The chemistry of perfluoroisobutylene (PFIB) with nitrone and nitroso spin traps: an EPR/Spin trapping study.

While applying electron paramagnetic resonance (EPR)/Spin Trapping techniques, several reactive intermediate species were identified in the reaction of perfluoroisobutylene (PFIB) with nitrone and nitroso spin trap agents: the carbon dioxide radical anion (CO2.-), a carbonyl fluoride intermediate (COF), and vinyl carbanions of PFIB. The reaction of PFIB with N-t-butyl-alpha-phenylnitrone (PBN) forms a dipolar ion which undergoes electron transfer reactions generating stable nitrone spin adducts. Nitroso compounds reacted with carbanions derived from PFIB, which raises the possibility that elec

Optimal EPR detection of weak nitroxide spin adduct and ascorbyl free radical signals.

We have investigated the optimal nominal power settings for the electron paramagnetic resonance detection of typical nitroxides, nitroxide spin adducts, and the ascorbyl free radical. In room temperature aqueous solution, we find that, for all the nitroxides examined, saturation effects begin at approx. 25 mW nominal power with maximum signal intensity achieved at approx. 100 mW power when using a TM110 cavity. For the ascorbyl free radical, we find that saturation effects begin at approx. 16 mW nominal power and that maximum peak-to-peak signal amplitude is obtained at approx. 40 mW microwave

Design and Investigation of Carbazole-Dithienylethene Compounds for Switchable Organic Radical Systems.

Photoswitchable radicals incorporating dithienylethene (DTE) moieties offer unique opportunities for dynamic control in various chemical and material applications. By exploiting light-triggered isomerization processes, researchers can achieve precise modulation of radical reactivity, enabling spatiotemporal control of chemical transformations. Despite growing interest in this field, challenges remain in controlling spin-orbit coupling and spin dynamics, which influence the electronic structure and transitions induced by light absorption as well as achieving photostability for repeated photoiso

Kondo Resonance of a Carbon-Centered Radical in a Single-Molecule Junction.

The low spin-orbit coupling and weak hyperfine interactions make organic radicals promising components used in molecular spintronics. Triphenylmethyl (TPM) is the first stable carbon-centered organic radical discovered more than a century ago. Here, we use scanning tunneling spectroscopy to study quantum transport through single TPM radicals that are attached to a Au(111) electrode via atomic contacts of Ni atoms. The transport exhibits a Kondo resonance, evidencing that the unpaired electron of the radical forms a spin singlet with the itinerant electrons of the electrode. Density functional

Conformational dynamics and distribution of nitroxide spin labels.

Long-range distance measurements based on paramagnetic relaxation enhancement (PRE) in NMR, quantification of surface water dynamics near biomacromolecules by Overhauser dynamic nuclear polarization (DNP) and sensitivity enhancement by solid-state DNP all depend on introducing paramagnetic species into an otherwise diamagnetic NMR sample. The species can be introduced by site-directed spin labeling, which offers precise control for positioning the label in the sequence of a biopolymer. However, internal flexibility of the spin label gives rise to dynamic processes that potentially influence PR

EPR spectroscopy of chlorpromazine-induced free radical formation in normal human melanocytes.

The purpose of this study was to estimate the effect of chlorpromazine on free radical concentration in HEMn-DP melanocytes using electron paramagnetic resonance (EPR) spectroscopy. It was found that chlorpromazine at concentrations of 1 × 10(-7) and 1 × 10(-6) M contributed to the formation of free radicals (g values ~2) in a dose-dependent manner. The increase in free radical formation was accompanied by an increase in cytotoxicity, as shown by a tetrazolium assay. Homogeneous broadening of EPR lines, slow spin-lattice relaxation processes, and strong dipolar interactions characterized all t

Expanding the Diversity of Nitroxide-Based Paramagnetic Probes Conjugated to Non-Canonical Amino Acids for Sdsl-Epr Applications.

Understanding protein structure requires studying its dynamics, which is critical to elucidating its functional role. Biophysical techniques have revolutionized this field over time, providing remarkable insights into structure-function relationships. Among these, Site-Directed Spin Labelling (SDSL) combined with Electron Paramagnetic Resonance (EPR) is a powerful method delivering structural data at the residue level, irrespective of protein size or environment. Traditional nitroxide labels targeting cysteine residues face limitations when these residues are essential for protein structure or

Synthesis and spin trapping properties of polystyrene supported trifluoromethylated cyclic nitrones.

Polystyrene supported fluorinated cyclic nitrone spin-traps: Resin-2-HFDMPO (2-hydroxymethyl-2-methyl-5-(trifluoromethyl)-3,4-dihydro-2H-pyrrole-1-oxide) and Resin-2-PFDMPO (2-(3-hydroxypropyl)-2-methyl-5-(trifluoromethyl)-3,4-dihydro-2H-pyrrole 1-oxide) containing a trifluoromethyl pyrroline-N-oxide core were developed to detect free radicals under flow conditions. A continuous flow EPR technique was used to evaluate the spin trapping properties of these tethered nitrones. While both resins trapped radicals, polymer supported nitrone Resin-2-PFDMPO with a longer and more flexible linker showe

Cu²⁺ as an ESR Probe of Protein Structure and Function.

Electron spin resonance (ESR) spectroscopy in combination with site-directed spin labeling has been widely used to determine the structure and dynamics of proteins and other biomolecules. The most popular spin label is a nitroxide-based radical which can be attached to a protein via a site-specific reaction with either native cysteines or cysteines engineered into the system via site-directed mutagenesis. Paramagnetic transition metals, including Cu(2+), often serve as cofactors of metalloproteins, and have already been realized as ESR probes to report structural information in these proteins.

Three dimensional electron spin resonance (ESR) imaging of internal organs in living mice.

In vivo electron spin resonance imaging (ESR imaging) was applied to living mice after peroral administration of a nitroxide radical spin probe. A 3D ESR imaging procedure was applied in vivo in order to obtain the exact distribution of the spin probe in a living animal. The imaging pictures demonstrated that the administered spin probe was firstly located in the stomach, then delivered to the liver, kidney and heart of the animal.

Synthesis of alpha-aryl N-adamant-1-yl nitrones and using them for spin trapping of hydroxyl radicals.

alpha-Aryl N-adamant-1-yl nitrones were synthesized and evaluated with respect to the stability of the hydroxyl radical adduct. The polarity and water solubility of nitrones were altered with changing the alpha-aryl groups. Introduction of adamantane ring instead of tert-butyl group resulted in a reasonable good stability of hydroxyl radical adduct for biological measurements.

New amino-nitroxide spin labels.

Stable free mono- and diradicals containing reactive primary or secondary amino groups in the side-chain have been synthesized by transesterification of amino-substituted esters with paramagnetic alcohols or from spin-labeled acid derivatives and amines. In the second approach the new radical 18 (1-oxyl-3-(2-bromoethoxycarbonyl)-2,2,5,5-tetramethylpyrroline) is proposed as an efficient alkylating species. The nitroxides described are pH-sensitive spin probes and spin labels potentially useful for a diversity of ESR applications in chemistry and biology. New spin-labeled tyramine 16 (N-(1-oxyl-

Symmetry Breaking and Spin-Orbit Coupling for Individual Vacancy-Induced In-Gap States in MoS2 Monolayers.

Spins confined to point defects in atomically thin semiconductors constitute well-defined atomic-scale quantum systems that are being explored as single-photon emitters and spin qubits. Here, we investigate the in-gap electronic structure of individual sulfur vacancies in molybdenum disulfide (MoS2) monolayers using resonant tunneling scanning probe spectroscopy in the Coulomb blockade regime. Spectroscopic mapping of defect wave functions reveals an interplay of local symmetry breaking by a charge-state-dependent Jahn-Teller lattice distortion that, when combined with strong (≃100 meV) spin-o

Unveiling the neglected role of alcohols and phenolic compounds in photocatalysis H2O2 generation chemistry.

Alcohols are promising sacrificial agents for improving the photocatalytic production of hydrogen peroxide (H2O2) through reaction with holes, inhibiting the electron-hole recombination. However, the side effects of alcohols on oxygen reduction and reactive oxygen species (ROS) generation remain controversial due to intrinsic formation of organic peroxides under irradiation in the presence of air or oxygen. To address this, we employ electron paramagnetic resonance (EPR), in-situ Fourier-transform infrared spectroscopy (FTIR) and 1H nuclear magnetic resonance (NMR) to directly monitor intermed

Observing extradiol dioxygenases in action through a crystalline lens.

Extradiol dioxygenases are a class of non-heme iron-dependent enzymes found in eukaryotes and prokaryotes that play a vital role in the aerobic catabolism of aromatic compounds. They are generally divided into three evolutionarily independent superfamilies with different protein folds. Our recent studies have shed light on the catalytic mechanisms and structure-function relationships of two specific extradiol dioxygenases: 3-hydroxyanthranilate-3,4-dioxygenase, a Type III enzyme essential in mammals for producing a precursor for nicotinamide adenine dinucleotide, and L-3,4-dihydroxyphenylalani

Influence of the ligand-field on EPR parameters of cis- and trans-isomers in MoV systems relevant to molybdenum enzymes: Experimental and density functional theory study.

The electron paramagnetic resonance (EPR) investigation of mononuclear cis- and trans-(L1O)MoOCl2 complexes [L1OH = bis(3,5-dimethylpyrazolyl)-3-tert-butyl-2-hydroxy-5-methylphenyl)methane] reveals a significant difference in their spin Hamiltonian parameters which reflect different equatorial and axial ligand fields created by the heteroscorpionate donor atoms. Excited state dynamics of a spontaneously generated TTF radical cation inside a water-soluble nanocage.

Supramolecular cavities have been traditionally used to stabilize reactive redox intermediates. Recently with the success of multiple new photoredox catalytic strategies that use supramolecular cages, there is a growing demand for photogeneration strategies of diverse reactive intermediates inside confined spaces, which will drive enzyme-like catalysis in real time. Here we report the excited state dynamics of a redox-active TTF radical cation and its corresponding dimethyl-derivative DiMeTTF inside a confined supramolecular cavity. We prepare the radical cation by spontaneous oxidation of neu

Measurement of Mitochondrial (Dys)Function in Cellular Systems Using Electron Paramagnetic Resonance (EPR): Oxygen Consumption Rate and Superoxide Production.

The oxygen consumption rate (OCR) and superoxide production are crucial when assessing mitochondrial function and/or dysfunction. EPR spectroscopy allows the measurement of both components either independently or simultaneously in a same cellular or mitochondrial preparation. OCR determination using EPR oximetry is based on the change in EPR linewidth of a paramagnetic oxygen sensing probe (a perdeuterated nitroxide) in the presence of oxygen consuming cells in a closed system. Superoxide production can be monitored by the oxidation of cyclic hydroxylamines into nitroxides. The contribution of

Low-frequency EPR of ferrimyoglobin fluoride and ferrimyoglobin cyanide: a case study on the applicability of broadband analysis to high-spin hemoproteins and to HALS hemoproteins.

An EPR spectrometer has been developed that can be tuned to many frequencies in the range of ca 0.1-15 GHz. Applicability has been tested on ferrimyoglobin fluoride (MbF) and ferrimyoglobin cyanide (MbCN). MbF has a high-spin (S = 5/2) spectrum with 19F superhyperfine splitting that is only resolved in X-band along the heme normal. Low-frequency EPR also resolves the splitting in the heme plane. Measurement of linewidth as a function of frequency provides the basis for an analysis of inhomogeneous broadening in terms of g-strain, zero-field distribution, unresolved superhyperfine splittings an

ESR spectroscopy as a tool to investigate the properties of self-assembled monolayers protecting gold nanoparticles.

Electron spin resonance (ESR) has emerged as a powerful spectroscopic technique to study the properties of metal nanoparticles (NPs) protected by a self-assembled monolayer (SAM) of organic molecules. This technique has been employed to explore the capacity of homoligand monolayers to bind to a hydrophobic probe or to "sense" the hydrophobicity of mixed-ligand monolayers. Moreover, spin labels anchored to the metal surface enable the investigation of the dynamic of the ligands that form the monolayer. Here we review these applications with the aim of unravelling the many features of monolayer-
