Scopolamine affects fear learning and social recognition in adult zebrafish.

Scopolamine is the secondary metabolite of the Datura stramonium and act as a muscarinic receptor antagonist. Previous studies showed that scopolamine caused attention and memory deficit. However, the effects of scopolamine on specific cognitive functions, such as fear learning and social recognition, remain poorly understood. Here, we demonstrate the effects of scopolamine on fear learning, social memory, and neural activity in zebrafish, providing a novel perspective on its impact on cognitive and social behaviors. Here, we used equal number of male and female zebrafish as an animal model an

Sugar-binding profiles of the mesothelial glycocalyx in frozen tissues of mice revealed by lectin staining.

The mesothelium is a non-adhesive protective surface that lines the serosal cavities and organs within the body. The glycocalyx is a complex structure that coats the outer layer of the mesothelium. However, due to the limitations of conventional fixation techniques, studies on glycans are limited. In this study, lectin staining of frozen tissues was performed to investigate the diversity of glycans in the glycocalyx of mesothelial cells in mice. Datura stramonium lectin (DSL), which recognizes lactosamine and binds to Galectin-3 and -1, was broadly bound to the mesothelial cells of the viscera

Herbal intoxication. Psychoactive effects from herbal cigarettes, tea, and capsules.

Herbal preparations, designed to be smoked or ingested for "health and happiness", are promoted as legal hallucinogens, euphoriants, and marihuana substitutes. A total of 25 psychoactive substances have been identified in these products, and a number of intoxications have resulted from their short- or long-term use. Physicians should be alerted to the nature of these effects when taking drug histories, and their possible role in the causation of medical complaints should be considered.

The immunoregulatory effects of natural products on psoriasis via its action on Th17 cells versus regulatory T cells balance.

Psoriasis is an incurable, chronic inflammatory disease, which brings a substantial burden on individuals and society. Currently, the treatment of psoriasis has entered the era of biologics, but its highly targeting of inflammatory mediators may enable the immune response to circumvent the blockade, leading to disease recurrence, or other clinical and immunological characteristics. Therefore, the discovery of new therapies that have the ability of multidirectional regulation on immunity and maintain the dynamic balance of immunity in psoriasis, may be the key to the treatment of the disease. Discrimination of Solanaceae taxa and quantification of scopolamine and hyoscyamine by ATR-FTIR spectroscopy.

Plant species of the Solanaceae family (nightshades) contain pharmacologically active anticholinergic tropane alkaloids, e.g., scopolamine and hyoscyamine. Tropane alkaloids are of special interest, either as active principles or as starting materials for semisynthetic production of other substances. For genetic evaluation, domestication, cultivation, harvest and post-harvest treatments, quantification of the individual active principles is necessary to monitor industrial processes and the resulting finished products. Up to now, frequently applied methods for quantification are based on high p

Phylogenetic patterns of tolerance to herbivory and underlying suite of traits: phylogenetic signal and trade-offs.

When herbivory is inevitable, plants can cope with it through tolerance, buffering its negative effects on plant fitness. Macroevolutionary patterns of tolerance to herbivory may improve our understanding about its evolution. Here, we explore the phylogenetic patterns of tolerance to herbivory and underlying functional traits, as well as the potential evolutionary constraints in the forms of fitness costs and a trade-off with resistance across the genus Datura. We performed a common garden experiment with simulated herbivory (defoliation) and analyzed data within a phylogenetic comparative exp

Jimson weed toxicity: management of anticholinergic plant ingestion.

Jimson weed is a hallucinogenic plant that is common in rural areas. Consumption of any part of the plant can result in severe anticholinergic toxicity. The clinical presentation of jimson weed toxicity is similar to that seen in cases of atropine poisoning. Treatment is aimed at removing plant material from the gastrointestinal tract, keeping the patient safe and reversing severe anticholinergic sequelae.

Bioherbicidal Evaluation of Methanol Extract of Sorghum halepense L. Rhizome and Its Bioactive Components Against Selected Weed Species.

Sorghum halepense (L.) Pers. (common name Johnson grass) is a perennial invasive weed that causes great harm worldwide, and its allelopathy has been demonstrated in a series of experiments. The present study offers new insights into its organ-specific phytochemical profiles using state-of-the-art metabolomic technology and explores the effects of a methanol extract of S. halepense rhizomes (ShER) and its major bioactive compounds (p-hydroxybenzoic acid and chlorogenic acid) on three noxious weed species. The phytotoxic effects of ShER are reflected through the inhibition of seed germination an

Development of a novel lectin-based gold nanoparticle point-of-care immunoassay for rapid diagnosis of patients with severe Dengue infection.

Rapid diagnosis of patients with severe Dengue infection can be useful for the efficient clinical management of cases caused by the Dengue virus. Lateral Flow Immunoassay (LFIA) have been broadly used for rapid Dengue diagnosis, because of their quick readouts with the human eye, simplicity of use, and affordability. Despite the availability of several commercial Dengue point-of-care assays, none has shown to be successful in discriminating between severe and nonsevere forms of Dengue infection. In the current study, for the first time, a novel lectin-based point-of-care assay for the early de

Label-free pharmacological profiling based on dynamic mass redistribution for characterization and authentication of hazardous natural products.

Natural products are becoming increasingly popular in multiple fields involving medicines, foods and beverages. However, due to the frequent occurrence of poisoning incidents, their toxicity and safety have caused a serious concern. Here we report a method of biosensor-based two-phase pharmacological profiling (BTPP) for discovery, monitor and control of receptor-targeted natural products. BTPP uses a resonant waveguide grating biosensor for label-free and non-invasive detection of intracellular dynamic mass redistribution (DMR), a phenomenon caused by protein relocalization after receptors re

Combined untargeted metabolomics and network pharmacology approaches to reveal the therapeutic role of withanolide B in psoriasis.

Psoriasis is a refractory inflammatory skin disorder in which keratinocyte hyperproliferation is a crucial pathogenic factor. Up to now, it is commonly acknowledged that psoriasis has a tight connection with metabolic disorders. Withanolides from Datura metel L. (DML) have been proved to possess anti-inflammatory and anti-proliferative properties in multiple diseases including psoriasis. Withanolide B (WB) is one of the abundant molecular components in DML. However, existing experimental studies regarding the potential effects and mechanisms of WB on psoriasis still remain lacking. Present stu

Toxic Plants Potentially Impacting the Health Status of Livestock in St. Kitts-A Surveillance Study.

Tropical regions are characterized by exceptional biodiversity, including extremely diverse flora. Many plants produce chemical compounds that function as defense mechanisms, some of which are toxic to mammals, including livestock species. However, documentation of toxic plants in the Caribbean region remains limited. The present study aimed to identify toxic and potentially toxic plant species affecting livestock on the island of Saint Christopher (St. Kitts) through surveys of a representative number of farms across all nine parishes. An extensive literature review was performed in order to

Monitoring the degradation of atropine and scopolamine in soil after spiking with naturally contaminated organic millet.

The spread of Datura sp. in European countries influences crop management and implies continuous food safety issues because of tropane alkaloids, atropine and scopolamine, the most relevant natural toxic compounds of this weed. These alkaloids can contaminate cereals to such a level that hampers food/feed related use and diverts batches of contaminated raw materials towards ultimate disposal such as burning. As no unambiguous information has been available on the fate of tropane alkaloids in soils, our study focused on the quantification and follow-up of these toxic residues in a soil experime

[Emergent drugs (III): hallucinogenic plants and mushrooms].

An increase in the consumption of vegetable substances with a hallucinogenic effect has been observed. Some of these substances are associated with ancestral religious ceremonies, while many of them are legal or are partially regulated. Salvia divinorum is a powerful kappa receptor agonist, with dissociative and hallucinogenic properties, which start quickly and have a short duration. Kratom (Mytragyna speciosa) has mitragynine as its principal alkaloid, with stimulating effects at low doses (coke-like effect), and sedative effects (opiate-like effect) at high doses. Composition of the sugar moiety of Tamm-Horsfall protein in patients with urinary diseases.

The sugar moiety of Tamm-Horsfall protein (THP) is altered by pathological conditions. The aim of this study was to investigate the composition of THP glycans in urinary diseases. THP was isolated from the urine of patients with urinary tract infection (group A), glomerulonephritis or interstitial nephritis (group B), and Bartter's syndrome (BS) (group C). Monosaccharides, N-glycan profile, THP reactivity with specific lectins and some other proteins were analyzed. THP of patients from groups A, B, and C showed lower amounts of N-acetylgalactosamine (P<0.05, P<0.005, and P<0.05, respectively)

N-glycosylated proteins are involved in efficient internalization of Klebsiella pneumoniae by cultured human epithelial cells.

Klebsiella pneumoniae obtained from patients with urinary tract infections is able to invade cultured human epithelial cells. The internalization process is dependent upon both microfilaments and microtubules. To better understand the interaction of these invasive bacteria with the host cell receptor(s), bladder, lung, and ileocecal epithelial cells were infected with K. pneumoniae in the presence of various lectins possessing multiple glycan specificities. It was found that the N-acetylglucosamine (GlcNAc)-specific lectins concanavalin A, Datura stramonium agglutinin, and wheat germ agglutini
