Molecular characterization and expression analysis of interleukin-1 beta in Japanese eel (Anguilla japonica).

Interleukin 1-beta (IL-1B) is a key proinflammatory cytokine involved in disease resistance. In recent years, il-1β genes from several teleosts have been cloned. The Japanese eel (Anguilla japonica) is an evolutionarily primitive fish widely farmed in East Asia. The il-1β gene has not yet been cloned from Japanese eel. In this study, the complete cDNA of il-1β was successfully sequenced from peripheral leukocytes through the rapid amplification of cDNA ends. The il-1β gene consists of five exons and four introns, and the full-length cDNA spans 1204 bp, comprising a 99-bp 5' untranslated region

Full-length transcriptome analysis of male and female gonads in Japanese Eel (Anguilla japonica).

BACKGROUND: The Japanese eel (Anguilla japonica) holds significant economic value in East Asia, but limitations in understanding its reproductive biology have hindered advancements in artificial breeding techniques. Previous research has primarily focused on conserved sex differentiation genes, offering limited insights into the broader molecular mechanisms driving gonadal development and sexual dimorphism. To address these limitations, this study aims to investigate key genes and pathways involved in gonadal development through a comprehensive transcriptomic analysis of male and female eel go

Progestogen is a potential trigger initiating sex inversion in rice field eel (Monopterus albus).

The rice field eel or swamp eel (Monopterus albus) is a hermaphroditic freshwater teleost fish that transitions from a functional female to a functional male. However, the biological mechanism of sex inversion in this species remains unclear. Here we clone the 5' proximal flanking region of the rice field eel dmrt1 gene and find that the progesterone response element (PRE) and androgen response element (ARE) uniquely exist in this region. More importantly, based on reporter gene analysis, in vitro tissue culture, and progestogen hormone level assays, we reveal that progestogen is the prerequis

A new species of the conger eel genus Macrocephenchelys from deep waters of the Arabian Sea.

A new species of the genus Macrocephenchelys is described herein based on a single specimen collected from the deep-sea trawl landing at Kalamukku fish landing centre, Kerala coast, Arabian Sea. The new species is distinguished by having a dorsal-fin origin behind the middle of pectoral fin, a larger head, shorter trunk, larger gill opening, dorsal surface of body with dark-brown colour and ventral surface of head and belly with numerous patches of melanophores before anus, vertebrae 14-30-151. The new species shares most of the characteristics with Macrocephenchelys brevirostris but differs f

Fatal Blowhole Obstruction by Eel in Common Bottlenose Dolphins (Tursiops truncatus) in Florida, USA.

Esophageal and pharyngeal obstruction are commonly reported in marine mammals, but asphyxiation from blowhole and nasal cavity obstruction has been reported only rarely: in two long-finned pilot whales (Globicephala melas), several harbor porpoises (Phocoena phocoena), and one common bottlenose dolphin (Tursiops truncatus). We describe two cases of blowhole obstruction and subsequent asphyxiation in bottlenose dolphins caused by eels. A whip eel (Bascanichthys scuticaris) was found obstructing the blowhole of a deceased dolphin from the Indian River Lagoon, Florida, US (2011) and a shrimp eel

How the European eel (Anguilla anguilla) loses its skeletal framework across lifetime.

European eels (Anguilla anguilla) undertake an impressive 5 000 km long migration from European fresh waters through the North Atlantic Ocean to the Sargasso Sea. Along with sexual maturation, the eel skeleton undergoes a remarkable morphological transformation during migration, where a hitherto completely obscure bone loss phenomenon occurs. To unravel mechanisms of the maturation-related decay of the skeleton, we performed a multiscale assessment of eels' bones at different life-cycle stages. Accordingly, the skeleton reflects extensive bone loss that is mediated via multinucleated bone-reso

Expression and ontogeny of growth hormone (Gh) in the protogynous hermaphroditic ricefield eel (Monopterus albus).

Growth hormone (GH) is a single-chain polypeptide hormone mainly secreted by somatotropes of the anterior pituitary gland and is an important regulator of somatic growth in vertebrates including teleosts. In this study, a polyclonal antiserum against ricefield eel Gh was generated and the expression of Gh at the mRNA and protein levels was analyzed. Both RT-PCR and western blot analysis showed that Gh was predominantly expressed in the pituitary glands of ricefield eels. The immunoreactive Gh signals were localized to the multicellular layers of the adenohypophysis adjacent to the neurohypophy

Mechanisms of guanylin action on water and ion absorption at different regions of seawater eel intestine.

Guanylin (GN) inhibited water absorption and short-circuit current (Isc) in seawater eel intestine. Similar inhibition was observed after bumetanide, and the effect of bumetanide was abolished by GN or vice versa, suggesting that both act on the same target, Na(+)-K(+)-2Cl(-) cotransporter (NKCC), which is a key player for the Na(+)-K(+)-Cl(-) transport system responsible for water absorption in marine teleost intestine. However, effect of GN was always greater than that of bumetanide: 10% greater in middle intestine (MI) and 40% in posterior intestine (PI) for Isc, and 25% greater in MI and 3

Structural and functional characterization of neuropeptide Y in a primitive teleost, the Japanese eel (Anguilla japonica).

In the present study, the first full-length cDNA encoding Neuropeptide Y (NPY) was cloned from the brain of Japanese eel (Anguilla japonica). The open reading frame of Japanese eel NPY gene is 294 bp in length, encoding a precursor protein of 97 amino acids, which contains a 36-amino-acid mature peptide. Sequence analysis showed that the Japanese eel NPY peptide is similar to that of other species. Real-time PCR revealed that NPY in Japanese eel is mainly expressed in the brain, especially in the hypothalamus and the optic tectum thalamus. The effect of a negative energy balance on NPY gene ex

First evidence for a direct inhibitory effect of kisspeptins on LH expression in the eel, Anguilla anguilla.

The kisspeptin system has emerged as one of the main puberty gatekeepers among vertebrates. The European eel (Anguilla anguilla) is a remarkable model due to its phylogenetical position at the basis of teleosts, and its unique life cycle with a blockade of puberty before reproductive migration. We cloned the full-length coding sequence of a kisspeptin receptor (Kissr) in the eel. Comparison of Kissr sequences assigned the eel Kissr to a basal position in a clade including most of the known teleost Kissr, in agreement with the eel phylogenetical position. Eel Kissr tissue distribution was analy

Acute ammonia toxicity and the protective effects of methionine sulfoximine on the swamp eel, Monopterus albus.

The objectives of this study were to examine how the swamp eel, Monopterus albus, defended against acute ammonia toxicity derived from the intraperitoneal injection with a sublethal dose (10 micromol g(-1) fish) of ammonium acetate (CH(3)COONH(4)) followed by 24 hr of emersion, and to elucidate the mechanisms of acute ammonia toxicity with respect to glutamine accumulation in the brain using L-methionine S-sulfoximine [MSO; a glutamine synthetase inhibitor]. When confronted with a sublethal dose of CH(3)COONH(4) followed by emersion, only a small fraction of the exogenous ammonia was excreted,

Hypertonicity stimulates Cl(-) transport in the intestine of fresh water acclimated eel, Anguilla anguilla.

Eel intestinal epithelium when bathed symmetrically with normal Ringer solution develops a net Cl(-) current (short circuit current, Isc) giving rise to a negative transepithelial potential (Vt) at the basolateral side of the epithelium, lower in fresh-water (FW)-acclimated animals with respect to sea-water (SW). The aim of the present work was to study the cell response to hypertonic stress of FW eel intestinal epithelium in relation to Cl(-) absorption. The hypertonicity of the external bathing solutions produced first a transient increase of Vt and Isc, then followed (after 10-15 min) by a

Immunological cross-reactivity between electric-eel acetylcholinesterase and rat-tail-tendon collagen.

Immunological cross-reactivity between acetylcholinesterase from the electric organ of the electric eel and rat tail tendon collagen was examined both on the cellular and humoral levels. 1. Guinea pigs immunized with rat tail tendon collagen displayed a strong delayed-type skin reaction when tested with the elongated acetylcholinesterase preparation (i.e. 14-S + 18-S molecular forms). However, when the glubular 11-S enzyme was tested, almost no cross-reactivity was obtained. Similarly, guinea pigs immunized with 14-S + 18-S preparation exhibited skin sensitization to rat tail tendon collagen.

Milt androgen profile and evaluation of sperm morpho-functional characteristics of wild-caught and farmed European eels (Anguilla anguilla).

Evaluating milt samples before their preservation and/or reproductive use is critical to correctly develop assisted reproductive strategies for teleost species. This is especially true in case of "critically endangered" species, like the European eel (Anguilla anguilla), whose life cycle is not closed yet, leading to the necessity of capturing younglings from natural populations for both research and productive purposes. Despite androgens being involved in spermatogenesis and playing pivotal roles in the pre-migratory morphological and physiological changes, their quantification in different b

Transcriptome analysis of host anti-Aeromonas hydrophila infection revealed the pathogenicity of A. hydrophila to American eels (Anguilla rostrata).

Aeromonas hydrophila is a commonly pathogenic bacterium in cultivated eels, but its pathogenicity to American eel (Anguilla rostrata) and the molecular mechanism of host anti-A. hydrophila infection remains uncertain. In this study, LD50 of A. hydrophila to American eels was determined and bacterial load in the liver and kidney of eels was assessed post 2.56 doses of LD50 of A. hydrophila infection. The results showed that the LD50 of A. hydrophila to American eels was determined to be 3.9 × 105 cfu/g body weight (7.8 × 106 cfu/fish), and the bacterial load peaked at 36 h post the infection (h

Bidirectional Jump Point Search Path-Planning Algorithm Based on Electricity-Guided Navigation Behavior of Electric Eels and Map Preprocessing.

The electric eel has an organ made up of hundreds of electrocytes, which is called the electric organ. This organ is used to sense and detect weak electric field signals. By sensing electric field signals, the electric eel can identify changes in their surroundings, detect potential prey or other electric eels, and use it for navigation and orientation. Path-finding algorithms are currently facing optimality challenges such as the shortest path, shortest time, and minimum memory overhead. In order to improve the search performance of a traditional A* algorithm, this paper proposes a bidirectio

Fatty acid profiles and lipid quality indices in canned European eels: Effects of processing steps, filling medium and storage.

This study evaluated the effects of the canning process, the filling medium (sunflower or olive oil), the spices included as ingredients and storage on the fatty acid profiles and related lipid quality indices in the European eel. The fatty acids did not undergo any changes during thawing-salting, because of the short salting periods. However, frying, sterilization and room storage greatly altered the fatty acid profiles. In general, the nutritional value and the lipid quality increased, owing to absorption of the oil used in the canning process. The fatty acid profiles of canned eels tended t

Role of Activin A in regulating ovarian development during vitellogenesis in Japanese eel (Anguilla japonica).

Activin A, a member of the TGF-β superfamily, plays pivotal roles in vertebrate reproductive regulation. This research investigated the biological functions and potential regulatory mechanisms of Activin A during vitellogenesis in Japanese eel. Analysis of the structural characteristics and evolutionary relationships confirmed that Japanese eel Activin A is highly conserved, providing a molecular basis for elucidating its role in ovarian development. Using a prokaryotic expression system, recombinant Japanese eel Activin A protein was successfully produced, and a series of in vitro functional

A preliminary whole-genome survey of longfin snake eel Pisodonophis cancrivorus (Richardson, 1848).

Longfin snake eel, Pisodonophis cancrivorus (Anguilliformes: Ophichthidae), is a typical snake eel widely distributed across brackish and marine waters of the Indo-Pacific oceans. As sensitivity to environmental change, it has become a potential bio-indicator for assessing habitat health in tropical coastal ecosystems, and plays a significant ecological role in nutrient cycling and benthic community dynamics. However, to our knowledge, no related genomic studies of P. cancrivorus have been reported until now. In this study, the preliminary genome information of P. cancrivorus was derived by us

An eclectic encounter: ticks feeding on an electric eel and the untapped potential of natural history collections.

Parasites are quite likely the most diverse guild of species on earth. Nevertheless, they remain under-documented despite their impact on the health and ecology of nearly every other species. Natural history collections offer a powerful, underutilized resource for uncovering the hidden diversity of parasites, particularly by preserving specimens that reveal unexpected host-parasite associations. Ticks, although known for their broad host range among terrestrial vertebrates, have never been documented parasitizing fish. Here, the author reports the first known instance of ticks associated with
