, DNA synthesis, and aggrecan mRNA expression were measured. The level of transforming growth factor-beta in supernatants from the culture system was also measured. Cell number, aggrecan mRNA expression, and uptake of [(35)S]-sulfate

Administration of somatostatin induced changes of functional activity in all the central and peripheral structures under study. Neuronal activity in the paraventricular nucleus decreased by the 3rd and 15th min after somatostatin injection whereas by the 20th min the firing rate increased. Amount of argenaffin cells was sharply reduced, at that, between the 4th and 15th min which suggested an active release of enterochromaffin cell granules into the blood.

Administration of enkephalin into the 3rd ventricle enhanced the excitability of the paraventricular nucleus neurosecretory cells, the excitability reaching its maximum within 10 min. By the 15th min after the enkephalin administration the number of the enterochromaffin cells increases suggesting the inhibition of the secretory granules contents release into the blood. Within next 15 min the excitability of the hypothalamic neurosecretory neurons returns to its normal level. A simultaneous sharp decrease of the enterochromaffin cells occurs suggesting an endogenous activation of the secretory
