Interleukin-6 is important for regulation of core body temperature during long-term cold exposure in mice.

Interleukin-6 (IL6) is a cytokine important for inducing the fever response during infection and has been reported to uphold core body temperature during acute cold exposure. Recently it has also been indicated that IL6 in serum increases in cold-exposed mice. The aim of the present study was to investigate if IL6 is important for core body temperature regulation following a long-term cold exposure in mice. Experiments were performed with global IL6 deficient (-/-) mice, mice with conditional IL6 receptor α (IL6Rα) knockdown in the central nervous system (CNS; IL6RαNesCre) and appropriate wild

Identification of complex regional pain syndrome in the upper limb: Skin temperature asymmetry after cold pressor test.

BACKGROUND: Skin temperature asymmetry (SkTA) may assist in early identification of complex regional pain syndrome (CRPS), but previous work has been limited by methodological shortcomings including failure to account for the cutaneous nerve distribution where temperature is measured and reliance on laboratory equipment not clinically available. Pilot work suggested that a cold pressor test (CPT) provided a consistent thermoregulatory stress and might increase sensitivity/specificity of SkTA measurements generated reliably by handheld infrared (IR) thermometers. AIMS: This study investigated

Cold temperature improves mobility and survival in Drosophila models of autosomal-dominant hereditary spastic paraplegia (AD-HSP).

Autosomal-dominant hereditary spastic paraplegia (AD-HSP) is a crippling neurodegenerative disease for which effective treatment or cure remains unknown. Victims experience progressive mobility loss due to degeneration of the longest axons in the spinal cord. Over half of AD-HSP cases arise from loss-of-function mutations in spastin, which encodes a microtubule-severing AAA ATPase. In Drosophila models of AD-HSP, larvae lacking Spastin exhibit abnormal motor neuron morphology and function, and most die as pupae. Adult survivors display impaired mobility, reminiscent of the human disease. Here,

An improved Peltier effect-based instrument for critical temperature threshold measurement in cold- and heat-induced urticaria.

BACKGROUND: Cold- and heat-induced urticaria are chronic physical urticaria conditions in which wheals, angioedema or both are evoked by skin exposure to cold and heat respectively. The diagnostic work up of both conditions should include skin provocation tests and accurate determination of critical temperature thresholds (CTT) for producing symptoms in order to be able to predict the potential risk that each individual patient faces and how this may be ameliorated by therapy. OBJECTIVE: To develop and validate TempTest(®) 4, a simple and relatively inexpensive instrument for the accurate det

Intra-individual variation of skin temperature recovery rate after cold water immersion among Japanese bush-cutter operators.

Recovery rates of skin temperature after cold water immersion tests and other measurements were recorded from 1978 to 1989 to assess the effects of vibration due to the use of bush-cutters on the health of 42 road-maintenance workers having an average age of 51.7 years. Their work and health conditions were controlled and checked systematically, and the incidence of white finger in 1989 was 2%. The recovery rate as well as the initial skin temperature was influenced greatly by ambient temperature and thus direct assessment of the changes in recovery rate had little value. The intra-individual

Overcoming cold chain challenges: Cryoprotection of self-amplifying mRNA polyplexes for long-term storage at ambient temperature.

The approval of mRNA and self-amplifying mRNA lipid-nanoparticle vaccines against COVID-19 has demonstrated the effectiveness and safety of these platforms and carriers. However, their low thermostability requires (ultra)cold storage conditions that hamper their distribution and accessibility, especially in low-income countries. Therefore, we investigated the ability and potential benefits of our linear polyethylenimine-random-polypropylenimine copolymer (L-PEIx-ran-PPIy) carrier in maintaining stable, functional saRNA-polyplexes after freezing and freeze-drying. First, a freeze-thaw study was

Temperature-Dependent Relationship of Autophagy and Apoptotic Signaling During Cold-Water Immersion in Young and Older Males.

Autophagy is a crucial cytoprotective mechanism preventing the accumulation of cellular damage, especially during external stimuli such as cold exposure. Older adults poorly tolerate cold exposure and age-related impairments in autophagy may contribute to the associated reductions in cold tolerance. The purpose of this investigation is to evaluate the effect of different intensities of in vivo cold-water immersion and in vitro cold exposure on autophagic and apoptotic signaling in young and older males. Peripheral blood mononuclear cells (PBMCs) are isolated at baseline, end-cold exposure, and

Hypoxia-ischemia-mediated effects on neurodevelopmentally regulated cold-shock proteins in neonatal mice under strict temperature control.

BACKGROUND: Neonates have high levels of cold-shock proteins (CSPs) in the normothermic brain for a limited period following birth. Hypoxic-ischemic (HI) insults in term infants produce neonatal encephalopathy (NE), and it remains unclear whether HI-induced pathology alters baseline CSP expression in the normothermic brain. METHODS: Here we established a version of the Rice-Vannucci model in PND 10 mice that incorporates rigorous temperature control. RESULTS: Common carotid artery (CCA)-ligation plus 25 min hypoxia (8% O2) in pups with targeted normothermia resulted in classic histopathologi

Validity of Core Temperature Measurements at 3 Rectal Depths During Rest, Exercise, Cold-Water Immersion, and Recovery.

CONTEXT: No evidence-based recommendation exists regarding how far clinicians should insert a rectal thermistor to obtain the most valid estimate of core temperature. Knowing the validity of temperatures at different rectal depths has implications for exertional heat-stroke (EHS) management. OBJECTIVE: To determine whether rectal temperature (Trec) taken at 4 cm, 10 cm, or 15 cm from the anal sphincter provides the most valid estimate of core temperature (as determined by esophageal temperature [Teso]) during similar stressors an athlete with EHS may experience. DESIGN: Cross-sectional study

Evaluation of dorper sheep semen quality preserved in milk egg yolk extender at room and cold temperatures.

BACKGROUND: Artificial insemination (AI) is widely used to improve genetic progress and reproductive efficiency in sheep production systems, including Dorper sheep. The success of AI programs for liquid semen depends on the ability to maintain semen quality during storage. Although milk-egg yolk extenders are commonly used for ram semen preservation, the optimal egg yolk concentration for maintaining sperm quality under different storage conditions remains unclear. AIM: This study evaluated the effects of 5% and 10% egg yolk extenders on the quality of Dorper ram semen stored at room temperat

Setting the limit: cold rather than hot temperatures limit intertidal distribution of a coastal foundation species.

Long-lasting restoration success of foundation species requires understanding their responses to climate change. For species with broad distributions, lower latitudes may serve as a proxy for future warming at higher latitudes. Such space-for-time substitutions are a powerful tool for developing climate change predictions for species distributed along steep elevational gradients. To understand climate resilience of a key coastal foundation species, we examined the upper elevational limit of the native Olympia oyster (Ostrea lurida) along its entire range at 26 sites spanning 21° latitude, from

The cold-sensing ion channel TRPM8 regulates central and peripheral clockwork and the circadian oscillations of body temperature.

AIM: Physiological functions in mammals show circadian oscillations, synchronized by daily cycles of light and temperature. Central and peripheral clocks participate in this regulation. Since the ion channel TRPM8 is a critical cold sensor, we investigated its role in circadian function. METHODS: We used TRPM8 reporter mouse lines and TRPM8-deficient mice. mRNA levels were determined by in situ hybridization or RT-qPCR and protein levels by immunofluorescence. A telemetry system was used to measure core body temperature (Tc). RESULTS: TRPM8 is expressed in the retina, specifically in choline

Warm and cold temperatures have distinct germline stem cell lineage effects during Drosophila oogenesis.

Despite their medical and economic relevance, it remains largely unknown how suboptimal temperatures affect adult insect reproduction. Here, we report an in-depth analysis of how chronic adult exposure to suboptimal temperatures affects oogenesis using the model insect Drosophila melanogaster. In adult females maintained at 18°C (cold) or 29°C (warm), relative to females at the 25°C control temperature, egg production was reduced through distinct cellular mechanisms. Chronic 18°C exposure improved germline stem cell maintenance, survival of early germline cysts and oocyte quality, but reduced

Application of High Pressure with Homogenization, Temperature, Carbon Dioxide, and Cold Plasma for the Inactivation of Bacterial Spores: A Review.

Formation of highly resistant spores is a concern for the safety of low-acid foods as they are a perfect vehicle for food spoilage and/or human infection. For spore inactivation, the strategy usually applied in the food industry is the intensification of traditional preservation methods to sterilization levels, which is often accompanied by decreases of nutritional and sensory properties. In order to overcome these unwanted side effects in food products, novel and emerging sterilization technologies are being developed, such as pressure-assisted thermal sterilization, high-pressure carbon diox

Rupatadine and its effects on symptom control, stimulation time, and temperature thresholds in patients with acquired cold urticaria.

BACKGROUND: Patients with acquired cold urticaria (ACU) show itchy wheals during cold exposure. This disturbing condition involves histamine and platelet-activating factor in its pathogenesis. Rupatadine is a dual antagonist of both histamine and platelet-activating factor. OBJECTIVE: To assess rupatadine efficacy in preventing reactions to cold challenge in patients with ACU. METHODS: A crossover, randomized, double-blind, placebo-controlled study in which 21 patients with ACU received rupatadine, 20 mg/d, or placebo for 1 week each is presented. The main outcome was the critical stimulatio

Influence of sarcoptic mange and cold and ambient temperature on blastogenic responses of lymphocytes and serum cortisol concentrations of pigs.

Blood samples from sarcoptic mite-infested pigs were evaluated for effects of mite infestation and cold and ambient temperatures on lymphocyte blastogenic responses and for effects of mite infestation on serum cortisol concentrations. In experiment 1, sarcoptic mite-infested and noninfested pigs were housed in cold (5 to 15 C fluctuating) and thermoneural (25 C) environmental chambers for 5 weeks. Differences were not observed (P greater than 0.10) in blastogenic responses to phytohemagglutin or pokeweed mitogen between lymphocytes from infested and noninfested pigs on postinfestation days (PI

Metabolic and functional changes in apheresis platelets during cold and room temperature storage: A pilot in vitro study.

BACKGROUND: Platelet concentrates are conventionally stored at room temperature (20-24°C), which preserves platelet viability but is associated with progressive metabolic deterioration and increased risk of bacterial contamination. Cold storage (4°C) has re-emerged as a potential alternative strategy; however, its impact on platelet metabolism, activation, and hemostatic function during extended storage remains incompletely characterized. AIM: To evaluate metabolic and functional changes in apheresis platelet concentrates during storage under cold and room temperature conditions. METHODS: Ap

A slow decay to extinction: cold temperatures and reduced microbial complexity prolong the persistence of planktonic Francisella tularensis.

UNLABELLED: Francisella tularensis is a zoonotic pathogen transmitted to humans through contact with infected animals, arthropod vectors, or contaminated environmental sources. Although replication occurs primarily within eukaryotic host cells, persistence outside a host is thought to be critical for long-term maintenance and transmission in natural ecosystems. To better understand persistence in aquatic habitats outside of mammalian hosts, we studied virulent strains of F. tularensis subspecies tularensis (type A) and subspecies holarctica (type B) incubated in a planktonic state (free living

Engineering Perovskite Hydroxide as a Cold-Adapted Oxidase Mimic for Construction of the Robust Low-Temperature Adaptive Biosensors.

Traditional biological detection methods rely on signal amplification strategies such as enzymatic catalysis or nucleic acid amplification. However, their efficiency decreases in low-temperature environments, compromising their detection sensitivity. To break the loss of enzyme catalytic activity at low temperatures, research on cold-adaptive nanozymes has attracted much attention. Till now, only a few nanozymes have been reported to have cold-resistant catalytic properties. Here, a new type of cold-adapted nanozyme was constructed by engineering a perovskite hydroxide. The nanozyme not only b
