Nuclear receptors and endocrine disruptors in fetal and neonatal testes: a gapped landscape.

During the last decades, many studies reported that male reproductive disorders are increasing among humans. It is currently acknowledged that these abnormalities can result from fetal exposure to environmental chemicals that are progressively becoming more concentrated and widespread in our environment. Among the chemicals present in the environment (air, water, food, and many consumer products), several can act as endocrine disrupting compounds (EDCs), thus interfering with the endocrine system. Phthalates, bisphenol A (BPA), and diethylstilbestrol (DES) have been largely incriminated, parti

The Relationship between Uterine Myoma Growth and the Endocrine Disruptor in Postmenopausal Women.

OBJECTIVES: To investigate the number of leiomyoma patients-exposed to bisphenol A (BPA) and to observe whether the serum concentration of BPA is related to leiomyoma growth. METHODS: A total of 158 patients were recruited for this study. Leiomyoma patients were divided into three groups, mild (n = 48), moderate (n = 32) and severe (n = 28), according to the size of leiomyomas. The control (n = 30) group was defined as having no leiomyomas. Transvaginal ultrasonography was used to identify and measure the leiomyomas. Serum BPA concentrations were measured by enzyme-linked immunosorbent assay.

Adverse effects of endocrine disruptors on the foetal testis development: focus on the phthalates.

There are great concerns about the increasing incidence of abnormalities in male reproductive function. Human sperm counts have markedly dropped and the rate of testicular cancer has clearly augmented over the past four decades. Moreover, the prevalence rates of cryptorchidism and hypospadias are also probably increasing. It has been hypothesized that all these adverse trends in male reproduction result from abnormalities in the development of the testis during foetal and neonatal life. Furthermore, many recent epidemiological, clinical and experimental data suggest that these male reproductiv

Reproductive effects of the endocrine disruptor fenarimol on a Baltic amphipod Monoporeia affinis.

An endocrine disruptor, the fungicide fenarimol, was investigated regarding its effects on reproduction and hormone (ecdysteroid) levels in the deposit-feeding amphipod Monoporeia affinis. In addition, the influence of food shortage, both by itself and in combination with fenarimol, on reproduction was examined. Field-collected amphipods were exposed in flow-through microcosms during the period of sexual maturation and mating in four treatment series: Control with low food, fenarimol with low food, control with high food, and fenarimol with high food. Fenarimol was added at a concentration of

Bioaccumulation and Metabolic Effects of the Endocrine Disruptor Methoprene in the Lobster, Homarus americanus.

Methoprene is a pesticide that acts as a juvenile hormone agonist. Although developed initially against insects, it has since been shown to have toxic effects on larval and adult crustaceans. Methoprene was one of several pesticides applied to the Western Long Island Sound (WLIS) watershed area during the summer of 1999; the other pesticides were malathion, resmethrin, and sumethrin. These pesticides were applied as part of a county-by-county effort to control the mosquito vector of West Nile Virus. Subsequently, the seasonal lobster catches from the WLIS have decreased dramatically. The letha

Changes in male reproductive health and effects of endocrine disruptors in Scandinavian countries.

Male reproductive health has deteriorated in many ways during the last decades. The incidence of testicular cancer has rapidly increased in Europe and European-derived populations. Sperm concentrations have declined and sperm motility and morphology have worsened in many areas. Both adverse trends have been shown to be associated with year of birth. Older birth cohorts have better reproductive health than the younger generations. Incidences of cryptorchidism and hypospadias have also increased according to several studies. The reasons for secular trends are unknown, but the rapid pace of the c

Endocrine disruptors and development of the reproductive system in the fetus and children: is there cause for concern?

Reports of decreased semen quality and increased rates of developmental abnormalities of the male reproductive tract along with increasing incidence of testicular cancer have focused attention on man-made chemicals as potential causative factors. A biologically plausible hypothesis has been advanced which suggests that man-made chemicals act as endocrine disruptors through interaction with the estrogen receptor resulting in altered development of the reproductive tract. Available evidence suggests that this mechanism may play only a minor role in the purported adverse effects described to date

Disruption of hormonal balance and the potential development of cancer - the impact of endocrine disruptors.

BACKGROUND: Endocrine-disrupting chemicals (EDCs) constitute a heterogeneous group of substances capable of interfering with hormonal regulation and thereby influencing the risk of initiation and progression of hormone-sensitive cancers. Their mechanisms of action include interactions with hormone receptors, modulation of hormone synthesis and metabolism, epigenetic alterations, and activation of specific signalling pathways. In recent years, the adverse outcome pathway (AOP) framework has gained prominence, as it enables linking molecular initiating events affected by EDCs to clinical outcome

Global research on endocrine disruptors as emerging hazards for human health and the environment.

Endocrine disruptors (EDs) contaminate nearly every ecosystem and are significantly associated with different neurological and neurodevelopmental disorders. To date, there is no comprehensive literature on global publication efforts. Since there are many unknown substances, modes of action, and risks of EDs, it is necessary to provide detailed insight into global publication patterns from temporal, regional, and socioeconomic perspectives. Hence, this review article provides background information for all stakeholders, from scientists to clinicians and policymakers. A disproportionate increase

The Impact of Endocrine Disruptors on the Female Genital Tract Microbiome: A Narrative Review.

BACKGROUND/OBJECTIVES: Endocrine disruptors (EDs) are xenobiotic chemicals that disrupt hormone signaling and homeostasis within the human body. Accumulative evidence proposes that EDs could affect systemic hormone balance and local microbial communities, including the female genital tract (FGT) microbiome. The FGT microbiome, and especially the vaginal microbiota, contributes significantly to reproductive health maintenance, defense against infection, and favorable pregnancy outcomes. Disruption of the delicate microbial environment is associated with conditions like bacterial vaginosis, infe

BPA Endocrine Disruptor Detection at the Cutting Edge: FPIA and ELISA Immunoassays.

BPA is a chemical commonly used in the production of polymer-based materials that can have detrimental effects on the thyroid gland and impact human reproductive health. Various expensive methods, such as liquid and gas chromatography, have been suggested for detecting BPA. The fluorescence polarization immunoassay (FPIA) is an inexpensive and efficient homogeneous mix-and-read method that allows for high-throughput screening. FPIA offers high specificity and sensitivity and can be carried out in a single phase within a timeframe of 20-30 min. In this study, new tracer molecules were designed

The impact of endocrine disruptor chemicals on oocyte/embryo and clinical outcomes in IVF.

The negative impact of endocrine-disrupting pesticides on human fertility is now a key issue in reproductive health. There are much fewer literature data about the impact of pesticide exposure on women than on men and very few studies of women participating in an in vitro fertilization (IVF) programme. In the present review, we found that (1) various pesticides with an endocrine-disrupting action are associated with poor oocyte maturation and competency, embryonic defects and poor IVF outcomes, and (2) some pesticide compounds are linked to specific causes of female infertility, such as premat

Effects of elevated glucocorticoids on reproduction and development: relevance to endocrine disruptor screening.

This article reviews the influence of the hypothalamo-pituitary-adrenocortical (HPA) axis on mammalian male and female reproduction and development of offspring and its potential impact on the identification of endocrine disruptive chemicals by in vivo assays. In the adult male rat and baboon, stress suppresses testosterone secretion via a direct inhibitory effect of elevated glucocorticoids on Leydig cells. In adult female sheep, stress disrupts reproductive function via multi-stage mechanisms involving glucocorticoid-mediated suppression of LH secretion, LH action on the ovary and the action

Effects of endocrine disruptors on genes associated with 17beta-estradiol metabolism and excretion.

In order to provide a global analysis of the effects of endocrine disruptors on the hormone cellular bioavailability, we combined 17beta-estradiol (E2) cellular flow studies with real-time PCR and Western blot expression measurements of genes involved in the hormone metabolism and excretion. Three endocrine disruptors commonly found in food were chosen for this study, which was conducted in the estrogen receptor (ER) negative hepatoblastoma HepG2 cell line: bisphenol A (BPA), genistein (GEN) and resveratrol (RES). We showed that 24 h after a single dose treatment with genistein, resveratrol or

Non-chemical, chemical, and biochemical, endocrine disruptors: biphasic health effects and pathophysiological insights.

Endocrine-disrupting chemicals (EDCs) interfere with hormone synthesis, signaling, and metabolism, leading to reproductive, metabolic, and neuroendocrine dysfunction. Non-chemical environmental endocrine stressors, including electromagnetic fields, noise, artificial light at night, and thermal stress, can disrupt endocrine homeostasis, alter neuroendocrine and hormonal function, affect signaling pathways, oxidative stress responses, and circadian rhythms. Unlike classical EDCs, which primarily exert their effects by direct binding to hormone receptors and metabolic enzymes, non-chemical endocr

Prenatal Dietary Exposure to Endocrine Disruptors and Its Lasting Impact on Offspring Health.

Environmental stressors during the crucial period of fetal development can have a substantial impact on long-term health outcomes. A major concern is dietary exposure to endocrine-disrupting chemicals (EDCs), which can readily cross the placenta and disrupt fetal hormonal signaling and developmental programming. Examples of these chemicals include bisphenols, phthalates, pesticides, and persistent organic pollutants (POPs). Prenatal exposure to EDC has been associated with long-term effects in children, including immune disruption, metabolic dysregulation, impaired neurodevelopment, and reprod

Per- and polyfluoroalkyl substances (PFASs) - New endocrine disruptors in polar bears (Ursus maritimus)?

Per- and polyfluoroalkyl substances (PFASs) are emerging in the Arctic and accumulate in brain tissues of East Greenland (EG) polar bears. In vitro studies have shown that PFASs might possess endocrine disrupting abilities and therefore the present study was conducted to investigate potential PFAS induced alterations in brain steroid concentrations. The concentrations of eleven steroid hormones were determined in eight brain regions from ten EG polar bears. Pregnenolone (PRE), the dominant progestagen, was found in mean concentrations of 5-47ng/g (ww) depending on brain region. PRE showed sign
