Role of neutrophils in the immune microenvironment of endometriosis.

Endometriosis (EMs) is a common systemic estrogen-dependent chronic inflammatory disease in gynaecology. Available evidence indicates that immune factors exert extremely important functions in the pathogenesis of EMs. As a key cell group in the immune environment, neutrophils have been found to be implicated in EMs progression in various studies. In this paper, the function and mechanism of neutrophils in the immune microenvironment of EMs are summarized, providing new research ideas for the etiology and pathogenesis of EMs.

Reprogramming the wound microenvironment: identity remodeling strategies for fibroblasts, keratinocytes, and macrophages.

The core pathology of chronic non-healing wounds is the dysfunction of wound-repair cells-a molecular defect that conventional passive therapies can hardly correct at its root. Cell reprogramming techniques, by actively rewriting cell identity, have therefore brought a paradigm shift to wound repair. Herein, we propose a systematic "four-dimensional technology toolbox" for wound cell reprogramming, comprising transcription factor-mediated, small-molecule-induced, epigenetic and metabolic regulation, and nanomaterial-assisted delivery. Using this toolbox as the main thread, we comprehensively i

Exploring the links between cancer and placenta development.

The development of metastatic cancer is a multistage process, which often requires decades to complete. Impairments in DNA damage control and DNA repair in cancer cell precursors generate genetically heterogeneous cell populations. However, despite heterogeneity most solid cancers have stereotypical behaviours, including invasiveness and suppression of immune responses that can be unleashed with immunotherapy targeting lymphocyte checkpoints. The mechanisms leading to the acquisition of stereotypical properties remain poorly understood. Reactivation of embryonic development processes in cells

The histophysiology and pathophysiology of the peritoneum.

The peritoneum is an extensive serous organ with both epithelial and mesenchymal features and a variety of functions. Diseases such as inflammatory peritonitis and peritoneal carcinomatosis can induce disturbance of the complex physiological functions. To understand the peritoneal response in disease, normal embryonic development, anatomy in healthy conditions and physiology of the peritoneum have to be understood. This review aims to summarize and discuss the literature on these basic peritoneal characteristics. The peritoneum is a dynamic organ capable of adapting its structure and functions

Mapping subcellular microenvironments using oligonucleotide-directed proximity labeling.

Understanding how cells compartmentalize their biomolecules into discrete structures remains one of the fundamental goals of cell biology. The proliferation of proximity labeling (PL) technologies has been invaluable toward this goal, enabling biochemical "dissection" of compartments that would be intractable to classical biochemical methods. While robust PL approaches have long been established for targeting proteins of interest, targeting nucleic acids-RNAs and genomic loci-has remained significantly more challenging. Here, we review recent advancements in the field that overcome this longst

The role of the renal immune microenvironment in multisystem diseases.

BACKGROUND: Dysregulation of the renal immune microenvironment contributes to kidney pathology and also exerts widespread effects on multiple systems, including the intestinal, respiratory, cardiovascular, and nervous systems, as well as tumors. Therefore, a deeper understanding of how the renal immune microenvironment influences multisystem disease progression is of great significance. METHODS: This review is based on relevant literature retrieved from databases including PubMed and Web of Science. It systematically summarizes and integrates research advances on the role of the renal immune

The intelligent podocyte: sensing and responding to a complex microenvironment.

Podocytes are key components of the glomerular filtration barrier - a specialized structure that is responsible for the filtration of blood by the kidneys. They therefore exist in a unique microenvironment exposed to mechanical force and the myriad molecules that cross the filtration barrier. To survive and thrive, podocytes must continually sense and respond to their ever-changing microenvironment. Sensing is achieved by interactions with the surrounding extracellular matrix and neighbouring cells, through a variety of pathways, to sense changes in environmental factors such as nutrient level

Can't handle the stress? Mechanobiology and disease.

Mechanobiology is a rapidly growing research area focused on how mechanical forces and properties influence biological systems at the cell, molecular, and tissue level, and how those biological systems, in turn, control mechanical parameters. Recently, it has become apparent that disrupted mechanobiology has a significant role in many diseases, from cardiovascular disease to muscular dystrophy and cancer. An improved understanding of this intricate process could be harnessed toward developing alternative and more targeted treatment strategies, and to advance the fields of regenerative and pers

Macrophage Plasticity: Phenotypic and Functional Profiles Across Pathological Microenvironments.

Macrophages are highly plastic innate immune cells that adopt context-dependent phenotypes along a continuum, integrating developmental origin with local microenvironmental cues rather than conforming to discrete M1/M2 states. This review delineates the molecular circuits shaping macrophage identity-TLR/cytokine signaling, microRNA networks, metabolic rewiring, and epigenetic mechanisms including histone lactylation-and traces how circulating monocyte subsets contribute to tissue macrophage diversity. We examine macrophage plasticity across a broad disease spectrum-oncology, autoimmune and rhe

Zinc-enriched pathological biominerals in the human kidney encode their anatomical microenvironments.

The fundamental mechanisms underlying kidney stone formation remain elusive despite extensive research, limiting prevention strategies and contributing to high recurrence rates. Traditionally understood to arise through systemic urine supersaturation and conceptualized as monolithic concretions, kidney stones exhibit marked structural and compositional heterogeneity across distinct renal microenvironments. We hypothesized that these heterogeneous biominerals preserve physicochemical signatures of their anatomical origin and spatial organization of zinc (Zn) reflects microenvironment-specific m

Nerves in Bone: Evolving Concepts in Pain and Anabolism.

The innervation of bone has been described for centuries, and our understanding of its function has rapidly evolved over the past several decades to encompass roles of subtype-specific neurons in skeletal homeostasis. Current research has been largely focused on the distribution and function of specific neuronal populations within bone, as well as their cellular and molecular relationships with target cells in the bone microenvironment. This review provides a historical perspective of the field of skeletal neurobiology that highlights the diverse yet interconnected nature of nerves and skeleta

"Pumping iron"-how macrophages handle iron at the systemic, microenvironmental, and cellular levels.

Macrophages reside in virtually every organ. First arising during embryogenesis, macrophages replenish themselves in the adult through a combination of self-renewal and influx of bone marrow-derived monocytes. As large phagocytic cells, macrophages participate in innate immunity while contributing to tissue-specific homeostatic functions. Among the key metabolic tasks are senescent red blood cell recycling, free heme detoxification, and provision of iron for de novo hemoglobin synthesis. While this systemic mechanism involves the shuttling of iron between spleen, liver, and bone marrow through

Cytokine Expression in Keratoconus and its Corneal Microenvironment: A Systematic Review.

Keratoconus (KC) is a progressive corneal ecstasia characterized by thinning and weakening of the cornea that leads to a cone-like appearance, scarring, and decreased vision. Despite the well-described clinical signs, the cause of KC is unknown. Nevertheless, various genes, proteinases, and environmental factors (eye-rubbing, contact lens wear, tear film composition) have been implicated in its etiology. Although classically defined as a predominantly degenerative disease, with mechanically induced trauma accelerating its course, accumulating evidence suggests a pivotal role for inflammation i

Distinct single-cell cellular states and ecosystems linked to HPV status distinguish therapeutic vulnerability of head and neck squamous cell carcinoma.

Human papillomavirus (HPV) infection plays a significant role in shaping the tumor microenvironment of head and neck squamous cell carcinoma (HNSCC). However, the heterogeneity of cellular states and ecosystems within the tumor microenvironment of HPV-infected HNSCC is still largely unknown. Using the EcoTyper framework, we perform an extensive evaluation of HPV-related cellular profiles and cellular ecotypes (CEs) in HNSCC. Single-cell RNA sequencing reveals 46 unique states across 12 principal cell types. Spatial transcriptomics confirm the spatial correlation of cellular states from the sam

Inflammatory Signatures in MDS: The Missing Link Between Genetics, Microenvironment, and Therapy.

Myelodysplastic syndromes are clonal hematopoietic neoplasms in which ineffective hematopoiesis arises within the context of chronic inflammation and immune dysregulation. Growing evidence indicates that aging-associated inflammaging and inflammation-driven remodeling of the bone marrow microenvironment are not secondary phenomena, but active forces that shape clonal selection, lineage commitment, and disease evolution. This narrative review integrates recent insights from translational immunology, stem cell biology, multi-omics analyses, and clinical studies to examine the reciprocal interpla

Osteoimmune microenvironmental failure in osteonecrosis of the femoral head: a necrosis-inflammation-repair loop and immune-editing-inspired framework for hip preservation.

Osteonecrosis of the femoral head (ONFH) is traditionally framed as structural failure after interrupted blood supply, yet this linear model does not explain why patients with similar radiographic stage may show markedly different pain trajectories, collapse kinetics, and hip-preservation outcomes. We propose that ONFH is better interpreted as an osteoimmune microenvironmental failure sustained by a self-reinforcing loop: ongoing cell-death input, maladaptive inflammatory reprogramming, bone-marrow niche dysfunction, and mechanical destabilization. In this framework, early sterile inflammation

Exosomes: a promising microenvironment modulator for spinal cord injury treatment.

Spinal cord injury (SCI) remains a severely disabling disorder that impacts millions globally by causing irreversible damage to the nervous system. Although cell - based therapies have shown notable progress, the post - injury microenvironment presents significant obstacles that hinder the survival and effectiveness of implanted cells, ultimately limiting sustained functional restoration. Exosomes have emerged as a promising cell - free therapeutic alternative due to their stability, low immunogenicity, and ability to carry bioactive molecules such as proteins, microRNAs, and lipids. Regulation of m6A methylation in the immune microenvironment in the development of diabetes mellitus.

Diabetes mellitus is a widespread metabolic disorder characterized by chronic hyperglycemia, driven primarily by insulin resistance and β-cell dysfunction. N6-methyladenosine (m6A), the most prevalent internal RNA modification in eukaryotes, has emerged as a critical regulator in diabetes pathogenesis. This review outlines how m6A methylation, mediated by writers (METTL3, METTL14), erasers (FTO, ALKBH5), and readers (YTHDFs, IGF2BPs), influences key diabetic processes including β-cell function, lipid metabolism, and insulin resistance. A key focus is on the role of m6A in modulating the immune

Microenvironment-responsive nanorobots for biomedical applications.

Conventional drug therapies face significant limitations in complex physiological environments, including poor targeting efficiency, passive diffusion mechanisms, and systemic toxicity. Inspired by the immune system, where immune cells are actively navigated by chemotactic gradients to reach infection sites, researchers have developed nanorobots capable of autonomous, microenvironment-responsive drug delivery. These nanorobots convert endogenous biochemical cues (e.g., pH, enzymes, reactive oxygen species) into directed motion and structure or shape changes, enabling deep tissue penetration an
