Drug interactions with ethanol. Effects on body temperature and motor impairment.

Single doses of d-amphetamine, chlorpheniramine or diazepam were combined with ethanol under two conditions: (i) in drug-naive mice and (ii) in mice which had been given a single dose of ethanol 72 hr previously. Ethanol was administered orally at doses of 6.0, 3.0 or 1.5 g/kg; doses of d-amphetamine, chlorpheniramine or diazepam were given intraperitoneally. Three parameters were measured; changes in rectal temperature, forced motor coordination, as evaluated by rotarod performance and concentrations of ethanol in blood. d-Amphetamine and chlorpheniramine attenuated the hypothermia induced by

Effect of physical and chemical properties on drug release from selected thermosoftening vehicles.

The release profile of several drugs, (chlorpheniramine maleate, salicylic acid, hydrochlorothiazide, p-hydroxy benzoic acid, sulphafurazole, anhydrous theophylline) and the marker (D&C yellow No. 10) was detailed to determine the effect of physical and chemical properties on release from selected thermosoftening matrices (Gelucire 50/02 and 50/13). At a concentration of drug or marker of 2.5% w/w, hydrochlorothiazide showed the slowest release from G50/02, due to its low aqueous solubility, while theophylline showed the highest release owing to its low mol. wt and moderate aqueous solubility.

Separation of cold medicine ingredients by capillary electrophoresis.

This study demonstrates the separation of cold medicine ingredients (e.g., phenylpropanolamine, dextromethorphan, chlorpheniramine maleate, and paracetamol) by capillary zone electrophoresis and micellar electrokinetic chromatography. Factors affecting their separations were the buffer pH and the concentrations of buffer, surfactant and organic modifiers. Optimum results were obtained with a 10 mM sodium dihydrogen-phosphate-sodium tetraborate buffer containing 50 mM sodium dodecyl sulfate (SDS) and 5% methanol (MeOH), pH 9.0. The carrier electrolyte gave a baseline separation of phenylpropano

The discriminative stimulus properties of mepyramine.

Rats were trained to discriminate i.p. injections of mepyramine (10.0 mg/kg) from saline. Correct responses on one of the two levers were reinforced with access to a solution of sweetened condensed milk. A level of at least 27 correct responses in the first 30 (90%) was required in consecutive saline and mepyramine training sessions before a test session was conducted. Amongst the antihistamines, mepyramine, tripelennamine and chlorpheniramine all produced dose-related responding on the mepyramine lever, reaching a maximum of over 90% mepyramine-appropriate responses. Both tripelennamine and c

Determination of amine ingredients in cough-cold liquids by reversed-phase ion-pair high-performance liquid chromatography.

The chromatographic behavior of phenylephrine, codeine, pseudoephedrine, phenylpropanolamine, methoxyphenamine, pheniramine, pyrilamine, dextromethorphan, and chlorpheniramine was examined by reversed-phase, ion-pair high-performance liquid chromatography. An isocratic chromatographic system was devised for the analysis of cough-cold liquids containing these amine drugs by optimization of the mobile phase ionic strength, buffer pH, pairing ion concentration, and secondary ion concentration. Quantitative recovery and excellent precision were demonstrated for the simultaneous determination of ph

Increased levels of extracellular dopamine in neostriatum and nucleus accumbens after histamine H1 receptor blockade.

The dopaminergic system plays a central role in the processing of reward or reinforcement since drugs that have reinforcing properties all share the ability to elevate dopamine (DA) levels in the nucleus accumbens or neostriatum. Histamine H1 receptor antagonists are known to have reinforcing effects in humans and laboratory rats. Here, we examined the effect of systemic (i.p.) treatment with two H1 antagonists, chlorpheniramine and pyrilamine, on the extracellular levels of DA and its metabolites dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) in the neostriatum and nucleus acc

Transepithelial transport of diphenhydramine across monolayers of the human intestinal epithelial cell line Caco-2.

PURPOSE: The transepithelial transport characteristics of the antihistamine, diphenhydramine, were studied in human intestinal Caco-2 cell monolayers to elucidate the mechanisms of its intestinal absorption. METHODS: The transepithelial transport and the cellular accumulation of diphenhydramine were measured using Caco-2 cell monolayers grown in Transwell chambers. RESULTS: The transepithelial transport of diphenhydramine from the apical to basolateral side was saturable, and the flux and cellular accumulation of diphenhydramine were dependent on the apical extracellular pH (pH 7.4 > 6.5 > 5

Effect of flurbiprofen, a cyclooxygenase inhibiting drug, on induced allergic rhinitis.

To test whether prostaglandins contribute to the nasal allergic reaction, we subjected allergic rhinitis patients to nasal allergen challenge after treatment with flurbiprofen, a cyclooxygenase inhibitor. We challenged 18 patients after two pretreatment doses of flurbiprofen, 75 mg, chlorpheniramine, 6 mg, both drugs combined, and placebo. All patients received all treatments. None of the treatments affected measured nasal airway resistance. Nasal secretion weight, number of sneezes, and overall subjective severity scores for all active treatments differed significantly from placebo treatment.

Successful treatment of chronic idiopathic urticaria and angioedema with cimetidine alone.

We have studied a 50-year-old white man with chronic urticaria and angioedema who has responded to treatment with cimetidine alone for over 2 yr. In a double-blind, placebo-controlled study, cimetidine alone was at least as effective as chlorpheniramine in relief of urticaria and angioedema. Additionally, cimetidine significantly inhibited (p less than 0.01) the wheal response to histamine when it was compared to placebo. The inhibition of wheal response to histamine by cimetidine was significantly higher (p less than 0.05) than chlorpheniramine. The presence of predominantly H2- rather than H

Anticholinergic toxidrome in a toddler with unwitnessed ingestion: diagnostic clues pointing to over-the-counter medication overdose.

We report a case of a previously healthy 3-year-old girl who developed acute altered consciousness, agitation, and visual hallucinations consistent with an anticholinergic toxidrome. Initial examinations were unremarkable, and urine immunoassay screening (SIGNIFY™ ER) was negative. Serum analysis by liquid chromatography-quadrupole time-of-flight mass spectrometry identified chlorpheniramine and scopolamine, while excluding other agents. Concentrations were 370 ng/mL for chlorpheniramine and 1 ng/mL for scopolamine, both near reported toxic ranges. An empty blister pack of 22 pediatric motion

Acute anticholinergic action in focal dystonia.

We have investigated the effects of the intravenous administration of three anticholinergic drugs (atropine, benztropine, and chlorpheniramine) with variable antihistaminic properties in comparison with saline in adult-onset focal dystonias. In many patients, the severity of the involuntary movements varied from one observation to the next. Patients with spasmodic torticollis, writer's cramp, and segmental dystonia showed little change with any of the active drugs. The occasional individual who improved by more than 20% did so at the expense of drug-induced drowsiness. We conclude that choline

Characterization of daytime sleepiness and psychomotor performance following H1 receptor antagonists.

BACKGROUND: While first generation H1-receptor antagonists are widely used, there are relatively few data describing their comparative effects on subjective daytime sleepiness and psychomotor performance. OBJECTIVE: To compare the effects of first generation H1 receptor antagonists on subjective daytime sleepiness and psychomotor performance. METHODS: We conducted two single-dose, cross-over studies. In the first, we validated our methodology in 18 healthy subjects by examining the response to diphenhydramine (50 mg), terfenadine (60 mg), and placebo. In the second trial, we evaluated the re

Complementary use of partial least-squares and artificial neural networks for the non-linear spectrophotometric analysis of pharmaceutical samples.

The complementary use of partial least-squares (PLS) multivariate calibration and artificial neural networks (ANNs) for the simultaneous spectrophotometric determination of three active components in a pharmaceutical formulation has been explored. The presence of non-linearities caused by chemical interactions was confirmed by a recently discussed methodology based on Mallows augmented partial residual plots. Ternary mixtures of chlorpheniramine, naphazoline and dexamethasone in a matrix of excipients have been resolved by using PLS for the two major analytes (chlorpheniramine and naphazoline)
