Loss of dystrophin reduces CB1 receptor expression and endocannabinoid-dependent synaptic plasticity in the cerebellar cortex.

Duchenne Muscular Dystrophy (DMD) is a debilitating degenerative condition with complex musculoskeletal and cognitive symptoms. The protein responsible, dystrophin, is expressed in both muscle tissue and within the central nervous system (CNS) where it localizes to inhibitory synapses. Recent work has shown that dystrophin loss in skeletal muscle leads to abnormalities in endocannabinoid signaling, particularly related to Cannabinoid Receptor Type 1 (CB1R) signaling pathways. CB1Rs are highly expressed throughout the CNS, and have been implicated in short- and long-term plasticity mechanisms.

Effect of Sodium Nitrite on the cerebellar cortex and the possible role of Quercetin in adult male albino rats.

The cerebellum is particularly vulnerable to intoxication and poisoning due to its complex structure. Sodium Nitrite is commonly used as a food additive, as well as in fertilizers and rodenticides. Quercetin, a widely distributed flavonoid in nature, has been investigated for its potential protective effects. This research aimed to evaluate the histological and biochemical changes in the cerebellar cortex of adult male albino rats resulting from Sodium Nitrite exposure, and to assess the protective role of Quercetin and the reversibility of these changes after stopping Sodium Nitrite. Upregulation of Neural Cell Adhesion Molecule 1 and Excessive Migration of Purkinje Cells in Cerebellar Cortex.

Purkinje cells (PCs) are large GABAergic projection neurons of the cerebellar cortex, endowed with elaborate dendrites that receive a multitude of excitatory inputs. Being the only efferent neuron of the cerebellar cortex, PCs project to cerebellar nuclei and control behaviors ranging from movement to cognition and social interaction. Neural cell adhesion molecule 1 (NCAM1) is widely expressed in the embryonic and postnatal development of the brain and plays essential roles in neuronal migration, axon pathfinding and synapse assembly. However, despite its high expression levels in cerebellum,

Time-dependent regulation of perineuronal nets in the cerebellar cortex during abstinence of cocaine-self administration.

RATIONALE: The probability of structural remodeling in brain circuits may be modulated by molecules of perineuronal nets (PNNs) that restrict neuronal plasticity to stabilize circuits. Animal research demonstrates that addictive drugs can remodel PNNs in different brain regions, including the cerebellum. OBJECTIVE: This study aimed to investigate the effects of short versus extended access to cocaine self-administration on PNN expression around Golgi interneurons in the cerebellar cortex after different periods of abstinence. METHODS: After 1 week of training (2 h/day), Sprague-Dawley rats s

Mutations in the Reelin pathway are associated with abnormal expression of microglial IgG FC receptors in the cerebellar cortex.

Microglia are the immune cells of the central nervous system involved in a variety of developmental processes, such as regulation of cell death and survival, spatial patterning, and contribute to the development of Purkinje cells (PCs) during migration. Microglia express immunoglobulin G Fc receptors (FcgRs). In this report, we describe microglial FcgR expression and its relation to abnormal PC migration in the cerebellum during development. To detect microglial FcgR, the direct anti-IgG (secondary antisera) and high concentrations of Triton X-100 were applied as a method for labeling microgli

Chronic alcohol consumption reduces the cortical layer volumes and the number of neurons of the rat cerebellar cortex.

Past work showed that chronic alcohol consumption induces massive degeneration in the rat cerebellar cortex. In order to provide details of these effects, the cortical cell numbers and the cerebellar layer volumes were studied in adult rats fed alcohol for 1, 3, 6, 12, and 18 months and compared with respective age-matched controls. Significant decreases of granular and molecular layer volumes were found after 3 and 6 months of alcohol feeding, respectively. No significant differences were found in the Purkinje cell layer volume. Neuron countings showed that granule cells and molecular layer i

Atypical neuropathological sCJD-MM phenotype with abundant white matter Kuru-type plaques sparing the cerebellar cortex.

We describe an atypical neuropatholgical phenotype of sporadic Creutzfeldt-Jakob disease (sCJD) in a 64-year-old man presenting with a 5-month history of rapidly progressive dementia, comprising behavioral disturbances, memory complaints, disorientation and language alterations. MRI showed diffuse atrophy and hyperintensities in parietal, occipital, temporal and frontal cortices and left caudate nucleus on T2-weighted and fluid-attenuated inversion recovery images. No typical EEG alterations were observed. Repeated 14-3-3 assay was positive after a first negative test. Neuropathology showed cl

Degeneration of posterior column nucleus, inferior olivary nucleus and cerebellar cortex: system degeneration of paraneoplastic disease?

Degeneration of the posterior column nucleus, inferior olive, and cerebellar cortex is reported in a cancer patient. A 70-year-old man developed an ataxic gait and rapidly progressive disturbance of deep sensation over six months, followed by an abasic state. Early well-differentiated tubular adenocarcinoma was detected and total gastrectomy was performed. He died due to pulmonary tuberculosis about two years and nine months after the operation. Degeneration of the posterior column nuclei was found, explaining the disturbance of deep sensation noted in the clinical course, although there were

Chronic alcohol consumption induces plastic changes in granule cell synaptic boutons of the rat cerebellar cortex.

The reduction of cerebellar cortex molecular layer afferents, as a consequence of an alcohol-induced granule cell degeneration, is followed by a remodeling activity which leads to marked structural changes. Using Golgi, ultrastructural and combined Golgi-ultrastructural methods, it was evaluated the role played by granule cell synaptic boutons in this plastic process and, furthermore, determined whether there were differences between the boutons located in the ascending portions of the axons and those belonging to parallel fibers. No significant differences were found in the number of boutons

Broad glucagon-like peptide-1 (GLP-1) receptor distribution enhances glutamatergic transmission throughout the mouse cerebellar cortex.

The gut hormone glucagon-like peptide 1 (GLP-1) is a key contributor to the controls of food intake and reward processing. GLP-1 receptors (GLP-1R) are widely expressed throughout the central nervous system where their signaling alters neuronal excitability, resulting in suppressed food intake and reward. Recent studies have shown GLP-1R expression in Purkinje cells (PCs) within the cerebellum, a region gathering increasing appreciation for its role in sensory processing and appetitive behaviors. Importantly, the cerebellum responds to feeding signals and directly projects to key mesolimbic nu

Cerebellar vermis and somatosensory-motor cortex differentially contribute to sex differences in acute pain perception in rats.

BACKGROUND: Pain is a universal experience, yet sex differences in acute pain perception remain unclear. Here, we examined sex-specific brain responses to acute pain evoked by tail clamping in rats. METHODS: A suitable isoflurane concentration for resting-state functional magnetic resonance imaging (rs-fMRI) was identified to ensure sedation without suppressing nociceptive responsiveness. rs-fMRI was performed to assess sex differences in brain activity and functional connectivity during acute pain, with control groups receiving touch stimulation or isoflurane-only exposure to identify pain-s

Cerebellar Purkinje cell firing promotes conscious recovery from anesthesia state through coordinating neuronal communications with motor cortex.

Background: The neurobiological basis of gaining consciousness from unconscious state induced by anesthetics remains unknown. This study was designed to investigate the involvement of the cerebello-thalamus-motor cortical loop mediating consciousness transitions from the loss of consciousness (LOC) induced by an inhalational anesthetic sevoflurane in mice. Methods: The neural tracing and fMRI together with opto-chemogenetic manipulation were used to investigate the potential link among cerebello-thalamus-motor cortical brain regions. Excitatory Amino acid transporter expression in the essential tremor dentate nucleus and cerebellar cortex: A postmortem study.

BACKGROUND: Genome-wide association studies have revealed a link between essential tremor (ET) and the gene SLC1A2, which encodes excitatory amino acid transporter type 2 (EAAT2). We explored EAAT biology in ET by quantifying EAAT2 and EAAT1 levels in the cerebellar dentate nucleus, and expanded our prior analysis of EAAT2 levels in the cerebellar cortex. OBJECTIVE: To quantify EAAT2 and EAAT1 levels in the cerebellar dentate nucleus and cerebellar cortex of ET cases vs. METHODS: We used immunohistochemistry to quantify EAAT2 and EAAT1 levels in the dentate nucleus of a discovery cohort of 1

Effects of prenatal paraquat and mancozeb exposure on amino acid synaptic transmission in developing mouse cerebellar cortex.

The goal of this study was to analyze the effects of prenatal exposure to the pesticides paraquat (PQ) and mancozeb (MZ) on the development of synaptic transmission in mouse cerebellar cortex. Pregnant NMRI mice were treated with either saline, 10 mg/kg PQ, 30 mg/kg MZ or the combination of PQ + MZ, between gestational days 12 (E12) and E20. Variation in the levels of amino acid neurotransmitters was determined by HPLC, between postnatal day 1 (P1) and P30. Motor coordination was assessed by locomotor activity evaluation of control and experimental pups at P14, P21 and P30. Significant reducti

Impact of varying maternal dietary folate intake on cerebellar cortex histomorphology and cell density in offspring rats.

The cerebellum has a long, protracted developmental period that spans from the embryonic to postnatal periods; as a result, it is more sensitive to intrauterine and postnatal insults like nutritional deficiencies. Folate is crucial for foetal and early postnatal brain development; however, its effects on cerebellar growth and development are unknown. The aim of this study was to examine the effects of maternal folate intake on the histomorphology and cell density of the developing cerebellum. Twelve adult female rats (rattus norvegicus) were randomly assigned to one of four premixed diet group

Intermittent Theta-Burst Stimulation for Stroke: Primary Motor Cortex Versus Cerebellar Stimulation: A Randomized Sham-Controlled Trial.

BACKGROUND: Stroke survivors with impaired balance and motor function tend to have relatively poor functional outcomes. The cerebellum and primary motor cortex (M1) have been suggested as targets for neuromodulation of balance and motor recovery after stroke. This study aimed to compare the efficacy and safety of intermittent theta-burst stimulation (iTBS) to the cerebellum or M1 on balance and motor recovery in patients with stroke. METHODS: In this randomized, double-blind, sham-controlled clinical trial, patients with subacute stroke were randomly divided into 3 groups: M1-, cerebellar-, a

Calbindin D28k distribution in neurons and reactive gliosis in cerebellar cortex of natural Rabies virus-infected cattle.

Rabies has been an enigmatic disease because microscopic findings in central nervous system tissues do not always correlate well with the severity of the clinical illness. Immunohistochemical staining of the calcium-binding protein calbindin (specifically CbD28k) seems to be the technique most used to identify Purkinje neurons under normal and pathological conditions. In the present work, we evaluated CbD28k immunoreactivity in the cerebellar cortex of normal and natural Rabies virus (RABV)-infected cattle. We examined brains from 3 normal cows and from 6 crossbreed cattle with a histologic di

Quantitative scanning transmission electron microscopy of ultrathin cryosections: subcellular organelles in rapidly frozen liver and cerebellar cortex.

Freeze-dried, ultrathin cryosections of directly frozen mouse liver and brain have been prepared and characterized by low-dose dark-field scanning transmission electron microscopy (STEM). These improved cryosections gave images comparable to those from conventional plastic sections. They were thin enough (< < 1.0 elastic mean free path) to use established dark-field techniques, modified for thickness-dependent nonlinearities, to measure the dry mass fraction of individual organelles, and hence to deduce their water content.
