Effect of disulfidptosis-related genes SLC3A2, SLC7A11 and FLNB polymorphisms on risk of autoimmune thyroiditis in a Chinese population.

PURPOSE: This study aimed to evaluate the associations between disulfidptosis related genes-SLC3A2, SLC7A11 and FLNB polymorphisms and risk of autoimmune thyroiditis (AIT). METHODS: Six SNPs in the SLC3A2, SLC7A11 and FLNB were genotyped in 650 AIT cases and 650 controls using a MassARRAY platform. RESULTS: Minor alleles of SLC3A2-rs12794763, rs1059292 and FLNB-rs839240 might lead to a higher risk of AIT (p < 0.001), while SLC7A11-rs969319-C allele tends to decrease the risk of the disease (p = 0.006). Genetic model analysis showed that SLC3A2-rs12794763, SLC3A2-rs1059292 and FLNB-rs839240 p

Disulfidptosis as an immunometabolic rheostat in gastrointestinal cancers: tuning the balance between T Cell exhaustion and immunogenic cell death.

Disulfidptosis is a recently characterized regulated cell death pathway driven by disulfide stress. However, its immunological consequences in the tumor microenvironment remain poorly defined. In this review, we propose a conceptual framework in which disulfidptosis functions as an immunometabolic rheostat, wherein the net outcome-T cell exhaustion versus immunogenic cell death-is critically dependent on stress intensity, kinetics, and cellular context. We hypothesize that in glucose-deprived gastrointestinal tumors, chronic sub-lethal disulfide stress may erode CD8+ T cell effector function t

A new gene signature associated with disulfidptosis that forecasts myasthenia gravis and suggests infiltration of immune microenvironment in thymoma patients.

INTRODUCTION: The mechanism of thymoma-associated myasthenia gravis (TAMG) is currently unknown, although patients with TAMG experience more severe myasthenic symptoms and have worse prognoses compared to regular thymoma patients. The objective of this research is to create a transcriptome map of TAMG using genes linked to disulfidptosis, detect possible biomarkers, and examine the disparities in the tumor immune microenvironment (TIME) among different thymoma patients. The findings will offer valuable knowledge for personalized treatment alternatives. METHODS: Thymoma samples' RNA-seq data,

Redox Metabolism and Metabolic Vulnerabilities in Diabetes-Driven Pancreatic Cancer: Targeting System Xc-, mTORC1, and Disulfidptosis as Therapeutic Strategy.

Type 2 diabetes mellitus (T2DM) is increasingly recognized as a significant risk factor for pancreatic cancer, with both diseases sharing complex metabolic and molecular underpinnings. Insulin resistance, chronic inflammation, and aberrant activation of the KRAS-mTORC1 signaling axis collectively foster a tumor-permissive microenvironment and disrupt glucose regulation. Recent advances have highlighted disulfidptosis, a novel NADPH-depletion and disulfide-accumulation-driven cell death, as a therapeutic vulnerability in KRAS-mutated, cystine-dependent tumors. This process is linked to the meta

TCM-Derived Small Molecules Targeting Metabolic Vulnerabilities in NSCLC: Ferroptosis-Centered Mechanisms and Emerging Cuproptosis- and Disulfidptosis-Related Vulnerabilities.

Non-small cell lung cancer (NSCLC) remains the leading cause of cancer-related mortality worldwide and is characterized by therapeutic resistance, metabolic plasticity, and immune evasion. Accumulating evidence indicates that metabolic reprogramming not only supports tumor growth but also creates exploitable vulnerabilities linked to regulated cell death. Traditional Chinese medicine (TCM)-derived small molecules have attracted increasing attention owing to their structural diversity, multitarget properties, and broad pharmacological activities. In this review, we summarize recent advances in

Rhodium(III)-Nitroxyl Radical Complex Triggers Dual-Pronged Disulfidptosis-Apoptosis in Hepatocellular Carcinoma via Metabolic Sabotage and Redox Catalysis.

Hepatocellular carcinoma (HCC) resists apoptosis-targeting therapies, necessitating the development of agents targeting alternative cell death pathways. Here, we report the discovery of a nitroxyl radical-conjugated Rh(III) complex (OG-Rh) that triggers dual disulfidptosis and apoptosis via synergistic metabolic sabotage and redox catalysis. OG-Rh inhibited glucose uptake, depleted NADPH, and induced disulfidptosis, a novel disulfide-stress-mediated death, by inducing actin cytoskeleton collapse via pathogenic disulfide over-crosslinking. Simultaneously, its tumor-selective superoxide dismutas

Construction of a disulfidptosis-related glycolysis gene risk model to predict the prognosis and immune infiltration analysis of gastric adenocarcinoma.

BACKGROUND: The pattern of cell death known as disulfidptosis was recently discovered. Disulfidptosis, which may affect the growth of tumor cells, represents a potential new approach to treating tumors. Glycolysis affects tumor proliferation, invasion, chemotherapy resistance, the tumor microenvironment (TME), and immune evasion. However, the efficacy and therapeutic significance of disulfidptosis-related glycolysis genes (DRGGs) in stomach adenocarcinoma (STAD) remain uncertain. METHODS: STAD clinical data and RNA sequencing data were downloaded from the TCGA database. DRGGs were screened us

Disulfidptosis-related genes define a prognostic signature and novel therapeutic targets in Ewing's sarcoma through transcriptomic analysis and experimental validation.

BACKGROUND: Ewing's sarcoma (ES) is a malignant osseous neoplasm characterized by a dismal prognosis, particularly in its metastatic variant. The significance of disulfidptosis-a newly identified cell death mechanism induced by cystine metabolic imbalance and mitochondrial dysfunction-has yet to be investigated in ES. Thus, the aim of this study was to assess the prognostic significance of disulfidptosis-related genes (DRGs) in this disease. METHODS: We analyzed four GEO datasets to examine nine DRGs through differential expression, co-expression networks, and functional enrichment analyses.

SLC7A11-mediated cystine accumulation and Rac1-WRC-Arp2/3-driven actin cytoskeletal folding synergistically induce disulfidptosis in acute myocardial infarction.

Disulfidptosis, a newly identified form of cell death, holds significant biological relevance; however, its role in cardiovascular diseases remains poorly characterized. To investigate this, disulfide bond formation was evaluated using non-reducing Western blot, while cytoskeletal alterations were assessed via phalloidin staining. In vivo experiments using a rat model of myocardial infarction (MI) and in vitro experiments using H9c2 cardiomyocytes subjected to oxygen‑glucose deprivation (OGD) both demonstrated upregulation of SLC7A11, NADPH depletion, and cystine accumulation under OGD or MI c
