Systematic determination of disulfide bond reduction potentials reveals a nonequilibrium redox hierarchy in cyanobacteria.

Disulfide bonds act as reversible switches that regulate cellular function in nearly all organisms. Their behavior is set by the midpoint potential (Em), yet Em is known for only a small number of sites, mostly in purified proteins studied away from their natural partners. We developed a mass-spectrometry workflow that measures Em directly from native cell lysates. By equilibrating proteins of the cyanobacterium Synechocystis sp. PCC 6803 in defined redox buffers and reading out the oxidation state of individual cysteines, we obtained 368 Em values across the proteome and validated them agains

The calcium-binding protein CSE links Ca2+ signaling with cell-cell communication in cyanobacteria.

Multicellular cyanobacteria have evolved sophisticated cell-cell communication machinery to exchange, synchronize, and coordinate the efforts of individual cells. Analogous to gap junctions that have traditionally been regarded as a eukaryotic feature, multicellular cyanobacteria coordinate their cell-cell communication via septal junctions (SJs). However, the signals that regulate cell-cell communication and septal-junction assembly are largely unknown. Lately, calcium signaling has been implicated in regulating cell junctions in eukaryotes. We recently discovered a Ca2+-binding protein, CSE,

Unraveling the mechanism of greenhouse gas emissions during allelochemical-triggered inhibition of cyanobacteria recovery.

Climate change amplifies cyanobacterial blooms, creating a feedback loop between warming and bloom intensity. This dynamic emphasizes the need to address greenhouse gas (GHG) emissions during in-situ cyanobacterial control, minimizing algicides use and its long-term climatic impacts. However, research on the dose-response of algicides on GHG emissions is limited. This study investigated the dose-dependent effects of artemisinin sustained-release algicides (ASAs) on GHG emissions and microbial dynamics during cyanobacteria recovery prevention. Microcosm experiments revealed that ASAs dosage res

Acute Toxicity of Carbon Nanotubes, Carbon Nanodots, and Cell-Penetrating Peptides to Freshwater Cyanobacteria.

Synthetic non-metallic nanoparticles (NMNPs) such as carbon nanotubes (CNTs), carbon nanodots (CNDs), and cell-penetrating peptides (CPPs) have been explored to treat harmful algal blooms. However, their strain-specific algicidal activities have been rarely investigated. Here we determined their acute toxicity to nine freshwater cyanobacterial strains belonging to seven genera, including Microcystis aeruginosa UTEX 2386, M. aeruginosa UTEX 2385, M. aeruginosa LE3, Anabaena cylindrica PCC 7122, Aphanizomenon sp. NZ, Planktothrix agardhii SB 1810, Synechocystis sp. PCC 6803, Lyngbya sp. CCAP 144

Stalling the Course of Neurodegenerative Diseases: Could Cyanobacteria Constitute a New Approach toward Therapy?

Neurodegenerative diseases (NDs) are characterized by progressive and irreversible neuronal loss, accompanied by a range of pathological pathways, including aberrant protein aggregation, altered energy metabolism, excitotoxicity, inflammation, and oxidative stress. Some of the most common NDs include Alzheimer's Disease (AD), Parkinson's Disease (PD), Multiple Sclerosis (MS), Amyotrophic Lateral Sclerosis (ALS), and Huntington's Disease (HD). There are currently no available cures; there are only therapeutic approaches that ameliorate the progression of symptoms, which makes the search for new

Dissolved organic nitrogen, dinoflagellates and cyanobacteria in two eutrophic lakes: Analysis by inferential modelling.

The Lake Suwa (Japan) has a history of non-N-fixing Microcystis blooms. Lake Kinneret (Israel) experienced multiannual periods of sole domination by the dinoflagellate Peridinium gatunense and periods dominated seasonally by P. gatunense or cyanobacteria. Extensive studies have been carried out in both lakes regarding the role of dissolved inorganic nitrogen and phosphorus as drivers of primary productivity. There is growing evidence that dissolved organic nitrogen (DON) compounds also influence not only biomass and structure of phytoplankton communities but also microcystin production. This s

The rice field cyanobacteria Anabaena azotica and Anabaena sp. CH1 express vanadium-dependent nitrogenase.

Anabaena azotica FACHB-118 and Anabaena sp. CH1, heterocystous cyanobacteria isolated from Chinese and Taiwanese rice fields, expressed vanadium-containing nitrogenase when under molybdenum deficiency. This is the second direct observation of an alternative nitrogenase in cyanobacteria. The vanadium nitrogenase-specific genes vnfDG are fused and clustered in a phylogenetic tree next to the corresponding genes of Methanosarcina. The expression of vnfH in cells cultured in Mo-free medium and of nifH in Mo-grown cells was shown for the first time by sequencing cDNA derived from cultures of A. azo

Cyanobacteria-derived near-infrared autofluorescent exosomes enabling synergistic brain lesion imaging and neuroprotection.

Precision diagnosis and treatment of central nervous system (CNS) diseases are hindered by limited probe penetration, toxicity risks, and low imaging signal-to-noise ratio (SNR). The blood-brain barrier (BBB) further restricts drug delivery, especially in stroke therapy. This study proposes and validates a natural exosome (sExos) from cyanobacteria, featuring intrinsic near-infrared-I (NIR-I) autofluorescence, with strong imaging and neuroprotective functions. As a theranostic nanoplatform, sExos enable integrated diagnosis and treatment of stroke and other brain disorders. Enriched with the f

Investigation of cyanobacteria-hosted antibiotic resistance genes in cyanoHAB-impacted drinking water sources.

Freshwater cyanobacterial blooms (cyanoHABs) are expanding across the world, and their frequency and severity are becoming more intense due to prevalent eutrophication and a changing climate. Traditionally, the concerns about cyanoHABs have mainly focused on cyanotoxins in water. CyanoHABs are also hypothesized to play a role in the antibiotic resistome, but whether cyanobacteria host clinically relevant antibiotic resistant genes (ARGs) in the environment is largely unknown. To investigate this emerging issue, we examined whether cyanobacteria host ARGs within the broader microbiome context.

Engineering Cyanobacteria for High-Yield Photosynthetic Isoprene Production With Long-Term Phenotypic Stability.

In light of the looming climate crisis, a key cornerstone for a sustainable bioeconomy is photosynthetic production of chemicals and fuels from CO2 and water, powered by sunlight. Isoprene is a five-carbon volatile hydrocarbon with industrial use as a feedstock for rubber production and chemical synthesis and is, at present, generated from crude oil sources. It is, however, possible to produce isoprene using photoautotrophic microorganisms such as cyanobacteria, heterologously expressing the plant enzyme isoprene synthase. Here, we have employed diverse metabolic engineering strategies to deve

Rare cyanobacteria drive nitrogen-fixation and cyanotoxin production in an Aphanizomenon-dominated bloom.

Late summer, recurring cyanobacterial blooms in Lake of the Woods (LOW) are polycyanobacterial and typically dominated by Aphanizomenon flos-aquae. LOW waters are typically nitrogen limited, relative to phosphorus. As such, the dominance of Aphanizomenon flos-aquae, a putative nitrogen-fixing cyanobacterium, suggests that its ability to fix nitrogen may be advantageous and aid in its ability to bloom. This study sought to quantify nitrogen fixation rates and identify cyanotoxin-producing species during the blooms. Throughout the 2021 season, we quantified nutrients, N-fixation rates, microbial

COVID-19 pharmaceuticals in aquatic matrices: The threatening effects over cyanobacteria and microalgae.

Water contamination by pharmaceuticals is a global concern due to their potential negative effects on aquatic ecosystems and human health. This study examined the presence of three repositioned drugs used for COVID-19 treatment: azithromycin (AZI), ivermectin (IVE) and hydroxychloroquine (HCQ) in water samples collected from three urban rivers in Curitiba, Brazil, during August and September 2020. We conducted a risk assessment and evaluated the individual (0, 2, 4, 20, 100 and 200 μg.L-1) and combined (mix of the drugs at 2 μg.L-1) effects of the antimicrobials on the cyanobacterium Synechoco

Cyanobacteria-based self-oxygenated photodynamic therapy for anaerobic infection treatment and tissue repair.

Photodynamic therapy (PDT) is an important technique to deal with drug-resistant bacterial infections in the post-antibiotic era. However, the hypoxic environment in intractable infections such as refractory keratitis and periodontitis, makes PDT more difficult. In this work, spontaneous oxygen-producing cyanobacteria were used as the carrier of photosensitizer (Ce6), and ultrasmall Cu5.4O nanoparticles (Cu5.4O USNPs) with catalase activity for infection and inflammation elimination and rapid tissue repair (CeCycn-Cu5.4O). The loading of Ce6 and Cu5.4O USNPs onto cyanobacteria surface were con

Glycogen Metabolism Supports Photosynthesis Start through the Oxidative Pentose Phosphate Pathway in Cyanobacteria.

Cyanobacteria experience drastic changes in their carbon metabolism under daily light/dark cycles. During the day, the Calvin-Benson cycle fixes CO2 and diverts excess carbon into glycogen storage. At night, glycogen is degraded to support cellular respiration. The dark/light transition represents a universal environmental stress for cyanobacteria and other photosynthetic lifeforms. Recent studies revealed the essential genetic background necessary for the fitness of cyanobacteria during diurnal growth. However, the metabolic processes underlying the dark/light transition are not well understo

Determination of FVIIa-sTF Inhibitors in Toxic Microcystis Cyanobacteria by LC-MS Technique.

The blood coagulation cascade involves the human coagulation factors thrombin and an activated factor VII (fVIIa). Thrombin and fVIIa are vitamin-K-dependent clotting factors associated with bleeding, bleeding complications and disorders. Thrombin and fVIIa cause excessive bleeding when treated with vitamin-K antagonists. In this research, we explored different strains of toxic Microcystis aeruginosa and cyanobacteria blooms for the probable fVIIa-soluble Tissue Factor (fVIIa-sTF) inhibitors. The algal cells were subjected to acidification, and reverse phase (ODS) chromatography-solid phase ex

Urlárins, chlorinated bis-indoles from a benthic mat of cyanobacteria in the West of Ireland.

Cyanobacteria are well-known for producing a diverse array of specialised metabolites, with increasing occurrences due to our changing climate. Though some cyanobacterial proliferations are harmless, others produce toxic compounds with detrimental effects to their surrounding environment including human health. A large cyanobacterial accumulation proliferating at Lough Urlár, Co. Mayo, Ireland, was selected for further chemical analysis owing to the presence of several polychlorinated metabolites detected by LC-MS in the methanolic fraction, which also displayed cytotoxic activity. After deep
