Machine Learning and Blood-Targeted Proteomics Enable Early Prediction and Etiological Discrimination of Hypertensive Pregnancy Disorders.

Imperfect first-trimester screening for hypertensive disorders of pregnancy (HDP) means many high-risk women miss the window for preventive aspirin, and the biological heterogeneity of HDPs is overlooked. This study aimed to leverage first-trimester serum proteomics to create a more precise tool for predicting preeclampsia (PE) and differentiating it from other HDPs. A prospective nested case-control study (n = 172) was conducted using targeted liquid chromatography-multiple reaction monitoring-mass spectrometry (LC-MRM-MS) proteomic profiling of 115 proteins. Machine learning (ML) methods wer

Precise discrimination of mycobacterial pulmonary diseases via multimodal machine learning integrating chest CT and clinical markers.

INTRODUCTION: Differentiating Mycobacterium tuberculosis lung disease (MTB-LD) from nontuberculous mycobacterial lung disease (NTM-LD) remains clinically challenging because of overlapping symptoms and imaging features, as well as the slow turnaround of conventional culture-based diagnostics. METHODS: This retrospective study enrolled 102 patients with microbiologically confirmed mycobacterial lung disease, including 53 patients with MTB-LD and 49 patients with NTM-LD. We developed and validated an interpretable multimodal machine-learning framework integrating clinical symptoms, hematologica

Machine Learning-Driven Multi-Emission Fluorescence Array for Simultaneous Size Discrimination and Quantification of Gold Nanoparticles.

Gold nanoparticles (AuNPs) exhibit size-dependent environmental behaviors and bioaccumulation risks, necessitating precise characterization of their hydrodynamic dimensions and concentrations for toxicity assessment. Existing analytical platforms are limited in their ability to perform efficient multidimensional analysis due to time-consuming procedures and the reliance on large, costly equipment. Herein, we developed a multiemission fluorescence sensing array that assisted with machine learning analysis for the simultaneous size discrimination and quantification of AuNPs. The sensor integrate

Discrimination of Rheumatoid and Psoriatic Arthritis Based on Raman and NIR Spectra Using Machine-Learning Algorithms.

Rheumatoid arthritis (RA) and psoriatic arthritis (PsA) are chronic autoimmune diseases. They share similar symptoms. The lack of specific markers can lead to misdiagnosis. Using spectroscopic information on the chemical composition of body fluids can effectively differentiate these diseases. The discriminant analysis results are presented based on Raman and near-infrared (NIR) spectra of freeze-dried blood sera. The performance of partial least squares discriminant analysis (PLS-DA) and counter-propagation artificial neural network (CP-ANN) techniques in differentiation between RA (n = 30) an

Discrimination of Dengue Diseases in Children Using Surface-Enhanced Raman Spectroscopy Coupled with Machine Learning Approaches.

This study introduces a novel approach to dengue diagnostics by leveraging surface-enhanced Raman spectroscopy (SERS) coupled to machine learning. This method addresses the critical need for rapid and accurate identification of dengue virus (DENV) infection and prediction of the disease severity. For the first time, a commercialized SERS substrate is applied to analyze plasma samples from 60 pediatric patients, equally distributed among other febrile illnesses (OFI), dengue fever (DF), and dengue hemorrhagic fever (DHF) cases. This innovative application of SERS technology captures unique mole

Toward Reagent-Free Discrimination of Alzheimer's Disease Using Blood Plasma Spectral Digital Biomarkers and Machine Learning.

BACKGROUND: Alzheimer's disease (AD) is the most prevalent neurodegenerative disease. The detection of early-stage AD is particularly desirable because it would allow early intervention. However, a minimally invasive, low-cost, and accurate discrimination or diagnostic method for AD is especially difficult in the earliest stage of AD. OBJECTIVE: The aim of this research is to discover blood plasma spectral digital biomarkers of AD, develop a novel intelligent method for the discrimination of AD and accelerate the translation of Fourier transform infrared (FTIR) spectral-based disease discrimi

Deep Learning for Discrimination of Hypertrophic Cardiomyopathy and Hypertensive Heart Disease on MRI Native T1 Maps.

BACKGROUND: Native T1 and radiomics were used for hypertrophic cardiomyopathy (HCM) and hypertensive heart disease (HHD) differentiation previously. The current problem is that global native T1 remains modest discrimination performance and radiomics requires feature extraction beforehand. Deep learning (DL) is a promising technique in differential diagnosis. However, its feasibility for discriminating HCM and HHD has not been investigated. PURPOSE: To examine the feasibility of DL in differentiating HCM and HHD based on T1 images and compare its diagnostic performance with other methods. STU

A Machine Learning-Based Approach to Discrimination of Tauopathies Using [18 F]PM-PBB3 PET Images.

BACKGROUND: We recently developed a positron emission tomography (PET) probe, [18 F]PM-PBB3, to detect tau lesions in diverse tauopathies, including mixed three-repeat and four-repeat (3R + 4R) tau fibrils in Alzheimer's disease (AD) and 4R tau aggregates in progressive supranuclear palsy (PSP). For wider availability of this technology for clinical settings, bias-free quantitative evaluation of tau images without a priori disease information is needed. OBJECTIVE: We aimed to establish tau PET pathology indices to characterize PSP and AD using a machine learning approach and test their validi

Unpredictable chronic mild stress differentially impairs social and contextual discrimination learning in two inbred mouse strains.

Alterations in the social and cognitive domain are considered important indicators for increased disability in many stress-related disorders. Similar impairments have been observed in rodents chronically exposed to stress, mimicking potential endophenotypes of stress-related psychopathologies such as major depression disorder (MDD), anxiety, conduct disorder, and posttraumatic stress disorder (PTSD). Data from numerous studies suggest that deficient plasticity mechanisms in hippocampus (HC) and prefrontal cortex (PFC) might underlie these social and cognitive deficits. Specifically, stress-ind

Measuring discrimination- and reversal learning in mouse models within 4 days and without prior food deprivation.

Many neurological and psychiatric disorders are characterized by deficits in cognitive flexibility. Modeling cognitive flexibility in mice enables the investigation of mechanisms underlying these deficits. The majority of currently available behavioral tests targeting this cognitive domain are reversal learning tasks that require scheduled food restriction, extended training periods and labor-intensive, and stress-inducing animal handling. Here, we describe a novel 4-day (4-d) continuously running task measuring discrimination- and reversal learning in an automated home cage (CognitionWall DL/

Impaired dimensional selection but intact use of reward feedback during visual discrimination learning in Parkinson's disease.

It has been suggested that Parkinson's disease (PD) impairs the ability to learn on the basis of reward or reinforcing feedback i.e., by trial-and-error. In many learning tasks, particular 'dimensions' of stimulus information are relevant whilst others are irrelevant; therefore, efficient performance depends on identifying the dimensions of these 'compound' stimuli and selecting the relevant dimension for further processing. We investigated the ability of patients with PD, as well as patients with Huntington's disease and patients with frontal or temporal lobe lesions, to learn visual discrimi

Hippocampus and the blood supply to TE: parahippocampal pial section impairs visual discrimination learning in monkeys.

In the base of each temporal lobe in 4 monkeys (Macaca fascicularis) we sectioned the pia mater in an antero-posterior line along the most medial part of the inferior temporal visual association cortex (area TE), lateral to the parahippocampal cortex. This pial section interrupts the blood flow in branches of the posterior cerebral artery which cross the parahippocampal gyrus en route to area TE. Histologically, the brains of 2 of these animals did not show any abnormalities in cresyl violet stained material, apart from the small cortical damage along the line of the pial section itself. Concept discrimination learning by preschool children using facial stimuli differing in age, sex, race, and eyeglasses.

In two experiments, the relative salience of the variables age, sex, race, and eyeglasses was determined with the use of a discrimination learning task and preschool-age Ss. The stimuli for the task were 112 frontal head and shoulder color photographs arranged in pairs. In both experiments, sex discrimination was the easiest to learn, followed by race and age, which did not differ from each other. Each of the variables of sex, age, and race, however, proved to be more salient than glasses which was not learned by any of the 19 Ss who were given a glasses discrimination problem in Experiment 1.

Mice with exonic RELN deletion identified from a patient with schizophrenia have impaired visual discrimination learning and reversal learning in touchscreen operant tasks.

The Reelin gene (RELN) encodes a large extracellular protein, which has multiple roles in brain development and adult brain function. It activates a series of neuronal signal transduction pathways in the adult brain that function in synaptic plasticity, dendritic morphology, and cognitive function. To further investigate the roles of Reln in brain function, we generated a mouse line using the C57BL/6 J strain with the specific Reln deletion identified from a Japanese patient with schizophrenia (Reln-del mice). These mice exhibited abnormal sociality, but the pathophysiological significance of

Deep learning discriminates thymic epithelial tumors' histological subtypes using digital pathology.

BACKGROUND: Thymic epithelial tumors (TETs) are rare malignancies that pose significant diagnostic challenges due to their heterogeneous histological patterns and substantial interobserver variability in classification. Despite standardized World Health Organization (WHO) classification criteria, diagnostic concordance remains suboptimal, particularly in nonexpert settings, where second-opinion reviews lead to diagnostic reclassification in up to 57% of cases. Deep learning may offer a tool to reduce diagnostic variability and improve the consistency of histological classification. MATERIALS

Detecting Anomalies in Retinal Diseases Using Generative, Discriminative, and Self-supervised Deep Learning.

IMPORTANCE: Anomaly detectors could be pursued for retinal diagnoses based on artificial intelligence systems that may not have access to training examples for all retinal diseases in all phenotypic presentations. Possible applications could include screening of population for any retinal disease rather than a specific disease such as diabetic retinopathy, detection of novel retinal diseases or novel presentations of common retinal diseases, and detection of rare diseases with little or no data available for training. OBJECTIVE: To study the application of anomaly detection to retinal disease

Role of the right temporal lobe in visual-cue learning during repeated pattern discriminations.

The ability to acquire visual cues for discriminating between two targets against a background of visually similar items was examined in 64 patients with unilateral temporal- or frontal-lobe excisions and 20 normal control subjects. The subjects were tested on two versions (letters and abstract designs) of a visual cue-learning task that was an adaptation and extension of RABBITT's (Am. J. Psychol. 80, 1-13, 1967) visual-search paradigm. For both versions, the material consisted of two packs of cards, each containing a different set of background letters or designs. Bridging Generative and Discriminative Noisy-Label Learning via Direction-Agnostic EM Formulation.

Although noisy-label learning is often approached with discriminative methods for simplicity and speed, generative modeling offers a principled alternative by capturing the joint mechanism that produces features, clean labels, and corrupted observations. However, prior work typically (i) introduces extra latent variables and heavy image generators that bias training toward reconstruction, (ii) fixes a single data-generating direction (Y →X or X →Y), limiting adaptability, and (iii) assumes a uniform prior over clean labels, ignoring instance-level uncertainty. Here, we propose a single-stage,
