Human endometrial stromal cell plasticity: Reversible sFlt1 expression negatively coincides with decidualization.

Preeclampsia (PE) is a major complication of pregnancy in which the placenta is known to have shallow implantation into the uterine decidua. Studies have implicated soluble fms-like tyrosine kinase-1 (sFlt1), a soluble vascular endothelial growth factor (VEGF) receptor protein, in the pathogenesis of PE. sFlt1 has the ability to bind to and neutralize the angiogenic functions of VEGF and placental growth factor (PlGF). The presence of sFlt1 and its action in the endometrium is yet to be determined. We hypothesize that endometrial stromal cells (ESC) at the maternal-fetal interface may play a r

Cell adhesion molecules in neural plasticity and pathology: similar mechanisms, distinct organizations?

Brain plasticity and the mechanisms controlling plasticity are central to learning and memory as well as the recovery of function after brain injury. While it is clear that neurotrophic factors are one of the molecular classes that continue to regulate brain plasticity in the adult central nervous system (CNS), less appreciated but equally profound is the role of cell adhesion molecules (CAMs) in plasticity mechanisms such as long term potentiation, preservation of neurons and regeneration. Ironically, however, CAMs can also reorganize the extra-cellular space and cause disturbances that drive

Cerebellar plasticity: modification of Purkinje cell structure by differential rearing in monkeys.

Dendritic branching in Purkinje and granule cells and the diameters of Purkinje cell somas were compared in several cerebellar areas of monkeys reared in isolation, with social experience, or in a large colony. In the colony-reared monkeys, spiny branchlets of Purkinje cells were more extensive in the paraflocculus and the nodulus than they were in the other two groups. Granule cell dendritic branching in the paraflocculus and nodulus did not differ across groups. In addition, Purkinje cell somas were larger in the uvula and the nodulus of the colony animals than in the other groups. These dat

Single-cell analysis identifies BASP1 as a driver of drug resistance and cell plasticity in oral squamous cell carcinoma.

Despite initial positive responses with chemotherapy, many cancer patients experience relapse, continued tumor growth, and metastatic spread due to drug resistance. It is well documented that a rare population of phenotypically heterogeneous cells contributes to intratumour heterogeneity and drug resistance. To date, these rare populations are poorly characterized. To identify the potential role of these rare populations in drug resistance, here we have performed single-cell RNA sequencing of human oral squamous cell carcinomas lines presenting with sensitive, early, and late cisplatin-resista

Intestinal Stem Cells: Regulation, Plasticity, and Therapeutic Potential.

Intestinal stem cells (ISCs) are central to the maintenance and regeneration of the gut epithelium, supporting its rapid turnover and adaptive capacity. Recent advances, including the identification of LGR5+ stem cells, 3D in vitro organoid technology, and single-cell transcriptomics, have uncovered the remarkable heterogeneity and plasticity of ISCs and their niche. The ISC niche comprises dynamic interactions among epithelial, stromal, immune, and microbial components, enabling responses to injury and disease. Notably, both active and reserve stem cell pools, as well as progenitor and differ

Plasticity, signaling, and metabolic rewiring in melanoma persister cells.

Despite advances in therapy, survival rates for metastatic melanoma remain low. Drug-tolerant persister cells (persisters) can facilitate cancer recurrence, yet their characteristics and vulnerabilities differ across cancers and even within melanoma, depending on mutational context and treatment strategy. Both preexisting phenotypic traits and drug-induced adaptations contribute to persister formation, linked by "primed" persisters that exhibit intermediate states observed in multiple studies. Compared with parental melanoma cells and other phenotypic variants such as melanoma stem cells or se

Experience-Dependent Plasticity of Periglomerular Cells in the Olfactory Bulb.

The olfactory bulb (OB) serves as the first relay station that processes odor information. Within the OB, odor representations are dynamically modulated by reciprocal synapses between excitatory mitral and tufted (M/T) cells and local inhibitory interneurons. Among these interneurons, periglomerular cells (PGCs) are GABAergic neurons located in the glomerular layer that provide inhibitory inputs to M/T cells. Although their anatomical and molecular properties have been extensively characterized, the in vivo dynamics of PGCs during odor processing and learning remain poorly understood. To addre

Regulatory T cells in the skin: Origin, function, and plasticity.

Skin is continually exposed to external stimuli, making this frontline barrier highly susceptible to inflammatory disorders. Foxp3+ regulatory T cells (Tregs) are considered guardians of skin homeostasis, with functions that extend beyond immune regulation to include neuroimmunological modulation, hair cycling, and wound healing. In the resting state, skin Tregs originate from the thymus; however, the composition of the Treg population under pathological conditions remains elusive. Notably, emerging studies show that Tregs can acquire proinflammatory characteristics during the development of a

The Power of Plasticity-Metabolic Regulation of Hepatic Stellate Cells.

Hepatic stellate cells (HSCs) are resident non-parenchymal liver pericytes whose plasticity enables them to regulate a remarkable range of physiologic and pathologic responses. To support their functions in health and disease, HSCs engage pathways regulating carbohydrate, mitochondrial, lipid, and retinoid homeostasis. In chronic liver injury, HSCs drive hepatic fibrosis and are implicated in inflammation and cancer. To do so, the cells activate, or transdifferentiate, from a quiescent state into proliferative, motile myofibroblasts that secrete extracellular matrix, which demands rapid adapta

Diversity and plasticity of microglial cells in psychiatric and neurological disorders.

Recent advanced immunological analyses have revealed that the diversity and plasticity of macrophages lead to the identification of functional polarization states (classically activated M1 type and alternatively activated M2 type) which are dependent on the extracellular environment. M1 and M2 polarization states of macrophages play an important role in controlling the balance between pro-inflammatory and anti-inflammatory conditions. Microglial cells are resident mononuclear phagocytes in the central nervous system (CNS), express several macrophage-associated markers, and appear to display fu

Progenitor cells confer plasticity to cardiac valve endothelium.

The endothelium covering the aortic, pulmonary, mitral, and tricuspid valves looks much like the endothelium throughout the vasculature, in terms of general morphology and expression of many endothelial markers. Closer examination, however, reveals important differences and hints of a unique phenotype that reflects the valvular endothelium's embryonic history, and potentially, its ability to maintain integrity and function over a life span of dynamic mechanical stress. A well-studied property that sets the cardiac valvular endothelium apart is the ability to transition from an endothelial to a

Functional adaptation: the key to plasticity of cardiovascular "stem" cells?

There is increasing evidence that cells of disparate phenotypes displaying various degrees of proliferative capacity engraft and function heterotopically in adult organisms. Efforts were made to reconcile these findings with the embryologic notions of pluripotent stem or progenitor cell, although the nature of the 'stemness' remained elusive. This topic is particularly important for the cardiovascular system, in which cytotrophoblasts, certain tumor cells, monocytes/macrophages, peritoneal mesothelial cells, and others acquire endothelial properties and/or perform endothelial functions. Here w

Epigenetic and metabolic plasticity of tumor cells in immunotherapy resistance.

Cancer immunotherapy has demonstrated remarkable efficacy across multiple tumor types. Despite their significant clinical benefits, most patients with solid tumors remain refractory to immunotherapy, and initial responders frequently develop acquired resistance. Tumor plasticity is a key driver of resistance to cancer immunotherapy. In this review, we discuss how epigenetic and metabolic plasticity of tumor contributes to immunotherapy resistance and highlight emerging strategies targeting these programs to overcome resistance and improve clinical outcomes.

[Adipose tissue-derived stem cells: hepatic plasticity].

Currently, the only effective treatment for end-stage liver disease is liver transplantation. The number of patients on the waiting list increases considerably each year, giving rise to a wide imbalance between supply and demand for healthy livers. Knowledge of stem cells and their possible use have awakened great interest in the field of hepatology, these cells being one of the most promising short-term alternatives. Hepatic stem cell therapy consists of the implantation of healthy cells capable of performing the functions that damaged cells are unable to carry out. Recent observations indica

Quercetin in oral squamous cell carcinoma: Integrating immunometabolic reprogramming, cancer stem cell plasticity, and precision therapeutic strategies.

Oral squamous cell carcinoma (OSCC) remains a major clinical challenge because of its recurrence, metastasis, and therapeutic resistance. Quercetin, a dietary flavonoid, has shown broad anticancer activity in experimental models of OSCC through the modulation of several processes, including apoptosis, autophagy, oxidative stress, epithelial-mesenchymal transition (EMT), cancer stem cell (CSC) phenotypes, metabolism, inflammation, and oncogenic signalling. However, the available evidence remains largely preclinical, and no clinical trial has demonstrated an improvement in progression-free survi

Single-cell multiomics reveals increased plasticity, resistant populations, and stem-cell-like blasts in KMT2A-rearranged leukemia.

KMT2A-rearranged (KMT2A-r) infant acute lymphoblastic leukemia (ALL) is a devastating malignancy with a dismal outcome, and younger age at diagnosis is associated with increased risk of relapse. To discover age-specific differences and critical drivers that mediate poor outcome in KMT2A-r ALL, we subjected KMT2A-r leukemias and normal hematopoietic cells from patients of different ages to single-cell multiomics analyses. We uncovered the following critical new insights: leukemia cells from patients <6 months have significantly increased lineage plasticity. Steroid response pathways are downreg

SEZ6 expression and lineage plasticity in small cell lung cancer and transformed non-small cell lung cancer.

INTRODUCTION: Small cell lung cancer (SCLC) and extrapulmonary neuroendocrine (NE) tumors are aggressive malignancies with limited treatment options. Seizure-related homolog 6 (SEZ6) is a potential therapeutic target, but its expression in these tumors remains poorly understood. Lineage plasticity contributes to resistance in non-small cell lung cancer (NSCLC), where some cases can undergo SCLC-transformation after targeted therapy. We aimed to characterize SEZ6 expression across NE tumors and presumed NSCLC-to-SCLC transformations. METHODS: DNA and RNA sequencing were performed for SCLC, NSC

Single-Cell Evolutionary Analysis Reveals Drivers of Plasticity and Mediators of Chemoresistance in Small Cell Lung Cancer.

UNLABELLED: Small cell lung cancer (SCLC) is often a heterogeneous tumor, where dynamic regulation of key transcription factors can drive multiple populations of phenotypically different cells which contribute differentially to tumor dynamics. This tumor is characterized by a very low 2-year survival rate, high rates of metastasis, and rapid acquisition of chemoresistance. The heterogeneous nature of this tumor makes it difficult to study and to treat, as it is not clear how or when this heterogeneity arises. Here we describe temporal, single-cell analysis of SCLC to investigate tumor initiati

Exosome-Mediated Bidirectional Signaling Regulates Glioma Stem-Cell Homeostasis and Radiation-Induced Plasticity.

BACKGROUND: Glioblastoma (GBM) is an aggressive brain malignancy characterized by therapeutic resistance and frequent recurrence. Glioma-initiating cells (GICs), also known as glioma stem cells (GSCs), contribute to these features through self-renewal and resistance to genotoxic stress. Radiation therapy can paradoxically replenish the GIC compartment by inducing stem-like properties in non-stem glioma cells. We investigated whether exosomes mediate bidirectional communication that maintains the balance between GIC and non-stem populations at steady state and following irradiation. METHODS: E

Hedgehog signaling drives glial cell plasticity and oncogenic reprogramming in gastroenteropancreatic neuroendocrine neoplasms.

UNLABELLED: Disruption of the Men1 locus in epithelial and endocrine tissues fails to generate the full spectrum of gastroenteropancreatic neuroendocrine tumors (GEP-NETs), raising the possibility of a potential stromal source for these cancers. Neural crest-derived glial cells were previously implicated in neuroendocrine tumors arising in the pituitary and pancreas, yet these studies lacked a clear mechanism for these events. Here, we investigated the hypothesis that Men1 -driven Hedgehog (HH) signaling redirects the glial cell fate to give rise to neuroendocrine tumors in the gastrointestina
