Inhibition increases response variability and reduces stimulus discrimination in random networks of cortical neurons.

Much of what is known about the contribution of inhibition to stimulus discrimination is due to extensively studied sensory systems, which are highly structured neural circuits. The effect of inhibition on stimulus representation in less structured networks is not as clear. Here we exercise a biosynthetic approach in order to study the impacts of inhibition on stimulus representation in non-specialized network anatomy. Combining pharmacological manipulation, multisite electrical stimulation and recording from ex-vivo randomly rewired networks of cortical neurons, we quantified the effects of i

Neuromodulation for psychological sequelae of traumatic brain injury.

Traumatic brain injury (TBI) frequently precipitates chronic psychological disorder, particularly anxiety, depression, post-traumatic stress disorder (PTSD), and increased suicide risk, that worsen with injury severity yet remain understudied relative to sensorimotor and cognitive deficits. As TBI severity increases, conventional interventions (pharmacotherapy, cognitive-behavioral therapy, exercise) lose efficacy, revealing an urgent need for novel treatments targeting affective and stress-related disorders. Neuromodulatory approaches such as transcranial magnetic stimulation (TMS), transcran

Combined Transcranial Direct Current Stimulation and Virtual Reality-Based Paradigm for Upper Limb Rehabilitation in Individuals with Restricted Movements. A Feasibility Study with a Chronic Stroke Survivor with Severe Hemiparesis.

Impairments of the upper limb function are a major cause of disability and rehabilitation. Most of the available therapeutic options are based on active exercises and on motor and attentional inclusion of the affected arm in task oriented movements. However, active movements may not be possible after severe impairment of the upper limbs. Different techniques, such as mirror therapy, motor imagery, and non-invasive brain stimulation have been shown to elicit cortical activity in absence of movements, which could be used to preserve the available neural circuits and promote motor learning. Feasibility of a Community-Based Boxing Program with Tailored Balance Training in Parkinson's Disease: A Preliminary Study.

BACKGROUND/OBJECTIVES: Persons with Parkinson's disease (PD) are at elevated risk of falling due to deficits in postural control, lower limb strength, and sensory integration. While community-based boxing programs (CBPs) have shown promise in improving strength and balance, their feasibility and potential role in addressing fall risk remain unclear. This preliminary, prospective cohort study explored the feasibility of a CBP enhanced with individualized balance training tailored to somatosensory deficits and explored early indications of potential impact on fall risk and related outcomes. Adipose tissue as a humoral-neuronal hub in metabolic regulation.

Adipose tissue has emerged as a dynamic endocrine organ that coordinates systemic energy balance and cardiometabolic health. This Review highlights the dual humoral and neuronal pathways through which adipose tissue regulates systemic metabolism. Humoral signals include peptide hormones, lipid mediators, metabolites, chemokines and exosomal microRNAs secreted by adipose depots. Neuronal circuits control adipose function rapidly and precisely: sympathetic efferents trigger lipolysis in white adipose tissue and thermogenesis in brown adipose tissue, whereas sensory afferents detect chemical, the

An integrated multi-electrode-optrode array for in vitro optogenetics.

Modulation of a group of cells or tissue needs to be very precise in order to exercise effective control over the cell population under investigation. Optogenetic tools have already demonstrated to be of great value in the study of neuronal circuits and in neuromodulation. Ideally, they should permit very accurate resolution, preferably down to the single cell level. Further, to address a spatially distributed sample, independently addressable multiple optical outputs should be present. In current techniques, at least one of these requirements is not fulfilled. In addition to this, it is inter

Using the Activity-based Anorexia Rodent Model to Study the Neurobiological Basis of Anorexia Nervosa.

Anorexia nervosa (AN) is a psychiatric illness characterized by excessively restricted caloric intake and abnormally high levels of physical activity. A challenging illness to treat, due to the lack of understanding of the underlying neurobiology, AN has the highest mortality rate among psychiatric illnesses. To address this need, neuroscientists are using an animal model to study how neural circuits may contribute toward vulnerability to AN and may be affected by AN. Activity-based anorexia (ABA) is a bio-behavioral phenomenon described in rodents that models the key symptoms of anorexia nerv

Gold Nanowire Sponge Electrochemistry for Permeable Wearable Sweat Analysis Comfortably and Wirelessly.

Electrochemistry-based wearable and wireless sweat analysis is emerging as a promising noninvasive method for real-time health monitoring by tracking chemical and biological markers without the need for invasive blood sampling. It offers the potential to remotely monitor human sweat conditions in relation to metabolic health, stress, and electrolyte balance, which have implications for athletes, patients with chronic conditions, and individuals for the early detection and management of health issues. The state-of-the-art mainstream technology is dominated by the concept of a wearable microflui

From Striatum to Prescription: An Evidence-Based and Bayesian Framework for Neuromotor Rehabilitation in Parkinson's Disease.

Parkinson's disease (PD) involves progressive basal ganglia dysfunction, with hyperexcitability of striatal indirect-pathway D2 medium spiny neurons (D2-MSNs) linked to motor impairment. Neuromotor rehabilitation is an important therapy, but its efficacy varies across interventions. This thematic review examines eleven rehabilitative strategies for PD, spanning forced and voluntary exercise (FE and VE respectively), non-invasive brain stimulation (rTMS, tDCS), and technology-based approaches such as exoskeletons, augmented reality, and dual-task training, within a framework distinguishing stri

Proteomic Profiling of Human Sweat Reveals Molecular Adaptations to Heat Acclimatization Induced by Isothermic Conditioning.

INTRODUCTION: Prolonged physical exertion in hot and/or humid environments can induce physiological strain and increase the risk of heat exhaustion or exertional heat stroke (EHS). Heat acclimatization (HA) optimizes an individual's ability to tolerate heat and is therefore widely adopted to improve exercise performance in thermally challenging environments. Sweat is a key thermoregulatory indicator reflective of physiological adaptations during HA and hence is regarded as a valuable resource for monitoring HA. However, knowledge of molecular-level changes and adaptations in sweat associated w

Vulnerable and Resilient Phenotypes in a Mouse Model of Anorexia Nervosa.

BACKGROUND: Increased physical activity is a common feature of anorexia nervosa (AN). Although high activity levels are associated with greater risk of developing AN, particularly when combined with dieting, most individuals who diet and exercise maintain a healthy body weight. It is unclear why some individuals develop AN while most do not. A rodent model of resilience and vulnerability to AN would be valuable to research. Dopamine, which is believed to play a crucial role in AN, regulates both reward and activity and may modulate vulnerability. METHODS: Adolescent and young adult female C57

The Assisted Decision-Making (Capacity) Bill 2013: content, commentary, controversy.

BACKGROUND: Ireland's Assisted Decision-Making (Capacity) Bill (2013) aims to reform the law relating to persons who require assistance exercising their decision-making capacity. When finalised, the Bill will replace Ireland's outdated Ward of Court system which has an all-or-nothing approach to capacity; does not adequately define capacity; is poorly responsive to change; makes unwieldy provision for appointing decision-makers; and has insufficient provision for review. AIMS: To explore the content and implications of the Assisted Decision-Making (Capacity) Bill. METHODS: Review of the cont

Metabolic inflammation at the adipose-brain axis.

Overweight and obesity have emerged as global health crises and are increasingly recognized as drivers of central nervous system (CNS) dysfunction. Beyond excess energy storage, white adipose tissue (WAT) functions as an active endocrine and immune organ that, during obesity, undergoes inflammatory remodeling and releases cytokines, lipid mediators, adipokines, and extracellular vesicles that influence brain physiology. These peripheral signals disrupt key brain interfaces, including the blood-brain barrier (BBB), perivascular and glymphatic clearance pathways, promoting endothelial dysfunctio

Alpha-Synuclein Neurobiology in Parkinson's Disease: A Comprehensive Review of Its Role, Mechanisms, and Therapeutic Perspectives.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra (SN) and the presence of intracellular α-synuclein (αSyn) aggregates known as Lewy bodies (LB). αSyn, a presynaptic protein, is believed to play a crucial role in synaptic function, neurotransmitter release, and neuronal plasticity. However, its misfolding and aggregation are thought to be central to PD pathogenesis. This review provides a comprehensive analysis of αSyn's role in PD, exploring its normal physiological functions, pathological mechanisms,

Assessment, pathophysiology and treatment of fatigue in multiple sclerosis.

This review will update current views of the physiopathology and treatment of fatigue in multiple sclerosis. Fatigue is a common symptom in multiple sclerosis, being reported by about a third of the patients. For many of them it is the most disabling symptom, with negative consequences on working activity and daily life. There are no objective measures of fatigue which is essentially based on subjective complaints. Even if fatigue may be influenced by motor disturbances and depression, it is largely independent from both. Peripheral mechanisms, such as muscular disuse and deconditioning, joint

Adenosine. An evaluation of its use in cardiac diagnostic procedures, and in the treatment of paroxysmal supraventricular tachycardia.

Adenosine (adenine riboside), administered either as the free base or as the 5'-triphosphate (ATP) by rapid intravenous bolus, depresses atrioventricular (AV) nodal conduction, resulting in transient AV block. Adenosine is the active agent and ATP is rapidly converted to adenosine after exogenous administration. By blocking the anterograde AV nodal limb of a re-entrant circuit, adenosine 6 to 12 mg (or ATP 10 to 20 mg) converts almost all episodes of paroxysmal supraventricular tachycardia (PSVT) involving the AV node within 30 seconds of administration. This is at least equivalent in efficacy

The effect of circuit resistance training, empagliflozin or "vegeterranean diet" on physical and metabolic function in older subjects with type 2 diabetes: a study protocol for a randomized control trial (CEV-65 trial).

BACKGROUND: Treatment of the older diabetic individual comprises a therapeutic challenge. Currently little scientific evidence exists depicting the best approach to type 2 diabetes treatment in this growing sub-population of patients. The purpose of this study is to assess the effects of a modified plant-based Mediterranean diet ("vegeterranean" diet), circuit resistance training (CRT) and empagliflozin, separately or in combination, on body composition and physical function in older subjects with type 2 diabetes. The rationale for this study is to assess three interventions associated with a
