Nucleolin inhibits porcine circovirus type 3 replication by activating the JAK-STAT1 signaling pathway.

Porcine circovirus type 3 (PCV3), a newly identified pathogen linked to multiple clinical diseases, threatens the global swine industry. Nucleolin, a multifunctional protein involved in various biological events, has been confirmed to suppresses PCV3 replication; however, the underlying molecular mechanism remains unclear. Herein, we demonstrate that nucleolin inhibits PCV3 replication by activating the type I interferon (IFN)/Janus kinase (JAK)-signal transducer and activator of transcription 1 (STAT1) pathway, a critical host antiviral defense axis. Mechanistically, nucleolin enhances STAT1/

VLP-based indirect ELISA enables reliable sero-diagnosis and epidemiological monitoring of duck circovirus.

INTRODUCTION: Duck circovirus (DuCV) is an emerging immunosuppressive virus that causes growth retardation, feather loss, and increased susceptibility to secondary infections in ducks, leading to significant economic losses in the duck industry. However, existing serological assays for DuCV detection often lack accuracy and reproducibility due to antigen instability, limiting effective disease surveillance. METHODS: To develop a more reliable diagnostic tool, the full-length capsid (Cap) gene of DuCV-2 was codon-optimized, expressed in Escherichia coli, purified under denaturing conditions, a

Tetraspanin CD81 serves as a functional entry factor for porcine circovirus type 2 infection.

Porcine circovirus type 2 (PCV2) is the primary causative agent of porcine circovirus-associated disease, clinically resulting in immunosuppression and co-infections with other pathogens in infected pigs. The mechanism of PCV2 infection remains unclear. In this study, we firstly found that the tetraspanin CD81 in PK-15 cells interacts with PCV2 Cap protein by using virus overlay protein-binding assay combined with mass spectrometry. Knockdown of the CD81 significantly reduces the levels of the viral Cap mRNA and protein, and viral internalization in PK-15 cells. The critical interaction region

Matrine Targets Intestinal Lactobacillus acidophilus to Inhibit Porcine Circovirus Type 2 Infection in Mice.

Porcine circovirus type 2 (PCV2) has caused huge economic losses to the pig industry across the world. Matrine is a natural compound that has been shown to regulate intestinal flora and has anti-PCV2 activity in mouse models. PCV2 infection can lead to changes in intestinal flora. The intestinal flora has proved to be one of the important pharmacological targets of the active components of Traditional Chinese Medicine. This study aimed to determine whether matrine exerts anti-PCV2 effects by regulating intestinal flora. In this study, fecal microbiota transplantation (FMT) was used to evaluate

Balantidium Coli in Pig Farms Suspected of Porcine Circovirus Type 2 (PCV2) Associated Enteritis.

INTRODUCTION: Diarrhoea in growing-finishing pigs is a common problem of commercial pig farms. Among many causative factors, porcine circovirus type 2 (PCV2) is one considered an important pathogen in modern pig production. The aim of the study was to verify if PCV2 was responsible for antibiotic non-responsive diarrhoea and wasting in pigs. MATERIAL AND METHODS: A total of 13 dead pigs aged between 12 and 15 weeks from three Polish farms with persistent herd symptoms suggestive of PCV2 infection were provided for evaluation. Sections of lymph nodes and intestines were analysed by in situ hyb

Generation and immunogenicity assessment of ELPylated virus-like particles of porcine circovirus type 2.

BACKGROUND: Porcine circovirus type 2 (PCV2) is an economically important pathogen affecting swine industry worldwide. The production of current PCV2 vaccines is time-consuming and expensive. Elastin-like polypeptides (ELP) undergo temperature-dependent inverse phase transition and ELPylated proteins can be purified simply by inverse transition cycling (ITC). METHODS: The Cap protein of PCV2b, together with the virus neutralizing (VN) epitopes of PCV2a, PCV2d and PCV2e, was expressed in E. coli as an ELPylated protein, and purified by ITC in the presence of mild detergents. For the control pu

Genomic characterization of a novel circovirus from a stranded Longman's beaked whale (Indopacetus pacificus).

Tissues from a juvenile Longman's beaked whale that stranded in Hawaii in 2010 were screened for viruses using a Next-Generation Sequencing (NGS) approach. From the NGS data, the full genome (1,849 bp) of a novel beaked whale circovirus (BWCV) was determined. Two open reading frames (ORF) were annotated, including ORF1 that encodes the capsid gene, ORF2 that encodes the replication-associated gene, and a 9-bp conserved nonamer on the apex of the open loop found in all circoviruses. Independent phylogenetic analyses based on amino acid sequence alignments of the two CV proteins supported the BW

Conjugation of chitosan oligosaccharides enhances immune response to porcine circovirus vaccine by activating macrophages.

Porcine circovirus type 2 (PCV2)-associated diseases have led to great economic losses to the pig industry. Our lab previously found that conjugation of chitosan oligosaccharides (COS) or via a carrier protein enhanced the immunogenicity of PCV2 vaccine against infectious pathogens. However, precise mechanisms and signal transduction pathways underlying the efficacy of COS conjugation remains poorly defined. In this study, to better understand the effects and mechanism of COS conjugates maintain the adjuvant potential in vivo, we investigated its augmentation of macrophage function, including

Comparative genetic characterization of Porcine Circovirus type 2 samples from German wild boar populations.

PCV-2 is involved in "postweaning multisystemic wasting syndrome" (PMWS), "porcine dermatitis and nephropathy syndrome" (PDNS), respiratory and reproductive disorders, and thereby plays a crucial role in today's swine production world-wide. The virus is apparently ubiquitous in domestic pigs and has also been demonstrated in wild pigs. Up to now, a characterization of PCV-2 samples from wild pigs, which might help to estimate the possible role of wild pigs as sources of domestic pig infection, has not been carried out. Spleen samples from 16 PCV-2-positive wild pigs from hunting grounds of fou

Characterization of a duck circovirus propagated in duck embryos: from genome to pathogenesis in SPF ducks.

Duck circovirus (DuCV) is a major pathogen in duck farming that causes immunosuppression and increases the susceptibility to secondary infections. This study focused on a Chinese DuCV strain, designated SDDC, which was isolated and characterized from Cherry Valley ducks in Shandong Province in 2022. The SDDC strain was successfully propagated in specific-pathogen-free (SPF) duck embryos and confirmed by transmission electron microscopy. Genomic analysis identified a complete genome of 1994 bp, with a critical amino acid mutation (I to T) at the 160th position in the Cap protein, potentially in

Identification and genomic analysis of a pathogenic circovirus associated with maricultured Scophthalmus maximus L. in China.

In China, a novel pathogen within the genus Circovirus has been identified as a causative agent of the 'novel acute hemorrhage syndrome' (NAHS) in aquacultured populations of turbot (Scophthalmus maximus L.). Histopathological examination using light microscopy revealed extensive necrosis within the cardiac, splenic, and renal tissues of the afflicted fish. Utilizing transmission electron microscopy (TEM), we detected the presence of circovirus particles within the cytoplasm of these cells, with the virions consistently exhibiting a spherical morphology of 20-40 nm in diameter. TEM inspections

Induction of Porcine Dermatitis and Nephropathy Syndrome in Piglets by Infection with Porcine Circovirus Type 3.

Porcine circovirus type 3 (PCV3) is an emerging porcine circovirus that has been associated with porcine dermatitis and nephropathy syndrome (PDNS)-like clinical signs, reproductive failure, cardiac pathologies, and multisystemic inflammation in piglets and sows. Many aspects of PCV3 infection biology and pathogenesis, however, remain unknown. Here, we used a PCV3 virus stock from the rescue of an infectious PCV3 DNA clone to intranasally inoculate 4- and 8-week-old specific-pathogen-free piglets for evaluation of PCV3 pathogenesis. For 4-week-old piglets, typical clinical signs resembling tho

Development of a Plexor real-time PCR assay for the detection of porcine circovirus type 2.

A novel, real-time PCR system for the detection of porcine circovirus type 2 (PCV2) was developed. The system employed Plexor technology and detected 10(8)-10(1) copies per reaction of PCV2 DNA within a recombinant plasmid. The examination of clinical material showed consistent diagnostic sensitivity when samples contained more than 10(3) viral copies per reaction. Specificity of Plexor real-time PCR was confirmed using the porcine viruses PCV1, PRRSV, CSFV, TTSuV1 and TTSuV2 employing the melting curve analysis of PCR products.
