Prophylactic oral diosgenin attenuates acute sleep deprivation-induced liver injury in mice via modulation of the gut microbiota and vascular barrier.

Acute sleep deprivation (SD) is an increasingly prevalent health risk, yet the early inflammatory processes associated with acute hepatic injury following SD remain largely elusive. Following a two-step screening of 265 natural products and bioactive small molecules, diosgenin (DG), a sapogenin found in Dioscorea species, was prioritized for subsequent in vivo evaluation. Under the rotating-platform paradigm in mice, 24-hour SD (SD24h) was associated with acute hepatic inflammation, increased overall intestinal permeability, gut vascular barrier (GVB)-associated alterations, gut microbiota dys

Diosgenin Promotes Bacterial Clearance in Bladder Epithelial Cells by Regulating Rab27b Expression: A Study Based on Virtual Screening and Experimental Validation.

INTRODUCTION: The annual rise in antimicrobial resistance (AMR) rates highlights the critical significance of exploring non-antibiotic therapies for infectious diseases. Urinary tract infection (UTI) is one of the most common bacterial infectious diseases, and uropathogenic Escherichia coli (UPEC), its primary pathogenic bacterium, invades bladder epithelial cells (BECs) to survive and proliferate intracellularly, thereby evading the host's immune clearance. Rab27b mediates the expulsion of intracellular bacteria, and upregulating Rab27b expression can effectively reduce the number of intracel

Enhanced Neuroprotection by Diosgenin and Pterostilbene Combination Against Neurotoxicity Induced by Amyloid-Β 1-42 in SH-SY5Y Differentiated Cell Models.

BACKGROUND: Alzheimer's disease (AD) is the predominant age-related neurodegenerative condition, characterised by the gradual and irreversible loss of neurons. Key pathological features include amyloid plaques and neurofibrillary tangles, which trigger a chronic inflammatory response in the brain, leading to microglial activation and proliferation. PURPOSE: This study evaluated the neuroprotective potential of diosgenin (DGN) and pterostilbene, two phytoconstituents with known antioxidant and anti-inflammatory properties, in amyloid-β 1-42 exposed SH-SY5Y cells. METHODS: Human neuroblastoma

Construction of ROS-Responsive Hyaluronic Acid Modified Paclitaxel and Diosgenin Liposomes and Study on Synergistic Enhancement of Anti-Ovarian Cancer Efficacy.

PURPOSE: Ovarian cancer is a fatal gynecologic malignancy with a high rate of abdominal metastasis. Chemotherapy still has a poor clinical prognosis for ovarian cancer patients, with cell proliferation and angiogenesis leading to invasion, migration, and recurrence. To overcome these obstacles, we constructed a novel HA-modified paclitaxel and diosgenin liposome (PEG-TK-HA-PDLPs) using two novel functional materials, DSPE-PEG2000-HA and DSPE-PEG2000-TK-PEG5000, to specifically deliver the drugs to the tumor site in order to reduce OC cell proliferation and anti-angiogenic generation, thereby i

Oral administration with diosgenin enhances the induction of intestinal T helper 1-like regulatory T cells in a murine model of food allergy.

Although the development of T helper (Th)1-like regulatory T (Treg) cells under Th1 inflammatory conditions has been reported, the role of Th1-like Treg cells in Th2 allergic responses remains mostly unclear. We previously demonstrated that diosgenin, the major sapogenin contained in the Chinese yam, attenuated food allergy and augmented Th1 and Treg immune responses. In this study, we hypothesized that diosgenin may enhance the induction of Th1-like Treg cells in the gut of mice with food allergy. Ovalbumin (OVA)-sensitized BALB/c mice were gavaged daily with diosgenin and received repeatedly

Diosgenin Alleviates Age-Related Sarcopenia by Promoting Satellite Cell Proliferation and Myogenic Differentiation via Activation of the SIRT1/PGC-1α Signaling Pathway.

Age-related sarcopenia is characterized by a progressive decline in skeletal muscle mass and function, with satellite cell dysfunction representing a central pathogenic mechanism. Diosgenin, a steroidal saponin derived from plants of the Dioscorea genus, has demonstrated potential anti-aging properties; however, its role in sarcopenia remains unclear. In this study, naturally aged C57BL/6J mice and a D-galactose (D-gal)-induced senescent C2C12 cell model were employed to systematically investigate the effects of diosgenin on muscle function, satellite cell dynamics, and the sirtuin 1 (SIRT1)/p

Diosgenin Ameliorates Insulin Resistance in High-Fat Diet-Induced Metabolic Dysfunction-Associated Steatotic Liver Disease Rats via the Adiponectin/ADIPOR1/APPL1 Pathway.

Insulin resistance (IR) is a central pathogenic driver of metabolic dysfunction-associated steatotic liver disease (MASLD), yet current therapeutic strategies remain insufficient. Diosgenin (DG), a well-characterized natural compound, has been reported to modulate IR-related mechanisms and, thus, holds significant promise for MASLD treatment. In this study, rodent models fed a high-fat diet and HepG2 cells treated with free fatty acids were employed to simultaneously evaluate systemic metabolic outcomes (including hepatic indices and glucose tolerance) and cellular responses (including lipid a

Identification and a phased pH control strategy of diosgenin bio-synthesized by an endogenous Bacillus licheniformis Syt1 derived from Dioscorea zingiberensis C. H. Wright.

Diosgenin is widely used as one precursor of steroidal drugs in pharmaceutical industry. Currently, there is no choice but to traditionally extract diosgenin from Dioscorea zingiberensis C. H. Wright (DZW) or other plants. In this work, an environmentally friendly approach, in which diosgenin can be bio-synthesized by the endophytic bacterium Bacillus licheniformis Syt1 isolated from DZW, is proposed. Diosgenin produced by the strain was identified by high-performance liquid chromatography (HPLC), nuclear magnetic resonance (NMR), and Fourier transform infrared spectroscopy (FTIR). The thermal

Diosgenin enhances transplanted human oligodendrocyte precursor cell-mediated myelination in preterm white matter injury by inhibiting matrix metalloproteinase-9-driven TGF-β/Smad2/3 signaling.

Preterm white matter injury (PWMI) is a major cause of long-term motor and cognitive disability, and there are no therapies that directly promote oligodendrocyte-lineage maturation. Here, we identify the natural steroid sapogenin diosgenin as a potent pro-myelinating modulator for transplanted human oligodendrocyte precursor cells (hOPCs) using a hypoxia-ischemia mouse model of PWMI, together with in vitro microglia-OPC co-culture systems. Diosgenin crosses the blood-brain barrier and promotes the differentiation of both transplanted hOPCs and endogenous OPCs into myelinating oligodendrocytes,

Systemic role of melatonin in enhancing temperature stress tolerance in fenugreek: coordination of antioxidant defense, hormonal regulation, energy status, sulfur metabolism, and diosgenin pathway genes.

BACKGROUND: Temperature stresses, encompassing heat and cold, significantly hinder the growth and productivity of fenugreek. Melatonin, known for its antioxidant and signaling properties, enhances plant stress tolerance, yet its specific role and mode of action in fenugreek under temperature stress remain largely uncharacterized. This study investigated the protective effects of exogenous melatonin application (0, 30, 60, and 90 ppm) on fenugreek plants exposed to temperature stress (10 °C and 42 °C). RESULTS: Melatonin, particularly at 60 ppm, substantially improved photosynthetic efficiency

Repeated evolution of cytochrome P450-mediated spiroketal steroid biosynthesis in plants.

Diosgenin is a spiroketal steroidal natural product extracted from plants and used as the single most important precursor for the world steroid hormone industry. The sporadic occurrences of diosgenin in distantly related plants imply possible independent biosynthetic origins. The characteristic 5,6-spiroketal moiety in diosgenin is reminiscent of the spiroketal moiety present in anthelmintic avermectins isolated from actinomycete bacteria. How plants gained the ability to biosynthesize spiroketal natural products is unknown. Here, we report the diosgenin-biosynthetic pathways in himalayan pari
