Chemotaxis of Miamiensis avidus to hyaluronic acid, a component of fish surface mucus.

Miamiensis avidus is a marine parasitic ciliate belonging to the order Scuticociliatida, the members of which are the causal agents of scuticociliatosis. This ciliate has resulted in considerable economic losses to the aquaculture industry in the Republic of Korea and Japan. Nevertheless, the development of vaccines and therapeutics has proven to be challenging, and there are currently no reports of such products commercially available in Korea or Japan. Furthermore, the aetiology and pathogenesis of M. avidus infection in fish remain unknown, as do the underlying reasons for the initial infec

In silico characterization of cysteine-stabilized αβ defensins from neglected unicellular microeukaryotes.

BACKGROUND: The emergence of multi-resistant pathogens have increased dramatically in recent years, becoming a major public-health concern. Among other promising antimicrobial molecules with potential to assist in this worldwide struggle, cysteine-stabilized αβ (CS-αβ) defensins are attracting attention due their efficacy, stability, and broad spectrum against viruses, bacteria, fungi, and protists, including many known human pathogens. RESULTS: Here, 23 genomes of ciliated protists were screened and two CS-αβ defensins with a likely antifungal activity were identified and characterized, usin

Experimental infections of Orchitophrya stellarum (Scuticociliata) in American blue crabs (Callinectes sapidus) and fiddler crabs (Uca minax).

Outbreaks of an unidentified ciliate have occurred on several occasions in blue crabs from Chesapeake Bay held during winter months in flow-through systems. The parasite was initially thought to be Mesanophrys chesapeakensis, but molecular analysis identified it as Orchitophyra stellarum, a facultative parasite of sea stars (Asteroidea). We investigated the host-parasite association of O. stellarum in the blue crab host. Crabs were inoculated with the ciliate, or they were held in bath exposures after experimentally induced autotomy of limbs in order to determine potential mechanisms for infec

Efficacy of quinine against ichthyophthiriasis in common carp Cyprinus carpio.

Ichthyophthiriasis, caused by Ichthyophthirius multifiliis, is an economically important worldwide parasitic disease that infects all freshwater fish. Since the banning of malachite green for use in food fish, there has been a great need for alternative therapeutants. The goal of this study was to investigate the efficacy of quinine against I. multifiliis. Parasite developmental stages from our laboratory-established life cycle in rainbow trout Oncorhynchus mykiss were exposed to quinine in vitro, and a dual fluorescent staining technique was used to allow a clear distinction between viable an

Tissue-invading ciliates associated with chronic colitis in a horse.

Ciliates resembling Polymorphella ampulla (Dogiel, 1929) were found in large numbers in the crypts and lamina propria of the caecum and colon of a one-year-old Thoroughbred gelding that had shown recurring bouts of chronic diarrhoea and weight loss over a 5-month period. A heavy burden of helminths, mainly cyathostomes (trichonemes) was present at post-mortem examination and no significant bacterial pathogens were recovered. It was considered that the Polymorphella (which is normally a commensal organism) had in this case assumed a secondary pathogenic role.

Comparative analysis of the expression patterns of IL-1β, IL-11, and IL-34 in golden pompano (Trachinotus ovatus) following different pathogens challenge.

Interleukins (ILs) are a subgroup of cytokines, which are molecules involved in the intercellular regulation of the immune system. These cytokines have been extensively studied in mammalian models, but systematic analyses of fish are limited. In the current study, 3 IL genes from golden pompano (Trachinotus ovatus) were characterized. The IL-1β protein contains IL-1 family signature motif, and four long helices (αA - αD) in IL-11 and IL-34, which were well conserved. All 3 ILs clustered phylogenetically with their respective IL relatives in mammalian and other teleost species. Under normal phy

Development of a Tetrahymena thermophila-Based Vaccine Expressing Miamiensis avidus Ciliary Proteins to Combat Scuticociliatosis.

Miamiensis avidus is a parasitic pathogen responsible for scuticociliatosis, a lethal infection affecting marine fish worldwide, including Japanese flounder. Immunisation with formalin-killed M. avidus has shown promise in inducing protective immunity, positioning it as a potential vaccine candidate against scuticociliatosis. However, challenges such as the high cost of producing sufficient cells and inconsistent quality due to the lack of cryopreservation methods hinder its development. In this study, we expressed M. avidus ciliary proteins in Tetrahymena, a culturable ciliate, and used these

AI-Driven Realtime Monitoring of Early Indicators for Ichthyophthirius multifiliis Infection of Rainbow Trout.

A novel video-based real-time system based on AI (artificial intelligence) was developed to detect clinical signs in fish exposed to pathogens. We selected a White Spot Disease model involving rainbow trout as the experimental animal and the parasitic ciliate Ichthyophthirius multifiliis as a pathogen. We compared two identical fish tank systems: one tank was infected by co-habitation, whereas the other tank was kept non-infected (sham infection). The two fish tanks were separately video monitored (full top and side view) during the course of infection, during which fish were removed whenever

[Immotile cilia syndrome. Presentation of 3 cases].

Immotile cilia syndrome is an uncommon disorder, with an autosomal recessive mode of inheritance; the incidence is about 1/25,000 and is a cause of chronic disease of both upper and lower airways, male infertility and, in 50%, situs inversus totalis. It's pathogenesis lies in a variety of genetically determined alteration of ciliary ultrastructure. We report three cases, two in siblings, who had a typical syndrome, all of them with situs inversus totalis (Kartagener syndrome), chronic nasal symptoms, sinusitis and serous otitis with hearing loss. Two had recurrent productive cough with chest X

Characteristics delineation of piscidin 5 like from Larimichthys crocea with evidence for the potent antiparasitic activity.

Several researches reported that piscidin members of teleosts owned strong antiparasitic activity. Cryptocaryon irritans, a type of ectoparasite, could infect most of the marine teleosts. Larimichthys crocea could severely suffer from marine white spot disease caused by C. irritans, and their mortality rate was significantly high. Concentrating on this problem, we have done many related works. Piscidin 5 like (termed Lc-P5L) was another piscidin member isolated from a comparative transcriptome of C. irritans-immuned L. crocea. In the paper, quantitative Real-time PCR (qRT-PCR) showed Lc-P5L wa

The hidden passengers: On the role of exotic crayfish in the spread of symbiotic and pathogenic organisms in Northern Iberian Peninsula.

The spread of invasive crayfish species poses a growing threat to freshwater ecosystems and public health, not only through direct ecological impacts but also by facilitating the transmission of symbionts and potential zoonotic pathogens. This study characterizes the symbiont diversity associated with the crayfish species that occur in northern Spain. In addition, the first comprehensive and comparative compilation of reported symbiont and parasite species of crayfish species reported in Iberia is provided. A combination of scanning electron and optical microscopy analysis and a review of the

Gastrointestinal parasites in pigs in Brazil: prevalence, risk factors, diagnosis, and extension activities.

Brazil is a major pig-producing country with production systems that include both industrial and family farms. In these facilities, parasitic diseases present an obstacle to production. This study aimed to estimate the prevalence and risk factors of the gastrointestinal parasites that infect pigs as well as to provide information for producers through extension activities. Visits were made to 15 pig farms in cities in the states of Rio de Janeiro and Minas Gerais. Fecal samples were collected, questionnaires administered, and extension activities conducted. A total of 1,148 samples were collec

Dinoflagellate bloom in tropical fish ponds of coastal waters of the South China Sea.

Red tide of dinoflagellate was observed in brackish water fish ponds of Terengganu along the coast of the South China Sea during the study period between January 1992 to December 1992. The nearby coastal moat water facing the South China Sea is the source of water for fish pond culture activities of sea bass during the study period. An examination of water quality in fish ponds during the study period indicated that both the organic nutrients were high during the pre-wet monsoon period. The source of the nutrients in coastal water was believed to be derived from the agro-based industrial efflu

Molecular characterization and antiparasitic activity analysis of a novel piscidin 5-like type 4 from Larimichthys crocea.

Cryptocaryon irritans is an obligate parasitic ciliate protozoan that can infect various commercially important mariculture teleosts and cause high lethality and economic loss, especially Larimichthys crocea. Current methods of controlling or preventing this parasite with chemicals or antibiotics are widely considered to be environmentally harmful. The antiparasitic activity of some antimicrobial peptides (AMPs) attracted extensive attention of scholars. In the study, a novel piscidin 5-like type 4 (termed Lc-P5L4) excavated from comparative transcriptome of C. irritans - immuned L. crocea was

Molecular identification and expression analysis of TLR5M and TLR5S from orange-spotted grouper (Epinepheluscoioides).

Toll-like receptor 5 (TLR5) is an important receptor that interacts with bacterial flagellin and regulates host immune response in mammal. Recent studies demonstrate that piscine contains two types of TLR5, namely membrane form of TLR5 (TLR5M) and soluble form of TLR5 (TLR5S), and both of which perform crucial role in flagellin response. In the present study, a TLR5M and a TLR5S sequence was cloned from orange-spotted grouper (Epinepheluscoioides), and their ORFs are respectively 2466 bp (821 aas) and 1935 bp (644 aas). EcTLR5M has the typical TLR structure of a LRR domain, a transmembrane reg

Anthropogenic nutrient discharge and microbial community shifts associated with typhoon-driven runoff in an artificial coastal lake.

This study investigated changes in nutrient sources and microbial community composition across a typhoon-associated runoff period in Lake Sihwa, an anthropogenically influenced artificial coastal lake. Stable isotope analyses (δ13CPOC, δ15NPN, δ15NNO₃, and δ18ONO₃) and water-column environmental DNA (eDNA) metabarcoding were combined to characterize nutrient sources and compare ecological conditions between the pre- and post-typhoon surveys. Before the typhoon-associated rainfall period, nitrate inputs were primarily associated with urban runoff, whereas particulate organic matter (POM) reflec

Integrated transcriptomic and immune enzymatic analyses uncover coordinated immunometabolic responses in large yellow croaker (Larimichthys crocea) to Metanophrys sp. infection.

Metanophrys sp. (Scuticociliatida) has recently emerged as a significant parasitic threat in large yellow croaker (Larimichthys crocea) aquaculture. To elucidate the host response, we conducted an experimental infection followed by an integrated analysis combining immune enzymatic profiling and transcriptome sequencing. Antioxidant and immune enzyme activities, including SOD, CAT, and MDA in the skin, gill, and liver, and LYZ and Na+/K+-ATPase in the skin and gill were monitored from 0 to 72 hours post-infection (hpi). The results revealed tissue- and time-specific significant changes, indicat

Growth, Toxin Content and Production of Dinophysis Norvegica in Cultured Strains Isolated from Funka Bay (Japan).

The successful cultivation of Dinophysis norvegica Claparède & Lachmann, 1859, isolated from Japanese coastal waters, is presented in this study, which also includes an examination of its toxin content and production for the first time. Maintaining the strains at a high abundance (>2000 cells per mL-1) for more than 20 months was achieved by feeding them with the ciliate Mesodinium rubrum Lohmann, 1908, along with the addition of the cryptophyte Teleaulax amphioxeia (W.Conrad) D.R.A.Hill, 1992. Toxin production was examined using seven established strains. At the end of the one-month incubatio
