Effects of anesthesia and blood sampling techniques on plasma metabolites and corticosterone in the rat.

Blood is routinely sampled from laboratory animals in biomedical research, and many of the commonly applied sampling techniques require anesthesia. Acute effects of many sampling and anesthesia procedures may confound the results, but those effects are incompletely characterized. We here compare the effects of four common anesthesia procedures (inhalation anesthesia with ether (EA) or isoflurane (IA) and intraperitoneal injection anesthesia with xylazin/ketamine (XKA) or medetomidine/midazolam/fentanyl (MMFA)) on plasma concentrations of glucose, lactate, non-esterified fatty acids (NEFAs), an

Plasma concentrations of corticosterone and buprenorphine in rats subjected to jugular vein catheterization.

The present study investigated the postoperative plasma concentrations of corticosterone and buprenorphine in male Wistar and Sprague-Dawley rats, treated with buprenorphine administered either through subcutaneous (SC) injection or through voluntary ingestion (VI). The animals were treated with buprenorphine for pre-emptive analgesia prior to surgical placement of a jugular catheter, followed by automated blood sampling during 96 h. Buprenorphine was administered on a regular basis throughout the experiment, and blood was collected on selected time points. Body weight was measured before and

Prodynorphin-derived peptides are critical modulators of anxiety and regulate neurochemistry and corticosterone.

Stress and anxiety are mainly regulated by amygdala and hypothalamic circuitries involving several neurotransmitter systems and providing physiological responses to peripheral organs via the hypothalamic-pituitary-adrenal axis and other pathways. The role of endogenous opioid peptides in this process is largely unknown. Here we show for the first time that anxiolytic parameters of explorative behavior in mice lacking prodynorphin were increased 2-4-fold in the open field, the elevated plus maze and the light-dark test. Consistent with this, treatment of wild-type mice with selective kappa-opio

Aldosterone-, corticosterone- and cortisol-secreting adrenocortical carcinoma in a dog: case report.

A 12-year-old, intact female beagle exhibited symptoms of polyuria-polydipsia and hyperorexia for two months. Blood tests showed elevated asparate aminotransferase, alanine aminotransferase, alkaline phosphatase and creatine kinase levels, as well as marked hypokalemia. The results of adrenocorticotropic hormone stimulation test showed elevated cortisol, aldosterone and corticosterone concentrations. Abdominal ultrasonography confirmed a mass in the left adrenal gland. Masses were also seen in the liver and caudal vena cava. Diagnosis was a tumor of the adrenal cortex with metastases. Trilosta

Effect of housing density on reproductive parameters and corticosterone levels in nursing mice.

The Guide for the Care and Use of Laboratory Animals (Guide) recommends minimum floor space per mouse based on weight, with no other factors considered. We conducted a randomized experiment to evaluate the effect of housing density on reproductive indices and corticosterone levels in lactating mice. Female mice matched for age, strain, and date-of-pregnancy were housed individually. At parturition the dams were randomly allocated to have litters culled or remain intact. The experimental group had litters culled to meet the Guide's space density requirement. Litters of the second group were mai

Timing is essential for rapid effects of corticosterone on synaptic potentiation in the mouse hippocampus.

Stress facilitates memory formation, but only when the stressor is closely linked to the learning context. These effects are, at least in part, mediated by corticosteroid hormones. Here we demonstrate that corticosterone rapidly facilitates synaptic potentiation in the mouse hippocampal CA1 area when high levels of the hormone and high-frequency stimulation coincide in time, but not when corticosterone is given either before or after repetitive stimulation. This effect could not be blocked by antagonists of the mineralocorticoid receptor and glucocorticoid receptor (spironolactone and RU 38486

Serum leptin and corticosterone levels after exposure to noise stress in rats.

Even though extensive studies have been conducted on the effect of noise exposure on hearing apparatus / auditory system, information on the effect of noise on the other body functions is sparse. The present study examined the effect of exposure of albino rats to acute and chronic noise stress on two important interlaced endocrine levels. In acute experiments the animals were exposed to 120 dB noise for a duration of 1, 2, 3 hrs. In chronic experiments the animals were exposed to noise for one hour daily for 30, 60 and 90 days. Plasma corticosterone and leptin levels were measured in these ani

Mineralocorticoid receptors are indispensable for nongenomic modulation of hippocampal glutamate transmission by corticosterone.

The adrenal hormone corticosterone transcriptionally regulates responsive genes in the rodent hippocampus through nuclear mineralocorticoid and glucocorticoid receptors. Via this genomic pathway the hormone alters properties of hippocampal cells slowly and for a prolonged period. Here we report that corticosterone also rapidly and reversibly changes hippocampal signaling. Stress levels of the hormone enhance the frequency of miniature excitatory postsynaptic potentials in CA1 pyramidal neurons and reduce paired-pulse facilitation, pointing to a hormone-dependent enhancement of glutamate-releas

Relative and combined effects of estradiol and prolactin on corticosterone secretion in ovariectomized rats.

In vivo and in vitro experiments were designed to assess the relationship of the estradiol (E2) and prolactin (PRL) on glucocorticoid secretion in ovariectomized (Ovx) rats. Female rats were Ovx for two weeks and then subcutaneously injected with oil or estradiol benzoate (EB) for 3 days before experimentation. Venous blood samples were collected from right jugular vein at 0, 30, 60, 90, and 120 min after challenge with adrenocorticotropin (ACTH). Adrenal zona fasciculata-reticularis (ZFR) cells from Ovx rats were isolated and incubated with E2 or PRL. In the morning and afternoon, EB enhanced

The role of corticosterone in food deprivation-induced reinstatement of cocaine seeking in the rat.

RATIONAL AND OBJECTIVES: Acute 1-day food deprivation stress reinstates heroin seeking in rats, but the generality of this effect to other drugs, and its underlying mechanisms, are largely unknown. Here we studied whether food deprivation would reinstate cocaine seeking and whether the stress hormone, corticosterone, is involved in this effect. METHODS: Rats were trained to press a lever for cocaine for 10-12 days (0.5-1.0 mg/kg per infusion, IV, 4 h/day) and were then divided into four groups that underwent different manipulations of plasma corticosterone levels: (1) bilateral adrenalectomy

Effects of adrenalectomy and corticosterone replacement on the hypothalamic-pituitary response to neural stimuli.

In this study we examined whether circulating glucocorticoids (GC) have a permissive facilitatory role in the hypothalamo-pituitary-adrenal (HPA) axis responses to neural or metabolic stimuli. In control sham operated rats the exposure to photic or acoustic neural stimuli and to either cytoglucopenia induced by 2-deoxyglucose (2-DG) or to hypoglycemia induced by insulin caused a significant 5-fold increase in serum ACTH as compared to basal non-stress levels. In adrenalectomized (Adex) rats tested under basal conditions at 4, 7 and 14 days post-Adex, serum ACTH gradually increased in a time-de

Circadian rhythm of Arg-vasopressin contents in the suprachiasmatic nucleus in relation to corticosterone.

Circadian rhythms of locomotor activity and adrenal glucocorticoid are controlled by the suprachiasmatic nucleus (SCN), the center of a biological clock, in mammals. Arg-vasopressin (AVP) contents in the SCN play a role in endogenous circadian rhythm during the absence of time cues. The AVP-containing neurons in the SCN are considered to transmit a circadian signal to the other parts of the brain. The circadian rhythms of AVP in the SCN in relation to the plasma corticosterone and locomotor activity were investigated. Under the light-dark cycle, plasma corticosterone levels were reciprocally c

The effect of corticosterone on reactivity to spatial novelty is mediated by central mineralocorticosteroid receptors.

Corticosterone, secreted by the adrenal glands, binds to central mineralocorticoid receptors with high affinity and to glucocorticoid receptors with a tenfold lower affinity. In previous studies we have shown that the selective activation of either mineralocorticoid receptors or glucocorticoid receptors exerts distinctly different behavioural effects. In this study we examined in particular the mineralocorticoid receptor-mediated effect of corticosterone on the control of the behavioural response of male Wistar rats to spatial novelty. This analysis was based on our observation that in adrenal

Mineralocorticoid receptors mediate the enhancing effects of corticosterone on convulsion susceptibility in mice.

With hippocampal slice electrophysiology, direct evidence that mineralocorticoid receptors (MR) mediate excitatory effects of corticosteroids on neuronal activity was found. The present experiments extended this hypothesis to a whole animal model. The effects of manipulations of MR binding on convulsions produced by pentylenetetrazol (PTZ), strychnine and kainic acid were examined. Moderate increases in plasma corticosterone levels resulted in enhanced susceptibility to several convulsion types; decreased levels attenuated the susceptibility. The proconvulsant effects of corticosterone were in

Effects of chronic stressors or corticosterone treatment on the septohippocampal cholinergic system of the rat.

The effects of prolonged (2 months) corticosterone (CORT) treatment on several cholinergic markers of various brain areas were compared to the effects of prolonged intermittent exposure to stress. CORT, but not stress, caused a significant reduction in the number of acetylcholinesterase-stained neurons in the medial septal area. Neither treatment resulted in any hippocampal pyramidal cell loss. It is concluded that a time-dependent degeneration of the septohippocampal cholinergic system follows 2 months of CORT administration but not chronic intermittent stress of this duration.

Regulation of the release of neuronally localized corticosterone in rat forebrain areas by catecholamines.

The influence of intralimbic and intrahypothalamic application of catecholamine-related compounds on the release of neuronally localized corticosterone in the central amygdala, ventral hippocampus and medial hypothalamus was investigated under in vivo and in vitro conditions. It is reported that agonists and antagonists of the alpha-adrenergic system did not affect the release of corticosterone in the brain areas studied. In contrast, the beta-adrenoreceptor agonist isoprenaline markedly enhanced the release of corticosterone. This response is prevented in the presence of the beta-adrenorecept

LPS-caused secretion of corticosterone is mediated by histamine through histidine decarboxylase.

Escherichia coli lipopolysaccharide (LPS) induced strong time- and dose-dependent secretion of corticosterone (CS) in C3H/HeN mice. In contrast, C3H/HeJ mice were very insensitive to LPS; 100,000 times more LPS was required with C3H/HeJ mice for producing a similar degree of CS secretion as that of C3H/HeN mice. However, C3H/HeJ mice could efficiently respond to other types of stressors, immobilization stress or injection of histamine, a possible mediator of LPS-induced CS secretion (28), leading to a striking increase in the serum levels of CS. Adoptive transfer of spleen cells from C3H/HeN m

The hippocampal corticosterone receptor system of the homozygous diabetes insipidus (Brattleboro) rat.

The binding of [3H] corticosterone to hippocampal cytosol receptors of Brattleboro rats homozygous for diabetes insipidus (Ho-Di) and of normal Brattleboro rats (Ho-No) was investigated at 24 h after removal of the adrenals. The apparent maximal binding capacity of the Ho-Di hippocampal corticosterone receptor system was about 30% less than that of the Ho-No rats. Substitution of the vasopressin deficient rats with 1E pitressin tannate in oil partially restores the hippocampal corticosterone receptor level towards that of the control animals.
