Neutrophil chemotactic activity produced by normal and activated human bronchoalveolar lavage cells.

Activated macrophages can secrete a number of mediators that can attract inflammatory cells and enhance secretion of phlogistic substances from these cells. The ultimate effect of activated bronchoalveolar lavage (BAL) cells may be fibrotic lung injury. Inasmuch as pulmonary sarcoidosis is a disease associated with spontaneous activation of macrophages and lymphocytes among BAL cells, cells obtained from patients with sarcoidosis were compared with normal cells. We report that adherent BAL cells in culture from patients with sarcoidosis (n = 21) release during a resting period in vitro more ch

Chemotactic reactivity of eosinophils obtained from bone marrow and peritoneal cavity of cyclophosphamide-treated, Toxocara canis-infected mice.

Chemotactic reactivity of eosinophils obtained from the bone marrow (BM-Eo) of cyclophosphamide-treated, Toxocara canis-infected mice was compared to that of eosinophils obtained from the peritoneal cavity (PEC-Eo). BM-Eo responded well to alanyl-tetrapeptide, a synthetic eosinophil chemotactic factor of anaphylaxis (ECF-A), whereas PEC-Eo did not. Both BM-Eo and PEC-Eo showed almost equally high chemotactic reactivity to parasite-derived ECF, ECF lymphokine and complement-derived ECF. Chemotactic reactivity of BM-Eo to synthetic ECF-A was deactivated by preincubation with ECF-A. Unresponsiven

A leukotactic factor in the stratum corneum of pustulosis palmaris et plantaris: a possible mechanism for the formation of intra-epidermal sterile pustules.

Previous studies on the chemotactic properties of soluble substances from psoriatic scales demonstrated the presence of complement-derived chemotactic factors (anaphylatoxins) that seem to play a crucial role in the production of typical psoriasiform tissue changes. The crude extract from the stratum corneum of lesions of pustulosis palmaris et plantaris (PPP) was also noted to contain highly chemotactic components for polymorphonuclear leukocytes (PMN's) as compared with a crude callus extract in the present study. The leukotactic factors contained in the major chemotactic fractions isolated

Hepatocyte growth factor is a predominant chemoattractant for gingival epithelial cells produced by radicular cyst-derived fibroblast-like cells.

To clarify whether fibroblasts could be involved in the pathogenesis of periodontal pocket formation, the chemotactic activity of radicular cyst-derived fibroblast-like cell (RCF)-conditioned medium (RCF-CM) for gingival epithelial cells was examined using a modified Boyden chamber assay. RCF-CM possessed significant chemotactic activity, which was decreased markedly by treatment with anti-human hepatocyte growth factor (HGF) antibody. Furthermore, the chemotactic activity of RCF-CM was well correlated with HGF content. These results show that the RCF secrete an HGF-like factor, and suggest th

Presence of chemotactic peptides other than C5a anaphylatoxin in scales of psoriasis and sterile pustular dermatoses.

A unique leukocyte chemotactic factor of around 12 kD molecular weight has been suggested to be responsible for the mechanisms involved in transepidermal migration of leukocytes observed in psoriasis and related sterile pustular dermatoses. Although we confirmed with radioimmunoassay the presence of a C5-cleavage product in the chemotactic fractions from psoriatic scale extract eluted by gel filtration HPLC, the neutrophil chemotactic activity demonstrated in the peak fraction was only partially inhibited by rabbit antiserum to human C5a, whereas that noted in the peak fraction, prepared from

Chemotactic defect in Wegener's granulomatosis.

Chemotaxis of the polymorphonuclear (PMN) leukocytes from nine Wegener's granulomatosis patients was examined in vitro. All the patients except one showed remarkable decrease in their chemotactic activity: This might be caused by abnormality in the PMN leukocytes themselves. It was confirmed that there was no increase in chemotactic inhibitory substance(s). There was chemotactic factor(s) in sufficient amount in the serum to react with the antigen-antibody complex for normal chemotaxis. As to the random mobility of PMN leukocytes there was no abnormality in these patients.

Production of chemotactic activity for polymorphonuclear leukocytes by cultured rat hepatocytes exposed to ethanol.

Acute alcoholic hepatitis is characterized by infiltration of the liver parenchyma with polymorphonuclear leukocytes. As a possible explanation for this phenomenon, we have found that ethanol stimulates cultured rat hepatocytes to generate potent chemotactic activity. Hepatocytes (greater than 99% pure), isolated from the livers of Sprague-Dawley rats, responded to incubation with ethanol (2.0-10 mM) by releasing chemotactic activity for human polymorphonuclear leukocytes into culture supernatants in a time- and concentration-dependent fashion. Chemotactic activity was maximal after incubation

[The role of eosinophils in inflammatory reactions].

Recent findings have contributed substantially to a better understanding of the ontogeny and function of eosinophil leukocytes. Together with other granulocytes, the cells originate in the bone marrow from a common stem cell. Their development is regulated by genetic factors and by products of T-lymphocytes. Chemotactic factors of the complement cascade (C5a), lymphokines and eosinophil chemotactic leukotrienes (ECL) stimulate their migration to tissue sites. Generation and biological effectiveness of eosinophil chemotactic factors can be modulated in numerous ways, thus changing the outcome o

Proliferative effects of heparin-binding epidermal growth factor-like growth factor on pterygium epithelial cells and fibroblasts.

PURPOSE: To investigate the growth promoting and chemotactic effects of heparin binding epidermal growth factor-like growth factor (HB-EGF), recently shown to be upregulated by ultraviolet irradiation in pterygium-derived epithelium cells (PECs) and pterygium fibroblasts (PFs). METHODS: PECs and PFs were incubated with various concentrations of HB-EGF. Cell proliferation was evaluated by measurement of [3H]thymidine incorporation. The potential chemotactic effect of HB-EGF on these two cell lines was assessed with migration assays, using modified Boyden chambers and checkerboard analysis. RE

Competing chemical gradients change chemotactic dynamics and cell distribution.

Cells are constantly exposed to diverse stimuli-chemical, mechanical, or electrical-that guide their movement. In physiological conditions, these signals often overlap, as seen during infections, where neutrophils and dendritic cells navigate through multiple chemotactic fields. How cells integrate and prioritize competing signals remains unclear. For instance, in the presence of opposing chemoattractant gradients, how do cells decide which direction to go? When should local signals dominate distant ones? A key factor in these processes is the precision with which cells sense each gradient, wh

Stimulation of the biosynthesis of interleukin 8 by interleukin 1 and tumor necrosis factor alpha in cultured human chorionic cells.

Human chorion, but not amnion, tissue explants produced substantial quantities of neutrophil chemoattractant in response to interleukin 1 (IL-1) and tumor necrosis factor alpha (TNF alpha). This suggested that chorion is one of the chemoattractant producing tissues. Therefore, the biochemical properties and the regulation of a chemoattractant in human chorionic cells were examined. IL-1 alpha and TNF alpha stimulated human chorionic cells to produce neutrophil chemotactic factor in both a dose- and time-dependent manner. This chemotactic factor was a heat-stable and trypsin-sensitive protein w

The relationship between bronchoalveolar neutrophil recruitment and bronchoconstriction induced by a soluble extract of cotton bracts.

Byssinosis is characterized by an obstructive bronchitis in some workers exposed to cotton dust. Mechanisms of airway alterations induced by cotton dust are unknown. To study mechanisms of these alterations, we administered an aqueous extract of cotton bract (CBE) to normal human subjects. After CBE inhalation, we demonstrated reproducible reductions in flow rates of variable magnitude. The percentage of polymorphonuclear leukocytes (PMN) recovered by bronchoalveolar lavage (BAL) and total PMN per milliliter BAL fluid were increased after challenge with CBE, and these parameters were correlate

Detection of a truly chemotactic lymphokine for human monocytes using millipore membranes.

The detection of the lymphokine monocyte chemotactic factor by determination of the leading front of monocyte migration is described. Stimulated and control supernatants were placed on the attractant side of cellulose ester filters with normal human monocytes as responder cells. Lymphokine activity was assessed by comparing the leading front of monocyte migration in response to control and stimulated culture supernatants. More monocytes migrated to all levels within the filters towards PHA stimulated culture supernatant than to control supernatant, indicating that the lymphokine acted on the e

Mediation of leukocyte components of inflammatory reactions by lipoxygenase products of arachidonic acid.

The leukocyte lipoxygenase products LTB4 and 5-HETE elicit human neutrophil and eosinophil chemotactic responses in vitro and in vivo (Fig. 4), and are present at elevated concentrations in the lesions of some human inflammatory diseases, such as rheumatoid arthritis and the spondyloarthritides. The chemotactic potency of LTB4 is similar to that of the minor fragment of the fifth component of human complement, termed C5a, and is 30- to 300-fold greater than that of 5-HETE and of other natural DHETE isomers. Human neutrophils possess distinct subsets of chemotactic factor receptors that are spe

Psoriasis and leukocyte chemotaxis.

Transepidermal migration of leukocytes, with resultant formation of microscopic or macroscopic sterile subcorneal pustules is a phenomenon characteristically noted in psoriasis and related sterile pustular dermatoses. It is natural to assume the presence of potent neutrophil chemotactic substances in the subcorneal portion of the lesional epidermis, because this location is the target of the in vivo leukocyte chemotaxis. In fact, crude psoriasis scale extracts show remarkably high neutrophil chemotactic and activating properties as compared with those of other non-psoriatic inflammatory dermat

Chemotactic heterogeneity of eosinophils in idiopathic pulmonary eosinophilia.

Heterogeneity in the chemotactic response of eosinophils to 5 T cell line eosinophilic chemotactic factors (ECFs) was assessed in 5 patients with idiopathic pulmonary eosinophilia. Eosinophils from 2 patients responded to all 5 ECFs (group 1), whereas eosinophils from the other 3 patients responded to ECF-PI 5, PI 6, PI 7 and PI 8 but failed to respond to ECF-PI 9 (group 2). It was further found that group 1 showed an elevated level of lactate dehydrogenase and a positive tuberculin reaction, whereas group 2 showed neither. The effects of steroid therapy on the chemotactic responses of eosinop

In vivo and in vitro studies on possible pathogenic mechanisms of Actinomyces viscosus.

Actinomycotic infections are characterized by long-term inflammatory lesions containing large numbers of polymorphonuclear leukocytes (PMNs) and mononuclear cells. The pathogenic mechanisms involved in these lesions are not understood. Homogenates of Actinomyces viscosus (AVIS) induce an acute inflammatory response with a predominance of PMNs within 6 h after injection into the footpads of nonimmunized mice. These homogenates, when tested in vitro, contain potent chemotactic activity for human PMNs. In vitro chemotactic activity for human monocytes is weak but statistically significant (P less

The mediation of tissue eosinophilia in hypersensitivity reactions. V. Comparative study of tissue eosinophilia in the skin lesions of local and systemic passive cutaneous anaphylactic reactions.

Tissue eosinophilia in active cutaneous anaphylaxis reactions is biphasic: the early phase (6 hr) is induced by a low molecular (mol. wt. 300) factor (early ECF), and the delayed phase (24 hr) is mediated by synergy of two different factors (delayed ECF-a and -b). In this study, we report the mediation of tissue eosinophilia in passive cutaneous anaphylactic (PCA) reaction sites. Tissue eosinophilia in systemic PCA showed two phases with peaks at 6 hr and 24 hr, while that in local PCA was monophasic and peaked at 12 hr. A dialysable eosinophil chemotactic factor was isolated from the early st

Enhancement of intestinal eosinophilia during Hymenolepis nana infection in mice.

The ability of Hymenolepis nana oncosphere extract to induce eosinophil chemotactic response was examined in vitro and in vivo. The extract showed a chemotactic activity specific for eosinophils but not for neutrophils. Partially purified eosinophil chemotactic factors (ECFs) from the oncosphere extract showed apparent molecular mass from 5.5 to 9.6kDa and 30 to 40kDa. These were resistant to heating and proteinase K digestion but sensitive to periodate oxidation. Peritoneal injection of the crude extract or partially purified ECFs to mice resulted in a preferential eosinophil infiltration. Th

Serum levels of neutrophil and eosinophil chemotactic activities in mastocytosis.

Neutrophil and eosinophil chemotactic activities (NCA and ECA) were measured in serum from twenty-two patients with urticaria pigmentosa or systemic mastocytosis. NCA was also measured after heating serum to 56 degrees C (heat-stable NCA). Although these factors were increased in about half of the patients there was no correlation with histamine release as estimated by the excretion of the main histamine metabolite methylimidazoleacetic acid (MelmAA) in urine. A significant increase in heat-stable NCA, however, was found in patients with pruritus and abnormal high values of MelmAA. It is concl
