Clinical course and outcomes of corneal blood staining in open-globe injury: Eye Injury Vitrectomy Study.

PURPOSE: To describe the clinical course and outcomes of corneal blood staining in open-globe injuries (OGIs) after vitrectomy without routine immediate allograft penetrating keratoplasty (PKP). METHODS: This study included 42 eyes of 42 patients with corneal blood staining secondary to OGIs. Clinical data were retrospectively collected from the Eye Injury Vitrectomy Study (EIVS). All eyes underwent anterior chamber washout at the start of pars plana vitrectomy (PPV). When visualization remained inadequate, temporary keratoprosthesis (TKP)-assisted PPV was performed, followed by replacement o

Acute high intraocular pressure induced by vitreous cavity infusion: an animal model to evaluate corneal endothelial injuries.

Elevated intraocular pressure (IOP) is a critical factor in corneal endothelial cells (CECs) injury, yet the precise mechanisms remain poorly understood due to confounding variables in clinical studies. This study developed an acute high IOP model in New Zealand rabbits by vitreous cavity infusion to investigate the effects of high IOP on CECs. The observed anterior chamber shallowing and anterior shift of the lens and iris suggest that this model mechanistically mimics acute angle-closure glaucoma (AACG), particularly those cases driven by a posterior pushing mechanism. The model induced stab

Fenofibrate microemulsion eyedrops for treating nitrogen mustard induced corneal injury.

Vesicants are highly toxic chemicals used as warfare agents, primarily absorbed through the skin, inhalation, or ocular surface. Nitrogen mustard (NM), a potent vesicant, often leads to blindness due to the inadequacies of existing therapies. This study explores the role of peroxisome proliferator-activated receptor-α (PPARα) in NM-induced ocular injury and evaluates the efficacy of fenofibrate (Feno), a PPARα agonist, in preventing corneal injury progression. Feno, an FDA-approved oral medication used to treat dyslipidemia, has shown off-labelled potential in promoting corneal wound healing.

Proteomics analysis of the mechanism of the treatment of corneal injury in dry-eye mice.

Dry eye disease (DED) is a common ocular surface disorder affecting millions globally. Clinical and experimental studies have shown that the traditional Chinese medicine formula Qingxuan Runmu Yin decoction (QXRMY) is effective in treating DED. This study aimed to explore the molecular mechanisms of corneal damage in DED and evaluate QXRMY's therapeutic effects. A total of 120 C57BL/6 mice were divided into control, DED model, and QXRMY treatment groups. DIA sequencing of corneal tissue identified 2411 differentially expressed proteins. Enrichment analysis revealed these proteins were involved

Rapamycin Mitigates Corneal Damage in a Mouse Model of Alkali Burn Injury.

Alkali burns to the cornea cause severe damage characterized by an intense inflammatory response driven by inflammatory cytokines, which orchestrate pathological processes, including neovascularization, fibrosis, apoptosis, abnormal cell proliferation, and disorganization of the extracellular matrix (ECM), often resulting in permanent vision impairment or loss. Rapamycin (RAPA), a well-known mTOR inhibitor with potent immunosuppressive activity and pleiotropic therapeutic effects, was investigated as a novel restorative modality for promoting corneal wound healing in a mouse model of alkali bu

Antagonizing NK-1R modulates pain perception following corneal injury.

Substance P (SP) expressed by corneal nerves, is an 11-amino acid long neuropeptide from the tachykinin family, encoded by the Tac1 gene, and binds to neurokinin receptors. SP overexpression is associated with various pathological responses in the cornea including vasodilation, pain, inflammation, and angiogenesis in the normally avascular tissue. This study investigates the role of neurokinin-1 receptor (NK-1R) mediated signaling in nociception, nerve regeneration, and neuronal activation following mechanical corneal injury in mice. Corneal injuries were induced in age- and sex-matched C57BL/

The corneal fibroblast: The Dr. Jekyll underappreciated overseer of the responses to stromal injury.

PURPOSE: To review the functions of corneal fibroblasts in wound healing. METHODS: Literature review. RESULTS: Corneal fibroblasts arise in the corneal stroma after anterior, posterior or limbal injuries and are derived from keratocytes. Transforming growth factor (TGF) β1 and TGFβ2, along with platelet-derived growth factor (PDGF), are the major modulators of the keratocyte to corneal fibroblast transition, while fibroblast growth factor (FGF)-2, TGFβ3, and retinoic acid are thought to regulate the transition of corneal fibroblasts back to keratocytes. Adequate and sustained levels of TGFβ1

Mouse strain variation in SMA(+) myofibroblast development after corneal injury.

The purpose of present study was to investigate differences in myofibroblast development after haze-generating injury in different commonly used strains of mice. The inbred mouse strains used in this study were Balb/c, C57BL/6, C3H/HeJ and DBA/1J. All mice had uniform irregular phototherapeutic keratectomy with an excimer laser according to a previously published method to generate stromal haze. DBA/1J mice generated significantly greater density of alpha smooth muscle actin (SMA)-positive myofibroblasts in the anterior stroma compared to Balb/c (p < 0.05), C57BL/6 (p < 0.05) and C3H/HeJ (p <

Role of matrix metalloproteinases in failure to re-epithelialize after corneal injury.

Delayed re-epithelialization of the cornea after injury usually precedes stromal ulceration. Previous findings using a rat thermal injury model suggested that re-epithelialization is impeded by products of resident corneal cells, which destroy adhesive structures at the basement membrane zone. In this study, we provide additional evidence for this concept. Failure to re-epithelialize was found to correlate with an increase in the amounts of gelatinolytic matrix metalloproteinases present in the rat cornea. One of these gelatinases, gelatinase B, is synthesized by the resident corneal cells, an

Corneal injury alters eicosanoid formation in the rabbit anterior segment in vivo.

The author injected [1-14C]arachidonic acid into the anterior chamber of the rabbit eye, and its metabolism was studied in vivo in the various eye tissues. Incorporation of the radioisotope was analyzed in individual phospholipids and neutral lipids, and its conversion to oxygenated metabolites was evaluated in the three corneal layers and the aqueous humor, iris-ciliary body, lens, vitreous, and "rest of eye." Three hours after injection, 30% of the label was in corneal stroma and 25% in iris-ciliary bodies. The posterior part of the eye retained 40%, and the remainder was distributed unequal

LM22B-10 promotes corneal nerve regeneration through in vitro 3D co-culture model and in vivo corneal injury model.

Corneal nerve wounding often causes abnormalities in the cornea and even blindness in severe cases. In this study, we construct a dorsal root ganglion-corneal stromal cell (DRG-CSC, DS) co-culture 3D model to explore the mechanism of corneal nerve regeneration. Firstly, this model consists of DRG collagen grafts sandwiched by orthogonally stacked and orderly arranged CSC-laden plastic compressed collagen. Nerve bundles extend into the entire corneal stroma within 14 days, and they also have orthogonal patterns. This nerve prevents CSCs from apoptosis in the serum withdrawal medium. The conditi
