Anopheles culicifacies Y-chromosome dimorphism indicates sibling species (B and E) with different malaria vector potential in Sri Lanka.

In Sri Lanka, malaria is transmitted mainly by Anopheles culicifacies Giles sensu lato (Diptera: Culicidae). In India, this nominal taxon comprises sibling species A, B, C, D and E, distinguished by their chromosome morphology. Species B (identified by polytene chromosome sequence Xab, 2g1 + h1) is not such an efficient vector of malaria as other members of the An. culicifacies complex in India. All specimens of An. culicifacies s.l. examined from Sri Lanka possess Xab, 2g1 + h1 polytenes, previously interpreted as species B, despite their important vector status. Recently, species E was descr

Genome assembly of Thaumatotibia leucotreta, a major polyphagous pest of agriculture in sub-Saharan Africa.

The false codling moth (FCM; Thaumatotibia leucotreta, Meyrick; Lepidoptera: Tortricidae) is a highly polyphagous, major agricultural pest indigenous to sub-Saharan Africa. With growing international trade, there is an increasing concern about introducing this pest into other countries. In South Africa, FCM poses a risk to multiple crops, and is currently suppressed through a combination of chemical, microbial, cultural, augmentative biological control, and the sterile insect technique. Compared with other lepidopteran agricultural pests, such as codling moth Cydia pomonella, genetic and other

Changes in cytoforms of Simulium damnosum sensu lato (Diptera: Simuliidae) and onchocerciasis transmission zones in northern Cameroon with possible implications for onchocerciasis transmission elimination.

Cytoforms (cytospecies and cytotypes) of the Simulium damnosum complex are the major onchocerciasis vectors in Africa; but they differ in vectorial efficiency. Transmission elimination of onchocerciasis requires understanding the vector dynamics and delineation of transmission zones, for monitoring and evaluation, and for stop-treatment decisions. This study sought to understand the vector population dynamics and to delineate Operational Transmission Zones in the 3 administrative regions (Far North, North, and Adamawa) of northern Cameroon. Using precontrol and relief maps, larvae were sampled

Identification of the sex chromosome system in a sand fly species, Lutzomyia longipalpis s.l.

In many animal species, sex determination is accomplished by heterogamety i.e., one of the sexes produces two types of gametes, which upon fertilization will direct the development toward males or females. Both male ("XY") and female ("ZW") heterogamety are known to occur and can be easily distinguished when the sex-chromosomes are morphologically different. However, this approach fails in cases of homomorphic sex chromosomes, such as the sand fly Lutzomyia longipalpis s.l. (Psychodidae, Diptera), which is the main vector of visceral leishmaniosis in Brazil. In order to identify the heterogame

Decoding cis-regulatory elements in the germline of the human malaria vector Anopheles gambiae.

Cis-regulatory elements (CREs) drive tissue- and cell-specific gene expression and are essential for safe, sustainable genetic control strategies in pest and vector insects, including the engineering of gene drives in the primary human-malaria vector Anopheles gambiae. Yet CREs remain poorly defined in mosquitoes due to limited computational tools and practical methods for identification and validation. We present a systematic in silico approach for CRE discovery, correlating targeted DNA-motif searches with gene expression, followed by frequency and distribution analysis within putative promo

Virus-like Particles from Wolbachia-Infected Cells May Include a Gene Transfer Agent.

Wolbachia are obligate intracellular bacteria that occur in insects and filarial worms. Strains that infect insects have genomes that encode mobile genetic elements, including diverse lambda-like prophages called Phage WO. Phage WO packages an approximately 65 kb viral genome that includes a unique eukaryotic association module, or EAM, that encodes unusually large proteins thought to mediate interactions between the bacterium, its virus, and the eukaryotic host cell. The Wolbachia supergroup B strain, wStri from the planthopper Laodelphax striatellus, produces phage-like particles that can be

Transcriptome-wide analysis of N6-methyladenosine uncovers its regulatory role in gene expression in the lepidopteran Bombyx mori.

N6-methyladenosine (m6A) methylation is the most common form of RNA modification in eukaryotes and has been shown to act as an important epitranscriptomic marker, which can regulate gene expression, messenger RNA stability, alternative splicing and translation efficiency. Recent progress on determining insect m6A function has been limited to the dipteran Drosophila melanogaster, in which m6A is involved in neuronal functions and sex determination. Methylation and function of m6A in other insects, however, remain unknown. Here, we investigated a transcriptome-wide profile of m6A in the lepidopt

Silencing subtelomeric VSGs by Trypanosoma brucei RAP1 at the insect stage involves chromatin structure changes.

Trypanosoma brucei causes human African trypanosomiasis and regularly switches its major surface antigen variant surface glycoprotein (VSG) to evade mammalian host immune responses at the bloodstream form (BF) stage. Monoallelic expression of BF Expression Site (BES)-linked VSGs and silencing of metacyclic VSGs (mVSGs) in BF cells are essential for antigenic variation, whereas silencing of both BES-linked and mVSGs in the procyclic form (PF) cells is important for cell survival in the midgut of its insect vector. We have previously shown that silencing BES-linked VSGs in BF cells depends on Tb

Sex-specific differences in meiotic chromosome segregation revealed by dicentric bridge resolution in mice.

The meiotic properties of paracentric inversion heterozygotes have been well studied in insects and plants, but not in mammalian species. In essence, a single meiotic recombination event within the inverted region results in the formation of a dicentric chromatid, which usually breaks or is stretched between the two daughter nuclei during the first meiotic anaphase. Here, we provide evidence that this is not the predominant mode of exchange resolution in female mice. In sharp contrast to previous observations in other organisms, we find that attempts to segregate the dicentric chromatid freque

A newly identified Y chromosome gene obp-Y is required for sperm storage in female Zeugodacus tau (Diptera: Tephritidae).

In the organisms with XX/XY sex chromosomes, Y chromosome is unique to males and plays an important role in male reproductive development. The study of Y chromosome genes will contribute to the development of pest genetic prevention and control technology. In this study, we identified 9 Y chromosome genes in Zeugodacus tau (Diptera: Tephritidae), including gene 16222. Protein structure analysis showed that 16222 was highly similar to odorant binding protein, and thus gene 16222 was named obp-Y. Obp-Y knockout (KO) significantly reduced hatching rate of offspring. Sperm detection results showed

Dichotomy of Dosage Compensation along the Neo Z Chromosome of the Monarch Butterfly.

Mechanisms of sex chromosome dosage compensation (SCDC) differ strikingly among animals. In Drosophila flies, chromosome-wide transcription is doubled from the single X chromosome in hemizygous (XY) males, whereas in Caenorhabditis nematodes, expression is halved for both X copies in homozygous (XX) females [1, 2]. Unlike other female-heterogametic (WZ female and ZZ male) animals, moths and butterflies exhibit sex chromosome dosage compensation patterns typically seen only in male-heterogametic species [3]. The monarch butterfly carries a newly derived Z chromosome segment that arose from an a

The species delimitation problem in the Simulium damnosum complex, blackfly vectors of onchocerciasis.

The Simulium damnosum Theobald complex (Diptera: Simuliidae) comprises 57 cytoforms grouped into six subcomplexes. Previous phylogenetic studies using gene sequences have not completely resolved the evolutionary relationships of the cytoforms. The present study investigated the systematics of the complex using a phylogeographic approach. The differentiation between eastern and western forms observed in the phylogenetic studies is confirmed in the estimated haplotype networks. However, haplotypes tend to group in geographical clades and not according to cytoforms. Spatial analyses of the molecu

Experimental hybridization between members of the Simulium damnosum complex.

Males of the Beffa form of Simulium soubrense Vajime and Dunbar were successfully crossed with female S.squamosum V. & D. and S.sanctipauli V. & D. Fertile eggs from both crosses were reared to larvae and, in the case of the Beffa x squamosum cross, to adults. Larval chromosomes from the Beffa x sanctipauli cross had normally paired homologues but those of the Beffa x squamosum cross showed extensive asynapsis and large heterozygous loops. The morphology of the larval and adult hybrids from the Beffa x squamosum cross were more characteristic of squamosum, but the adult males had scutal patter

A cytogenetic analysis of the Simulium ochraceum species complex (Diptera: Simuliidae) in Central America.

A cytobiotaxonomic study of the medically important insect vector Simulium ochraceum s.l. revealed two sibling species and one cytotype from various endemic and nonendemic zones of human onchocerciasis in Guatemala and Mexico. Polytene chromosome maps and idiograms as well as notes on the biology of the three taxa designated S. ochraceum A, S. ochraceum B, and S. ochraceum C within the subgenus Psilopelmia are presented. All three taxa exhibit distinct sex chromosomes and taxon-specific suites of autosomal inversion polymorphisms. Simulium ochraceum C differs from both S. ochraceum A and S. oc

A comprehensive gene expression atlas of sex- and tissue-specificity in the malaria vector, Anopheles gambiae.

BACKGROUND: The mosquito, Anopheles gambiae, is the primary vector of human malaria, a disease responsible for millions of deaths each year. To improve strategies for controlling transmission of the causative parasite, Plasmodium falciparum, we require a thorough understanding of the developmental mechanisms, physiological processes and evolutionary pressures affecting life-history traits in the mosquito. Identifying genes expressed in particular tissues or involved in specific biological processes is an essential part of this process. RESULTS: In this study, we present transcription profiles

A conserved zinc-finger gene BmZFO721 regulates meiosis and female fertility in silkworm, Bombyx mori.

Accurate meiotic progression during oogenesis is fundamental to oocyte maturation and female fertility in sexually reproducing organisms. In lepidopteran insects, however, the upstream regulators governing female meiotic progression, particularly ovary-specific functional genes, remain largely uncharacterized, and their in vivo roles in oocyte development are poorly understood. Here, we identified a gene encoding an ovary-specific C2H2-type zinc-finger protein (ZFP), designated BmZFO721 (Bombyx mori Zinc-Finger Ovary gene 721). Phylogenetic and genomic synteny analyses reveal that BmZFO721 is

Molecular analysis and chromosome mapping of the H2A, H3 and H4 histone genes from the malaria vector Anopheles gambiae.

In this article we report the cloning and analysis of PCR generated fragments that encode H2A, H3 and H4 histone genes from the malaria vector An. gambiae. Sequence analysis indicated that some conservative changes are present in the An. gambiae H2A and H4 genes as compared with histone genes from other organisms. Divisional mapping showed that these genes map in division 20 on the left arm of the second chromosome. Southern blot experiments and the molecular characterization of the genomic fragment containing the H2A, H2B, H3 and H4 genes showed that they are organized in a cluster with an or

Rapid neo-sex chromosome evolution and incipient speciation in a major forest pest.

Genome evolution is predicted to be rapid following the establishment of new (neo) sex chromosomes, but it is not known if neo-sex chromosome evolution plays an important role in speciation. Here we combine extensive crossing experiments with population and functional genomic data to examine neo-XY chromosome evolution and incipient speciation in the mountain pine beetle. We find a broad continuum of intrinsic incompatibilities in hybrid males that increase in strength with geographic distance between reproductively isolated populations. This striking progression of reproductive isolation is c
