A new non-human primate model of desiccating stress-induced dry eye disease.

Dry eye disease (DED), a multifactorial ocular surface disease, is estimated to affect up to 34% of individuals over 50 years old. Although numerous animal models, including rodents and rabbits, have been developed to mimic the pathophysiologic mechanisms involved in dry eye, there is a lack of non-human primate (NHP) models, critical for translational drug studies. Here, we developed a novel desiccating stress-induced dry eye disease model using Rhesus macaque monkeys. The monkeys were housed in a controlled environment room for 21 to 36 days under humidity, temperature, and airflow regulatio

Desiccating Stress Significantly Increases the Risk for Chronic Ocular Graft-versus-Host-Disease.

Desiccating stress (DS) is known to induce dry eye disease but has not been studied in the context of ocular graft-versus-host disease (oGVHD). Patients undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT) are exposed to DS on transplantation wards, which are highly climate-regulated for hygienic purposes. Because oGVHD demonstrates features of dry eye disease, this retrospective study aimed to analyze DS as a risk factor for chronic oGVHD. A total of 444 patients undergoing allo-HSCT were investigated with a maximum follow-up of 5.8 years post-transplantation. Relative hu

Restarting the Healing Process of Chronic Wounds Using a Novel Desiccant: A Prospective Case Series.

INTRODUCTION: Chronic skin wounds represent a major global health problem and financial burden. The blocked healing process of chronic wounds involves excess inflammatory proteins, persistent microbial burden, and often, drug-resistant biofilm on the wound bed. Wound-bed debridement is considered crucial to restart the healing process. OBJECTIVE: The authors developed a novel desiccant (desiccating agent A) to serve as a new form of chemical debridement. The objective is to establish the working mechanism of desiccating agent A. MATERIALS AND METHODS: Desiccating agent A was exposed to 7 pat

Therapeutic effect of topical adiponectin in a mouse model of desiccating stress-induced dry eye.

PURPOSE: To investigate the therapeutic effect of topical adiponectin in a mouse model of experimental dry eye (EDE). METHODS: EDE was created by desiccating stress in 6- to 8-week old female C57BL/6 mice. Eye drops consisting of 0.0001%, 0.001%, or 0.01% adiponectin, or balanced salt solution (BSS), were applied in EDE. Tear volume and corneal irregularity score were measured at 5 and 10 days after treatment. Levels of interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, interferon (IFN)-γ, and monokine induced by interferon-γ (MIG) were measured in the conjunctiva and lacrimal gland us

Desiccating environmental stress exacerbates autoimmune lacrimal keratoconjunctivitis in non-obese diabetic mice.

The non-obese diabetic (NOD) mouse is prone to develop autoimmune disease, including Sjögren's syndrome. The purpose of this study was to determine if desiccating environmental stress exacerbates the development of Sjögren's syndrome-like lacrimal keratoconjunctivitis in the NOD.B10.H2(b) mouse. Four-week-old male mice were used as young controls. Sixteen-week-old male mice were untreated or subjected to desiccating stress with a fan alone or with a fan plus subcutaneous injections of the anticholinergic agent scopolamine for 5 or 10 days to inhibit tear production. Mice spontaneously develope

Desiccating stress induces T cell-mediated Sjögren's Syndrome-like lacrimal keratoconjunctivitis.

Chronic dry eye syndrome affects over 10 million people in the United States; it is associated with inflammation of the lacrimal gland (LG) and in some cases involves T cell infiltration of the conjunctiva. We demonstrate that environmental desiccating stress (DS) elicits T cell-mediated inflammation of the cornea, conjunctiva, and LG, but not other organs in mice. The lacrimal keratoconjunctivitis (LKC) was mediated by CD4(+) T cells, which, when adoptively transferred to T cell-deficient nude mice, produced inflammation in the LG, cornea, and conjunctiva, but not in any other organ. Adoptive

How melanised lichens survive drying: transcriptomic insights into desiccation and UV-B cross-tolerance of Lobaria pulmonaria.

Lichens are known for their remarkable ability to cope with a wide range of environmental stresses, particularly desiccation. Under natural conditions, lichens often experience both desiccation and ultraviolet (UV) irradiation simultaneously. Such a combination of stresses may promote the development of cross-tolerance mechanisms, but there have been few studies on these mechanisms in lichens. Previously, we reported that in the lichen Lobaria pulmonaria, UV-B induces synthesis of the dark pigment melanin and changes in gene expression and energy metabolism. The aim of the current study was to

Melatonin Ameliorates Desiccation Stress-Induced Ocular Inflammation in an In Vitro Model by Activating the Nrf2 Pathway.

Dry eye disease (DED) is a multifactorial ocular disorder marked by tear film instability, oxidative stress, and inflammation. Although melatonin is known to exert antioxidant and anti-inflammatory effects, its precise role in regulating desiccation-induced stress responses at the ocular surface has not been well defined. In particular, the involvement of the Nrf2 signalling pathway in melatonin-mediated protection against corneal epithelial inflammation under tear film instability remains unexplored. In this study, we evaluated the therapeutic potential of melatonin in a human corneal epithel

Inter- and intraspecific variation in resistance to desiccation by adult Aedes (Stegomyia) spp. (Diptera: Culicidae) from Indonesia.

Desiccation survival times of adult mosquitoes were studied for yellow fever mosquito, Aedes aegypti (L.) (6 strains), Asian tiger mosquito, Ae. albopictus (Skuse) (5 strains), and Ae. paullusi Stone & Farmer (1 strain) colonized from South Sulawesi, Indonesia. At both 90 and 70% RH, Ae. aegypti males and females outlived the other species. The forest species Ae. paullusi was least resistant to desiccation. Strains of Ae. aegypti and Ae. albopictus from urban areas were more resistant to desiccation than conspecific strains from rural villages. When water was accessible, Ae. albopictus females

Electrophysiologic and histologic observations of chronic atrioventricular block induced by closed-chest catheter desiccation with radiofrequency energy.

Direct-current or laser energy has been used to induce atrioventricular (AV) block, but certain complications associated with this type of energy have been reported. We have previously documented that radiofrequency (RF) energy can effectively and safely induce acute AV block in closed-chest dogs during the 4-7 days of follow-up. This study was undertaken to determine if the ablation was permanent and to define the chronic pathology and site of AV block. Complete AV block was successfully achieved in four dogs immediately after ablation with a bipolar "standard" RF output (750 kHz) delivered b

The architecture of resilience: a genome assembly of Myrothamnus flabellifolia sheds light on desiccation tolerance and sex determination.

Myrothamnus flabellifolia is a dioecious resurrection plant endemic to southern Africa that has become an important model for understanding desiccation tolerance. Despite its ecological and medicinal significance, genomic and transcriptomic resources for the species are limited. We generated a chromosome-level, haplotype-resolved reference genome assembly and annotation for M. flabellifolia and conducted transcriptomic profiling across a natural dehydration-rehydration time course in the field. Genome architecture and sex determination were characterized, and co-expression network and cis-regu

Effects and molecular mechanism of sugar transporter ESA_RS15745 on desiccation resistance, motility, and biofilm formation of Cronobacter sakazakii.

Cronobacter sakazakii, an important Gram-negative foodborne pathogen, can cause neonatal meningitis and sepsis with high rates of infection and death. Gene ESA_RS15745 encodes a sugar transporter protein, which is not only essential for osmotic pressure maintenance during bacterial growth and reproduction but also associated with their desiccation tolerance, motility, and biofilm formation. Here, a mutant strain of ESA_RS15745 (ΔESA_RS15745) and the complementation strain (cpESA_RS15745) were constructed using a suicide vector knockout and gene complementation. ΔESA_RS15745 was found to have a

Effect of Artificial Tear Formulations on the Metabolic Activity of Human Corneal Epithelial Cells after Exposure to Desiccation.

Artificial lipid-containing tear formulations are developed to reduce tear evaporation by the restoration of a deficient tear lipid layer. Artificial tear formulations that prevent cell desiccation will result in ocular surface protection and the maintenance of cell metabolic activity. During dehydration, cells undergo the process of loss of metabolic activity and subsequently cell death. This work describes a method for assessing the efficacy of artificial tear formulations. The metabolic dye (i.e., alamarBlue) changes from a low fluorescent molecule resazurin to a fluorescent molecule resoru

Directional selection to improve the sterile insect technique: Survival and sexual performance of desiccation resistant Anastrepha ludens strains.

The sterile insect technique (SIT) is an effective, environmentally friendly method for insect control whose success depends on the sexual performance and survival of sterile males. These two parameters are influenced by environmental conditions of target areas, and releasing insects with a higher tolerance to stressful environments can improve SIT efficiency. Directional selection can be used to produce insect strains with higher tolerance to extreme environmental conditions, such as low humidity, for extended periods. We evaluated, under field cage conditions, the sexual competitiveness, sex

A small heat-shock protein (Hsp20) regulated by RpoS is essential for cyst desiccation resistance in Azotobacter vinelandii.

In Azotobacter vinelandii, a cyst-forming bacterium, the alternative sigma factor RpoS is essential to the formation of cysts resistant to desiccation and to synthesis of the cyst-specific lipids, alkylresorcinols. In this study, we carried out a proteome analysis of vegetative cells and cysts of A. vinelandii strain AEIV and its rpoS mutant derivative AErpoS. This analysis allowed us to identify a small heat-shock protein, Hsp20, as one of the most abundant proteins of cysts regulated by RpoS. Inactivation of hsp20 did not affect the synthesis of alkylresorcinols or the formation of cysts wit

Pain perception and management during chemical debridement with a topical desiccating agent: a case-series analysis.

OBJECTIVE: A new topical desiccating agent (TDA; DEBRICHEM, DEBx Medical BV, the Netherlands) based on methanesulfonic acid (MSA) and dimethyl sulfoxide (DMSO) has proved to be an effective biofilm- and necrosis-removing chemical debridement option. However, its application can be temporarily accompanied by sharp pain perception, depending on the individual patient. This study aimed to assess application-associated pain and two strategies to manage this. METHOD: A single-centre, retrospective, case-series study design was used to assess pain associated with MSA/DMSO treatment. To lower pain s

Development of an Ex Vivo Model of Focal Corneal Desiccation to Investigate the Pathophysiology of Corneal Dellen and Ulcers.

PURPOSE: To determine the maximum extent of corneal thinning induced solely by dehydration and to establish the threshold at which corneal lysis begins, thereby refining therapeutic indications, with the aim of improving clinical management. METHODS: Dehydration was induced in 28 corneas, after 4 ex vivo experimental models. Corneal thickness was regularly measured using swept-source optical coherence tomography (OCT). Histological analysis was performed on 14 of the corneas, with computer-based counting of collagen lamellae. The remaining 14 corneas were rehydrated and remeasured using OCT.

Topical desiccating agent (DEBRICHEM): an accessible debridement option for removing biofilm in hard-to-heal wounds.

It is now assumed that all hard-to-heal wounds contain biofilm. Debridement plays a key role in wound-bed preparation, as it can remove biofilm along with the devitalised tissue, potentially leaving a clean wound bed that is more likely to progress towards healing. The gold standard methods of debridement (surgical and sharp) are the least used, as they require specialist training and are often not readily available at the point of need. Most other methods can be used by generalists but are slower. They all need regular applications. The topical desiccating agent DEBRICHEM is an innovative alt

New porous amine-functionalized biochar-based desiccated coconut waste as efficient CO2 adsorbents.

Climate change caused by the greenhouse gases CO2 remains a topic of global concern. To mitigate the excessive levels of anthrophonic CO2 in the atmosphere, CO2 capture methods have been developed and among these, adsorption is an especially promising method. This paper presents a series of amine functionalized biochar obtained from desiccated coconut waste (amine-biochar@DCW) for use as CO2 adsorbent. They are ethylenediamine-functionalized biochar@DCW (EDA-biochar@DCW), diethylenetriamine-functionalized biochar@DCW (DETA-biochar@DCW), triethylenetetramine-functionalized biochar@DCW (TETA-bio
