HIV Replication Under High-Level Cabotegravir Is Associated with the Appearance of 3'-PPT Mutations, Circular DNA Transcription and Recombination.

The HIV integrase inhibitor, dolutegravir (DTG), in the absence of eliciting integrase (int) resistance, has been reported to select mutations in the virus 3'-polypurine tract (3'-PPT) adjacent to the 3'-LTR U3. An analog of DTG, cabotegravir (CAB), has a high genetic barrier to drug resistance and is used in formulations for treatment and long-acting pre-exposure prophylaxis. We examined whether mutations observed for DTG would emerge in vitro with CAB. HIV-1IIIB was cultured in paired experiments of continuous high (300 nM) CAB initiated 2 h or 24 h after infection. After eight months of CAB

Differential HBV replicative markers and covalently closed circular DNA transcription in immune-active chronic hepatitis B with and without HBeAg.

BACKGROUND AND AIMS: Molecular processes driving immune-active chronic hepatitis B (CHB) with and without hepatitis B e antigen (HBeAg) remain incompletely understood. This study aimed to investigate expression profiles of serum and intrahepatic HBV markers and replicative activity of HBV in CHB patients with or without HBeAg. METHODS: This study recruited 111 untreated immune-active CHB (60 HBeAg-positive and 51 HBeAg-negative) patients and quantified intrahepatic covalently closed circular DNA (cccDNA), pre-genomic RNA (pgRNA), total HBV DNA (tDNA), and replicative intermediates as well as

DNA, a target of mixed chelate copper(II) compounds (Casiopeinas®) studied by electrophoresis, UV-vis and circular dichroism techniques.

Casiopeinas are a family of mixed chelate copper(II) complexes with antiproliferative and antineoplastic activities, results that have positioned them as an alternative for cancer treatment. Because DNA is one of their principal targets, it is of our interest to find out the effect of substituents on the diiamine ligands over mode of interaction. Therefore, we studied 21 Casiopeinas upon DNA by gel electrophoresis, UV-vis and circular dichroism (CD) spectroscopic techniques, previously studied by DFT calculations and Quantitative Structure-Activity Relationship (QSAR). According to electrophor

Droplet digital PCR technique is ultrasensitive for the quantification of covalently closed circular DNA in the blood of chronic HBV-infected patients.

BACKGROUND: Covalently closed circular DNA (cccDNA) is a stable, episomal form of HBV DNA. cccDNA is a true marker for the intrahepatic events in controlled CHB infection. Quantifying cccDNA is critical for monitoring disease progression, and efficacy of anti-viral therapies. METHODS: To standardize the method, total HBV DNA was isolated from HepAD38 cells and digested with three exonuclease enzymes to remove linear and relaxed circular HBV DNA. Purified cccDNA quantification used ddPCR with specific primers. Treatment-naive chronic hepatitis B virus patients (nCHBV, n=36) with detectable HBV

Recombinant covalently closed circular DNA of hepatitis B virus induces long-term viral persistence with chronic hepatitis in a mouse model.

UNLABELLED: Covalently closed circular DNA of hepatitis B virus (HBV) is critical for viral persistence in vivo. We recently reported a technique involving recombinant covalently closed circular DNA (rcccDNA) of HBV by site-specific DNA recombination. Using hydrodynamic injection, rcccDNA induces a temporarily prolonged HBV antigenemia in immunocompetent mice, similar to acute resolving HBV infection. In this study, we simulated the pathophysiological impact of chronic hepatitis to reproduce rcccDNA persistence in mouse models. We showed that rcccDNA achieved long-lasting persistence in the pr

Type III Druantia two-component antiphage defense depends on the DruH-DruE interaction for inhibiting viral DNA circularization and replication.

Bacteria have evolved multiple immune systems to resist phage invasion, however, only a small part of the defensive mechanisms have been clearly uncovered. In this study, we report a type III Druantia two-component defense system, DruH-DruE, identified from Mycolicibacterium smegmatis (M. smegmatis). The DruH-DruE prevents phage DNA circularization and replication. DruE can be replaced from the defense system by either homolog in M. tuberculosis or M. smegmatis. The physical interaction between these two components is essential for fighting against phage infection. Mutations in the interaction

Identification and characterization of a G-quadruplex structure in the pre-core promoter region of hepatitis B virus covalently closed circular DNA.

Approximately 250 million people worldwide are chronically infected with the hepatitis B virus (HBV) and are at increased risk of developing cirrhosis and hepatocellular carcinoma. The HBV genome persists as covalently closed circular DNA (cccDNA), which serves as the template for all HBV mRNA transcripts. Current nucleos(t)ide analogs used to treat HBV do not directly target the HBV cccDNA genome and thus cannot eradicate HBV infection. Here, we report the discovery of a unique G-quadruplex structure in the pre-core promoter region of the HBV genome that is conserved among nearly all genotype

SIRT7 Promotes Hepatitis B Virus Transcription via the Upregulation of DDB1 Expression and Facilitation of DDB1 Recruitment to Covalently Closed Circular DNA.

BACKGROUND: Transcription of the hepatitis B virus (HBV) is regulated by the acetylation status of H3/H4 histones bound to the covalently closed circular DNA (cccDNA) minichromosome. Thus, this study aimed to investigate whether sirtuin 7 (SIRT7), an NAD⁺-dependent acetylated lysine 18 of histone H3 (H3K18Ac) deacetylase, represses HBV transcription by catalyzing H3K18 deacetylation on the cccDNA minichromosome. METHODS: We investigated the effects of SIRT7 overexpression/knockdown on HBV transcription and histone acetylation in HepG2.2.15 cells, which constitutively express HBV, and HBV-infe

Quantification and epigenetic evaluation of the residual pool of hepatitis B covalently closed circular DNA in long-term nucleoside analogue-treated patients.

Hepatitis B virus (HBV) covalently closed circular (ccc)DNA is the key genomic form responsible for viral persistence and virological relapse after treatment withdrawal. The assessment of residual intrahepatic cccDNA levels and activity after long-term nucleos(t)ide analogues therapy still represents a technical challenge. Quantitative (q)PCR, rolling circle amplification (RCA) and droplet digital (dd)PCR assays were used to quantify residual intrahepatic cccDNA in liver biopsies from 56 chronically HBV infected patients after 3 to 5 years of telbivudine treatment. Activity of residual cccDNA

The genome of Oryctes rhinoceros nudivirus provides novel insight into the evolution of nuclear arthropod-specific large circular double-stranded DNA viruses.

The Oryctes rhinoceros nudivirus (OrNV) is a dsDNA virus with enveloped, rod-shaped virions. Its genome is 127,615 bp in size and contains 139 predicted protein-coding open reading frames (ORFs). In-depth genome sequence comparisons revealed a varying number of shared gene homologues, not only with other nudiviruses (NVs) and baculoviruses, but also with other arthropod-specific large dsDNA viruses, including the so-called Monodon baculovirus (MBV), the salivary gland hypertrophy viruses (SGHVs) and white spot syndrome virus (WSSV). Nudivirus genomes contain 20 baculovirus core gene homologues

Zinc finger protein ZBTB7A suppresses hepatitis B virus replication by attenuating FXRα-mediated HBV transcription and lowering covalently closed circular DNA levels.

Chronic hepatitis B virus (HBV) infection remains a major global public health challenge, partly because of significant gaps in our understanding of its pathogenesis. Host factors play critical roles in regulating specific stages of the HBV life cycle, including replication. ZBTB7A, a member of the POK family of zinc-finger transcription factors, is involved in carcinogenesis and the modulation of certain viral infections. However, its role in HBV replication remains unclear, and this study aims to address this. We observed that ZBTB7A mRNA levels were higher in the immune-control phase than i

A New Role of Class A Hepatitis B Virus Capsid Assembly Modulators in Core Protein Dynamics and Covalently Closed Circular DNA Replenishment.

BACKGROUND & AIMS: Current treatments of chronic hepatitis B are rarely curative. Capsid assembly modulators target capsid formation by the hepatitis B virus core protein. Class A capsid assembly modulators induce abnormal core protein assembly, but how they affect core protein dynamics and inhibit hepatitis B virus replication is only partially understood. METHODS: Human liver chimeric mice and hepatitis B virus-susceptible cells were infected to monitor dynamic changes and the fate of core protein and assembled capsids. We analyzed hepatitis B virus and protein dynamics over 4 weeks under t

Intranuclear paraspeckle-circular RNA TACC3 assembly forms RNA-DNA hybrids to facilitate MASH-related hepatocellular carcinoma growth in an m6A-dependent manner.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) is anticipated to become the leading cause of hepatocellular carcinoma (HCC). Accumulating evidence indicates that N6-methyladenosine (m6A)-modified circular RNAs (circRNAs) play key roles in tumor malignant progression. However, the precise molecular mechanisms by which circRNAs and their m6A modification regulatory networks respond to metabolic reprogramming, such as lipid overload stress, to drive malignant tumor progression in the context of MASH-related HCC remain unclear. This study aimed to investigate the role and regu

The Translocation of Circular DNA with Variable Single/Double Strand Ratios through Solid-State Nanopores: Transition from Entropic Barriers to Crowding-Dominated Kinetics.

Solid-state nanopores serve as powerful single-molecule analytical platforms for characterizing the physicochemical signatures of nucleic acids and proteins, yet prior work has focused almost exclusively on linear DNA, leaving circular DNA translocation poorly understood, particularly how alternating single-/double-stranded (ss/ds) segments modulate transport kinetics. Here, a series of circular DNAs with discrete ssDNA fractions (0%, 33%, 66%, and 100%) was constructed and systematically interrogated for their voltage-dependent translocation through solid-state nanopores. Under fixed bias, io

SUMO Modification of Hepatitis B Virus Core Mediates Nuclear Entry, Promyelocytic Leukemia Nuclear Body Association, and Efficient Formation of Covalently Closed Circular DNA.

Persistence of hepatitis B virus (HBV) infection is due to a nuclear covalently closed circular DNA (cccDNA), generated from the virion-borne relaxed circular DNA (rcDNA) genome in a process likely involving numerous cell factors from the host DNA damage response (DDR). The HBV core protein mediates rcDNA transport to the nucleus and likely affects stability and transcriptional activity of cccDNA. Our study aimed at investigating the role of HBV core protein and its posttranslational modification (PTM) with SUMO (small ubiquitin-like modifiers) during the establishment of cccDNA. Current Status and Challenges in Anti-Hepatitis B Virus Agents Based on Inactivation/Inhibition or Elimination of Hepatitis B Virus Covalently Closed Circular DNA.

There has been over half a century since the discovery of hepatitis B virus (HBV) to now, but approximately 300 million patients with chronic hepatitis B (CHB) still live in the world, resulting in about one million deaths every year. Although currently approved antivirals (e.g., nucleoside analogues) are effective at reducing HBV replication, they have almost no impact on the existing HBV covalently closed circular DNA (cccDNA) reservoir. HBV cccDNA is a critical obstacle to the complete elimination of the virus via antiviral therapy. The true cure of HBV infection requires the eradication of

Encoding gene RAB3B exists in linear chromosomal and circular extrachromosomal DNA and contributes to cisplatin resistance of hypopharyngeal squamous cell carcinoma via inducing autophagy.

Cisplatin (DDP) resistance is an important factor that decreases the effect of chemotherapy, thus leading to local recurrence and lymph node metastasis of hypopharyngeal squamous cell carcinoma (HSCC). We aimed to explore the role and mechanism of extrachromosomal circular DNA (eccDNA) in the DDP resistance of HSCC. In our research, the HSCC cell line FaDu and the DDP-resistant cell line FaDu/DDP were used as subjects. eccDNA sequencing and whole transcriptome sequencing were conducted, followed by a combined analysis of the two sequencing profiles.
