Pseudophosphorylation of Tau at distinct epitopes or the presence of the P301L mutation targets the microtubule-associated protein Tau to dendritic spines.

Alzheimer's disease is characterized by the accumulation of amyloid-β (Aβ) and Tau in the brain. In mature neurons, Tau is concentrated in the axon and found at lower levels in the dendrite where it is required for targeting Fyn to the spines. Here Fyn mediates Aβ toxicity, which is vastly abrogated when Tau is either deleted or a truncated form of Tau (Tau(1-255)) is co-expressed. Interestingly, MAP2, a microtubule-binding protein with mainly dendritic localization that shares Fyn-binding motifs with Tau, does not mediate Aβ's synaptic toxicity in the absence of Tau. Here we show in hippocamp

[Quantitative study of the apical dendritic spines in the cingulate lamina-V-pyramid neurons of the rat following oral application of alcohol].

The question answered in this paper is whether the apical dendritic spines-values of lamina-V-pyramidal cells of the gyrus cinguli of the rat are changed as a result of the oral application of ethanol. Alcohol-application causes a statistically significant increase of apical spines, especially at the main dendrite when given during the early postnatal development phase, where the brain is still impressible. The results are discussed as compensatory achievement of the not degeneratively changed neurons, while other pyramids show signs of degeneration. The conclusions were compared with the resu

Functional and structural characterization of dendritic spine pathology in a mouse model of tauopathy.

Abnormal deposition of the microtubule-associated protein tau has long been associated with neurodegenerative diseases. While spine loss and neuronal death are hallmarks of tauopathy, how pathological tau affects synaptic activity in vivo and whether functional properties of individual synapses dictate the survival fate of dendritic spines remain elusive. Here we examined the visual response properties of dendrites and spines of layer 2/3 primary visual cortical neurons, using longitudinal two-photon calcium imaging in P301S mouse model of tauopathy. We found that neuronal outputs in tau mutan

Cocaine and habit training cause dendritic spine rearrangement in the prelimbic cortex.

Successfully navigating dynamic environments requires organisms to learn the consequences of their actions. The prelimbic prefrontal cortex (PL) formulates action-consequence memories and is modulated by addictive drugs like cocaine. We trained mice to obtain food rewards and then unexpectedly withheld reinforcement, triggering new action-consequence memory. New memory was disrupted by cocaine when delivered immediately following non-reinforcement, but not when delayed, suggesting that cocaine disrupted memory consolidation. Cocaine also rapidly inactivated cofilin, a primary regulator of the

Dendritic spine membrane proteome and its alterations in autistic spectrum disorder.

Dendritic spines are small protrusions stemming from the dendritic shaft that constitute the primary specialization for receiving and processing excitatory neurotransmission in brain synapses. The disruption of dendritic spine function in several neurological and neuropsychiatric diseases leads to severe information-processing deficits with impairments in neuronal connectivity and plasticity. Spine dysregulation is usually accompanied by morphological alterations to spine shape, size and/or number that may occur at early pathophysiological stages and not necessarily be reflected in clinical ma

Semaphorin3F Drives Dendritic Spine Pruning Through Rho-GTPase Signaling.

Dendritic spines of cortical pyramidal neurons are initially overproduced then remodeled substantially in the adolescent brain to achieve appropriate excitatory balance in mature circuits. Here we investigated the molecular mechanism of developmental spine pruning by Semaphorin 3F (Sema3F) and its holoreceptor complex, which consists of immunoglobulin-class adhesion molecule NrCAM, Neuropilin-2 (Npn2), and PlexinA3 (PlexA3) signaling subunits. Structure-function studies of the NrCAM-Npn2 interface showed that NrCAM stabilizes binding between Npn2 and PlexA3 necessary for Sema3F-induced spine p

CRMP2 mediates Sema3F-dependent axon pruning and dendritic spine remodeling.

Regulation of axon guidance and pruning of inappropriate synapses by class 3 semaphorins are key to the development of neural circuits. Collapsin response mediator protein 2 (CRMP2) has been shown to regulate axon guidance by mediating semaphorin 3A (Sema3A) signaling; however, nothing is known about its role in synapse pruning. Here, using newly generated crmp2-/- mice we demonstrate that CRMP2 has a moderate effect on Sema3A-dependent axon guidance in vivo, and its deficiency leads to a mild defect in axon guidance in peripheral nerves and the corpus callosum. Surprisingly, crmp2-/- mice dis

Loss of EPAC2 alters dendritic spine morphology and inhibitory synapse density.

EPAC2 is a guanine nucleotide exchange factor that regulates GTPase activity of the small GTPase Rap and Ras and is highly enriched at synapses. Activation of EPAC2 has been shown to induce dendritic spine shrinkage and increase spine motility, effects that are necessary for synaptic plasticity. These morphological effects are dysregulated by rare mutations of Epac2 associated with autism spectrum disorders. In addition, EPAC2 destabilizes synapses through the removal of synaptic GluA2/3-containing AMPA receptors. Previous work has shown that Epac2 knockout mice (Epac2-/-) display abnormal soc

PINK1 Silencing Modifies Dendritic Spine Dynamics of Mouse Hippocampal Neurons.

PTEN-induced kinase 1 (PINK1) mutations can cause early-onset Parkinson's disease and patients are likely to develop cognitive decline, depression, and dementia. Several neurophysiological studies have demonstrated PINK1 deficiency impairs striatal and hippocampal presynaptic plasticity. Dendritic spine postsynaptic abnormalities are common in neurological diseases; however, whether PINK1 silencing modifies dendritic spine dynamics of hippocampal neurons is unclear. To address this question, confocal images of mouse cultured hippocampal neurons transfected with plasmids to silence PINK1 were a

Rac GEF Dock4 interacts with cortactin to regulate dendritic spine formation.

In neuronal development, dendritic spine formation is important for the establishment of excitatory synaptic connectivity and functional neural circuits. Developmental deficiency in spine formation results in multiple neuropsychiatric disorders. Dock4, a guanine nucleotide exchange factor (GEF) for Rac, has been reported as a candidate genetic risk factor for autism, dyslexia, and schizophrenia. We previously showed that Dock4 is expressed in hippocampal neurons. However, the functions of Dock4 in hippocampal neurons and the underlying molecular mechanisms are poorly understood. Here we show t

Dendritic spine loss and neurodegeneration is rescued by Rab11 in models of Huntington's disease.

Huntington's disease (HD) is a fatal neurodegenerative disorder caused by expansion of a polyglutamine tract in the huntingtin protein (htt) that mediates formation of intracellular protein aggregates. In the brains of HD patients and HD transgenic mice, accumulation of protein aggregates has been causally linked to lesions in axo-dendritic and synaptic compartments. Here we show that dendritic spines - sites of synaptogenesis - are lost in the proximity of htt aggregates because of functional defects in local endosomal recycling mediated by the Rab11 protein. Impaired exit from recycling endo

L-methionine decreases dendritic spine density in mouse frontal cortex.

Schizophrenia postmortem brain is characterized by gamma aminobutyric acid downregulation and by decreased dendritic spine density in frontal cortex. Protracted L-methionine treatment exacerbates schizophrenia symptoms, and our earlier work (Tremolizzo et al. and Dong et al.) has shown that L-methionine decreases reelin and GAD67 transcription in mice which is prevented by co-administration of valproate. In this study, we observed a decrease in spine density following L-methionine treatment, which was prevented by co-administration of valproate. Together with our earlier findings conducted und

Regulation of NMDA receptor interactions with the actin cytoskeleton in dendritic spine development.

In the central nervous system, the majority of excitatory synapses exist on small actin-enriched structures protruding from dendrites, known as dendritic spines. Actin cytoskeletal rearrangements drive dynamic changes in spine shape, size, and density depending on developmental stage and synaptic activity. Spines initially emerge from the dendritic shaft as dynamic protrusions known as filipodia-like spines, which serve as precursors to mature dendritic spines. The formation and maturation of dendritic spines facilitate information transfer in neural circuits, underlying cognitive processes, s

Nootropic benzothiazoles promote dendritic spine formation by targeting fascin-1.

Identifying new drug targets to enhance memory and learning is essential for treating neurodegenerative diseases, such as Alzheimer's disease. We previously showed that benzothiazole amphiphiles can improve memory and learning by increasing dendritic spine and synaptic density in both WT mice and in a transgenic Alzheimer's model mice. The cellular target for this class of compounds, however, was unknown. Using a photoaffinity-labeling approach, we identify fascin-1 as the major protein target in neurons for these compounds. These compounds enhance spine density by directly modulating actin dy

Dendritic retraction contributes to spine density recovery following denervation in vitro.

Neurodegenerative diseases cause local brain lesions as well as denervation of neurons in areas connected to the lesion site. In part, the denervation-induced damage associated with brain lesions can be homeostatically compensated by network reorganization. To study the dynamics of compensatory changes occurring in denervated brain areas, we established an in vitro denervation model based on organotypic slice cultures of mouse entorhinal cortex and hippocampus. In these cultures, the entorhino-dentate projection was transected and denervation-induced changes of tdTomato-labeled dentate granule

Regional Specificity of Cortical Layer 3 Dendritic Spine Deficits in Schizophrenia.

IMPORTANCE: Schizophrenia (SZ) is characterized by deficits in visual-spatial working memory, a function dependent on a distributed cortical network that includes nodes in the primary visual (V1), posterior parietal (PPC), and dorsolateral prefrontal (DLPFC) cortices. The connections across these regions involve excitatory synapses on the dendritic spines of pyramidal neurons in layer 3 (L3). OBJECTIVE: To assess whether SZ is associated with regional differences in the magnitude or nature of L3 spine alterations. DESIGN, SETTING, AND PARTICIPANTS: This case-control study examined brain tiss

Direct evidence for dendritic spine compensation and regeneration in Alzheimer's disease models.

INTRODUCTION: Dendritic spine loss in Alzheimer's disease (AD) strongly correlates with cognitive decline, whereas spine preservation is associated with cognitive resilience. Yet, whether and how neurons compensate for spine loss in AD remains largely unknown. METHODS: We developed a chromophore-assisted light inactivation (CALI) strategy to selectively eliminate dendritic spines to model this key feature of AD. Two-photon microscopy was used to monitor the structural plasticity of spines over time after spine elimination. Validation experiments were conducted in amyloid beta (Aβ)-driven mode

nArgBP2 regulates excitatory synapse formation by controlling dendritic spine morphology.

Neural Abelson-related gene-binding protein 2 (nArgBP2) was originally identified as a protein that directly interacts with synapse-associated protein 90/postsynaptic density protein 95-associated protein 3 (SAPAP3), a postsynaptic scaffolding protein critical for the assembly of glutamatergic synapses. Although genetic deletion of nArgBP2 in mice leads to manic/bipolar-like behaviors resembling many aspects of symptoms in patients with bipolar disorder, the actual function of nArgBP2 at the synapse is completely unknown. Here, we found that the knockdown (KD) of nArgBP2 by specific small hair
