A cell culture technique for quantitative analysis of radiation-induced chromosome aberrations in somatic cells has been developed and used for the comparison of chromosomal sensitivity of skin cells of mouse and man to 60Co-gamma-rays. This includes culture of irradiated tissues or cells in culture in arginine and isoleucine-deficient medium and subsequent refeeding with complete medium (CM). With this technique, radiation-induced chromosome aberrations can be analyzed selectively in the cells exposed in G1 phase and recovered at their first post-irradiation mitosis. When tested on the human

Somatic cells can be reprogrammed into iPS (induced pluripotent stem) cells through the ectopic expression of defined transcription factors. However, the inefficiency and amount of time needed limited the potential application of iPS cells. We report an efficient method to generate iPS cells from MEF (mouse embryonic fibroblasts) through folate-depriviatoin, which was used to change the methylation of MEF. Without folate for 3 days, the induction efficiency is enhanced fivefold. Karyotype analysis showed that transient folate-depriving treatment did not negatively affect properties of iPS cell

Transplantation is the treatment of choice for many diseases that result in organ failure, but its success is limited by organ rejection. Stem cell therapy has emerged in the last years as a promising strategy for the induction of tolerance after organ transplantation. Here we discuss the ability of different stem cell types, in particular mesenchymal stromal cells, neuronal stem/progenitor cells, hematopoietic stem cells and embryonic stem cells, to modulate the immune response and induce peripheral or central tolerance. These stem cells have been studied to explore tolerance induction to sev

The tumor marker carcinoembryonic antigen (CEA) is predominantly expressed in epithelial cells along the gastrointestinal tract and in a variety of adenocarcinomas. As a basis for investigating its in vivo regulation and for establishing an animal model for tumor immunotherapy, transgenic mice were generated with a 33-kilobase cosmid clone insert containing the complete human CEA gene and flanking sequences. CEA was found in the tongue, esophagus, stomach, small intestine, cecum, colon, and trachea and at low levels in the lung, testis, and uterus of adult mice of independent transgenic strain

Y-chromosome AZFc deficiency, which includes the DAZ gene family, is one of the most common genetic causes of severe male infertility. Epidemiological and molecular evidence suggest a developmental link between abnormalities in germ cell development and testicular germ cell tumor (TGCT), but whether and how DAZ deficiency is associated with TGCT susceptibility remains unclear. Using a specific human embryonic stem cell (hESC) model, we constructed and characterized a DAZ-deleted cell line (SKLRMe001-A) and compared its efficiency of induction into human primordial germ cell-like cells (hPGCLCs

It has been demonstrated that naturally derived osteogenic proteins extracted and purified from allogeneic bone matrices, reconstituted and implanted in the rectus abdominis muscle of recipient non-human primates of the species Papio ursinus, induce bone as a recapitulation of embryonic development. There is the induction of cartilage anlagen, angiogenesis, and vascular invasion, chondrolysis with the induction of osteoblast differentiation and bone matrix deposition. Recombinant human osteogenic protein-1 (hOP-1) also induces large ossicles in the rectus abdominis muscle of Papio ursinus. Our

Background: Hyaluronan (HA) is a critical extracellular matrix component that we have demonstrated to be important for embryonic endochondral bone formation and postnatal synovial joint formation, supporting normal articular cartilage (AC) architecture and chondrocyte function. Although the embryonic requirement for Hyaluronan Synthase 2 (Has2), the main HA-producing enzyme in skeletal tissues, has been extensively investigated, the cartilage-cell-specific roles of Has2 and HA in maintaining postnatal cartilage and joint integrity are not well-defined. Methods: In this study, we used a tamoxif

The fibroblast growth factor receptors (FGFRs) are a family of receptor protein tyrosine kinases that have been shown to mediate a variety of cellular processes including angiogenesis, wound healing, tumorigenesis, and embryonic development. Distinct FGFR mutations in individuals with autosomal dominant disorders of bone growth and development provide a unique opportunity to determine the function of FGFRs during embryonic development. To determine the consequences of these mutations on receptor function, we have made mutations in Xenopus FGFR1 (XFGFR1) and FGFR2 (XFGFR2) that correspond to se

In order to determine the time window for induction of lateral line placodes in the axolotl, we performed two series of heterotopic and isochronic transplantations from pigmented to albino embryos at different stages of embryogenesis and assessed the distribution of pigmented neuromasts in the hosts at later stages. First, ectoderm from the prospective placodal region was transplanted to the belly between early neurula and mid tailbud stages (stages 13-27). Whereas grafts from early neurulae typically differentiated only into epidermis, grafts from late neural fold stages on reliably resulted

In eggs of Xenopus laevis, dorsal development is activated on the future dorsal side by cortical rotation, after fertilization. The immediate effect of cortical rotation is probably the transport of a dorsal determinant from the vegetal pole to the equatorial region on the future dorsal side. However, the identity and action of the dorsal determinant remain problematic. In the present experiments, individual isolated cortices from various regions of the unfertilized eggs and embryos were implanted into one of several positions of a recipient 8-cell embryo. The incidence of secondary axes was u

Embryonic stem cells possess the remarkable ability to self-organize into blastocyst-like structures upon induction. These stem cell-based embryo models serve as invaluable platforms for studying embryogenesis and therapeutic developments. Nevertheless, the specific intrinsic regulators that govern this potential for blastoid formation remain unknown. Here we demonstrate an intrinsic program that plays a crucial role in both blastoids and blastocysts across multiple species. We first establish metrics for grading the resemblance of blastoids to mouse blastocysts, and identify the differential

Fate decisions in the embryo are controlled by a plethora of microenvironmental interactions in a three-dimensional niche. To investigate whether aspects of this microenvironmental complexity can be engineered to direct myogenic human-induced pluripotent stem cell (hiPSC) differentiation, we here screened murine cell types present in the developmental or adult stem cell niche in heterotypic suspension embryoids. We identified embryonic endothelial cells and fibroblasts as highly permissive for myogenic specification of hiPSCs. After two weeks of sequential Wnt and FGF pathway induction, these

The aim of this study was to assess placental changes and reproductive outcomes in neonatally induced mild diabetic dams and fetal development in their offspring. At birth, female rats were assigned either to control or diabetic group (100 mg of streptozotocin/Kg, subcutaneously). At adulthood, the female rats were mated. During pregnancy, the blood glucose levels and glucose and insulin tolerance tests were performed. At term, maternal reproductive outcomes, fetal and placental weight, and placental morphology were analyzed. Diabetic rats had smaller number of living fetuses, implantations an

In ruminants, more than 30% of the embryonic losses observed after artificial insemination (AI) have an early origin, coincident with a marked elongation of the trophoblast which occurs before implantation. Several observations provide clear evidence that early elongation of the conceptus relies on cell multiplication, cell growth and cell shape remodeling. Recent results indicating an intense multiplication of a non-fully differentiated trophoblast, which still expresses some epiblast genes, has to be considered at the onset of elongation. It has also been shown in the last two years that gen

Neurogenin 3 (Ngn3) is key for endocrine cell specification in the embryonic pancreas and induction of a neuroendocrine cell differentiation program by misexpression in adult pancreatic duct cells. We identify the gene encoding IA1, a zinc-finger transcription factor, as a direct target of Ngn3 and show that it forms a novel branch in the Ngn3-dependent endocrinogenic transcription factor network. During embryonic development of the pancreas, IA1 and Ngn3 exhibit nearly identical spatio-temporal expression patterns. However, embryos lacking Ngn3 fail to express IA1 in the pancreas. Upon ectopi

Expression of AP-2 transcription factors has been detected previously in embryonic renal tissues. We show here that AP-2beta -/- mice complete embryonic development and die at postnatal days 1 and 2 because of polycystic kidney disease. Analyses of kidney development revealed that induction of epithelial conversion, mesenchyme condensation, and further glomerular and tubular differentiation occur normally in AP-2beta-deficient mice. At the end of embryonic development expression of bcl-X(L), bcl-w, and bcl-2 is down-regulated in parallel to massive apoptotic death of collecting duct and distal

The RNA-binding molecule Bicaudal-C regulates embryonic development in Drosophila and Xenopus. Interestingly, mouse mutants of Bicaudal-C do not show early patterning defects, but instead develop polycystic kidney disease (PKD). To further investigate the molecular mechanism of Bicaudal-C in kidney development, we analyzed its function in the developing amphibian pronephros. Bicaudal-C mRNA was present in the epithelial structures of the Xenopus pronephros, the tubules and the duct, but not the glomus. Inhibition of the translation of endogenous Bicaudal-C with antisense morpholino oligomers (

The Australian plague locust Chortoicetes terminifera (Walker) exhibits facultative embryonic diapause during autumn. To approximate natural photoperiod changes during late summer and autumn, locust nymphs were reared under different total declines in laboratory photophase (-0.5, -0.75, -1.0, -1.25, -1.5, -1.75, -2 h each lowered in 15 min steps) in a 24 h photoperiod to quantify any effect on the subsequent production of diapause eggs. Induction of diapause eggs was significantly affected by accumulated photoperiod decline experienced by the parental generation throughout all development stag

The domesticated silkworm Bombyx mori is both a major sericultural species and a powerful model for studying how environmental cues are converted into developmental decisions. A central seasonal trait in this species is embryonic diapause, a maternally programmed developmental arrest whose incidence is shaped by genetic background and environmental conditions experienced during the maternal generation. Recent genome-editing studies have provided new functional evidence for the role of circadian clock genes in this process, particularly the two silkworm cryptochromes, BmCRY1 and BmCRY2. BmCRY1
