Ionized calcium concentrations in squid axons.

Values for ionized [Ca] in squid axons were obtained by measuring the light emission from a 0.1-mul drop of aequorin confined to a plastic dialysis tube of 140-mum diameter located axially. Ionized Ca had a mean value of 20 x 10(-9) M as judged by the subsequent introduction of CaEGTA/EGTA buffer (ratio ca. 0.1) into the axoplasm, and light measurement on a second aequorin drop. Ionized Ca in axoplasma was also measured by introducing arsenazo dye into an axon by injection and measuring the Ca complex of such a dye by multichannel spectrophotometry. Values so obtained were ca. 50 x 10(-9) M as

Sustainable and Integral Valorization of Dosidicus gigas Pen Waste: Combined Production of Chitosan with Antibacterial Properties and Human and Marine Probiotics.

This study details a biorefinery approach to valorize Dosidicus gigas squid pen waste. The process starts with the enzymatic deproteinization of squid pens, which prove effective with both Alcalase and Novozym, with the latter exhibiting a slightly higher efficiency to yield a material with 73% chitin content. Subsequent alkaline hydrolysis produces highly deacetylated chitosan (>90% degree of deacetylation), followed by controlled depolymerization to obtain polymers with molecular weights ranging from 50 to 251 kDa. Both native and depolymerized chitosan exhibit antimicrobial activity against

A Japanese Case of Food Protein-induced Enterocolitis Syndrome Caused by Multiple Seafoods.

Food protein-induced enterocolitis syndrome (FPIES) caused by fish and others is prevalent in the Mediterranean regions but is less frequently reported in Japan. This case report describes a 3-year-old Japanese girl who developed FPIES triggered by multiple seafoods, including swordfish, cod, and squid. The diagnosis was confirmed through oral food challenge tests (OFC), which led to repeated vomiting and an increase in thymus and activation-regulated chemokine (TARC) levels. This case highlights the importance of considering fish-induced FPIES in the differential diagnosis of recurrent vomiti

Occurrence of larval ascaridoid nematodes in the Argentinean short-finned squid Illex argentinus from the Southwest Atlantic Ocean (off Falkland Islands).

The Argentinean short-finned squid (Illex argentinus) is an oceanic, neritic species widely distributed off the east coast of South America, representing the most abundant commercially exploited squid species in these waters. Despite the great commercial importance of Argentinean short-finned squid as a food resource, and as frozen product exported to Europe, the presence of zoonotic anisakid nematodes, especially in the mantle of the squid, is poorly known. The occurrence and site of infection of larval ascaridoid nematodes in 70 I. argentinus caught off the Falkland Islands were investigated

Physicochemical characterization of bionanocomposite films based on tuna skin gelatin (Thunnus tonggol), squid ink melanin, and earthworm-derived glycolipoprotein G-90.

This study presents a sustainable strategy for developing multifunctional biopolymer-based composite films from marine and terrestrial bioresources. Gelatin extracted from Thunnus tonggol skin, melanin isolated from squid ink, and glycolipoprotein G-90 derived from earthworms were employed to enhance resource utilization within a circular bioeconomy framework. Four types of films namely gelatin, gelatin-melanin, gelatin-G-90, and gelatin-melanin-G-90 were fabricated and characterized for their structural, mechanical, physicochemical, and biological properties. ATR-FTIR spectra confirmed the su

Protective Effects of Squid Gonad Phospholipids Against Hepatotoxicity Induced by Multi-Glycoside From Tripterygium wilfordii (GTW): Inhibition of Inflammatory Response and Boost of GTW Metabolism in Zebrafish.

Multi-glycoside from Tripterygium wilfordii Hook.f. (GTW) has shown clinical efficacy in suppressing immunological dysfunction, but is associated with severe hepatotoxicity. Phospholipids derived from marine organisms, particularly enriched in long-chain polyunsaturated fatty acids, possess various beneficial bioactivities and significant medicinal value. However, the antagonistic action of phospholipids against GTW-induced hepatotoxicity remains unclear. In this study, we explored the protective effect of squid gonads-derived phospholipids (SPLs) against GTW-induced hepatotoxicity using a zeb

Chemical analysis of flotsam ambergris.

The natural product ambergris is only found rarely on beaches, as jetsam. Even more scarce, or even absent, are accounts of flotsam ambergris. Here, we report the chemical analysis of a rare, large piece (>100kg) of flotsam found in the Atlantic in 2019. About 95% of subsamples from the outside of the coprolith was soluble in dichloromethane. Of this, FTIR spectroscopy, APCI-MS and GC-MS indicated the presence of ambrein. Radiocarbon dating indicated that the sample was post 1950s in age. The 13C/12C isotope ratio (-22.5 ‰) was typical of those reported to date for whale 'body' ambergris. Meta

Effect of Temperature and pH on the Secondary Structure and Denaturation Process of Jumbo Squid Hepatopancreas Cathepsin D.

BACKGROUND: Cathepsin D is a lysosomal enzyme that is found in all organisms acting in protein turnover, in humans it is present in some types of carcinomas, and it has a high activity in Parkinson's disease and a low activity in Alzheimer disease. In marine organisms, most of the research has been limited to corroborate the presence of this enzyme. It is known that cathepsin D of some marine organisms has a low thermostability and that it has the ability to have activity at very acidic pH. Cathepsin D of the Jumbo squid (Dosidicus gigas) hepatopancreas was purified and partially characterized

An approach to the study of intracellular proteins related to the excitability of the squid giant axon.

The technique for covalently labeling proteins with 125I-labelled Bolton-Hunter reagent was used to determine the quantities of proteins released from the axoplasmic side of the squid axon membrane. The reagent could be introduced into the interior of the axon by the technique of intracellular perfusion, the radioiodination reaction being carried out in situ. Alternatively, the reaction could be carried out in vitro, i.e., by mixing the reagent with samples of proteins dissolved in the intracellular perfusion fluid collected from the axon. This technique was found to be sensitive enough to per

Environmental Viscosity Modulates Interbacterial Killing during Habitat Transition.

Symbiotic bacteria use diverse strategies to compete for host colonization sites. However, little is known about the environmental cues that modulate interbacterial competition as they transition between free-living and host-associated lifestyles. We used the mutualistic relationship between Eupyrmna scolopes squid and Vibrio fischeri bacteria to investigate how intraspecific competition is regulated as symbionts move from the seawater to a host-like environment. We recently reported that V. fischeri uses a type VI secretion system (T6SS) for intraspecific competition during host colonization.

Melatonin as a proconvulsive hormone in humans.

The pineal gland and melatonin exert a major influence in the control of brain electrical activity and have been shown to be involved in seizure and sleep mechanisms. Since pinealectomy has been reported to result in seizures in experimental animals, it is assumed that melatonin has anticonvulsant properties. Indeed, limited studies in humans with temporal lobe epilepsy indicate that melatonin attenuates seizure activity. In the present communication we present evidence, based on magnetoencephalographic (MEG) brain measurements, that melatonin may exert proconvulsive activity in humans as well

Hepatotoxicity and hepatocarcinogenicity in rats fed squid with or without exogenous nitrite.

The popular seafood squid contains high levels of naturally occurring amines such as dimethylamine (DMA) trimethylamine and trimethylamine-N-oxide (TMAO). The hepatotoxicity and hepatocarcinogenicity of squid with or without exogenous nitrite were investigated in rats. Acute necrosis including polymorphogenic neutrophil infiltration, haemorrhage and cholangiofibrosis were observed in the livers of most rats fed squid. Hepatocellular carcinoma (HCC) was induced in two out of 12 rats (16%) by feeding 10% squid in Purina rat chow for 10 months. The incidence of HCC was increased to four out of 10

Stage-specific microbial dynamics and transmission pathways in salted seafood processing plants: A comparison of salted pollock roe and salted squid.

This study investigated microbial transmission dynamics and community structures across processing stages of two salted seafood products: salted pollock roe and salted squid. Samples of raw materials, environmental samples, and final products were collected from two processing plants (n = 87), and both culture-based analyses and 16S rRNA gene sequencing were performed. SourceTracker was used to track potential sources contributing to the final products. Microbial dynamics exhibited contrasting patterns between the two products due to differences in salting intensity and processing methods. In

Ionic crosslinking encapsulation of grapefruit essential oil by chitosan: Process optimization, characterization, and application in hairtail preservation.

Hairtail (Trichiurus haumela) is prone to spoilage due to lipid oxidation and microbial activity during cold storage, necessitating the development of new preservatives to maintain their quality. In this study, grapefruit essential oil microcapsules (GEOs) were prepared by ionic crosslinking of chitosan and sodium tripolyphosphate. The optimal GEOs preparation parameters were established through sequential single-factor experiments followed by response surface methodology. Researchers systematically characterized GEOs and assessed their efficacy in prolonging the shelf life of refrigerated hai

Stem-Cell-Regenerative and Protective Effects of Squid (Symplectoteuthis oualaniensis) Skin Collagen Peptides against H2O2-Induced Fibroblast Injury.

Recently, there has been a growing interest in collagen peptides derived from marine sources for their notable ability to protect skin cells against apoptosis induced by oxidants. Therefore, the current study aimed to investigate the fundamental properties of collagen peptides, including their physicochemical, thermal, structural, stem-cell-regenerative, and skin-cell-protective effects, in comparison to commercial collagen peptides. The acid-soluble (ASC) and pepsin-soluble (PSC) collagens exhibited three distinct bands on SDS-PAGE, namely α (α1 and α2), β, and γ chains, confirming a type I p

Effects of chemical preservation on bulk and amino acid isotope ratios of zooplankton, fish, and squid tissues.

RATIONALE: It is imperative to understand how chemical preservation alters tissue isotopic compositions before using historical samples in ecological studies. Specifically, although compound-specific isotope analysis of amino acids (CSIA-AA) is becoming a widely used tool, there is little information on how preservation techniques affect amino acid δ15 N values. METHODS: We evaluated the effects of chemical preservatives on bulk tissue δ13 C and δ15 N and amino acid δ15 N values, measured by gas chromatography/isotope ratio mass spectrometry (GC/IRMS), of (a) tuna (Thunnus albacares) and squi

Identification of the receptor for toxin II alpha from scorpion Leirus quinquestriatus in isolated lobster and squid nerve membranes.

1. Isolated membranes from lobster and squid nerves were labeled with Leirus quinquestriatus toxin II alpha. 2. Dinitrofluorophenylazide 125I-ScTx derivative was synthesized and covalently bound to the membranes. 3. It was found that the membranes of these nonmyelinated nerves present a single polypeptide located at approximately 220-230 Kd range labeled by the ScTx. 4. Rat brain membrane used for comparison presents two polypeptidic zones at 220 and 50 Kd labeled by the toxin.
