Drug metabolism and disposition: the biological fate of chemicals2026eng
Proteolysis targeting chimeras (PROTACs) have emerged as a promising modality over the past decade, with only a few advancing to clinical trials. As "beyond rule-of-five" compounds, PROTACs pose significant challenges for absorption, distribution, metabolism, and excretion, often necessitating iterative in vitro absorption, distribution, metabolism, and excretion assay optimization or reliance on in vivo pharmacokinetic studies, particularly to understand oral absorption. In this study, in vitro-in vivo correlations from clinically advanced PROTACs were leveraged to develop a novel classificat

Targeted chimeras have reshaped therapeutic design by converting target recognition into active degradation. As one of the most clinically established therapeutic modalities, antibodies provide high affinity and specificity to the protein of interest. Antibody-associated targeted chimeras (AbTACs) are emerging as an important class of biologic degraders for targeted protein elimination, particularly of extracellular and membrane-associated proteins. AbTACs offer a strategy to extend its recognition capacity beyond target binding by coupling defined degradation pathway. In these systems, antibo

INTRODUCTION: Friedreich's ataxia (FRDA) is a prototypical repeat expansion disorder in which large intronic guanine‑adenine‑adenine (GAA) tracts at the frataxin (FXN) gene locus induce heterochromatin formation, impaired transcriptional elongation, and reduced FXN expression, driving progressive neurodegeneration and cardiomyopathy. Epigenetic therapies that restore endogenous FXN transcription have therefore emerged as a coherent disease‑modifying strategy focused on reversing repeat‑associated gene silencing at its root.
AREAS COVERED: This review summarizes the evolution of FXN protein‑re

Proteolysis Targeting Chimera (PROTAC) is an innovative drug discovery strategy that utilizes bifunctional small molecules to bring a target protein (POI) into proximity with an E3 ubiquitin ligase, thereby inducing ubiquitination of the pathogenic protein and its subsequent degradation by the proteasome. PROTACs exhibit a novel mechanism of action, catalytic efficiency, high selectivity, and broad applicability. They have achieved significant breakthroughs, particularly in oncology, with several molecules currently in phase II/III clinical trials. The application of PROTAC technology in antiv

Chronic hepatitis B virus (HBV) infection remains a major global health challenge, largely because current antiviral therapies seldom achieve sustained clearance of the viral surface antigen (HBsAg), a central driver of immune tolerance and viral persistence. Here, we present a programmable, nanobody-based targeted protein degradation platform, termed Nanobody-Targeting Chimera (Nab-TAC), designed for the intracellular clearance of HBV antigens. To systematically identify functional degradation modules, we developed a split-luciferase complementation-based quantitative screening platform that

The dysregulation of CK1 isoforms is linked to various types of diseases, including neurodegeneration and different types of neoplasia such as colon, pancreatic, breast, and ovarian cancer. For CK1 isoforms, a plethora of effective small molecule inhibitors are available. However, only a few degraders of CK1α and, more recently, proteolysis targeting chimeras (PROTACs) for CK1δ/CK1ε have been reported. In this study, we applied the PROTAC concept by harnessing molecular modelling to design and synthesize a series of candidate CK1δ-targeting PROTACs based on a highly specific and potent benzoth

A ribonuclease-targeting chimera (RiboTAC) is a heterobifunctional compound that binds to an RNA target and recruits a ribonuclease to cleave the bound RNA. This study investigates the impact of linker length and composition on RiboTAC potency in leukemia and breast cancer cellular models. Increasing linker length from two polyethylene glycol units to eight increased RiboTAC potency, while longer linker lengths decreased potency. The optimized RiboTAC reduced the abundance of oncogenic LGALS1 transcripts in THP-1 leukemia cells (∼95%) and in MDA-MB-231 triple negative breast cancer cells (∼70%

Recent medical research involving human-monkey chimeras, human brain organoids in rats, and the transplantation of a gene-edited pig heart and gene-edited pig kidneys in living human beings have intensified the debate about whether we should create human-animal chimeras for biomedical purposes and, if so, how we should treat them. Influential views in the debate frequently appeal to assumptions regarding how people will react to such chimeras. It has, for example, been argued that the most important objection against creating such chimeras is that this will result in inexorable moral confusion

Introduction: The new category of chimeric antigen receptor T - cell raised hopes for a more effective treatment of large B cell lymphoma and acute lymphoblastic leukemia. Nevertheless, their soaring acquisition costs will stretch the fiscal capacity of the health systems worldwide. To this direction, the scope of this study is to provide a systematic review of their economic evaluations. Areas covered: A systematic review of the economic evaluations of tisagenlecleucel and axicabtagene was performed. Expert opinion: The available data indicate that these products demonstrate a potentially fav

UNLABELLED: The coronavirus membrane (M) protein is the central actor in virion morphogenesis. M organizes the components of the viral membrane, and interactions of M with itself and with the nucleocapsid (N) protein drive virus assembly and budding. In order to further define M-M and M-N interactions, we constructed mutants of the model coronavirus mouse hepatitis virus (MHV) in which all or part of the M protein was replaced by its phylogenetically divergent counterpart from severe acute respiratory syndrome coronavirus (SARS-CoV). We were able to obtain viable chimeras containing the entire

BACKGROUND: Mixed chimerism is an effective approach for tolerance induction in transplantation. Strategies to achieve mixed chimerism with relatively low toxicity have significantly expanded the clinical use of chimerism.
METHODS: Allogeneic bone marrow transplants were performed between B6 (H2(b)) and BALB/c (H2(d)) mice. Recipient B6 mice were nonmyeloablatively conditioned with anti-αβ-T-cell receptor, anti-CD154, or rapamycin alone or in different combinations. A total of 15 × 10(6) BALB/c bone marrow cells were transplanted after varying doses of cGy of total body irradiation.

We describe general methods to detect and quantify small numbers of specific molecules. We redirected self-splicing protein inteins to create 'tadpoles', chimeric molecules comprised of a protein head covalently coupled to an oligonucleotide tail. We made different classes of tadpoles that bind specific targets, including Bacillus anthracis protective antigen and the enzyme cofactor biotin. We measured the amount of bound target by quantifying DNA tails by T7 RNA polymerase runoff transcription and real-time polymerase chain reaction (PCR) evaluated by rigorous statistical methods. These assay

Human ecto-ATPase (ecto-nucleoside triphosphate diphosphohydrolase 2 [eNTPDase2], also known as CD39L1) has been expressed and characterized in COS cells. It exhibits some unusual enzymology that is similar to a few members of this class of proteins but different from the majority of the family members. Hydrolysis of ATP by human ecto-ATPase is nonlinear with time, and its activity is stimulated/stabilized by both the lectin concanavalin A and the chemical cross-linking agent disuccinimidyl suberate. Like other members of the eNTPDase family, the human ecto-ATPase is a tetramer, the activity o

Tolerance to major histocompatibility antigens as well as the ability to mount a cytotoxic response to hapten-modified cells of bone marrow donor and host origin was studied in allogeneic radiation chimeras. Lethally irradiated (C57BL/6 X DBA/2)F1 hosts reconstituted with anti-Thy 1.2 + C-treated bone marrow from (C57BL/6 X CBA)F1 mice showed tolerance to the MHC antigens of the three parental strains as measured by MLC and CML assay. The chimeras responded normally to unrelated allogeneic cells. Chimeric animals generated a cytotoxic response to hapten-modified cells of both donor (CBA) and h

The chimera combining the peptide nucleic acids (PNAs) and peptides represent a promising bifunctional strategy by concurrently binding with protein catalytic pocket and its associated RNA template, effectively disrupting protein's function. Conventional designs face challenges due to non-optimized peptide-protein interactions and empirical PNA sequence selection, which lack thermodynamic or computational refinement, compromising selectivity and efficacy. Here, we present an artificial intelligence (AI) and molecular simulations-driven framework for the de novo design of a high-affinity PNA-pe

PTP1B, a promising target for insulin sensitizers in type 2 diabetes treatment, can be effectively degraded using proteolysis-targeting chimera (PROTAC). This approach offers potential for long-acting antidiabetic agents. We report potent bifunctional PROTACs targeting PTP1B through the E3 ubiquitin ligase cereblon. Western blot analysis showed significant PTP1B degradation by PROTACs at concentrations from 5 nM to 5 μM after 48 h. Evaluation of five highly potent PROTACs revealed compound 75 with a longer PEG linker (23 atoms), displaying remarkable degradation activity after 48 and 72 h, wit

Visceral leishmaniasis (VL) is a neglected tropical disease caused by protozoan parasites of the genus Leishmania. Systemic VL is fatal if untreated and there are no prophylactic human vaccines available. Several studies suggest that Th1 cell-mediated immunity plays a major role in protecting against VL. In this chapter we describe a method for designing recombinant chimera vaccines in silico based on the prediction of T cell epitopes within protein antigens identified as potential protective immunogens. Development of a recombinant chimera protein (RCP) vaccine using T cell epitope peptides i

The NaV1.7 voltage-gated sodium channel is implicated in human pain perception by genetics. Rare gain of function mutations in NaV1.7 lead to spontaneous pain in humans whereas loss of function mutations results in congenital insensitivity to pain. Hence, agents that specifically modulate the function of NaV1.7 have the potential to yield novel therapeutics to treat pain. The complexity of the channel and the challenges to generate recombinant cell lines with high NaV1.7 expression have led to a surrogate target strategy approach employing chimeras with the bacterial channel NaVAb. In this rep

The M2 channel of influenza A is a target of the adamantane family antiviral drugs. Two different drug-binding sites have been reported: one inside the pore, and the other is a lipid-facing pocket. A previous study showed that a chimera of M2 variants from influenza A and B that contains only the pore-binding site is sensitive to amantadine inhibition, suggesting that the primary site of inhibition is inside the pore. To obtain atomic details of channel-drug interaction, we determined the structures of the chimeric channel with and without rimantadine. Inside the channel and near the N-termina
