Further investigation of the stimulus properties of chlordiazepoxide and zolpidem. Agonism and antagonism by two novel benzodiazepines.

The effects of Ro 16-6028 and Ro 17-1812, two novel benzodiazepines with mixed agonist and antagonist properties, were studied in rats trained to discriminate either chlordiazepoxide or the benzodiazepine receptor ligand zolpidem. In rats discriminating 5 mg/kg chlordiazepoxide from saline both Ro 16-6028 and Ro 17-1812 produced responding on the drug lever. At a dose of 10 mg/kg both compounds gave rise to 100% responding on the lever associated with chlordiazepoxide. The dose-response curve produced by Ro 16-6028 was flatter than that for Ro 17-1812, however. The discriminative cue produced

Fear is not critical to classically conditioned analgesia: the effects of periaqueductal gray lesions and administration of chlordiazepoxide.

A high correlation between fear and analgesia classically conditioned to footshock in rats has been reported in the literature. However, it has never been directly tested whether or not fear is in fact causal to the production of conditioned analgesia. We therefore tested whether conditioned analgesia could be elicited in the absence of fear by employing two independent methods of fear suppression. First, areas of the periaqueductal gray (PAG) previously implicated in fear were selectively lesioned. Lesions of the dorsolateral PAG significantly attenuated conditioned analgesia and markedly dec

Effects of chlordiazepoxide on food anticipation, drinking and other behaviors in food-deprived and satiated rats.

Two groups of rats, Deprived and Satiated, were presented with food according to a fixed time 60-sec schedule. They were then injected with saline, 5, 10, and 15 mg/kg of chlordiazepoxide hydrochloride according to a Latin square design. During saline administration time spent visiting the food tray, time spent drinking, number of tray entries and the amount of water ingested were always greater in the Deprived than in the Satiated group; whereas the opposite was true for grooming. As chlordiazepoxide dose increased time spent visiting the food tray increased in both groups, but the effect was

Chlordiazepoxide-induced ataxia, muscle relaxation and sedation in the rat: effects of muscimol, picrotoxin and naloxone.

Chlordiazepoxide-induced ataxia was potentiated by muscimol (0.5 mg/kg) and antagonised by picrotoxin (2 and 4 mg/kg) and by naloxone (2 and 4 mg/kg). The muscle-relaxant effects of chlordiazepoxide were not significantly altered by any of these three drugs and nor was the chlordiazepoxide-induced decrease in exploratory head-dipping. However, the sedative effects of chlordiazepoxide, measured by a reduction in spontaneous motor activity, were antagonised by picrotoxin.

Variable-interval schedules of timeout from avoidance: effects of chlordiazepoxide, CGS 8216, morphine, and naltrexone.

Rats were trained on concurrent schedules in which pressing one lever postponed shock and pressing the other occasionally (variable-interval schedule) produced a 2-min timeout during which the shock-postponement schedule was suspended and its correlated stimuli were removed. These procedures provided a baseline for studying the effects of drugs on behavior maintained by different sources of negative reinforcement (shock avoidance and timeout from avoidance). Experiment 1 studied a benzodiazepine agonist, chlordiazepoxide, and antagonist, CGS 8216. Chlordiazepoxide (2.5-30 mg/kg) had little eff

Should chlordiazepoxide and diazepam be avoided when treating alcohol withdrawal syndrome in patients with hepatic insufficiency?

INTRODUCTION: Chlordiazepoxide and diazepam are first-line treatments for patients with alcohol withdrawal. However, their use is discouraged in hepatic insufficiency due to concerns that impaired metabolism may prolong their half-lives and those of their active metabolites, increasing the risk of prolonged sedation to an unacceptable level. THE ACTIVITY OF CHLORDIAZEPOXIDE AND ITS METABOLITES: Findings presented here suggest that chlordiazepoxide has minimal sedative activity-its effect is primarily dependent on its metabolites. Chlordiazepoxide metabolism occurs through hepatic oxidation, s

Simultaneous determination of imipramine hydrochloride and chlordiazepoxide in pharmaceutical preparations by spectrophotometric, RP-HPLC, and HPTLC methods.

A binary mixture of imipramine HCl and chlordiazepoxide was determined by three different methods. The first involved determination of imipramine HCl and chlordiazepoxide using the first derivative spectrophotometric technique at 219 and 231.5 nm over the concentration ranges of 1-20 and 2-24 microg/mL with mean accuracies of 99.47 +/- 0.78 and 101.43 +/- 1.20%, respectively. The second method utilized RP-HPLC with methanol-acetonitrile-0.065 M ammonium acetate buffer (45 + 25 + 30, v/v/v, pH adjusted to 5.6 +/- 0.02 with phosphoric acid) as the mobile phase pumped at a flow rate of 1.0 mL/min

Termination of pseudopregnancy in the rat alters the response to progesterone, chlordiazepoxide, and MK-801 in the elevated plus-maze.

RATIONALE: Allopregnanolone, a neurosteroid-reduced metabolite of progesterone, is a well-documented positive modulator of the gamma-aminobutyric type A (GABA(A)) receptor. As has been reported for other positive modulators of the GABA(A) receptor, chronic exposure to neurosteroids is hypothesized to decrease GABA(A) receptor function. Drawing from the literature on chronic exposure to benzodiazepines or alcohol, putative changes in N-methyl-D-aspartate (NMDA) receptor function are also expected after chronic neurosteroid exposure. OBJECTIVES: To assess the sensitivity of the GABA(A) and NMDA

Prior test experience compromises the anxiolytic efficacy of chlordiazepoxide in the mouse light/dark exploration test.

It is now well established that prior test experience can alter behavioural baselines and attenuate/abolish the anxiolytic efficacy of benzodiazepines in the elevated plus-maze paradigm. In view of evidence that different models of anxiety measure qualitatively distinct forms of anxiety-like behaviour, it is important to establish whether the effects of prior experience extend to other widely-used tests. The present study assessed the behavioural and pharmacological sequelae of a single undrugged prior exposure to the light/dark exploration (L/D) test in mice, using ethological scoring methods

Evidence that the anxiolytic-like effects of chlordiazepoxide on the elevated plus maze are confounded by increases in locomotor activity.

In exploratory animal models of anxiety, such as the elevated plus maze, the anxiogenic- and anxiolytic-like effects of drugs may be confounded by changes in locomotor activity. In the present experiments, the sensitivity of several measures of anxiety and locomotor activity in the elevated plus maze were assessed. Both chlordiazepoxide hydrochloride (CDP, 7.5 mg/kg) and d-amphetamine sulphate (AMP, 0.75, 1.5 mg/kg) increased the percent time on the open arms and doses of 7.5 mg/kg and 1.5 mg/kg CDP and AMP, respectively, increased the number of entries into the open arms. Effects of intracranial infusions of chlordiazepoxide on spatial learning in the Morris water maze. II. Neuropharmacological specificity.

In the preceding paper it was found that infusions of chlordiazepoxide (CDP) into the medial septal region, but not several other regions possessing a high density of benzodiazepine receptors, impaired spatial learning, but not cue learning or swim speed, in the Morris water maze. The present investigation sought to further characterize the neuropharmacological profile of this effect. Initially, it was reconfirmed that systemically administered CDP impaired spatial learning, but not cue learning or swim speed, in the water maze. Additionally, it was found that systemically administered scopola

Double blind study on the efficacy and safety of tetrabamate and chlordiazepoxide in the treatment of the acute alcohol withdrawal syndrome.

1. Efficacy and safety of tetrabamate and chlordiazepoxide in the treatment of the acute or Primary Alcohol Withdrawal Syndrome (PAWS) were assessed during a randomized double blind clinical trial, carried out on sixty male alcoholic in-patients. 2. The two drugs were administered four times a day in double dummy conditions, according to a fixed-flexible decreasing dosage schedule (six days basic regimen). 3. Drug efficacy was measured daily throughout the study period using a battery of standard instruments for collecting quantitative clinical, behavioral, psychopathological and laboratory da

The effects of chlordiazepoxide on synaptic transmission and amino acid neurotransmitter release in slices of rat olfactory cortex.

The rat olfactory cortex slice has been used to investigate the effects of chlordiazepoxide on evoked field potentials and the release of endogenous amino acid neurotransmitters (aspartate, glutamate, GABA and possibly taurine) which accompany electrical stimulation of the lateral olfactory tract. When single, low frequency stimuli were employed, chlordiazepoxide (2 microM-1 mM) depressed the amplitude of the field potential correlate of the depolarizing actions of the lateral olfactory tract excitatory transmitter (aspartate?) although aspartate release was unaffected. The field potential cor

Chlordiazepoxide against signalling, receptor and regulatory proteins of breast cancer: a structure-based in-silico approach.

Among the most prevalent forms of cancer are breast, lung, colon-rectum, and prostate cancers, and breast cancer is a major global health challenge, contributing to 2.26 million cases with approximately 685,000 deaths worldwide in 2020 alone, typically beginning in the milk ducts or lobules that produce and transport milk during lactation and it is becoming challenging to treat as the tissues are developing resistance, which makes urgent calls for new multitargeted drugs. The multitargeted drug design provides a better solution, simultaneously targeting multiple pathways, even when the drug re

GABAergic drugs can enhance or attenuate chlordiazepoxide-induced sleep time in a heterogeneous strain of mice.

Evidence supports the notion that differences between the Long-Sleep and Short-Sleep selectively-bred lines of mice are attributable to differences in brain excitability and that these differences are mediated by activity of the GABAergic system. The general applicability of this hypothesis to other populations of mice was tested by using an outbred strain of mice. Specifically, a heterogeneous strain of mice was administered several doses of the hypnotic chlordiazepoxide. Additionally, the indirect GABA agonist AOAA, and the GABA antagonists bicuculline, picrotoxin and pentylenetetrazol were

Anxiety- and activity-related effects of diazepam and chlordiazepoxide in the rat light/dark and dark/light tests.

We have investigated, through factor analysis, anxiety- and activity-related variables in rats placed in the light/dark box. Thus, vehicle-, diazepam (DZ)-, and chlordiazepoxide (CDP)-treated rats were submitted 30 min later to 5-min light/dark or dark/light tests (initial placements in light or dark, respectively). Following this test, the animals were tested for 5 min in an automated activity monitor. Doses of DZ (0.75-3.0 mg/kg) and CDP (2.5-10.0 mg/kg) were based on preliminary evidence for 1.5 mg/kg of DZ and 5 mg/kg of CDP being anxiolytic in the elevated plus-maze. In the light/dark tes

Effects of intracranial infusions of chlordiazepoxide on spatial learning in the Morris water maze. I. Neuroanatomical specificity.

The present investigation sought to determine the neuroanatomical locus through which the amnesic and anxiolytic effects of the benzodiazepine agonist chlordiazepoxide are mediated. Rats were infused with either chlordiazepoxide (60 nmol/microliters) or artificial CSF (1 microliter) into either the frontal cortex, nucleus basalis magnocellularis/substantia innominata, amygdala, medial septum, hippocampus, or cerebellum and run in the open field to assess anxiety as thigmotaxia and in the Morris water maze to assess spatial learning.
