Bleeding Complication in a Patient with Concomitant Use of Rivaroxaban and Saffron Supplement: A Case Report.

BACKGROUND: Direct oral anticoagulants (DOACs) carry a lower potential risk of food/herb and drug interactions compared to oral vitamin K antagonists. However, as a new class of medications, drug interactions of DOACs have not been fully known. CASE PRESENTATION: We herein present the case of a 64-year old male with the complaint of acute onset epistaxis and bleeding gums following the concomitant use of rivaroxaban and saffron supplement. It seems that coadministration of DOACs and saffron supplements should be avoided due to the potential drug-herbal interactions and possible risk of subseq

Chemical analysis of saffron by HPLC based crocetin estimation.

Saffron is one of the most expensive and valuable spice having tremendous commercial value in food industry and thus its quality check is of utmost importance. Crocins are unique group of extremely hydrophilic apocarotenoids which are main components of saffron. Crocetin is an aglycone of crocins which do occur naturally in saffron, and is produced in biological system as a bioactive metabolite via hydrolytic cleavage of crocins. Crocins are unstable and tend to undergo isomerisation/ inter-conversions, and therefore their quantitative estimation is difficult. Herein, we have established for t

Saffron: Chemical Composition and Neuroprotective Activity.

Crocus sativus L. belongs to the Iridaceae family and it is commonly known as saffron. The different cultures together with the geoclimatic characteristics of the territory determine a different chemical composition that characterizes the final product. This is why a complete knowledge of this product is fundamental, from which more than 150 chemical compounds have been extracted from, but only about one third of them have been identified. The chemical composition of saffron has been studied in relation to its efficacy in coping with neurodegenerative retinal diseases. Accordingly, experimenta

Metabolic engineering of rice endosperm for crocin biosynthesis.

Crocins, the main active ingredients in Crocus sativus, have anti-inflammatory, antioxidant, hypolipidemic, hypoglycemic, and antitumor properties. Due to the harsh growing conditions and the limited yield of C. sativus, sustainable alternative methods for crocin production need to be explored. In this study, a new rice germplasm resource, "crocin rice", was developed by expressing eight genes of the crocin synthesis pathway in rice endosperm. The content of crocins in rice seeds was up to 9.25 μg/g dry weight, and it was also rich in a variety of carotenoids, including α-carotene, lutein, and

In vitro starch digestibility and fate of crocins in pasta enriched with saffron extract.

This work aims to study the effect of the addition of saffron extract on fresh pasta in-vitro digestibility. Fresh pasta was formulated with different concentrations of saffron extracts (0.2 and 0.4 %w/w), cooked at two different times (1.5 and 3 min), and in vitro digested (oral, gastric and intestinal stages). Oil was added to pasta before digestion to evaluate the presence of lipids on starch and crocin bioaccessibility. Saffron enrichment and oil addition slowed down the digestion of starch, thus, decreasing the glycemic index of pasta. Concentration of saffron and oil addition contributed

Efficacy of Hydroponically Cultivated Saffron in the Preservation of Retinal Pigment Epithelium.

Saffron treatment is a broad-spectrum therapy used for several retinal diseases, and its effectiveness depends on a particular molecular composition (REPRON® saffron). Its production requires specific crops and procedures that, together with low yields, make this spice expensive. To reduce costs, the use of hydroponic crops is gradually increasing. In this study, we tested the protective properties of a hydroponic saffron (sH) batch in models of retinal pigmented epithelium (RPE) degeneration. ARPE-19 cells were pretreated with 40 µg/mL saffron and exposed to different types of damage: excess

Saffron Extract Attenuates Anxiogenic Effect and Improves Cognitive Behavior in an Adult Zebrafish Model of Traumatic Brain Injury.

Traumatic brain injury (TBI) has the highest mortality rates worldwide, yet effective treatment remains unavailable. TBI causes inflammatory responses, endoplasmic reticulum stress, disruption of the blood-brain barrier and neurodegeneration that lead to loss of cognition, memory and motor skills. Saffron (Crocus sativus L.) is known for its anti-inflammatory and neuroprotective effects, which makes it a potential candidate for TBI treatment. Zebrafish (Danio rerio) shares a high degree of genetic homology and cell signaling pathways with mammals. Its active neuro-regenerative function makes i

Cytotoxic effect against 3T3 fibroblasts cells of saffron floral bio-residues extracts.

For every kilogram of saffron spice produced, about 63 kg of floral bio-residues (FB) (tepals, stamens and styles) are thrown away. Extracts of these bio-residues in water (W1), water:HCl (100:1, v/v) (W2), ethanol (E3), ethanol:HCl (100:1, v/v) (E4), dichloromethane (D5) and hexane (H6) were prepared. Their composition in flavonols and anthocyanins, and their effect on cell viability were determined. W1 was the richest in kaempferol 3-sophoroside (30.34 mg/g dry FB) and delphinidin 3,5-diglucoside (15.98 mg/g dry FB). The highest tested concentration (900 μg/ml) of W1, W2, E4, D5 and H6 did n

Exploring the therapeutic potential of crocin, a bioactive component of saffron, in neurocognitive deficit and pulmonary impairment induced by inhalational exposure to petroleum vapours.

BACKGROUND: Long-term recurrent exposure to petroleum fumes poses a substantial occupational risk, specifically among individuals in gasoline stations. They are routinely exposed to the inherent toxic constituents of petrol and diesel, such as hydrocarbons, VOCs, and trace elements, during their routine activities, including fuel dispensing, handling, storage, transport, and refining processes. Due to inadequate safety measures allow these toxicants into the lungs, cross the pulmonary barrier, enter the circulation and subsequently reach the brain through disrupting the lung-brain axis (LBA).

Effect of saffron, black seed, and their main constituents on inflammatory cytokine response (mainly TNF-α) and oxidative stress status: an aspect on pharmacological insights.

Tumor necrosis factor-α (TNF-α), an inflammatory cytokine, is produced by monocytes and macrophages. It is known as a 'double-edged sword' because it is responsible for advantageous and disadvantageous events in the body system. The unfavorable incident includes inflammation, which induces some diseases such as rheumatoid arthritis, obesity, cancer, and diabetes. Many medicinal plants have been found to prevent inflammation, such as saffron (Crocus sativus L.) and black seed (Nigella sativa). Therefore, the purpose of this review was to assess the pharmacological effects of saffron and black s
