The two sites of fusion of the neural folds and the two neuropores in the human embryo.

BACKGROUND: Since reports on a pattern of multiple sites of fusion of the neural folds in the mouse appeared, it has been widely assumed that a similar pattern must be valid for the human. In the absence of embryological evidence, claims have been made that such a pattern can be discerned by classifying neural tube defects. METHODS: The neural folds and tube, as well as the neuropores, were reassessed in 98 human embryos of Stages 8-13; 61 were controlled by precise graphic reconstructions. RESULTS: Careful study of an extensive series of staged human embryos shows that two de novo sites of

Anatomical relationships between testis and epididymis during the fetal period in humans (10-36 weeks postconception).

OBJECTIVE: To determine the anatomy of the epididymis and its relationship with the testis during the fetal period in normal individuals. METHODS: We studied bilaterally 146 testes and epididymides taken from 73 normal fresh human fetuses ranging in age from 10 to 36 weeks postconception. The epididymal anatomy was classified in six types: type I: the epididymis is connected to the testis by its head and tail and the epididymal body is separated from the testis; type II: the epididymis is completely attached to the testis; type III: the epididymis is attached to the testis only by its head; t

Environmental and genetic determinants of neural migration and postmigratory survival.

The study of genetic/epigenetic/environmental factors underlies all therapeutic and preventive approaches in fetal, perinatal and paediatric neurology, including rehabilitation. In this paper, we selected a few targets of environmental determinants of brain development leading to underlying priorities for protection of the developing brain. Preparation of the neural germinative epithelium has to be protected against noxious pharmacological agents. New tools have been developed to improve early neural teratology, including the whole-embryo culture method. The neopallial astrocytic precursors ha

The subependymal zone: "brain marrow".

From all of the studies of developmentally regulated molecules as well as the impressive proliferation of cells within the SEZ, we would like to propose that the SEZ of the adult brain, from the lateral ventricles of the cerebrum to the central canal of the spinal cord, represents a potential "brain marrow" from which stem and progenitor cells can be further studied and exploited for cell replacement and circuitry repair paradigms for neurological disease. Without question the SEZ is not as regenerative or pleuripotential as bone marrow or other hematopoietic systems, but there are definitely

Development. The science of damage limitation.

The building of a normal embryo is not accomplished by following a series of complex instructions; rather a number of critical processes enable the cells involved in the process of development to follow pathways which subsequently constrain their growth, mobility and differentiation. There is considerable redundancy in the system, both in terms of mechanisms and permissible outcomes. The way that these processes have become established in evolution gives us a number of insights into how these processes may fail and information from developmental biology informs us about a number of other patho

Septation of the respiratory and digestive tracts in human embryos: crucial role of the tracheoesophageal sulcus.

Esophageal atresia and tracheoesophageal fistula, common malformations of the respiratory and digestive tracts, are of unsettled pathogenesis. Part of the difficulty in understanding these abnormalities arises from the uncertainties about the normal developmental processes in the region. This study examined the development and fate of the tracheoesophageal septum. Normal human embryos from the Carnegie Embryological Collection and fetuses from the Hopkins Pathology Collection were examined, and reconstructions of selected specimens were made from photomicrographs of serial histologic sections.

Current topic: confined placental mosaicism and intrauterine fetal development.

Chromosomal mosaicism is the presence of two or more cell lines with different chromosomal complements in an individual derived from a single zygote (which is distinguishable from chimaerism in which an individual is derived from more than one zygote). Mosaicism may involve a number of different karyotypic abnormalities including those for whole sets of chromosomes (polyploidy, including triploidy and tetraploidy), trisomies, sex chromosomal anomalies and chromosomal rearrangements. Mosaicism itself may be present in all the tissues of the body and placenta, or it may only be seen in certain t

Type IV collagen in developing human lung: a comparison between normal and hypoplastic fetal lungs.

Human lung tissue obtained from 24 aborted fetuses of varying gestational ages (beyond 24 weeks) was examined for immunolocalisation of type IV collagen, using monoclonal antibody. Twelve cases of lung hypoplasia associated with oligohydramnios were compared with 12 normally developed lungs over the same gestational age range acting as control. Type IV collagen was located in alveolar septa and subepithelial basement membranes in all the lungs examined but showed less prominently in hypoplastic lungs than in control lungs of a similar gestational age. The findings suggest a potential role of t

Evaluation of fetal growth by ultrasonography in twin pregnancy: a comparison between individual and cross-sectional growth curve standards.

Cross sectional curves and individual fetal growth curves standards from the Rossavik growth model [P = c(t)(k + s(t]] were generated for abdominal and head circumferences, femur diaphysis length and estimated fetal weight from a sample of 59 women with twin pregnancy. These curves were compared to their counterparts in singleton pregnancies. Cross sectional curves of the four fetal anatomic parameters under study fell progressively below the curves for singletons during the last trimester of gestation. In contrast, there were few differences between individual fetal groWth curve standards for

Does heat damage fetuses?

Temperature affects phenotypic variation during critical developmental stages in all forms of life that have been studied thus far. In animal studies of heat teratogenicity, adverse effects have ranged from disruption of the normal cell cycle leading to decreased numbers of cells, to the induction of developmental abnormalities by means of embryonic cell death. The heat shock response is a universal cellular stress reaction in which the transcriptional and translational mechanisms of the cell are pre-empted by preferential induction of heat shock protein synthesis. Occurrence of such a phenome

Fetal foot length--a new parameter for assessment of gestational age.

Ultrasonographic measurement of fetal foot length, a new parameter, was correlated with the gestation age. One hundred and five ultrasonographic measurement of fetal foot length was performed between 13 and 42 weeks gestation. Comparison of linear regression of foot length versus gestational age demonstrated a strong correlation with an r2 value of 0.84 (P less than 0.001). Ninety-five percent confidence intervals at each week compared favorably with both biparietal diameter and femur length data. Mean foot lengths at each week of gestation compared favorably with data based on pathological sp

Deregulation of the hedgehog signalling pathway: a possible role for the PTCH and SUFU genes in human rhabdomyoma and rhabdomyosarcoma development.

The naevoid basal cell carcinoma syndrome (NBCCS) is caused by mutations in the hedgehog receptor PTCH gene. It is characterized by developmental defects and a predisposition to the development of certain tumours, such as basal cell carcinoma, medulloblastoma and meningioma, and potentially fetal rhabdomyomas and embryonal rhabdomyosarcomas. This study aimed to analyse PTCH status in an NBCCS patient with fetal rhabdomyoma and to investigate whether deregulation of hedgehog signalling, as shown by altered expression of hedgehog pathway components and/or genetic imbalances, is a general finding

Pathways of vitamin A delivery to the embryo: insights from a new tunable model of embryonic vitamin A deficiency.

Circulating retinoids (vitamin A and its derivatives) are found predominantly as retinol bound to retinol-binding protein (RBP), which transports retinol from liver stores to target tissues, or as retinyl ester incorporated in lipoproteins of dietary origin. The transport of retinoids from maternal to fetal circulation is poorly understood, especially under conditions of inadequate dietary vitamin A intake. Here we present RBP-/- mice as a tunable model of embryonic vitamin A deficiency. This model has enabled us to analyze metabolic links between maternal nutrition and retinoid delivery to th

Embryonic and fetal morphology of the black agouti.

Understanding embryonic and fetal development is fundamental to interpreting life-history strategies associated with neonatal survival in mammals. This study provides a detailed characterization of the intrauterine development of the hystricognath rodent, the black agouti (Dasyprocta fuliginosa), one of the most hunted species in the Amazon. The developmental patterns are compared with those of other precocial and altricial mammals, offering insights into species-specific reproductive strategies. A total of 90 conceptuses (2 embryos and 88 fetuses) from 49 pregnant females collected from subsi

Long-bone growth in fetuses with Down syndrome.

Short stature is a well-recognized component of Down syndrome. The femur lengths of affected fetuses have been observed to be shorter than normal, with a ratio of actual to expected femur length of less than 0.91 indicating a high risk of trisomy. To further evaluate this finding we have determined the relationship between limb lengths and gestational age in 37 postmortem fetal specimens with trisomy 21. Control values were obtained from 174 normal fetuses. Measurements of the femur, tibia, fibula, humerus, radius, and ulna were made from roentgenograms or by direct measurement of the bone. Mo

Taurine and taurine-deficiency in the perinatal period.

Taurine, a non-protein sulfur amino-acid, is the most abundant free amino-acid in the body and plays an important role in several essential biological processes. Apart from its role in cholesterol degradation, it acts as neurotransmitter, and has a function as osmoregulator and antioxidant in most body tissues. During pregnancy, taurine accumulates in the maternal tissues, to be released in the perinatal period to the fetus via the placenta and to the newborn via the maternal milk. It is accumulated especially in the fetal and neonatal brain. Low maternal taurine levels result in low fetal tau

The course of the buccal nerve: relationships with the temporalis muscle during the prenatal period.

The aim of this study was to describe the course of the buccal nerve and its relationships with the temporalis muscle during the prenatal period. Serial sections of 90 human fetal specimens ranging from 9 to 17 wk development were studied by light microscopy. Each fetal specimen was studied on both right and left sides, making a total of 180 cases for study. A 3-D reconstruction of the region analysed in one of the specimens was made. In 89 cases the buccal nerve was located medial to the temporalis muscle; in 73 cases it penetrated the muscle; in 15 cases it lay in a canal formed by the muscl

[Phenytoin influence on human metanephros].

The influence of Phenytoin on the human embryo is knowing to produce the fetal hydantoin syndrome. Even this syndrome is knowing to produce microcephaly, mental retardation, eyelid ptosis etc it is not mentioned the influence of Phenytoin on the human metanephros. This is the reason of our study, made on human embryos. Their mothers received during pregnancy Phenytoin. In one studied embryo we have remarked a malformation in the metanephros. Even we can not make a strong correlation between the malformation and the Phenytoin we consider that pregnant women should not use Phenytoin during pregn

[New aspects on preventive iron supplementation in normal pregnancy].

The paper gives an overview of iron metabolism and supplementation in pregnancy. Pregnancy leads to an increased demand for iron which is not normally met by the iron content in food in Norway and other western countries. The authors discuss the possible advantages of supplementation, as well as adverse reactions. Iron supplementation may have beneficial effects on cerebral function in neonates. High doses should not be given because of possible adverse effects. Low doses of iron (50-100 mg a day) should be offered from the beginning of the 20th week to pregnant women with an S-ferritin concen
