Formulation strategies for the development of high drug-loaded amorphous solid dispersions.

Amorphous solid dispersions (ASD) have gained tremendous attention over the past two decades as one of the most promising techniques for enhancing the solubility of poorly water-soluble drugs. However, low drug loading is one of the major challenges of ASD technology that limits its commercialization to only a few drug candidates. Increasing the drug loading increases the risk of recrystallization during storage (solid state) and/or during dissolution (solution state). Various formulation and process-related strategies have been explored that open the possibility of formulating high drug-loade

A review on the treatment of intimal hyperplasia with perivascular medical devices: role of mechanical factors and drug release kinetics.

INTRODUCTION: Intimal hyperplasia (IH) is a significant factor limiting the success of revascularization surgery for blood flow restoration. IH results from a foreign body response and mechanical disparity that involves complex biochemical reactions resulting in graft failure. The available treatment option utilizes either different pharmacological interventions or mechanical support to the vascular grafts with limited success. AREAS COVERED: This review explains the pathophysiology of IH, responsible mechanical and biological factors, and treatment options, emphasizing perivascular devices.

Fabrication, GSH-responsive drug release, and anticancer properties of thioctic acid-based intelligent hydrogels.

Injectable hydrogels are potential local drug delivery systems since they contain plenty of water and soft like biological tissues. Such hydrogels could be injected directly into the tumor site where the drug is released under the tumor microenvironment. However, drug loaded hydrogels for cancer treatment based on lipoic acid (natural small molecule) have not been exploited. Here, a novel poly(lipoic acid)-poly(ethylene glycol) (PEG-PTA) hydrogels were prepared through a two-step reaction. The hydrogels contained disulfide bonds, so they could be degraded via the thiol exchange reaction with t

Potential Assessment of Topical Felbinac-Loaded Cubosomal Gel in Soft Tissue Injury in Albino Rats.

Muscle strain is one of the most common injuries with high intermittence rate. Due to diverseness of strain injuries, different experimental animal models are employed to investigate such injuries with reproducible results. Cubosomes, an emerging nano drug delivery tool, are considered ideal carriers for the topical delivery of lipophilic drugs to treat local inflammations with reduced frequency of application for prolonged periods. This work describes the development of Felbinac-loaded cubosomal gel and investigated the treatment of inflammation and tissue injury in vivo. Sciatic Function Ind

Self-healing gelatin-based shape memory hydrogels via quadruple hydrogen bonding and coordination crosslinking for controlled delivery of 5-fluorouracil.

Gelatin-UPy based on gelatin with ureidopyrimidinone (UPy) side chains was prepared with varying content of UPy units. On increasing the UPy content, the glass transition temperature, crystallinity and swelling decreased. Gelatin-UPy demonstrated self-healing properties as the UPy units could reversibly form dimers. At the same time, the gelatin-UPy and gelatin-UPy hydrogels demonstrated thermal responsive shape memory behaviors. The introduction of coordination crosslinking by introducing Fe3+ in gelatin-UPy hydrogels not only enhanced the crosslinking degree of gelatin-UPy and decreased the

Dental implants modified with drug releasing titania nanotubes: therapeutic potential and developmental challenges.

The transmucosal nature of dental implants presents a unique therapeutic challenge, requiring not only rapid establishment and subsequent maintenance of osseointegration, but also the formation of resilient soft tissue integration. Key challenges in achieving long-term success are sub-optimal bone integration in compromised bone conditions and impaired trans-mucosal tissue integration in the presence of a persistent oral microbial biofilm. These challenges can be targeted by employing a drug-releasing implant modification such as TiO2 nanotubes (TNTs), engineered on titanium surfaces via elect

Survival of worm masses of Onchocerca ochengi in gerbils and hamsters: implications for the development of an in vivo macrofilaricide screening model.

Onchocerciasis, the second leading infectious cause of blindness, afflicts approximately 21 million people globally. Its control is limited to the use of the microfilaricidal drugs, ivermectin and moxidectin. Both drugs are unable to kill the adult worms which can survive for up to 15 years in patients, justifying the urgent need for potent and novel macrofilaricides that kill adult worms. The development of such drugs has been hindered by the lack of an appropriate small laboratory animal model to evaluate potential drug candidates in vivo. This study assessed the survival of O. ochengi femal

[Non allergic skin reactions of drugs (author's transl)].

Non allergic skin reactions are differentiated in the following way: overdose, idiosyncrasy, intolerance and side effects. An intoxication caused by an overdose of a drug may be initiated by an increased resorption through the skin (e. g. salicylic acid or the obsolete boric acid). An overdose of a drug often leads to coma and in many cases, if the patients are lying unattended (e. g. at home). ischemic skin reactions, such as blisters or necrosis occur at pressure areas. Intolerance is an undesirable reaction, produced by a normal therapeutic dose of the drug. Reactions of special interest ar

Mechanistic Insights into pH-Responsive Drug Release and Antitumor Effects of an Albumin-Pirarubicin Conjugate.

PURPOSE: This study provides mechanistic insight into the behavior of HSA-THP, a human serum albumin-pirarubicin conjugate linked via a pH-sensitive hydrazone bond. Building upon our previous synthesis, we aimed to elucidate its intracellular uptake, and pH-responsive drug release behavior under acidic intracellular and extracellular conditions in vitro, and to evaluate its in vivo pharmacological properties. METHODS: Intracellular uptake of HSA-THP was evaluated in colon 26 monolayer cultures using fluorescence imaging and HPLC. Cytotoxic effects were compared in 2D and 3D spheroid cultures.

Smart Hydrogel Dressing Enhances the Healing of Chronic Infectious Diabetic Wounds through Dual-Barrier Drug Delivery Action.

Chronic diabetic wounds struggle to heal due to drug-resistant bacterial infections, oxidative stress microenvironment, and immune dysfunction. At present, the disease has become a huge clinical challenge. Multifunctional hydrogels with antibacterial, antioxidant, and anti-inflammatory properties are becoming an emerging trend in the treatment of chronic wounds. However, matching different bioactive functions with the wound healing process to sequentially exert antibacterial, antioxidant, anti-inflammatory, and immunomodulatory functions remains a significant challenge. In this research, a hyd

Defining treatment refractoriness in schizophrenia.

The criteria for treatment refractoriness for the use of clozapine are necessarily set at a high level because of the potential risk of agranulocytosis with this treatment. However, a more liberal definition should be considered to better appreciate the limitations of current therapies and prepare for switching to the next generation of clozapine-like drugs that do not cause agranulocytosis. It is suggested that failure of therapy to bring about the best premorbid level of functioning be considered as a starting point for treatment-resistance.

Making a medicine out of MDMA.

From its first use 3,4,-methylenedioxymethamphetamine (MDMA) has been recognised as a drug with therapeutic potential. Research on its clinical utility stopped when it entered the recreational drug scene but has slowly resurrected in the past decade. Currently there is enough evidence for MDMA to be removed from its Schedule 1 status of 'no medical use' and moved into Schedule 2 (alongside other misused but useful medicines such as heroin and amphetamine). Such a regulatory move would liberate its use as a medicine for patients experiencing severe mental illnesses such as treatment-resistant p

Cyclodextrin-based in situ gel systems: Their benefits and limitations for drug delivery.

The inherent limitations of conventional dosage forms, particularly for ocular and nasal administration, necessitate advanced drug delivery systems. In situ gelling systems and cyclodextrins (CDs) individually offer significant advantages, while their combination provides a synergistic platform. CD-based in situ gels combine the inclusion complexation ability of CDs with the sol-gel transition properties of stimuli-responsive polymers, enhancing the solubility, stability, and residence time of poorly soluble drugs across various mucosal routes. CDs improve drug availability and protect labile

Theoretical calculations of monolayer PtS2 as a drug delivery carrier for ursolic acid.

Ursolic acid (UA) exhibits anti-inflammatory and anti-tumor properties. Developing a nanodrug delivery system for UA can improve its bioavailability. Transition metal sulfides, owing to their distinctive physicochemical characteristics, have emerged as the predominant two-dimensional nanostructures utilized in the advancement of nanodrug delivery systems in recent years. This study employs first-principles calculations to assess the viability of monolayer PtS2 as a carrier for UA. The results indicate that monolayer PtS2 exhibits structural stability as a UA carrier, with an adsorption energy

Tuning bio-aerogel properties. Part 3: Exploring silica-pectin composite aerogels for drug delivery.

The release of the model drug theophylline from silica-pectin aerogels was investigated. The composite aerogels were prepared via impregnation of pectin alcogels with silica sol, followed by in situ silica gelation and drying with supercritical CO2. The structural and physico-chemical properties of the aerogels were tuned via the preparation conditions (type of silica sol, calcium crosslinking of pectin or not). Theophylline was loaded via impregnation and its release into simulated gastric fluid was studied during 1 h followed by release into simulated intestinal fluid. The swelling, mass los

Development of Antiplasmodial Peptide-Drug Conjugates Using a Human Protein-Derived Cell-Penetrating Peptide with Selectivity for Infected Cells.

Malaria continues to impose a global health burden. Drug-resistant parasites have emerged to each introduced small-molecule therapy, highlighting the need for novel treatment approaches for the future eradication of malaria. Herein, targeted drug delivery with peptide-drug conjugates (PDCs) was investigated as an alternative antimalarial therapy, inspired by the success of emerging antibody-drug conjugates utilized in cancer treatment. A synthetic peptide derived from an innate human defense molecule was conjugated to the antimalarial drug primaquine (PQ) to produce PDCs with low micromolar po
