Oxylipin-mediated metabolic signatures of symbiosis homeostasis and thermal stress in a model sea anemone.

Oxylipins are oxygenated products of fatty acids proposed to exert a regulatory role in cnidarian-dinoflagellate symbiosis; however, this has not been investigated in detail. We integrated physiological measurements and molecular phenotyping with comparative transcriptome mining to examine how the symbiotic cnidarian model, the sea anemone Aiptasia (i.e. Exaiptasia diaphana), and its dinoflagellate symbiont Breviolum minutum respond to symbiosis and thermal stress. We performed lipidomics in combination with the quantification of oxylipins, including octadecanoids, eicosanoids, and docosanoids

The disruption of a symbiotic sea anemone by light pollution: Non-linear effects on zooxanthellae and molecular indicators.

Artificial Light at Night (ALAN) is a pervasive stressor that may affect coastal organisms, particularly sessile forms associated with photosynthetic symbionts. We examined the effects of ALAN upon the symbiotic snakelocks anemone (Anemonia viridis)'s relationship with photosynthetic zooxanthellae, as well as molecular indicators of oxidative stress and metabolism. Anemones were exposed to natural daylight/night or either mild or strong ALAN intensities for four weeks, before quantifying zooxanthellae, superoxide dismutase (SOD) enzymes, and respiration rates. In comparison to natural conditio

Divergent responses of an armored and an unarmored dinoflagellate to ocean acidification.

Dinoflagellates, both armored and unarmored, with distinct cell wall difference, are being affected by elevated CO2-induced ocean acidification (OA). However, their specific responses to OA are not well understood. In this study, we investigated the physiological and molecular response of the armored species Prorocentrum obtusidens and the unarmored species Karenia mikimotoi to OA over a 28-day period. The results show that the two species responded differently to OA. Cell growth rate, particulate organic carbon (POC) content, and the activities of C4 pathway enzymes decreased in P. obtusidens

Effects of the toxic dinoflagellate Protoceratium reticulatum and its yessotoxins on the survival and feed ingestion of Argopecten purpuratus veliger larvae.

The effects of yessotoxins (YTXs) produced by the dinoflagellate Protoceratium reticulatum in the early stages of bivalves have not been studied in detail. The present study evaluates the effects of P. reticulatum and YTXs on the survival and feed ingestion of veliger larvae of Argopecten purpuratus. Larvae were 96 h-exposed to 500, 1000 and 2000 P. reticulatum cells mL-1, and their equivalent YTX extract was prepared in methanol. Results show a survival mean of 82 % at the highest density of dinoflagellate, and 38 % for larvae with the highest amount of YTX extract. Feed ingestion is reduced

Okadaic acid as a major problem for the seafood safety (Mytilus galloprovincialis) and the dynamics of toxic phytoplankton in the Slovenian coastal sea (Gulf of Trieste, Adriatic Sea).

This article presents the first results on shellfish toxicity in the Slovenian sea (Gulf of Trieste, Adriatic Sea) since the analytical methods for the detection of biotoxins (PSP, ASP, DSP and other lipophilic toxins) in bivalve molluscs were included in the national monitoring program in 2013. In addition to toxins, the composition and abundance of toxic phytoplankton and general environmental characteristics of the seawater (surface temperature and salinity) were also monitored. During the 2014-2019 study period, only lipophilic toxins were detected (78 positive tests out of 446 runs), of w

Biokinetics and biotransformation of paralytic shellfish toxins in different tissues of Yesso scallops, Patinopecten yessoensis.

Paralytic shellfish toxins (PSTs) are a group of natural toxic substances often found in marine bivalves. Accumulation, anatomical distribution, biotransformation and depuration of PSTs in different tissues of bivalves, however, are still not very well understood. In this study, we investigated biokinetics and biotransformation of PSTs in six different tissues, namely gill, mantle, gonad, adductor muscle, kidney, and digestive gland, in Yesso scallops Patinopecten yessoensis exposed to a toxic strain of dinoflagellate Alexandrium pacificum. High daily accumulation rate (DAR) was recorded at th

Species-specific PCR assays for Gambierdiscus excentricus and Gambierdiscus silvae (Gonyaulacales, Dinophyceae).

The two most toxic Gambierdiscus species identified from the Caribbean are G. excentricus and G. silvae. These species are the primary causes of ciguatera fish poisoning and likely contribute disproportionately to the toxicity of marine food webs. While Gambierdiscus species are difficult to distinguish using light or scanning electron microscopy, reliable species-specific molecular identification methods have been developed and used successfully to identify a number of other Gambierdiscus species. Corresponding species-specific assays are not yet available for G. excentricus and G. silvae, wh

A review of karenia mikimotoi: Bloom events, physiology, toxicity and toxic mechanism.

Karenia mikimotoi is a worldwide bloom-forming dinoflagellate in the genus Karenia. Blooms of this alga have been observed since the 1930s and have caused mass mortalities of fish, shellfish, and other invertebrates in the coastal waters of many countries, including Japan, Norway, Ireland, and New Zealand. This species has frequently bloomed in China, causing great financial losses (more than 2 billion yuan, Fujian Province, 2012). K. mikimotoi can adapt to various light, temperature, salinity, and nutrient conditions, which together with its complex life history, strong motility, and density-

The inhibitory effect of a non-yessotoxin-producing dinoflagellate, Lingulodinium polyedrum (Stein) Dodge, towards Vibrio vulnificus and Staphylococcus aureus.

The increased bacterial resistance to antibiotics has caused global concern, prompting the search for new compounds. Because of their abundance and diversity, marine phytoplankton are an important potential source of such compounds. Research on dinoflagellates has led to the discovery of inhibitors of bacterial growth. The marine dinoflagellate Lingulodinium polyedrum blooms in different regions of the world, including Mexico, and is also known to regulate the growth of other species in coastal waters. Here, we investigated the taxonomy of this dinoflagellate and characterized the ability of i

Diversification of the light-harvesting complex gene family via intra- and intergenic duplications in the coral symbiotic alga Symbiodinium.

The light-harvesting complex (LHC) is an essential component in light energy capture and transduction to facilitate downstream photosynthetic reactions in plant and algal chloroplasts. The unicellular dinoflagellate alga Symbiodinium is an endosymbiont of cnidarian animals, including corals and sea anemones, and provides carbohydrates generated through photosynthesis to host animals. Although Symbiodinium possesses a unique LHC gene family, called chlorophyll a-chlorophyll c2-peridinin protein complex (acpPC), its genome-level diversity and evolutionary trajectories have not been investigated.

Characteristic profiles of Ciguatera toxins in different strains of Gambierdiscus spp.

Ciguatera fish poisoning characterizes the intoxication caused by consumption of fish from tropical and subtropical areas, which have accumulated ciguatoxins (CTXs). The observed pattern of ciguatoxins in fish highly depends on the marine region and the causative organisms. It is evident that differences exist between ciguatoxins produced by certain strains of the dinoflagellate Gambierdiscus toxicus and other Gambierdiscus spp. In this context cultured strains purchased from the Provasoli-Guillard National Center for Culture of Marine Phytoplankton (CCMP) and strains from Vietnam were analyze

Harmful algal toxins of the Florida red tide (Karenia brevis): natural chemical stressors in South Florida coastal ecosystems.

The Florida red tide is a descriptive name for high concentrations of the harmful marine alga, Karenia brevis. Although most prevalent along the south-west Florida coast, periodic blooms have occurred throughout the entire US and Mexico Gulf coasts and the Atlantic coast to North Carolina. This dinoflagellate produces a suite of polyether neurotoxins, called brevetoxins, that cause severe impacts to natural resources, as well as public health. These naturally produced biotoxins may represent one of the most common chemical stressors impacting South Florida coastal and marine ecosystems. Impact

Bioavailability of dissolved organic phosphorus compounds to typical harmful dinoflagellate Prorocentrum donghaiense Lu.

The bioavailability of dissolved organic phosphorus (DOP) compounds to harmful alga Prorocentrum donghaiense Lu and its alkaline phosphatase activity (APA) were studied. Results showed that P. donghaiense could utilize the test DOP compounds such as glucose-6-phosphate (G-6-P), adenosine triphosphate (ATP) and ribonucleic acid (RNA) to sustain its growth. Nutrition between the test DOP compounds and orthophosphate was comparable. P. donghaiense could utilize intracellular phosphorus to sustain growth under depletion of dissolved phosphorus. Variation of APA in different test DOP compounds was

Pfiesteria and the skin: a practical update for the clinician.

Skin complaints, including an episodic burning sensation on contact with river water, were common among 13 persons with exposure to Maryland's Pocomoke River in the summer of 1997. While the majority of findings on dermatologic examination were unrelated to toxic dinoflagellate exposure, a subset of patients demonstrated otherwise unexplained erythematous, edematous papules on the trunk or extremities. Histopathologic findings were suggestive of an inflammatory, toxic, or allergic process. It may be speculated that these otherwise unexplained cutaneous findings represent a cutaneous reaction t

Biodiversity and spatial distribution of potentially ciguatera-causing dinoflagellates using DNA metabarcoding.

Ciguatera poisoning (CP) is a foodborne disease caused by eating seafood containing ciguatoxins from benthic dinoflagellates of the genus Gambierdiscus. Moreover, the related Fukuyoa, which often coexists with Gambierdiscus, also produces potent marine toxins. Though CP is endemic to tropical regions, it is increasingly being reported elsewhere, including the Canary Islands (NE Atlantic). In the Balearic Islands (W Mediterranean), potentially ciguatoxin-producing species have been found frequently in recent years, but so far without documented CP. Since toxin production varies greatly among Ga

A 15-Year Retrospective Review of Ciguatera in the Madeira Islands (North-East Atlantic, Portugal).

The first ciguatera fish poisoning (CFP) in Portugal dates from 2008 when 11 people reported CFP symptoms after consuming a 30 kg amberjack caught around the Selvagens Islands (Madeira Archipelago). Since then, 49 human poisonings have been reported. The emergence of a new threat challenged scientists and regulators, as methods for toxic microalgae analyses and ciguatoxin (CTX) detection were not implemented. To minimise the risk of ciguatera, the Madeira Archipelago authorities interdicted fisheries in Selvagens Islands and banned the capture of amberjacks weighing more than 10 kg in the enti

Effects of copper on the dinoflagellate Alexandrium minutum and its allelochemical potency.

The dinoflagellate Alexandrium minutum produces toxic compounds, including paralytic shellfish toxins, but also some unknown extracellular toxins. Although copper (Cu) is an essential element, it can impair microalgal physiology and increase their toxic potency. This study investigated the effect of different concentrations of dissolved Cu (7 nM, 79 nM and 164 nM) on A. minutum allelochemical potency, here defined as negative effects of a protist on competing protists through the release of chemicals. This was studied in relation to its physiology. The effects of Cu were assessed on A. minutum

Variation in allelopathy of extracellular compounds produced by Cylindrotheca closterium against the harmful-algal-bloom dinoflagellate Prorocentrum donghaiense.

Allelopathy between algae is an ecological strategy that can facilitate or inhibit the occurrence of algal blooms. The role of allelopathic effects of marine microalgae Cylindrotheca closterium in other phytoplankton population dynamics are still limited. In the current study, the effects of cell-free filtrates of diatom Cylindrotheca closterium on two common dinoflagellates (Prorocentrum donghaiense and Prorocentrum cordatum), a chrysophyceae (Isochrysis galbana) and a diatom (Chaetoceros curvisetus) were investigated within controlled laboratory experiments. Accumulation and Biotransformation of Dinophysis Toxins by the Surf Clam Mesodesma donacium.

Surf clams, Mesodesma donacium, were shown to accumulate toxins from Dinophysis acuminata blooms. Only pectenotoxin 2 (PTX2) and some of its derivatives were found, and no toxins from the okadaic acid group were detected. PTX2 seems to be transformed to PTX2 seco-acid (PTX2sa), which was found in concentrations more than ten-fold those of PTX2. The seco-acid was transformed to acyl-derivatives by esterification with different fatty acids. The estimated amount of these derivatives in the mollusks was much higher than that of PTX2. Most esters were originated by even carbon chain fatty acids, bu

Development of a LC-MS/MS method for the quantification of goniodomins A and B and its application to Alexandrium pseudogonyaulax strains and plankton field samples of Danish coastal waters.

An external standard of goniodomin A (GDA) was prepared from a strain of Alexandrium pseudogonyaulax originating from New Zealand and its chemical structure was confirmed by nuclear magnetic resonance (NMR) spectroscopy. Using the GDA standard, an ultra-performance liquid chromatography-tandem mass spectrometric (UPLC-MS/MS) method in selected reaction monitoring (SRM) mode was developed for separation and quantification of GDA. This method was successfully applied to planktonic field samples collected during an oceanographic expedition conducted with R/V Uthörn along the Danish west coast, Li
