Possible role of dentin matrix in region-specific deposition of cellular and acellular extrinsic fibre cementum.

The mechanism whereby a region-specific deposition of the two types of cementum (cellular cementum and acellular extrinsic fibre cementum) is regulated on the growing root surface was tested using bisphosphonate-affected teeth of young rats and guinea pigs. The animals were injected subcutaneously with 8 or 10 mg P x kg body weight(-1) x day(-1) of 1-hydroxyethylidene-1,1-bisphosphonate (HEBP) for 1 or 2 weeks. In rat molars, HEBP prevented mineralization of newly formed root dentin matrix and totally inhibited de novo deposition of acellular extrinsic fibre cementum. Instead, thick cellular c

Tooth eruption and cementum formation in the Runx2/Cbfa1 heterozygous mouse.

Cleidocranial dysplasia (CCD) is an autosomal dominant human disorder that affects development of bones and teeth. The dental disorders in CCD patients include formation of supernumerary teeth, delayed tooth eruption, and lack of formation of cellular cementum in permanent teeth. This disorder involves a mutation in the osteoblast-specific transcription factor Runx2/Cbfa1, leading to haploinsufficiency of the Runx2/Cbfa1 protein. Here, we examined if Runx2/Cbfa1 heterozygous mice (with one functional allele for Runx2/Cbfa1) exhibit similar changes in tooth eruption, and dental cementum formati

Quality of periodontal healing. IV: Enzyme histochemical evidence for an osteoblast origin of reparative cementum.

The aim of the present study was to map the distribution of healing gingival and periodontal connective tissue well as osteoblastic activity in marginal periodontal wound healing using activity of alkaline phosphatases as markers. Activity of non-specific alkaline phosphatase (NSAP) was abundant in periodontal but not in gingival connective tissue. Activity of vanadate-resistant alkaline phosphatase (VRAP) had a preferential localisation to periosteal and endosteal bone surfaces but was not detectable in cementoblasts along root surfaces. However, following spontaneous marginal healing of expo

Hypercementosis Associated with ENPP1 Mutations and GACI.

Mineralization of bones and teeth is tightly regulated by levels of extracellular inorganic phosphate (Pi) and pyrophosphate (PPi). Three regulators that control pericellular concentrations of Pi and PPi include tissue-nonspecific alkaline phosphatase (TNAP), progressive ankylosis protein (ANK), and ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1). Inactivation of these factors results in mineralization disorders affecting teeth and their supporting structures. This study for the first time analyzed the effect of decreased PPi on dental development in individuals with generalized art

Extracellular vesicles at the periodontal immune-regenerative interface: coupled mechanisms and translational challenges.

Periodontitis is a chronic immune-inflammatory disease in which dysbiotic microbial communities and sustained host responses disrupt the gingiva, periodontal ligament (PDL), cementum, and alveolar bone. Because these tissues form an integrated attachment apparatus, effective regeneration requires coordinated control of inflammation and reconstruction of soft and mineralized tissues. Extracellular vesicles (EVs) function as intercellular mediators at this immune-regenerative interface by transferring proteins, lipids, and nucleic acids among immune, stromal, vascular, and bone-remodeling cells.

Restoration of Cementoblast Mineralization by Glutamine Under Inflammatory Conditions.

INTRODUCTION AND AIMS: Cementoblasts are critical for cementum formation and periodontal regeneration during inflammatory diseases such as periodontitis and apical periodontitis (AP). Bacterial infection leads to metabolic changes in cementoblasts and reduces mineralization capability. This study aimed to fully elucidate the metabolic changes of cementoblasts in inflammatory mineralization and explore therapeutic strategies targeting metabolism. METHODS: Mouse cementoblasts cell line OCCM-30 were infected with Porphyromonas gingivalis (P.g) to model microbial infection stress. Metabolic profi

Age estimation using Olze's method in an adult Brazilian population.

OBJECTIVES: Age estimation is important to set an anthropological profile in human remains, as well as in legal issues where the suspect or victim doesn't have documents that prove the real age. The age estimation, in adults, is a challenge, and it can be analyzed through some dental stages. This study aimed to test Olze's method in a Brazilian sample composed by adults, aged between 20 and 70 years old, both sex. METHODS: For experiments, 306 orthopantomography from Brazilian individuals between 20 and 70 years were selected. The dental involution through four different dental parameters wer

Central role of pyrophosphate in acellular cementum formation.

BACKGROUND: Inorganic pyrophosphate (PP(i)) is a physiologic inhibitor of hydroxyapatite mineral precipitation involved in regulating mineralized tissue development and pathologic calcification. Local levels of PP(i) are controlled by antagonistic functions of factors that decrease PP(i) and promote mineralization (tissue-nonspecific alkaline phosphatase, Alpl/TNAP), and those that increase local PP(i) and restrict mineralization (progressive ankylosis protein, ANK; ectonucleotide pyrophosphatase phosphodiesterase-1, NPP1). The cementum enveloping the tooth root is essential for tooth function

The role of epithelium in the development, structure and function of the tissues of tooth support.

The roles of epithelium in the development, structure and function of the tissues of tooth support are reviewed. Epithelium is involved in initiating odontogenesis which includes the tissues of tooth support and this role is discussed. Particular attention is paid to Hertwig's epithelial root sheath and its participation in the formation of the hyaline layer on the root surface as well as its possible role in initiating the differentiation of cementoblasts. The possible functions of the epithelial cell rests are reviewed and it is concluded that as yet no function can be ascribed to them. Evid

Effect of topical alendronate on root resorption of dried replanted dog teeth.

Alendronate (ALN) is a third generation bisphosphonate with demonstrated osteoclast inhibitory activity that may slow down the resorptive process after severe traumatic injuries. Eighty-two premolar roots of five mongrel dogs were endodontically treated and restored, extracted and treated as follows: 70 roots were bench dried for either 40 or 60 min. Thirty-eight of these roots were then soaked for 5 min in a 1 mM solution of ALN in Hanks' Balanced Salt Solution (HBSS) and replanted. Thirty-two roots were soaked for 5 min in HBSS and replanted. In the remaining 12 roots which were not exposed

Cementomimetics-constructing a cementum-like biomineralized microlayer via amelogenin-derived peptides.

Cementum is the outer-, mineralized-tissue covering the tooth root and an essential part of the system of periodontal tissue that anchors the tooth to the bone. Periodontal disease results from the destructive behavior of the host elicited by an infectious biofilm adhering to the tooth root and left untreated, may lead to tooth loss. We describe a novel protocol for identifying peptide sequences from native proteins with the potential to repair damaged dental tissues by controlling hydroxyapatite biomineralization. Using amelogenin as a case study and a bioinformatics scoring matrix, we identi

Dynamics of orthodontic root resorption and repair in human premolars: a light microscopy study.

The purpose of the study was to investigate the relationship between root resorption and repair in human premolars that had been orthodontically intruded. The objective was to examine these processes related to time and root development. Seventy-six premolars were divided into subgroups: 33 teeth were intruded and then extracted (G1); 25 teeth were intruded and then left in situ for varying periods before extraction (G2); 18 teeth served as the controls (G3). All teeth were examined by light microscopy. Using non-parametric statistical analysis, differences between the groups were examined wit

Developmental appearance and distribution of bone sialoprotein and osteopontin in human and rat cementum.

BACKGROUND: Bone sialoprotein (BSP) and osteopontin (OPN), two major noncollagenous proteins (NCPs) in collagen-based mineralized tissues, have been implicated in mineral deposition and cell- and matrix-matrix interactions during root development. However, their role in cementogenesis is still a subject of debate. Since distribution of proteins is indicative of function, we have analyzed their temporo-spatial appearance in relation to that of cementum collagen. METHODS: Human premolars and rat molars at various stages of root development characterized by differing rates of formation were fixe

Effect of enamel matrix proteins on the periodontal connective tissue-material interface after wound healing.

The periodontal ligament has the potential to regenerate a complete periodontal connective tissue attachment, starting with the deposition of cementum, on pathologically exposed root surfaces as well as several materials including titanium oxide. However, most commonly used dental materials result in a fibrous encapsulation or a chronic inflammatory response after periodontal wound healing rather than the formation of a periodontal connective tissue attachment. Recently, an extract of porcine enamel matrix (Emdogain(R), EMD) has been reported inductive of cementum formation in both in vivo and

Differential effects of parathyroid hormone, parathyroid hormone-related protein, and abaloparatide on collagen 1 expression by mouse cementoblasts and mouse tooth root density.

INTRODUCTION: Parathyroid hormone (PTH) plays an important role in maintaining mineral homeostasis by regulating calcium and phosphate levels. Clinical trials have shown that peptides of PTH (1-34), PTH-related protein (PTHrP 1-36), and the new peptide modeled on PTHrP, abaloparatide, can have different anabolic effects on osteoporotic subjects, but the underlying mechanisms are still unclear. The prevalence of moderate and major gingival recession has been shown to be higher in postmenopausal women with osteoporosis. In addition, there is a significant association between osteoporosis and too
