Efficacy and Safety of Non-brain Penetrating H1-Antihistamines for the Treatment of Allergic Diseases.

H1 receptor antagonists, known as H1-antihistamines (AHs), inactivate the histamine H1-receptor thereby preventing histamine causing the primary symptoms of allergic diseases, such as atopic dermatitis, pollinosis, food allergies, and urticaria. AHs, which are classified into first-generation (fgAHs) and second-generation (sgAHs) antihistamines, are the first line of treatment for allergic diseases. Although fgAHs are effective, they cause adverse reactions such as potent sedating effects, including drowsiness, lassitude, and cognitive impairment; anticholinergic effects, including thirst and

Receptor-mediated release of inositol phosphates in the cochlear and vestibular sensory epithelia of the rat.

Various neurotransmitters, hormones and other modulators involved in intercellular communication exert their biological action at receptors coupled to phospholipase C (PLC). This enzyme catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PtdInsP2) to inositol 1,4,5-trisphosphate (InsP3) and 1,2-diacylglycerol (DG) which act as second messengers. In the organ of Corti of the guinea pig, the InsP3 second messenger system is linked to muscarinic cholinergic and P2y purinergic receptors. However, nothing is known about the InsP3 second messenger system in the vestibule. In this stud

Nicotine effects in mouse hippocampus are blocked by mecamylamine, but not other nicotinic antagonists.

Previous data indicated that bath-application of nicotine to mouse hippocampal slices resulted in a concentration-dependent increase in the amplitude of the orthodromic population spike and the appearance of multiple population spikes in the CA1 pyramidal cell layer. d-Tubocurarine (4-100 microM), alpha-bungarotoxin (10-160 microM), and atropine (40-200 microM) had similar effects, although for alpha-bungarotoxin these excitatory effects were transient. Mecamylamine (1.6-3.2 mM) inhibited the population spike, while hexamethonium (3.2 mM) had no effect. These cholinergic antagonists were teste

Pinnatoxins' Deleterious Effects on Cholinergic Networks: From Experimental Models to Human Health.

Pinnatoxins (PnTXs) are emerging neurotoxins that were discovered about 30 years ago. They are solely produced by the marine dinoflagellate Vulcanodinium rugosum, and may be transferred into the food chain, as they have been found in various marine invertebrates, including bivalves. No human intoxication has been reported to date although acute toxicity was induced by PnTxs in rodents. LD50 values have been estimated for the different PnTXs through the oral route. At sublethal doses, all symptoms are reversible, and no neurological sequelae are visible. These symptoms are consistent with impai

Loss of endogenous analgesia leads to delayed recovery from incisional pain in a rat model of chronic neuropathic pain.

BACKGROUND: Preoperative pain and impaired endogenous analgesia are risk factors of chronic postsurgical persistent pain (CPSP). A Chronic neuropathic pain model induced by spinal nerve ligation (SNL6W) shows impaired endogenous analgesia and delayed recovery from incisional pain. Repeated amitriptyline treatment can restore the endogenous analgesia, but its effects on delayed recovery are not clear. METHODS: A plantar incision was made on the side contralateral to the nerve ligation in SNL6W rats. Withdrawal thresholds were measured by von Frey filament test until 28 d after surgery. Amitrip

Pharmacological evidence of a cholinergic contribution to elevated impulsivity and risky decision-making caused by adding win-paired cues to a rat gambling task.

BACKGROUND: Pairing rewards with sensory stimulation, in the form of auditory and visual cues, increases risky decision-making in both rats and humans. Understanding the neurobiological basis of this effect could help explain why electronic gambling machines are so addictive, and inform treatment development for compulsive gambling and gaming. Numerous studies implicate the dopamine system in mediating the motivational influence of reward-paired cues; recent data suggest the cholinergic system also plays a critical role. Previous work also indicates that cholinergic drugs alter decision-making

Physiological role of alpha-adrenoceptors in salivary secretion.

The submaxillary gland (SM) of rat is innervated by both branches of the autonomic nervous system. Secretion is mediated by the activation of both muscarinic-cholinergic and alpha/beta adrenergic receptors. Studies of the relative affinity of pharmacological agonists and antagonists have warranted a subclassification of alpha adrenoceptors into types alpha 1 and alpha 2. Our studies involve an analysis of the physiologic role of both types of alpha adrenoceptors in salivary secretion. Dose response curves (DRC) to noradrenaline (NA) following administration of alpha adrenoceptor antagonists, i

Biochemical indentification of the mammalian muscarinic cholinergic receptor.

The muscarinic cholinergic antagonist 3-quinuclidinyl benzilate (QNB) binds avidly but reversibly to the muscarinic cholinergic recptor of mammalian brain and peripheral tissues. [3H]QNB binding provides a simple, sensitive, specific assay for the muscarinic cholinergic receptor binding. Inhibition of [3H]QNB binding to homogenates of brain and guinea pig ileum by muscarinic drugs correlates with their pharmacologic potencies, while nicotinic agents and noncholinergic drugs have neglibible affinity. The regional distribution of [3H]QNB binding throughout rat and monkey brain parallels to a maj

Histamine and related substances influence neurotransmission in the semicircular canal.

Histamine and other imidazole-containing substances were found to increase ampullar nerve afferent firing rate while both H1 and H2 histamine antagonists effectively inhibited ampullar nerve activity. A specific inhibitor of histidine decarboxylase, the enzyme which catalyses the synthesis of histamine, reduced ampullar nerve firing in a dose-dependent manner. These observations suggest a physiological role for histamine in the inner ear. Maintenance of a response to histamine after de-efferentation of the crista ampullaris supports the hypothesis that the site of action is the hair cell; anta

A role for vagal activity in preventing the suppression of glucagon secretion by GLP-1 during hypoglycemia.

Glucagon-like peptide 1 (GLP-1) is generally safe against hypoglycemia, although it stimulates insulin and inhibits glucagon secretion. One explanation is that glucagon secretion is not inhibited by GLP-1 during hypoglycemia. We aimed at understanding the lack of suppression of glucagon secretion by GLP-1 by exploring the paracrine and neural regulation of pancreatic hormone secretion during hypoglycemia. Isolated rat pancreas (A) and an organ block comprising pancreas and stomach (B) were perfused. We performed 1) dose-response studies with GLP-1 (7-36) at hypoglycemia; 2) studies with GLP-1

The effects of cholinergic drugs and cholinergic-rich foetal neural transplants on alcohol-induced deficits in radial maze performance in rats.

Chronic alcohol (20% v/v in drinking water for 28 weeks) impaired acquisition of radial maze spatial and associative tasks by increasing both within-trial working and long-term reference memory errors; animals with high (above the median of 100 mg/100 ml) blood alcohol concentrations (BACs) during treatment were significantly more impaired than those with BACs below the median. Alcohol-treated rats showed improvements in radial maze performance after treatment with cholinergic agonists (arecoline and nicotine) and disruption with antagonists (scopolamine and mecamylamine) at low doses which di

Differential response to cholinergic stimulation in psychogenitically selected rat lines.

Male and female rats of two lines psychogenetically selected for bipolar extremes in shuttle box avoidance were evaluated for tremor, salivation, chromodacryoorhea, and hypothermia following treatment with the muscarinic cholinergic agonist oxotremorine. Roman Low-Avoidance (RLA/Verh) rats exhibited more pronounced oxotremorine-induced tremor, chromodacryorrhea, and hypothermia than Roman High-Avoidance (RHA/Verh) rats. There was a sex difference only for a chromodacryorrhea response, with females exhibiting a greater response following oxotremorine than males. In a subsequent experiment using

Neuropharmacological profile of Meige's disease: overview and a case report.

A patient with Meige's disease was given several pharmacological trials, including muscle relaxants, dopaminergic antagonists, monoamine depletors, anticholinergics, dopamine agonists, cholinergics, benzodiazepines, tricyclic antidepressants, and lithium. Improvement of dystonic movements was best obtained by a combined treatment, haloperidol-benztropine. A review of the literature on Meige's disease is presented. The hypothesis of a simultaneous dopaminergic and cholinergic overactivity is discussed and the use of physostigmine for diagnostic purposes is suggested.

Selective Induction of Human Autonomic Neurons Enables Precise Control of Cardiomyocyte Beating.

The autonomic nervous system (ANS) regulates tissue homeostasis and remodelling through antagonistic effects of noradrenergic sympathetic and cholinergic parasympathetic signalling. Despite numerous reports on the induction of sympathetic neurons from human pluripotent stem cells (hPSCs), no induction methods have effectively derived cholinergic parasympathetic neurons from hPSCs. Considering the antagonistic effects of noradrenergic and cholinergic inputs on target organs, both sympathetic and parasympathetic neurons are expected to be induced. This study aimed to develop a stepwise chemical

Anticholinergic drug atropine diminishes newly formed fear memory in male rats.

Post-traumatic stress disorder (PTSD) is a condition which is triggered shortly after experiencing traumatic events. PTSD is complicated by the fact that people with PTSD often develop additional disorders such as phobias, addiction, depression, panic disorder and obsessive-compulsive disorder. Beta-adrenergic and cholinergic system both are involved in memory formation as well as in emotional response associated with memory. It is reported that the administration of beta-adrenergic and cholinergic antagonist results in the impairment in memory formation. Here, we examined the potential of bet

A role for histamine in cardiovascular regulation in late stage embryos of the red-footed tortoise, Chelonoidis carbonaria Spix, 1824.

A chorioallantoic membrane artery in embryos of the red-footed tortoise, Chelonoidis carbonaria was occlusively cannulated for measurement of blood pressure and injection of drugs. Two age groups of embryos in the final 10 % of incubation were categorized by the ratio of embryonic body to yolk mass. All embryos first received cholinergic and β-adrenergic blockade. This revealed that β-adrenergic control was established in both groups whereas cholinergic control was only established in the older group immediately prior to hatching. The study then progressed as two series. Series one was conduct

Opiate peptides control growth hormone through a cholinergic mechanism in man.

Opiate peptides are known to elevate circulating GH in man, and data in other animals suggest that acetylcholine may be involved. We have administered the long-acting met-enkephalin analogue DAMME to 6 normal males, and have shown that the resulting elevation in GH is blocked by the specific cholinergic antagonist, pirenzepine. It seems likely that a cholinergic mechanism may interact with opiate-induced changes in GH in man.

Evidence for the coupling of muscarinic M3 receptor to cyclic AMP formation and poly-phosphatidylinositol turnover in rat salivary glands.

The biochemical events which are coupled to the subtypes of muscarinic cholinergic receptors in rat salivary glands were studied. The subtype property of this receptor system was characterized by the use of 3H-QNB binding to glandular membrane homogenates. The Kd and Bmax values of this binding were found to be 0.2 nM and 210 fmole/mg protein respectively. In the drug-displacement study on the 3H-QNB binding the following potency order was obtained (based on the IC50 values calculated from each individual dose-response curve): Atropine greater than 4-DAMP greater than HHSiD greater than Pirenz
