Childhood parasitic infections endemic to the United States.

Endemic parasitic infections in the United States are more frequent than is commonly perceived. Intestinal parasitic infection with Cryptosporidium, Dientamoeba, and Giardia occurs most often in children in northern states during the summer months. Zoonotic Toxocara and Toxoplasma parasitic infections are more frequent in southern states, in African Americans, and in populations with lower socioeconomic status. Approximately 300, 000 people in the United States have Trypanosoma cruzi infection. Local, vector-borne transmission of T cruzi and Leishmania infections has been documented in souther

Amebiasis and "nonpathogenic" intestinal protozoa.

Infection with single or multiple species of intestinal protozoa is common in humans and can result in either asymptomatic colonization or symptoms of intestinal disease. Entamoeba histolytica serves as a paradigm for invasive colonic protozoal infection. The key to diagnosis and treatment of amebiasis is knowledge of the epidemiologic risk factors and clinical manifestations, a rational approach to diagnosis, and an understanding of the sites of action and uses of anti-amebic drugs. This knowledge of treatment provides a context for consideration of intestinal infection with less common proto

Molecular identification and characterization of waterborne protozoa among stranded Dugong dugon.

The Persian Gulf hosts the second-largest population of Dugongs. The International Union for the Conservation of Nature (IUCN) lists them as Vulnerable to Extinction species. Incidental fishing, vessel strikes, environmental pollution, habitat degradation, and infectious diseases currently threaten these sirenians. While research focused on the abovementioned threats, little is known about pathogen infections, particularly waterborne parasites. The current study aimed to investigate the prevalence of the waterborne parasites, namely Blastocystis, Cryptosporidium, Dientamoeba fragilis, Giardia

A zymodeme study of Entamoeba histolytica in a group of South African schoolchildren.

Using a biphasic culture medium, stocks of intestinal amoebae were isolated from a group of children attending school in Durban, South Africa. These were compared with stocks collected in other areas of the world already characterized using the electrophoretic patterns of four enzymes: glucose phosphate isomerase (GPI), phosphoglucomutase (PGM) L-malate: NADP+ oxido-reductase (oxalacetate-decarboxylating) (ME) and hexokinase (HK). 33% of 94 samples grew Entamoeba histolytica, only one of which gave a pattern indicative of a pathogenic stock. Entamoeba hartmanni, Dientamoeba fragilis and Entamo

Occasional parasitic infections of man in Papua New Guinea and Irian Jaya (New Guinea).

Most parasites of man in New Guinea are cosmopolitan species, widely distributed and highly prevalent in the island. Reports of other parasite infections are reviewed. This impoverished fauna includes anthroponotic species normally occurring at low prevalences, e.g. Isospora belli, Dientamoeba fragilis, Trichomonas hominis and a few zoonotic forms with pigs as reservoir hosts, notably Balantidium coli and Entamoeba polecki. Other zoonoses are very few and extremely rare. This may be due to the reduced placental mammal fauna. The potential for the spread of Taenia solium infection is highlighte

Associations between Gut Microbiota and Common Luminal Intestinal Parasites.

The development and integration of DNA-based methods in research and clinical microbiology laboratories have enabled standardised and comprehensive detection and differentiation of the microbes colonising our guts. For instance, the single-celled parasites Blastocystis and Dientamoeba appear to be much more common than previously thought, especially so in healthy individuals. While increasing evidence appears to suggest limited pathogenicity of these parasites, next-generation-sequencing-based studies have helped us to appreciate links between parasite colonisation and certain host phenotypica

Enteric pathogenic protozoa in homosexual men from San Francisco.

The prevalence of enteric protozoa was studied in a survey conducted among 150 male homosexual patients in San Francisco. All patients were from a private practice in internal medicine. Each was asked to complete a questionnaire and to submit multiple stool specimens for examination. Of this group, 47% were positive for one or more potentially pathogenic intestinal protozoa; Entamoeba histolytica was found in 36%, Entamoeba hartmanni in 35%, Giardia lamblia in 5%, and Dientamoeba fragilis in 1.3%. Symptoms were unreliable as a diagnostic index of intestinal protozoan infection. Colonization ra

Intestinal protozoan infections: prevalence in the San Francisco Bay Area.

A sample of 415 members of the Kaiser Foundation Health Plan (KFHP) were tested for intestinal protozoa. This parasite survey group was found to be comparable in age distribution, sex and ethnic background with the membership at large. Gastrointestinal complaints were no more numerous in this group than in a large series of KFHP members taking multiphasic examinations. Because KFPH membership included, at the time of the survey, approximately 27 percent of all residents of the San Francisco Bay Area, the membership may be considered approximately representative of the entire area. Fourteen per

Comparison of commercial and in-house real-time PCR platforms for 15 parasites and microsporidia in human stool samples without a gold standard.

INTRODUCTION: A test comparison of in-house and commercial real-time PCR (qPCR) kits for the detection of human parasites and microsporidia in stool samples was conducted without a gold standard. Three different commercial kits were included in the comparison, with a range of 3-15 different PCR targets, while 14 targets were covered by in-house testing, so not all 16 target pathogens were covered by all assays. METHODS: Residual materials from nucleic acid extractions of stool samples with very high likelihood of being colonized or infected by at least one enteric parasite species or microspo

What is the importance of zoonotic trichomonads for human health?

Trichomonads are common parasites of many vertebrate and invertebrate species, with four species classically recognized as human parasites: Dientamoeba fragilis, Pentatrichomonas hominis, Trichomonas vaginalis, and Trichomonas tenax. The latter two species are considered human-specific; by contrast, D. fragilis and P. hominis have been isolated from domestic and farm mammals, demonstrating a wide host range and potential zoonotic origin. Several new studies have highlighted the zoonotic dimension of trichomonads. First, species typically known to infect birds and domestic mammals have been ide

Detection of intestinal protozoa in the clinical laboratory.

Despite recent advances in diagnostic technology, microscopic examination of stool specimens remains central to the diagnosis of most pathogenic intestinal protozoa. Microscopy is, however, labor-intensive and requires a skilled technologist. New, highly sensitive diagnostic methods have been developed for protozoa endemic to developed countries, including Giardia lamblia (syn. G. intestinalis/G. duodenalis) and Cryptosporidium spp., using technologies that, if expanded, could effectively complement or even replace microscopic approaches. To date, the scope of such novel technologies is limite

The epidemiology of Entamoeba histolytica in a rural and an urban area of Mexico. A pilot survey II.

Stocks of intestinal amoebae grown in monoxenic culture, were compared against each other and against those previously reported from Mexico City. These were isolated from subjects in San Cristobal de las Casas, Chiapas (rural area) and hospital patients in Merida, Yucatan (urban area). Electrophoretic patterns of the four enzymes: glucose phosphate isomerase (GPI), phosphoglucomutase (PGM), L-malate: NADP+ oxido-reductase (oxalacetate-decarboxylating) (ME) and hexokinase (HK) demonstrated the presence of five groups (zymodemes) of Entamoeba histolytica already described from Mexico City, toget

Irritable bowel syndrome in Egyptian patients: plausible risk factors and association with intestinal protozoa.

The pathogenesis of irritable bowel syndrome (IBS) is not yet clear. Our study suggested parasitic infection and other plausible risk factors among Egyptian IBS patients. We studied 40 IBS patients diagnosed according to Rome III criteria (Group I), 40 with other gastrointestinal symptoms (Group II) and 40 healthy controls (Group III). Stool samples were examined using direct wet smear, sedimentation technique, trichrome stain and immune-chromatographic tests for Cryptosporidium parvum. IBS patients displayed a significantly greater percentage of Blastocystis hominis infection (45%) than non-I
