Our earlier research has shown that mono-substituted N-phenyl-2, 2-dichloroacetamide exhibited much higher anti-cancer activity than the lead compound sodium dichloroacetate (DCA). In this paper, a variety of multi-substituted N-phenyl-2, 2-dichloroacetamides were synthesized and biologically evaluated. The results showed that 3, 5-disubstituted N-phenyl-2, 2-dichloroacetamide analogues had satisfactory potency. Among them, N-(3, 5-diiodophenyl)-2, 2-dichloroacetamide had an IC50 of 2.84 micromol x L(-1) against non-small cell lung cancer cell line A549 and could induce cancer cell apoptosis.

A series of 1,3-disubstituted 2-iminobenzimidazolines as well as a number of their tautomeric analogs were synthesized. The disodium salt of 4-diethyl aminophenyl-4',4''-bis(3-sulfobenzyl ethyl aminophenyl)acetonitrile was made and studied. It was found to release cyanide lineraly with exposure to ionizing radiation. When administered ip to mice, it was absorbed in significant amounts and retained its ability to cleave upon irradiation. Based upon this, we gathered evidence and proposed that it is feasible to design a non-toxic compound which when exposed to ionizing radiation would yield predictable reactive end products that would remain localized and augment the effects of irradiation upon a neoplasm.

DESIGN, SETTING AND PARTICIPANTS: Community-based prospective observational study of serologically confirmed HCV antibody-negative PWID enrolled in six Sydney neighbourhoods located in three distinct regions between 10 November 2008 and 31 October 2011.
