First Gammaherpesvirus detection in a free-living Mediterranean bottlenose dolphin.

Recently, herpes viruses have been detected in different cetacean species from the Atlantic and in Mediterranean striped dolphins (Stenella coeruleoalba). While pathogens such as cetacean morbillivirus have been widely studied following recent epizootics, herpesvirus (HV) distribution and pathogenic effects in cetaceans are still understudied. This study reports the first molecular identification of a Gammaherpesvirus in the genital mucosa of a free-living Mediterranean bottlenose dolphin (Tursiops truncatus) stranded off the coast of central Italy. Sequenced herpesviral PCR product was closel

Biliary cirrhosis caused by Campula spp. in a dolphin and four porpoises.

Biliary cirrhosis produced by Campula spp. is described in 1 striped dolphin Stenella coeruleoalba and 4 harbour porpoises Phocoena phocoena. The hepatic lesions consisted of severe proliferation of fibrous connective tissue with loss of the lobular pattern, nodular regeneration of the hepatic tissue, bile duct hyperplasia and severe inflammatory infiltrate composed of eosinophils, macrophages, lymphocytes and plasma cells. These lesions were associated with severe infestation by Campula spp. Although inflammatory and degenerative hepatic lesions are frequently found in stranded dolphins, bili

Development of an indirect immunofluorescence technique for the diagnosis of toxoplasmosis in bottlenose dolphins.

The diagnosis of toxoplasmosis is often complicated by the lack of specific clinical symptoms or postmortem features, in humans and other animals. The only diagnostic test described so far for the serological diagnosis of Toxoplasma gondii in marine mammals is the modified agglutination test (Dubey et al., Am J Vet Res 48(8):1239-1243, 1987). The development of more sensible and specific immunological techniques requires specific antibodies, which are currently unavailable in the scientific market. Indirect immunofluorescence (IIF) is one of the most widely used methods for the diagnosis of to

Pulmonary ultrasound findings in a bottlenose dolphin Tursiops truncatus population.

Lung disease is common among wild and managed populations of bottlenose dolphins Tursiops truncatus. The purpose of the study was to apply standardized techniques to the ultrasound evaluation of dolphin lungs, and to identify normal and abnormal sonographic findings associated with pleuropulmonary diseases. During a 5 yr period (2005 to 2010), 498 non-cardiac thoracic ultrasound exams were performed on bottlenose dolphins at the Navy Marine Mammal Program in San Diego, California, USA. Exams were conducted as part of routine physical exams, diagnostic workups, and disease monitoring. In the ma

Mycobacterium abscessus pneumonia in an Atlantic bottlenose dolphin (Tursiops truncatus).

Mycobacterium abscessus pneumonia was diagnosed antemortem in a 23-yr-old male Atlantic bottlenose dolphin (Tursiops truncatus). Clinical signs included lethargy, hyporexia, coughing, and bloody respiratory discharge. Diagnostic findings included neutrophilic leukocytosis, anemia, elevated erythrocyte sedimentation rate, and repeated forceful exhaled breath (sputum) cytology, with acute inflammatory cells and acid-fast positive beaded rods. The bacteria were initially identified free in the sputum sample and subsequently were seen within neutrophils. A culture was positive for a rapidly growin

Molecular diagnosis of lobomycosis-like disease in a bottlenose dolphin in captivity.

We report the diagnosis and molecular characterization of lobomycosis-like lesions in a captive bottlenose dolphin. The clinical picture and the absence of growth in conventional media resembled the features associated with Lacazia loboi. However sequencing of ribosomal DNA and further phylogenetic analyses showed a novel sequence more related to Paracoccidioides brasilensis than to L. loboi. Moreover, the morphology of the yeast cells differed from those L. loboi causing infections humans. These facts suggest that the dolphin lobomycosis-like lesions might have been be caused by different a d

Microcystic meningioma in a dolphin (Delphinus delphis): immunohistochemical and ultrastructural study.

A wild common dolphin was found stranded on the French Atlantic coast. At necropsy, an intracranial grey- to tan-coloured mass (7 x 5 x 4 cm) was found at the right cerebellopontine angle, compressing the right cerebellar hemisphere, the brainstem and the occipital lobe of the right cerebral hemisphere. Microscopically, the tumour was composed of small lobules of polygonal to elongated neoplastic cells with multifocal areas of stellate and vacuolated cells. Neoplastic cells strongly expressed vimentin, S-100 protein and neuron-specific enolase. They were rarely positive for cytokeratin. Ultras

Investigating bottlenose dolphin physiology using near-infrared spectroscopy.

Cetaceans spend considerable periods of time underwater, but much about the physiological response of these animals during a dive remains unknown. We present an approach that combines near-infrared spectroscopy (NIRS) with kinematic measurements to investigate bottlenose dolphin hemodynamics in managed settings during voluntary breathing at the surface, diving to a submerged target and free swimming. For all conditions, oxygenated hemoglobin initially increased following respiration as oxygen saturation increased and then decreased until the following breath. Deoxygenated hemoglobin remained r

Systemic mucormycosis caused by Rhizopus microsporus in a captive bottlenose dolphin.

A 6-year-old female bottlenose dolphin (Tursiops truncatus) kept in dolphinarium died after a 3.5-month period of lethargy and inappetence despite antibiotics and supportive care. At necropsy, gross findings included diffuse varying-sized nodules in the lungs and scattered nodules throughout the heart, spleen, mesenteric and hilar lymph node and kidney. Microscopically, the lesions were characterised by disseminated fungal pyogranulomas with numerous intralesional Mucor-like fungi. The fungi structures were demonstrated by Periodic acid-Schiff and Gomori methenamine silver stain. Molecular ana

Lobomycosis in man and lobomycosis-like disease in bottlenose dolphin, Venezuela.

We report 1 case of lobomycosis caused by Lacazia loboi in a fisherman and 1 case of lobomycosis-like disease in a bottlenose dolphin (Tursiops truncatus) along the coast of Venezuela. These findings suggest that the marine environment is a likely habitat for L. loboi and a reservoir for infection.

Congenital toxoplasmosis in a Indo-Pacific bottlenose dolphin (Tursiops aduncus).

Toxoplasma gondii was identified in tissues of a stillborn late-term fetus from an Indo-Pacific bottlenose dolphin (Tursiops aduncus). Myocardial necrosis and nonsuppurative inflammation in the heart and nonsuppurative necrotizing encephalitis were associated with tachyzoites and tissue cysts. The diagnosis was confirmed by immunohistochemical staining with anti-T. gondii-specific polyclonal rabbit serum.

Time, pattern, and heterochrony: a study of hyperphalangy in the dolphin embryo flipper.

The forelimb of whales and dolphins is a flipper that shows hyperphalangy (numerous finger bones). Hyperphalangy is also present in marine reptiles, including ichthyosaurs and plesiosaurs. The developmental basis of hyper-phalangy is unclear. Kükenthal suggested that phalanx anlagen split into three pieces during cetacean development, thereby multiplying the ancestral number. Alternatively, Holder suggested that apical ectodermal ridge (AER)-directed limb outgrowth might be prolonged by a timing shift (heterochrony), leading to terminal addition of extra phalanges. We prepared a series of whol

Anatomy of the cranial nerve foramina in the Risso's dolphin (Grampus griseus).

In the present study, we examined the cranial nerve foramina of Risso's dolphin (Grampus griseus). There were two distinguishable characters in the cranial nerve foramina compared with terrestrial mammals. One was that the foramen infraorbitale was composed of several holes, but not a single hole. They should therefore be termed foramina infraorbitales. The infraorbital nerves ran through these foramina, went into the 'melon' and then branched in a complicated fashion. The facial nerve innervated the muscles surrounding the melon. A well-developed infraorbital and facial nerve complex may cont

Human infection due to Mycobacterium marinum after a dolphin bite.

A young man employed at the local aquarium was bitten by a bottlenose dolphin (Tursiops truncatus) during a training session, receiving a slight injury which healed rapidly. Some two months later fluctuant swellings appeared in the region of the bite, which developed into indolent ulcers which have not completely healed seven months after the original bite. Cultures taken on two occasions have yielded a pure growth of Mycobacterium marinum.

Molecular characterization of four Helicobacter cetorum strains from dolphins compared to human Helicobacter pylori.

Helicobacter species colonize the stomachs of many aquatic and terrestrial mammals, including Helicobacter pylori in humans and Helicobacter cetorum in dolphins. There are several H. cetorum genome sequences in databases, but a detailed molecular characterization of these bacteria is missing. Here, we compared four H. cetorum isolates from dolphins with H. pylori strains using electron microscopy as well as structural and functional analyses. All strains expressed similarly high urease activity and were hemolytic to erythrocytes. Western blots revealed conserved expression of flagellin-A, neut
