Crystal structure and computational study of 1-hy-droxy-3,6,7-trimeth-oxy-2,8-bis-(3-methyl-but-2-en-1-yl)-9H-xanthen-9-one (fuscaxanthone C).

The title xanthone derivative, C26H30O6, extracted from the bark of Cratoxylum glaucum, adopts an almost planar fused ring system with a maximum deviation of 0.078 (1) Å. In the crystal, mol-ecules are linked into inversion dimers by pairwise C-H⋯O inter-actions. These dimers are further linked into chains by C-H⋯π and C-O⋯π inter-actions. Computational chemistry revealed that the overall stabilization energy of the packing is dominated by dispersion energy. The conformations of related xanthones are also surveyed.

Molecular basis of opioid receptor signaling.

Opioids are used for pain management despite the side effects that contribute to the opioid crisis. The pursuit of non-addictive opioid analgesics remains unattained due to the unresolved intricacies of opioid actions, receptor signaling cascades, and neuronal plasticity. Advancements in structural, molecular, and computational tools illuminate the dynamic interplay between opioids and opioid receptors, as well as the molecular determinants of signaling pathways, which are potentially interlinked with pharmacological responses. Here, we review the molecular basis of opioid receptor signaling w

Discovery of Alternative Chemotherapy Options for Leishmaniasis through Computational Studies of Asteraceae.

Leishmaniasis is a complex disease caused by over 20 Leishmania species that primarily affects populations with poor socioeconomic conditions. Currently available drugs for treating leishmaniasis include amphotericin B, paromomycin, and pentavalent antimonials, which have been associated with several limitations, such as low efficacy, the development of drug resistance, and high toxicity. Natural products are an interesting source of new drug candidates. The Asteraceae family includes more than 23 000 species worldwide. Secondary metabolites that can be found in species from this family have b

Mechanistic Insights into SAM-Dependent Methyltransferases: A Review of Computational Approaches.

Methylation reactions catalyzed by S-adenosylmethionine (SAM)-dependent methyltransferases are essential to numerous biological functions, including gene expression regulation, epigenetic modifications, and biosynthesis of natural products. Dysregulation of these enzymes is associated with diseases, including cancer and neurodevelopmental disorders, making them attractive drug targets. This review explores the contribution of computational methods, particularly quantum chemical calculations and molecular dynamics (MD) simulations, in elucidating the mechanisms of SAM-dependent methyltransferas

A photogenerated triplet aura-nitrene for gold-mediated nitrene transfer.

The isolation and characterization of metal-stabilized, low-valent nitrogen species remain central challenges in advancing nitrogen-atom transfer chemistry. Although gold has emerged as a powerful catalyst in C-N bond formation, proposed mechanisms have consistently bypassed gold nitrenes as key intermediates-leaving their chemistry largely uncharted. Here we report the characterization of a bona fide aura-nitrene, formed via photochemical N2 extrusion from a gold(III) azide complex. In crystallo and solid-state analyses reveal a diverse reactivity landscape, including a nitrene-to-nitro trans

Copper-Catalyzed Enantioselective Synthesis of Chiral Selenium-Based Versatile Synthons.

Enantioenriched organoselenium compounds have found diverse applications across organic synthesis, asymmetric catalysis, and medicinal chemistry, driving a growing demand for efficient catalytic approaches to access novel chiral organoselenium molecules. However, in contrast to their oxygen- and sulfur-based counterparts, catalytic asymmetric routes to selenium-based chiral compounds remain underdeveloped, and no catalytic method has been reported for preparing chiral selenium-based molecules capable of serving multiple functional roles. Here, we describe a mild and simple Cu-(II)/diamine cata

Extracellular-Matrix-Anchored Click Motifs for Specific Tissue Targeting.

Local presentation of cancer drugs by injectable drug-eluting depots reduces systemic side effects and improves efficacy. However, local depots deplete their drug stores and are difficult to introduce into stiff tissues, or organs, such as the brain, that cannot accommodate increased pressure. We present a method for introducing targetable depots through injection of activated ester molecules into target tissues that react with and anchor themselves to the local extracellular matrix (ECM) and subsequently capture systemically administered small molecules through bioorthogonal click chemistry.

XPlex: An Effective, Multiplex Cross-Linking Chemistry for Acidic Residues.

Cross-linking/Mass spectrometry (XLMS) is a consolidated technique for structural characterization of proteins and protein complexes. Despite its success, the cross-linking chemistry currently used is mostly based on N-hydroxysuccinimide (NHS) esters, which react primarily with lysine residues. One way to expand the current applicability of XLMS into several new areas is to increase the number of cross-links obtainable for a target protein. We introduce a multiplex chemistry (denoted XPlex) that targets Asp, Glu, Lys, and Ser residues. XPlex can generate significantly more cross-links with rea

"Metal-based compounds in antiviral and Antiparasitic research: Mechanistic insights from HIV NCp7 and leishmaniasis case studies".

Metallodrugs have long occupied a significant position in medicinal chemistry, yet their application to infectious diseases has historically lagged the extensive development observed in cancer therapy. The COVID-19 pandemic, together with the increased spread of parasitic diseases driven by climate change and human migration, has renewed interest in metal-based compounds as antiviral and antiparasitic agents. These efforts have been further enabled by advances in computational modeling and mechanistic analysis, allowing rational design strategies that extend beyond in vitro drug screening. Thi

HIP: Hessian Interatomic Potentials without derivatives.

Molecular Hessians, the second derivatives of the potential energy, are fundamental to many workflows in computational chemistry. Usually, accurate Hessians are computationally expensive to calculate and scale poorly with system size, whether computed using quantum chemistry methods or machine learning interatomic potentials. Here we introduce Hessian Interatomic Potentials (HIPs), a deep learning model that directly predicts Hessians without relying on automatic differentiation or finite differences. To do so, we construct SE(3)-equivariant, symmetric Hessians from irreducible representation

Chiral sodium complex-catalyzed asymmetric cascade cycloaddition/hydration/1,2-rearrangement.

The research on the coordination chemistry and catalytic applications of sodium ions is of great importance. Inspired by the sodium ions-activated enzymes, we explored a sodium complex based on the synergistic coordination of polyfluorophosphate anions and water molecules. X-ray crystallography and computational chemistry revealed its unique spatial structure and highlighted the activation state of the water molecules within the chiral cavity. Furthermore, we have successfully applied the sodium complex in the catalysis of α-ketoimine-type 1,2-rearrangement, overcoming challenges of background

Oxidative Fluorination of Heteroatoms Enabled by Trichloroisocyanuric Acid and Potassium Fluoride.

In synthetic method development, the most rewarding path is seldom a straight line. While our initial entry into pentafluorosulfanyl (SF5 ) chemistry did not go according to plan (due to inaccessibility of reagents such as SF5 Cl at the time), a "detour" led us to establish mild and inexpensive oxidative fluorination conditions that made aryl-SF5 compound synthesis more accessible. The method involved the use of potassium fluoride and trichloroisocyanuric acid (TCICA)-a common swimming pool disinfectant-as opposed to previously employed reagents such as F2 , XeF2 , HF, and Cl2 . Thereafter, cu

Boradigermaallyl: (4+3) Cycloaddition-Initiated Boron Insertion into Benzene.

The 2π electron 1,3-dipole boradigermaallyl, valence-isoelectronic to an allyl cation, is synthesized from a bis(germylene). It reacts with benzene at room temperature by insertion of a boron atom into the benzene ring. Computational investigation of the mechanism shows the boradigermaallyl reacting with a benzene molecule in a concerted (4+3) or [π4s+π2s] cycloaddition reaction. Thus, the boradigermaallyl acts as a highly reactive dienophile in this cycloaddition reaction with nonactivated benzene as diene unit. This type of reactivity provides a novel platform for ligand assisted borylene in

Catalytic and Mechanistic Approach to the Metal-Free N-Alkylation of 2-Aminopyridines with Diketones.

N-alkylation of amines is an important catalytic reaction in synthetic chemistry. Herein, we report a simple strategy for the N-alkylation of 2-aminopyridines with 1,2-diketones using BF3·OEt2 as a catalyst. The reaction proceeds under aerobic conditions, leading to the formation of a diverse range of substituted secondary amines in good to excellent yields. A close inspection of the mechanistic pathway using various spectroscopic techniques and the computational study revealed that the reaction proceeds through the formation of an iminium-keto intermediate with the liberation of CO2.

Direct Stereodivergent Olefination of Carbonyl Compounds with Sulfur Ylides.

The reactivity of phosphorus and sulfur ylides toward carbonyl compounds constitutes a well-known dichotomy that is a common educational device in organic chemistry─the former gives olefins, while the latter gives epoxides. Herein, we report a stereodivergent carbonyl olefination that challenges this dichotomy, showcasing thiouronium ylides as valuable olefination reagents. With this method, aldehydes are converted to Z-alkenes with high stereoselectivity and broad substrate scope, while N-tosylimines provide a similarly proficient entry to E-alkenes. A chromium terephthalate-based solid with unusually large pore volumes and surface area.

We combined targeted chemistry and computational design to create a crystal structure for porous chromium terephthalate, MIL-101, with very large pore sizes and surface area. Its zeotype cubic structure has a giant cell volume (approximately 702,000 cubic angstroms), a hierarchy of extra-large pore sizes (approximately 30 to 34 angstroms), and a Langmuir surface area for N2 of approximately 5900 +/- 300 square meters per gram. Beside the usual properties of porous compounds, this solid has potential as a nanomold for monodisperse nanomaterials, as illustrated here by the incorporation of Keggi

Tetra- and penta-coordinated aluminum complexes with bis-amidine ligands and their reactivity for efficient CO2 transformation to cyclic carbonates.

The development of efficient catalysts for CO2 transformation into value-added products is crucial for sustainable chemistry. In this work, we report the synthesis and characterization of a new series of tetra- and penta-coordinated aluminum complexes supported by bis-amidine ligands featuring a rigid naphthalene bridge and their application as catalysts in the reaction of carbon dioxide with epoxides to form cyclic carbonates. Interestingly, while tetra- and penta-coordinated aluminum complexes bearing methyl groups are efficient when used with TBAI as a co-catalyst, their iodo analogs proved
