Baroreceptor-induced decrease in muscle blood flow upon propranolol administration.

The acute effects of propranolol, 1 mg/kg i.v., were studied in chloralosed, vagotomized cats. The vascularly isolated but innervated calf muscles were perfused from another animal. In one group of experiments, the carotid baroreceptors were exposed to ambient arterial pressure. Here, propranolol caused a fall in heart rate and an increase in resistance of the isolated muscle bed. In other experiments, the carotid sinuses were perfused at a constant pressure. In these animals, no increase in muscle flow resistance was observed after the drug. It is concluded that the increase in total peripher

[Changes in the central and peripheral circulation and in the acid-base equilibrium of the blood in dogs during a passive orthostatic test].

Integral changes in circulation and acid-base equilibrium of blood were studied on urethane-chloralose anesthesized dogs during a 20 min tilt test. Against the background of a relatively stable mean pressure in the aorta, the blood flow in the aorta and carotid artery diminished, pressure in the right atrium decreased and the heart rate increased. The phase of isometric contraction of the left ventricle increased, ejection time, mechanical systole and intrasystolic index decreased whereas the myocardial contractility increased. The blood flow in the liver, spleen and hind limb skin reduced and

Evidence for a non-classical splanchnic neural control of the feline pylorus.

Electrical stimulation of the divided splanchnic nerves proximal to the celiac ganglion in chloralosed cats induced a dilation of the pylorus in the majority of animals. However, in some cats a pure contractile motor response or a biphasic motor response (contraction followed by dilation) was elicited suggesting that the splanchnic nerves convey excitatory fibres to the pylorus as well. The pyloric and gastric contraction at such stimulation was sensitive to atropine, while the pyloric dilation was not sensitive to cholinergic, adrenergic or ganglionic blocking agents in contrast to the adrene

Interstitiospinal action on forelimb, hindlimb, and back motoneurons.

Responses of motoneurons supplying muscles of the forelimbs, hindlimbs, and back to stimulation of the interstitial nucleus of Cajal (INC) were recorded intracellularly in cats under chloralose anesthesia. Stimulation of the ipsilateral and contralateral INC evoked predominantly excitatory postsynaptic potentials in these motoneurons. Response latencies and properties of responses to multiple shock stimuli indicated that the responses were evoked by a di- or polysynaptic pathway. Stimulation of the anterior MLF (P2), which should have activated the entire interstitiospinal tract, but few retic

Hyperkalemia and cardiovascular collapse after verapamil and dantrolene administration in swine.

The cardiovascular and neuromuscular interactions of verapamil and dantrolene were evaluated in 20 chloralose-anesthetized swine. The animals were randomly divided into three groups. Group I, ten animals, received a bolus intravenous injection of 0.1 mg . kg-1 of verapamil followed by the continuous infusion of 5 micrograms . kg-1 . min-1. This group was then randomly divided into two equal subgroups. Five of these animals, Group Ia, continued to receive the verapamil infusion alone. The other five animals, Group Ib, received dantrolene in incremental doses of 1.0, 3.3, and 5.6 mg . kg-1 while

Halogenated Ether, Alcohol, and Alkane Anesthetics Activate TASK-3 Tandem Pore Potassium Channels Likely through a Common Mechanism.

The TWIK-related acid-sensitive potassium channel 3 (TASK-3; KCNK9) tandem pore potassium channel function is activated by halogenated anesthetics through binding at a putative anesthetic-binding cavity. To understand the pharmacologic requirements for TASK-3 activation, we studied the concentration-response of TASK-3 to several anesthetics (isoflurane, desflurane, sevoflurane, halothane, α-chloralose, 2,2,2-trichloroethanol [TCE], and chloral hydrate), to ethanol, and to a panel of halogenated methanes and alcohols. We used mutagenesis to probe the anesthetic-binding cavity as observed in a T

Association of anemia with reduced central respiratory drive in the piglet.

Central respiratory drive was studied in 13 piglets of both sexes varying in age from 19 to 67 days. The distal trachea was cannulated and the maximum rate of isometric inspiratory pressure change (dP/dt)(max), was measured at the airway. Curves were constructed relating this measurement to changes in arterial PCO(2) during carbon dioxide rebreathing. Data were obtained at intervals corresponding to stepwise reductions in central respiratory drive produced by added chloralose anaesthesia. Laryngeal reflex activation was achieved by electrical stimulation of the superior laryngeal nerves (SLN).

Electrical stimulation in restricted area of abdominal skin increased glucose level via the pancreatic branch of the vagal nerve in the rat.

Electrical stimulation (5 V, 50 Hz, 10 min.) of the middle upper abdominal area (Chuin acupuncture point) increased the blood sugar level in rats anesthetized with chloralose (60 mg/kg) and urethane (0.6 g/kg). Stimulation of both sides of middle abdominal skin area also increased the blood sugar level. However, electrical stimulation of other areas of the abdominal skin or the front paw, lower leg or thigh did not alter blood sugar level. Electrical stimulation of the middle upper abdomen inhibited spontaneous firing of the vagal nerve that innervates the pancreas, and enhanced neuronal disch

Plasma vasopressin response to haemorrhage in the anaesthetized rabbit.

In chloralose-urethane anaesthetized rabbits the acute circulatory and plasma vasopressin (pAVP) responses to moderate haemorrhage of 6 mL/kg body weight (10% blood volume) were followed after serial section of the aortic, vagus, and carotid sinus nerves. With all nerves intact, haemorrhage resulted in significant increases in pAVP, accompanied by decreases in systemic arterial pressure and right atrial pressure. With subsequent section of each afferent nerve, pAVP still increased in response to haemorrhage regardless of the order of nerve section. These results suggest that, in the anaestheti

Degeneration secretion of saliva in the rat following sympathectomy.

1. After previous extirpation of the superior cervical ganglion saliva was found to flow from the cannulated submaxillary duct of rats under chloralose anaesthesia. The secretion started 13-14.5 hr after sympathectomy, increased gradually in rate and then slowed down and ceased, lasting for about 12 hr.2. Injection of alpha- or beta-adrenoreceptor blocking drugs reduced the flow and a combination of both types of drugs abolished it. It is concluded that the secretion was caused by noradrenaline released from the degenerating nerves, and that the phenomenon is an example of the degeneration act

5-HT2 receptor agonists increase spontaneous sympathetic nerve discharge.

The selective 5-HT2 agonist 1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane (DOI) produced a marked increase in spontaneous sympathetic nerve discharge recorded from the inferior cardiac nerve in chloralose anesthetized cats. DOI (0.01-1.0 mg/kg i.v.) increased sympathetic nerve discharge to a maximum of 1750% of control values. The increase in sympathetic nerve discharge produced by DOI was reversed by the 5-HT2 antagonists ketanserin and LY 53857. In addition, pretreatment with ketanserin, but not prazosin, completely prevented the increase in sympathetic nerve discharge produced by DOI. These

A lack of potency for the delta-opioid antagonist naltrindole after microinjection into the rostral ventrolateral medulla of rabbits.

1. Naltrindole is a novel delta-opioid antagonist which is highly potent in vitro. We examined the effects on arterial pressure of naltrindole (0.3-300 pmol) after microinjection into the pressor area of the rostral ventrolateral medulla of chloralose-anaesthetized rabbits. 2. Naltrindole itself had no significant effects on arterial pressure, and only slightly attenuated the hypotensive effect of an exogenous agonist ( [D-Ala2, D-Leu5]-enkephalin). This is in contrast to previous demonstration of a marked pressor effect following another delta-antagonist, ICI 174,864. 3. Thus, naltrindole, te

Adrenergic receptor subtype activation by (+)-, (-)- and (+/-)-norephedrine in the pithed rat.

The ability of (+/-)-norephedrine (phenylpropanolamine) and its component isomers, (+)-and (-)-norephedrine, to activate adrenergic receptor subtypes in the cardiovascular system of the urethane/chloralose-anaesthetized pithed rat has been investigated. At all adrenoceptor subtypes, (-)-norephedrine was the most potent agonist followed by (+/-)- then (+)-norephedrine. The greatest activity was observed at the alpha 1-receptor, with little activity observed at either beta 1 or beta 2-adrenoceptors. Reserpinization shifted the (-)-norephedrine dose-response curve slightly to the right, indicatin

Cardiovascular effects of two potential beta-hydroxylase inhibitors.

The purpose of this study was to evaluate the cardiovascular effects of two potential beta-hydroxylase inhibitors, a dithiocarbamate and a thiourea derivative of d-amphetamine, 1 and 2, respectively. These compounds when given intravenously elicited a significant dose-dependent depressor effect in normotensive rats and dogs anesthetized with chloralose-urethane. The hypotensive effect of these compounds appears to be the result of a combination of two action components: (a) the major component, which can be blocked by atropine, exerted through the parasympathetic division and (b) a decreased a

Effect of phenytoin on cardiac slowing induced by cholinergic stimulation.

Phenytoin was tested for its ability to counteract cholinergic-induced slowing in sinus rate in chloralose-anesthetized cats subjected to bilateral cervical vagotomy and spinal cord transection. The cholinergic stimuli consisted of electrical stimulation of the cervical vagus nerves and injected acetylcholine. Phenytoin administered as a continuous infusion of 0.5 and 1.0 mg/kg/min was found to significantly reduce the cardiac slowing produced by both modes of cholinergic stimulation. Blockade of the acetylcholine response suggested that the inhibitory effect of phenytoin was on the postsynapt

General anesthetics hyperpolarize neurons in the vertebrate central nervous system.

The effect of general anesthetics on frog motoneurons and rat hippocampus pyramidal cells was examined with sucrose gap and intracellular recording, respectively. A number of volatile and intravenous anesthetics directly hyperpolarized the motoneurons. The potency of these agents in hyperpolarizing motoneurons was strongly correlated with their anesthetic potency. While the responses to barbiturates and alpha-chloralose were blocked by gamma-aminobutyric acid antagonists and were dependent on the chloride gradient, the responses to all the other anesthetics tested were generated by a separate
