Optimal routing to cerebellum-like structures.

The vast expansion from mossy fibers to cerebellar granule cells (GrC) produces a neural representation that supports functions including associative and internal model learning. This motif is shared by other cerebellum-like structures and has inspired numerous theoretical models. Less attention has been paid to structures immediately presynaptic to GrC layers, whose architecture can be described as a 'bottleneck' and whose function is not understood. We therefore develop a theory of cerebellum-like structures in conjunction with their afferent pathways that predicts the role of the pontine re

Unsupervised nonmotor learning in the human cerebellum.

A core motor function of the cerebellum is error-based learning: it uses prediction errors-mismatches between predicted and actual sensory feedback-to refine actions. One provocative hypothesis is that the cerebellum performs similar computations in cognitive domains. Here, we ask whether error-based learning computations in the cerebellum extend to a passive statistical learning task that requires neither action nor decision-making. Human participants viewed sequences of visual stimuli with varying probabilistic stimulus-stimulus transitions while undergoing fMRI. Subjects reported no explici

A vector calculus for neural computation in the cerebellum.

Null space theory predicts that neurons generate spikes not only to produce behavior but also to prevent the undesirable effect of other neurons on behavior. In this work, we show that this competitive cancellation is essential for understanding computation in the cerebellum. In marmosets, we identified a vector for each Purkinje cell (P cell) along which its spikes displaced the eyes. Two spikes in two different P cells produced superposition of their vectors. In the resulting population activity, the spikes were canceled if their contributions were perpendicular to the intended movement. Mos

Characterization of Perineuronal Nets in the Human Cerebellum.

Perineuronal nets (PNNs) are condensed extracellular matrix structures that regulate synaptic plasticity and neuronal stability. While their distribution and function have been well characterized in rodents, PNNs remain largely understudied in the human cerebellum. Here, we provide an anatomical characterization of PNNs in the post-mortem human cerebellum and compare their distribution and cellular phenotypes with those in mouse and macaque cerebellum. Using immunofluorescence for both Wisteria floribunda lectin (WFL) and aggrecan, we observed that PNNs in humans were exclusively localized in

Updating the physiology of hypnotizability: Cerebellum and insula.

This chapter describes the physiological correlates of hypnotizability associated with the morpho-functional aspects of the cerebellum and insula, which are brain structures neglected in physiological research on hypnotizability. Both structures are involved in sensorimotor and interoceptive networks that play a cooperative role in sensorimotor and cognitive-emotional control. Hypnotizability-related cerebellar and insular characteristics may underlie observed differences in postural and visuomotor control, motor cortex excitability, functional equivalence between actual and imagined perceptio

Neurophysiology of the cerebellum and clinical correlations: a review.

The cerebellum is a complex structure tightly connected to the cerebral cortex, brainstem, and spinal cord. It plays an important role in movement coordination and motor planning. Lately, it has been also recognized as a key component in cognitive circuits. The specific motor functions of the cerebellum include the control of the initiation, execution, and velocity of movements, as well as the maintenance of balance, motor coordination, and muscle tonus. Cerebellar lesions typically result in ataxia but can also lead to other manifestations, such as abnormal eye movements, severe vertigo, or i

Cellular development and evolution of the mammalian cerebellum.

The expansion of the neocortex, a hallmark of mammalian evolution1,2, was accompanied by an increase in cerebellar neuron numbers3. However, little is known about the evolution of the cellular programmes underlying the development of the cerebellum in mammals. In this study we generated single-nucleus RNA-sequencing data for around 400,000 cells to trace the development of the cerebellum from early neurogenesis to adulthood in human, mouse and the marsupial opossum. We established a consensus classification of the cellular diversity in the developing mammalian cerebellum and validated it by sp

Linking the cerebellum to Parkinson disease: an update.

Parkinson disease (PD) is characterized by heterogeneous motor and non-motor symptoms, resulting from neurodegeneration involving various parts of the central nervous system. Although PD pathology predominantly involves the nigral-striatal system, growing evidence suggests that pathological changes extend beyond the basal ganglia into other parts of the brain, including the cerebellum. In addition to a primary involvement in motor control, the cerebellum is now known to also have an important role in cognitive, sleep and affective processes. Over the past decade, an accumulating body of resear

Structure, Function, and Genetics of the Cerebellum in Autism.

Autism spectrum disorders are common neurodevelopmental disorders that are defined by core behavioral symptoms but have diverse genetic and environmental risk factors. Despite its etiological heterogeneity, several unifying theories of autism have been proposed, including a central role for cerebellar dysfunction. The cerebellum follows a protracted course of development that culminates in an exquisitely crafted brain structure containing over half of the neurons in the entire brain densely packed into a highly organized structure. Through its complex network of connections with cortical and s

Characteristics of Tremor Induced by Lesions of the Cerebellum.

It is a clinical experience that acute lesions of the cerebellum induce pathological tremor, which tends to improve. However, quantitative characteristics, imaging correlates, and recovery of cerebellar tremor have not been systematically investigated. We studied the prevalence, quantitative parameters measured with biaxial accelerometry, and recovery of pathological tremor in 68 patients with lesions affecting the cerebellum. We also investigated the correlation between the occurrence and characteristics of tremor and lesion localization using 3D T1-weighted MRI images which were normalized a

The volume of the cerebellum in the second semester of gestation.

BACKGROUND AND AIMS: The cerebellum ("little brain"), the largest part of hind brain, lies in the posterior cranial fossa, beneath the occipital lobe and dorsal to the brainstem. It develops over a long period: it is one of the first structures in the brain to begin to differentiate, but one of the last to mature. The use of ultrasonography has significantly improved the evaluation of fetal growth and development and has permitted prenatal diagnosis of a variety of congenital malformations. The aim of our study was to evaluate the cerebellar growth and development using 2 different measuring te

The cerebellum's contribution to beat interval discrimination.

From expert percussionists to individuals who cannot dance, there are widespread differences in people's abilities to perceive and synchronize with a musical beat. The aim of our study was to identify candidate brain regions that might be associated with these abilities. For this purpose, we used Voxel-Based-Morphometry to correlate inter-individual differences in performance on the Harvard Beat Assessment Tests (H-BAT) with local inter-individual variations in gray matter volumes across the entire brain space in 60 individuals. Analysis revealed significant co-variations between performances

The role of the cerebellum in the regulation of language functions.

The present paper is a review of studies on the role of the cerebellum in the regulation of language functions. This brain structure until recently associated chiefly with motor skills, visual-motor coordination and balance, proves to be significant also for cognitive functioning. With regard to language functions, studies show that the cerebellum determines verbal fluency (both semantic and formal) expressive and receptive grammar processing, the ability to identify and correct language mistakes, and writing skills. Cerebellar damage is a possible cause of aphasia or the cerebellar mutism syn

Thalamic interactions of cerebellum and basal ganglia.

Cerebellum and basal ganglia are reciprocally interconnected with the neocortex via oligosynaptic loops. The signal pathways of these loops predominantly converge in motor areas of the frontal cortex and are mainly segregated on subcortical level. Recent evidence, however, indicates subcortical interaction of these systems. We have reviewed literature that addresses the question whether, and to what extent, projections of main output nuclei of basal ganglia (reticular part of the substantia nigra, internal segment of the globus pallidus) and cerebellum (deep cerebellar nuclei) interact with ea

Oscillations, Timing, Plasticity, and Learning in the Cerebellum.

The highly stereotyped, crystal-like architecture of the cerebellum has long served as a basis for hypotheses with regard to the function(s) that it subserves. Historically, most clinical observations and experimental work have focused on the involvement of the cerebellum in motor control, with particular emphasis on coordination and learning. Two main models have been suggested to account for cerebellar functioning. According to Llinás's theory, the cerebellum acts as a control machine that uses the rhythmic activity of the inferior olive to synchronize Purkinje cell populations for fine-tuni

[The role of the cerebellum in auditory process and tinnitus].

Cerebellum connects with each part of the auditory pathway directly or indirectly, forming the anatomical basis for a cerebellar role in auditory process. Functional imaging studies and clinical observations provide more abundant support for this view. Tinnitus is one of the most common clinical manifestations when the auditory system is impaired and remains unsolved. Basic science researches in neurotransmitter chemistry and molecular biochemistry, together with functional imaging studies, indicate that cerebellum might contribute to the pathophysiology of tinnitus. In this review we summariz

Effects of Ethanol on the Cerebellum: Advances and Prospects.

Alcohol abuse causes cerebellar dysfunction and cerebellar ataxia is a common feature in alcoholics. Alcohol exposure during development also impacts the cerebellum. Children with fetal alcohol spectrum disorder (FASD) show many symptoms associated specifically with cerebellar deficits. However, the cellular and molecular mechanisms are unclear. This special issue discusses the most recent advances in the study of mechanisms underlying alcoholinduced cerebellar deficits. The alteration in GABAA receptor-dependent neurotransmission is a potential mechanism for ethanol-induced cerebellar dysfunc
