Vitamin E prevents cell death induced by mild oxidative stress in chicken skeletal muscle cells.

Apoptosis and necrosis are two forms of cell death that can occur in response to various agents and oxidative damage. In addition to necrosis, apoptosis contributes to muscle fiber loss in various muscular dystrophies as well participates in the exudative diathesis in chicken, pathology caused by dietary deficiency of vitamin E and selenium, which affects muscle tissue. We have used chicken skeletal muscle cells and bovine fibroblasts to study molecular events involved in the cell death induced by oxidative stress and apoptotic agents. The effect of vitamin E on cell death induced by oxidants

Unravelling the role of PANoptosis in diseases: from emerging molecular mechanisms-to-therapeutic implications.

PANoptosis represents a distinct form of programmed cell death (PCD) that is characterized by the integration of key features from apoptosis, pyroptosis, and necroptosis but cannot be fully explained by any single cell death pathway. A key trigger of PANoptosis is the assembly and activation of the PANoptosome. To date, four types of PANoptosomes have been identified, including PANoptosomes nucleated by Z-DNA binding protein 1 (ZBP1), absent in melanoma 2 (AIM2), receptor-interacting protein kinase 1 (RIPK1), and NOD-like receptor protein 12 (NLRP12). This review summarizes current advances in

PANoptosis in atherosclerotic plaques: Molecular mechanisms, clinical significance, and therapeutic potential.

Atherosclerosis (AS) is a leading cause of cardiovascular disease and occurs in the setting of chronic inflammation and lipid accumulation in the artery walls. A major driver of cardiovascular events is plaque vulnerability, characterized by large lipid-rich necrotic cores and thin fibrous caps. PANoptosis is a hybrid form of apoptosis, pyroptosis and necroptosis and has recently been identified as a critical regulator of plaque instability. This process is executed by the multi-protein PANoptosome complex, with key molecules such as Z-DNA binding protein 1 (ZBP1), receptor-interacting serine/

DLL4+ neutrophils promote Notch1-mediated endothelial PANoptosis to exacerbate acute lung injury in sepsis.

Neutrophils play a critical role in sepsis-induced acute lung injury (ALI). Extracellular cold-inducible RNA-binding protein (eCIRP), a damage-associated molecular pattern, promotes neutrophil heterogeneity. While delta-like ligand 4 (DLL4) expression has been studied in various cell populations, its expression in neutrophils and impact on inflammation remain unknown. Here, we discovered that eCIRP induces DLL4+ neutrophils. These neutrophils trigger PANoptosis, a novel proinflammatory form of cell death initiated by Z-DNA-binding protein-1 (ZBP1) in pulmonary vascular endothelial cells (PVECs

Novel WNT1 mutations in children with osteogenesis imperfecta: Clinical and functional characterization.

INTRODUCTION: Biallelic mutations in WNT1 can give rise to a rare form of moderate to severe OI. Here we report on 12 children (age 2 to 16 years; 5 girls) with biallelic WNT1 mutations. METHODS: Genomic DNA was analyzed either by targeted next-generation sequencing or Sanger sequencing. Mutations were modeled on the WNT1 protein structure. The in vitro functional effect of WNT1 mutations on WNT signaling was assessed in HEK293 cells using the topflash reporter assay system. RESULTS: All patients had lower extremity deformities and vertebral compression fractures. Seven individuals had upper

Integration of multi-omics and single-cell analysis reveals ZBP1 as a prognostic biomarker of renal cell carcinoma.

Z-DNA binding protein 1 (ZBP1) is a critical factor that induces various forms of cell death and inflammatory responses. However, it has not been comprehensively analyzed in malignancies. The present study aimed to elucidate the oncogenic role of ZBP1 in human malignancies and examine its prospective function in renal cell carcinoma. The pathogenic significance of ZBP1 in malignancies was clarified using a multi-omics analysis and single-cell analysis utilizing bioinformatics methods. The function of ZBP1 in renal cell carcinoma was validated using in vivo and in vitro studies. In this study,

Targeting the Zbp1-Ripk3 axis attenuates ischemic neuronal injury by suppressing PANoptosis.

Cerebral ischemia-reperfusion injury (CIRI) significantly exacerbates neuronal damage following ischemic stroke. Emerging evidence implicates PANoptosis, a novel inflammatory form of programmed cell death, in CIRI pathogenesis. However, the role of Z-DNA-binding protein 1 (Zbp1), a key regulator of PANoptosis and innate immune responses, remains poorly understood in this context. Here, we establish a middle cerebral artery occlusion/reperfusion (MCAO/R) mouse model and an oxygen-glucose deprivation/re-oxygenation (OGD/R) model in which HT-22 cells are used to simulate CIRI and investigate the

Mitochondria-associated programmed cell death in colorectal cancer: molecular mechanisms, metabolic plasticity, and therapeutic opportunities.

Mitochondria integrate bioenergetics, redox homeostasis, calcium signaling, metabolite synthesis, organelle quality control, innate immune sensing, and regulated cell death. In colorectal cancer (CRC), mitochondrial function is not simply suppressed by aerobic glycolysis; rather, tumor cells dynamically redistribute flux between glycolysis, oxidative phosphorylation, glutaminolysis, fatty-acid metabolism, and the mevalonate pathway to meet stage- and treatment-specific demands. This metabolic plasticity determines whether mitochondrial stress is buffered or converted into a lethal signal. Here

Bavachinin exerts anti-tumor effects by activating TLR4/STING axis-dependent PANoptosis and synergistically enhances chemosensitivity in endometrial cancer.

The incidence and mortality rates of endometrial cancer (EC), a malignancy originating from endometrium, have been increasing globally. Currently, there are no effective therapeutic options for patients with recurrent, chemoresistant, and metastatic forms of this disease. Through compound library screening, we identified that bavachinin (BVC) has a killing effect on EC cells. BVC is a bioactive small molecule with potential pharmacological effects derived from the traditional Chinese herb Proralea corylifolia L, but the specific mechanisms are unclear. We first discovered that BVC induces ZBP1

The KCNE genes in hypertrophic cardiomyopathy: a candidate gene study.

BACKGROUND: The gene family KCNE1-5, which encode modulating β-subunits of several repolarising K+-ion channels, has been associated with genetic cardiac diseases such as long QT syndrome, atrial fibrillation and Brugada syndrome. The minK peptide, encoded by KCNE1, is attached to the Z-disc of the sarcomere as well as the T-tubules of the sarcolemma. It has been suggested that minK forms part of an "electro-mechanical feed-back" which links cardiomyocyte stretching to changes in ion channel function. We examined whether mutations in KCNE genes were associated with hypertrophic cardiomyopathy

Epithelial mitochondrial fission-mediated PANoptosis is crucial for ulcerative colitis and its inhibition by saquinavir through Drp1.

Ulcerative colitis (UC) is characterized by increased cell death in intestinal epithelial cell (IEC), which compromises gut barrier function and activates inflammation. Aberrant mitochondrial dynamics have been implicated in various forms of cell death, but it is currently unclear if they play a role in IEC death and colitis pathogenesis. This study aims to investigate the contribution of aberrant mitochondrial dynamics to colitis progression using cellular models, animal models, and clinical samples. The results revealed that IEC in mice with Dextran sulfate sodium salt (DSS)-induced colitis

Synthesis and X-ray crystal structure of trans,cis-[Pt(OAc)2I2(en)]: a novel type of cisplatin analog that can be photolyzed by visible light to DNA-binding and cytotoxic species in vitro.

An original approach intended to facilitate the intratumoral activation of Pt(IV) diamines by illumination with visible light to form photolysis products that irreversibly bind to DNA and are cytotoxic to human cancer cells is reported. The novel Pt(IV) complex trans,cis-[Pt(OAc)2I2-(en)] was prepared by the acetylation of trans,cis-[Pt(OH)2I2(en)] with acetic anhydride in CH2-Cl2; trans,cis-[Pt(OH)2I2(en)] was synthesized by oxidation of [PtI2(en)] with 30% aqueous H2O2. trans,cis-[Pt(OAc)2I2(en)] crystallized from methanol as deep-red needles with a = 9.029(4) A, b = 11.443(2) A, c = 12.822(

Differentiated sex chromosomes, karyotype evolution, and spontaneous triploidy in carphodactylid geckos.

Geckos exhibit derived karyotypes without a clear distinction between macrochromosomes and microchromosomes and intriguing diversity in sex determination mechanisms. We conducted cytogenetic analyses in six species from the genera Nephrurus, Phyllurus, and Saltuarius of the gecko family Carphodactylidae. We confirmed the presence of a female heterogametic system with markedly differentiated and heteromorphic sex chromosomes in all examined species, typically with the W chromosome notably larger than the Z chromosome. One species, Nephrurus cinctus, possesses unusual multiple Z1Z1Z2Z2/Z1Z2W sex

Identification of a novel heterozygous mutation in exon 50 of the COL1A1 gene causing osteogenesis imperfecta.

UNLABELLED: A 19-year-old woman was diagnosed with osteogenesis imperfecta (OI). She had sustained numerous low-trauma fractures throughout her childhood, including a recent pelvic fracture (superior and inferior ramus) following a low-impact fall. She had the classical blue sclerae, and dual energy X-ray absorptiometry (DEXA) bone scanning confirmed low bone mass for her age in the lumbar spine (Z-score was -2.6). However, despite these classical clinical features, the diagnosis of OI had not been entertained throughout the whole of her childhood. Sequencing of her genomic DNA revealed that s

Glutamate oxidative injury to RGC-5 cells in culture is necrostatin sensitive and blunted by a hydrogen sulfide (H2S)-releasing derivative of aspirin (ACS14).

Oxidative stress to RGC-5 cells in culture was delivered by exposure to a combination of glutamate (Glu) and buthionine-S,R-sulfoximine (BSO). The effect of the insult on cell survival was quantified by the resazurin-reduction and a dead/live assays. Moreover, breakdown of DNA, the localisation of phosphatidylserine and reactive radical species (ROS) and its quantification were determined. In addition, various proteins and mRNAs were studied using Western blot, real time PCR and immunocytochemistry. ACS14, its sulfurated moiety ACS1 and aspirin were tested for their ability to blunt the negati

ZBP1 and IFI16: novel innate sensors and inflammation regulation under infection stress.

The innate immune system is the first line of defense against infection, using pattern recognition receptors to sense pathogens and abnormal host-derived nucleic acids. Among these, Z-DNA-binding protein 1 (ZBP1) and interferon gamma-inducible protein 16 (IFI16) are two important nuclear and cytosolic nucleic acid sensors attracting recent attention. These sensors play key roles in detecting abnormal nucleic acids from viruses, bacteria, and damaged host cells. They trigger downstream innate immune responses to regulate inflammation. Despite much progress, key questions remain about how they a

Biological Response of Human Cancer Cells to Ionizing Radiation in Combination with Gold Nanoparticles.

In the context of improving radiation therapy, high-atomic number (Z) metallic nanoparticles and, more importantly, gold-based nanostructures are developed as radiation enhancers/radiosensitizers. Due to the diversity of cell lines, nanoparticles, as well as radiation types or doses, the resulting biological effects may differ and remain obscure. In this multiparameter study, we aim to shed light on these effects and investigate them further by employing X-irradiation and three human cancer cell lines (PC3, A549, and U2OS cells) treated by multiple techniques. TEM experiments on PC3 cells show

Identification of a Hexenal Reductase That Modulates the Composition of Green Leaf Volatiles.

Green leaf volatiles (GLVs), including six-carbon (C6) aldehydes, alcohols, and esters, are formed when plant tissues are damaged. GLVs play roles in direct plant defense at wound sites, indirect plant defense via the attraction of herbivore predators, and plant-plant communication. GLV components provoke distinctive responses in their target recipients; therefore, the control of GLV composition is important for plants to appropriately manage stress responses. The reduction of C6-aldehydes into C6-alcohols is a key step in the control of GLV composition and also is important to avoid a toxic b
